[Therapy against relapse in generalized lymphoblastic and immunoblastic lymphosarcomas in complete remission].

The authors present the results of the COP therapy of 85 patients with generalized lymphoblastic and immunoblastic lymphosarcomas. Antirelapse therapy with cyclophosphamide and reinduction courses according to the COP scheme and COP reinduction courses only increase the period of remission. A necessity of antirelapse therapy after achieving a complete remission is substantiated.

Histone H1 in nuclei of butyrate-treated murine lymphosarcoma cells has increased affinity for heparin.

Nuclei from butyrate-treated murine lymphosarcoma cells were incubated with different amounts of the polyanion heparin, which is known to interact predominantly with chromatin-associated histones. Unlike isolated histone H1, histone H1 in the nuclei of butyrate-treated cells was found to display an enhanced affinity for the binding to heparin as compared to histone H1 from control cells. Dephosphorylation of histone H1 as a result of butyrate treatment of the cells is discussed as a possible factor involved in the observed higher affinity of the protein for heparin.

Levamisole as adjuvant to chemotherapy for canine lymphosarcoma.

Ninety-eight dogs with previously untreated lymphosarcoma were selected for study. All dogs were clinically staged and stratified on the basis of stage. All dogs were treated with combination chemotherapy (vincristine, L-asparaginase, cyclophosphamide, and methotrexate) and randomized to either a levamisole or placebo group. There was no significant difference in either remission time or survival time between the levamisole and placebo groups. Clinical stage, age, and body weight had no influence on remission or survival time. The most significant prognostic factor was sex: Female dogs had significantly enhanced remission time (p = 0.004) and survival time (p = less than 0.001).

[Thiamine diphosphate absorption by the cells of Pliss lymphosarcoma and Ehrlich ascitic cancer].

Concentration difference of vitamin B1 coenzyme in blood and in ascitic fluid was found in mice with the Ehrlich ascites tumour. The thiamine diphosphate deficiency was detected in the liver of rats with the Pliss lymphosarcoma. The introduction of cocarboxylase to animals was accompanied by the active absorption of this coenzyme by tumours. It is observed that experimental tumours disturb the vitamin B1 balance in the organism.

Characterisation of a cell line from a human lung lymphosarcoma. Abundance of vimentin-type intermediate filaments.

A cell line (HCL82) was established from a human lung lymphosarcoma. Cells exhibited a stable phenotype and remained nearly diploid with a modal chromosome number of 45-48, showing no significant aberrations. Cultures consisted of single elongated cells and irregular colonies. Ultrastructurally, mitochondria, golgi complexes, and large bundles of intermediate filaments were well developed; some cells contained virus-like particles. Filaments were composed of vimentin identified by immunofluorescence and gel electrophoresis. Reaction with monoclonal antibody OKT4 suggested a close relationship to T-helper cells. Cells did grow at clonal density but did not form tumors in nude mice.

[Possibilities of using polychemotherapy according to the COP protocol in lymphosarcoma of the gastrointestinal tract].

Based on the use in 119 patients, the COP scheme can be regarded as an effective method of drug therapy of lymphosarcoma of the gastrointestinal tract: the total efficacy amounts to 65.4% and is practically identical in primary (61.8%) and metastatic (70%) involvement of the alimentary tube into the pathological process. However, the attainment of complete remissions was stated in an insignificant number of patients (16%). The five-year survival is equal to 60.2%.

[Cationic protein content in the leukocytes of rats with Pliss' lymphosarcoma and 7,12-dimethylbenz(a)anthracene-induced sarcoma].

The level of intraleukocytic cationic proteins is studied in rats with the transplanted Pliss lymphosarcoma and sarcoma induced by 7,12-dimethylbenz(a)anthracene (DMBA). The amount of cationic proteins was found to decrease in granulocytes both of mongrel and Wistar rats with transplantable tumours. The phenomenon is typical of the rats with the DMBA-induced tumour.

On the structure of cytolipin R, a ceramide tetrahexoside hapten from rat lymphosarcoma.

Cytolipin R, a ceramide tetrahexoside isolated from rat lymphosarcoma, was studied by sequential hydrolysis with specific glycosidases which revealed the anomeric configurations of the glycosidic bonds. Sugar linkages were established by combined gas-liquid chromatography and mass spectrometry of the partially methylated alditol acetates prepared after permethylation and hydrolysis of the intact lipid. Results indicated the structure of cytolipin R to be N-acetylgalactosaminyl(beta1-->3)galactosyl(alpha1-->3) galactosyl(beta1-->4)glucosyl ceramide. Cytolipin K (globoside I) differs in having a -galactosyl(alpha1-->4)galactosyl- internal linkage, and this difference must account for the immunological differences between cytolipin K and cytolipin R.

