Comparison of effect of vacuum-condensed and ultrafiltered milk on cheddar cheese.

The objective of this study was to compare the effects of vacuum-condensed (CM) and ultrafiltered (UF) milk on some compositional and functional properties of Cheddar cheese. Five treatments were designed to have 2 levels of concentration (4.5 and 6.0% protein) from vacuum-condensed milk (CM1 and CM2) and ultrafiltered milk (UF1 and UF2) along with a 3.2% protein control. The samples were analyzed for fat, protein, ash, calcium, and salt contents at 1 wk. Moisture content, soluble protein, meltability, sodium dodecyl sulfate-PAGE, and counts of lactic acid bacteria and nonstarter lactic acid bacteria were performed on samples at 1, 18, and 30 wk. At 1 wk, the moisture content ranged from 39.2 (control) to 36.5% (UF2). Fat content ranged from 31.5 to 32.4% with no significant differences among treatments, and salt content ranged from 1.38 to 1.83% with significant differences. Calcium content was higher in UF cheeses than in CM cheeses followed by control, and it increased with protein content in cheese milk. Ultrafiltered milk produced cheese with higher protein content than CM milk. The soluble protein content of all cheeses increased during 30 wk of ripening. Condensed milk cheeses exhibited a higher level of proteolysis than UF cheeses. Sodium dodecyl sulfate-PAGE showed retarded proteolysis with increase in level of concentration. The breakdown of alphas1- casein and alphas1-I-casein fractions was highest in the control and decreased with increase in protein content of cheese milk, with UF2 being the lowest. There was no significant degradation of beta-casein. Overall increase in proteolytic products was the highest in control, and it decreased with increase in protein content of cheese milk. No significant differences in the counts of lactic starters or nonstarter lactic acid bacteria were observed. Extent as well as method of concentration influenced the melting characteristics of the cheeses. Melting was greatest in the control cheeses and least in cheese made from condensed milk and decreased with increasing level of milk protein concentration. Vacuum condensing and ultrafiltration resulted in Cheddar cheeses of distinctly different quality. Although both methods have their advantages and disadvantages, the selection of the right method would depend upon the objective of the manufacturer and intended use of the cheese.

Influence of condensed sweet cream buttermilk on the manufacture, yield, and functionality of pizza cheese.

Compositional changes in raw and pasteurized cream and unconcentrated sweet cream buttermilk (SCB) obtained from a local dairy were investigated over 1 yr. Total phospholipid (PL) composition in SCB ranged from 0.113 to 0.153%. Whey protein denaturation in pasteurized cream over 1 yr ranged from 18 to 59%. Pizza cheese was manufactured from milk standardized with condensed SCB (approximately 34.0% total solids, 9.0% casein, 17.8% lactose). Effects of using condensed SCB on composition, yield, PL recovery, and functional properties of pizza cheese were investigated. Cheesemilks were prepared by adding 0, 2, 4, and 6% (wt/wt) condensed SCB to part-skim milk, and cream was added to obtain cheesemilks with approximately 11.2 to 12.7% total solids and casein:fat ratio of approximately 1. Use of condensed SCB resulted in a significant increase in cheese moisture. Cheese-making procedures were modified to obtain similar cheese moisture contents. Fat and nitrogen recoveries in SCB cheeses were slightly lower and higher, respectively, than in control cheeses. Phospholipid recovery in cheeses was below 40%. Values of pH and 12% trichloro-acetic acid-soluble nitrogen were similar among all treatments. Cheeses made from milk standardized with SCB showed less melt and stretch than control cheese, especially at the 4 and 6% SCB levels. Addition of SCB significantly lowered free oil at wk 1 but there were no significant differences at wk 2 and 4. Use of SCB did not result in oxidized flavor in unmelted cheeses. At low levels (e.g., 2% SCB), addition of condensed SCB improved cheese yield without affecting compositional, rheological, and sensory properties of cheese.

Behavior of Listeria monocytogenes in a Pseudomonas putida biofilm on a condensate-forming surface.

Listeria monocytogenes has been isolated from condensate-forming surfaces in food processing plants. The objective of this research was to observe the behavior of L. monocytogenes on condensate-covered stainless steel with a Pseudomonas putida biofilm. L. monocytogenes-containing biofilms, either with or without added chicken protein, were incubated in a high humidity chamber at 12 degrees C to allow formation of condensate. Samples were analyzed for attached and unattached L. monocytogenes and total plate count periodically for 35 days. Samples were also taken for microscopic observation of Listeria and bacterial extracellular polymeric substances (EPS). L. monocytogenes attached in significantly greater numbers (> 3-log difference) to surfaces with preexisting P. putida biofilms than to Pseudomonas-free surfaces. L. monocytogenes survived in the presence or absence of P. putida with no added nutrients for 35 days, with numbers of survivors in the range of 3 to 4 log CFU/cm2 in the presence of P. putida and less than 2.9 log CFU/cm2 in pure culture. Attached and unattached L. monocytogenes were at similar levels throughout the incubation under all conditions studied. The addition of protein to the biofilms allowed growth of L. monocytogenes in pure culture during the first 7 days of incubation. Numbers of L. monocytogenes were not affected by the presence of P. putida when protein was present. Unattached L. monocytogenes were at levels of 3.6 to 6.7 log CFU/cm2 on the protein-containing surfaces. Microscopic observation of the condensate-covered biofilms indicated that L. monocytogenes formed microcolonies embedded within an EPS matrix over a 28-day period. This research demonstrates that L. monocytogenes can survive on condensate-forming stainless steel in low and high nutrient conditions, with or without the presence of Pseudomonas biofilm. The Listeria can detach and, therefore, have the potential to contaminate product.

[Genotoxicity of stack gas condensates of Bavarian waste incineration plants. II. Suitability of fast bacterial tests of emission monitoring].

The genotoxicity of stack gas condensates of 21 waste incineration plants (located in Bavaria) was examined in the years 1990-1995 using two bacterial short time tests. The SOS chromotest was carried out with the tester strains Escherichia coli PQ37, PQ243 and PQ300. In addition, for the purpose of comparison, the Ames-Test was performed for selected examples with the tester strains Salmonella typhimurium TA97, TA98, TA100 and TA1537. In a pilot study, carried out in the years 1990 to 1991, the stack gas condensates from five plants were examined. They showed clear genotoxic and mutagenic effects. On the other hand, in subsequent tests we generally discovered only weak inductions for 9 of 18 crude and 24 of 78 clean gas condensate extracts, mostly after metabolic activation. Four plants were tested continuously in the years 1992 to 1995. Three of them showed a clear reduction of the detectable genotoxic potential. The fourth one gave negligible SOS inducing emissions in the whole examining period. On the other hand, for 6 of the 21 tested plants we found chromotest positive results even at the last test point. Correlations between the SOS inducing potential of the stack gas condensates and the analytical parameters detected at the same time (6 summary parameters, 24 inorganic and 63 organic chemical parameters) were not evident. Only the two highest emissions of nitropyrenes were associated with SOS inductions. Organic substances which are not analytically detected or synergistic effects might be responsible for the SOS inducing potency of the other genotoxic stack gas condensates.

Condensed chromatin and cell inactivation by single-hit kinetics.