Value of biopsy in diagnosis of primary lymphosarcoma of marrow.

We report the clinical, hematologic, and survival characteristics of 12 patients with lymphosarcoma primarily involving the marrow but without substantial nodal disease. The importance of the biopsy in establishing this diagnosis is stressed, since in 7 of the 12 cases the diagnosis would not have been made had only a marrow aspiration been performed.

Chemoimmunotherapy for canine lymphosarcoma.

A total of 157 dogs with lymphosarcoma were available for study; 67 were treated. All of the treated dogs were given 4 drug combinations and 20 of them also were given autogenous vaccine. Sixty (90%) of the dogs treated with multiple drugs improved clinically. Of the dogs with clinical improvement, 48 (80%) had either complete or partial remission; of these, 32 (67%) had complete remission. Clinical staging proved useful in increasing the accuracy of prognosis, whereby dogs in less advanced stages of disease responded better to therapy, with a higher percentage of complete clinical remissions and longer survival. The mean survival time of the 47 dogs treated with drugs alone was 138 days, which compared with a mean survival time of 30 days for 34 nontreated dogs. Dogs subjected to chemotherapy and immunotherapy had a mean survival time of 341 days. Dogs in complete remission at time of vaccination survived significantly (P less than 0.01) longer than did dogs treated with drugs and vaccinated while not incomplete remission.

Distribution and pharmacokinetics of methotrexate in localized chemotherapy of solid Gardner's lymphosarcoma.

Pharmacokinetics and organe distribution of Methotrexate (MTX) in Gardner lymphosarcoma bearing C3H mice was investigated following two ways of drug administration: 1. intraperitoneal injection, 2. intratumoral implantation of 2-hydroxyethylmethacrylate gel with sorbed Methotrexate (localized chemotherapy). The highest level of MTX in blood appeared 2 hours after intrperitoneal injection but 7 hr after localized intratumorous application. Following intraperitoneal application the drug level in tumor reached its maximum two hours after injection; after 7 hr the drug could not be detected any longer. The localized chemotherapy led to six times higher concentration of the drug in the tumors as compared with the intraperitoneal application. This high level persisted for 17 hr and decreased moderately for 48 hr. MTX was accumulated in liver after both modes of administration with a half life 6.1 hr after intraperitoneal injection and 12.4 hr after the localized chemotherpay, respectively.

[Lymphoblastic lymphosarcoma of the stomach (author's transl)].

A case of primary lymphoblastic lymphosarcoma of the stomach, concerning a 15 year old boy, is described. The problematic of such cases is discussed in the light of the respective literature together with the own observation. Diagnostic, therapy and course of the case are reported on. It is emphasized that benign pseudolymphoma of the stomach must be considered in the differential diagnosis. The necessity of an optimal cooperation between internal gastroenterology, roentgenology and pathology in such cases is stressed. It is pointed out that the quality of the preoperative diagnosis mainly depends on endoscopy and histology.

Effect of glucocorticoid hormones in vitro on the structural integrity of nuclei in corticosteroid-sensitive and -resistant lines of lymphosarcoma P1798.

Exposure of rat thymus cells to glucocorticoids leads to a decreased ability of nuclei to survive the lysis of whole cells by hypotonic shock. In this study, a similar glucocorticoid-induced increase in "nuclear fragility" was found in both corticoid-sensitive and -resistant lines of P1798 mouse lymphosarcoma cells. In corticoid-sensitive cells a small increase in nuclear fragility is seen after a 2-hr exposure to cortisol (10(-6) M); by 3 hr it is 20 to 40% above control values. This effect appears to be a specific glucocorticoid response. Both cortisol and dexamethasone at 10(-7) M produce an effect, 10(-6) M testosterone is inactive, and cortexolone, which binds to glucocorticoid receptors, reduces the effect. Cycloheximide, at concentrations that inhibit protein synthesis, also blocks this effect. While the corticoid-resistant line also demonstrates an effect of similar magnitude, it requires a much longer exposure to the hormone (6 hr). Distinct differences in the "hardiness" of the two cell lines (nuclei of the corticoid-resistant line are less fragile) measurable in the absence of hormones appears to account for the differential susceptibility to steroids. On this basis a new theory of resistance is advanced where the emergence of resistance is related to structural differences in the cells.

Behaviour of murine lymphosarcoma cells during prolonged cultivation in vivo.

Transplantable strain of murine lymphosarcoma growing as a free suspension in the peritoneal cavity of isologous hosts was isolated after fractionated irradiation of C57BL mice. A 14-year cultivation led to changes in its properties; the mean survival time of the hosts was shortened, the modal number of chromosomes changed from 41--42 to 39 and their radioresistance increased slightly. Strain specificity remained unchanged.