Mammalian cells are extremely sensitive to gamma rays at mitosis, the time at which their chromatin is maximally condensed. The radiation-induced killing of mitotic cells is well described by single-hit inactivation kinetics. To investigate if radiation hypersensitivity by single-hit inactivation correlated with chromatin condensation, Chinese hamster ovary (CHO) K1 (wild-type) and xrs-5 (radiosensitive mutant) cells were synchronized by mitotic shake-off procedures and the densities of their chromatin cross sections and their radiosensitivities were measured immediately and 2 h into G1 phase. The chromatin of G1-phase CHO K1 cells was dispersed uniformly throughout their nuclei, and its average density was at least three times less than in the chromosomes of mitotic CHO K1 cells. The alpha-inactivation co-efficient of mitotic CHO K1 cells was approximately 2.0 Gy(-1) and decreased approximately 10-fold when cells entered G1 phase. The density of chromatin in CHO xrs-5 cell chromosomes at mitosis was greater than in CHO K1 cell chromosomes, and the radiosensitivity of mitotic CHO xrs-5 cells was the greatest with alpha = 5.1 Gy(-1). In G1 phase, CHO xrs-5 cells were slightly more resistant to radiation than when in mitosis, but a significant proportion of their chromatin was found to remain in condensed form adjacent to the nuclear membrane. These studies indicate that in addition to their known defects in DNA repair and V(D)J recombination, CHO xrs-5 cells may also be defective in some process associated with the condensation and/or dispersion of chromatin at mitosis. Their radiation hypersensitivity could result, in part, from their DNA remaining in compacted form during interphase. The condensation status of DNA in other mammalian cells could define their intrinsic radiosensitivity by single-hit inactivation, the mechanism of cell killing which dominates at the dose fraction size (1.8-2.0 Gy) most commonly used in radiotherapy.

Novel inhibitors of the condensing enzymes of the type II fatty acid synthase of pea (Pisum sativum).

The type II fatty acid synthases (FASs) of higher plants (and Escherichia coli) contain three condensing enzymes called beta-ketoacyl-ACP synthases (KAS), where ACP is acyl-carrier-protein. We have used novel derivatives of the antibiotic thiolactomycin to inhibit these enzymes. Overall de novo fatty acid biosynthesis was measured using [1-(14)C]acetate substrate and chloroplast preparations from pea leaves, and [1-(14)C]laurate was used to distinguish between the effects of the inhibitors on KAS I from those on KAS II. In addition, the activities of these enzymes, together with the short-chain condensing enzyme, KAS III, were measured directly. Six analogues were tested and two, both with extended hydrocarbon side chains, were found to be more effective inhibitors than thiolactomycin. Incubations with chloroplasts and direct assay of the individual condensing enzymes showed that all three compounds inhibited the pea FAS condensing enzymes in the order KAS II > KAS I > KAS III. These results demonstrate the general activity of thiolactomycin and its derivatives against these FAS condensation reactions, and suggest that such compounds will be useful for further detailed studies of inhibition and for use as pharmaceuticals against Type II FASs of pathogens.

Electron microscopy and biological properties of pBR322 DNA condensed with the trivalent cations spermidine and hexammine cobalt (III).

Electron microscopy and the biological properties of susceptibility to DNase I, genetic transcription, and transformation of pBR322 DNA compacted with spermidine or hexammine cobalt (III), were analyzed in order to characterize the association of DNA in its compacted form with these two different trivalent cations. Spermidine and hexammine cobalt (III) produced an average 4-fold reduction of the DNA perimeter in compact DNA forms, which were doughnut-shaped toroids and cylinders. Both compacted DNAs were resistant to the hydrolytic activity of DNase I. However, spermidine-condensed pBR322 DNA was 10-fold and 4 to 6-fold more active in transcription and transformation, respectively, than naked pBR322. I. Hexammine cobalt (III)-condensed pBR322 was inactive in both biological properties. An inhibitory effect of hexammine cobalt (III) on RNA polymerase and genetic transformation activities was discarded because at higher ionic strength, in which DNA is not compacted by hexammine cobalt (III), transcription and transformation were similar to those observed with naked DNA. This information showed that the interaction of hexammine cobalt (III) with the DNA converted the pBR322 DNA into an inert molecule. In contrast, pBR322 did not loose its biological properties after its interaction with the polyamine spermidine; i.e., experimental condensation of pBR322 DNA by spermidine produced compacted DNA that is more similar to compact native genomes than relaxed DNA. These experiments led us to conclude that spermidine-condensed DNA can be used to study the roll of the native supercoiling of DNA in the regulation of genetic replication and transcription, as well as to study the mechanisms that allow the accessibility of the supercoiled or condensed DNA substrate for enzymes.

The stereochemistry at carbon 3 of pyruvate lyase condensation products. 2-Keto-3-deoxygluconate-6-phosphate aldolase.

In a condensation between [3-3H3]pyruvate and D-glyceraldehyde-3-P as catalyzed by 2-keto-3-deoxygluconate-6-P aldolase (EC 4.1.2.14) of Pseudomonas putida, C--C synthesis occurred appreciably faster than C--3H bond breaking. Since tritium is present in tritiated pyruvate in tracer amounts, this result showed hydrogen isotope discrimination in pyruvate deprotonation and suggests enolpyruvate generation to be at least partially rate-limiting in the condensation reaction. Consequently, in a condensation reaction between [3-3H, 2H,H]pyruvate of known chirality and D-glyceraldehyde-3-P, the newly synthesized C--C bond would be enriched for at what was the C--H bond of chiral pyruvate, discriminating against the C--2H and C--3H bonds. Additional studies showed that condensations between (3S)-[3-3H, 2H,H]- or (3R)-[3-3H, 2H,H]pyruvate and D-glyceraldehyde-3-P yielded predominantly (3S)- or (3R)-2-keto-3-deoxy[3-3H, 2H]gluconate-6-P, respectively. By comparison with sterochemical models, it was concluded that condensation occurred with retention of configuration at C-3. Thus in the turnover of substrates as catalyzed by this enzyme, both the exchanging proton from water and D-glyceraldehyde-3-P attack the same face of the enzyme-bound pyruvyleneamine.

Microleakage of packable composites used in post spaces condensed using different methods.

The aim of this in vitro study was to evaluate the apical and coronal microleakage of a packable composite material compacted by either hand or ultrasonic condensers when placed in post spaces. Forty-two extracted anterior teeth were used. After conventional obturation of the root canals, parallel post spaces 5 mm in depth and 2 mm in diameter were prepared using Gates Glidden and diamond burs. A primer and a dentin bonding agent (Prime & Bond NT) was applied to the etched surface for 30 seconds and light polymerized for 40 seconds. Surefil packable composite resin was inserted in four equal increments and each polymerized for 40 seconds. Half of the specimens were compacted with an ultrasonic tip and the other half with a mechanical hand compactor. After immersion in the dye, the teeth were sectioned longitudinally and dye penetration was evaluated using a stereomicroscope and microleakage recorded in mm. A One-Way Analysis of Variance indicated the two condensing methods were not statistically different (p>0.05). When microleakage was compared within groups, there was also no statistical difference between coronal or apical microleakage (p>0.05). In an in vitro environment, ultrasonic condensers do not offer any advantage over hand condensers. Both the coronal and apical portions of the root had microleakage indicating that regardless of the condensation method and the use of a packable composite, there is still microleakage that could potentially create further clinical problems.

Markers of pulmonary diseases in exhaled breath condensate.

Exhaled breath condensate has been more and more extensively used as a novel and non-invasive method to study airway inflammation. It is simple to perform, very well tolerated by patients and no adverse events have been reported so far. Serial measurements can be made with no harmful effects on patients, which is of extreme value in occupational medicine. Exhaled breath condensate has been obtained from both adult and children patients suffering from various pulmonary diseases such as asthma, cystic fibrosis, chronic obstructive pulmonary disease, and interstitial lung diseases. Several markers and mediators are detectable in breath condensate: hydrogen peroxide, thiobarbituric acid-reactive substances, isoprostanes, prostaglandins and leukotrienes. Nitric oxide-related markers have also been studied in the condensate. There is increasing body of evidence that changes in condensate markers reflect local abnormalities of airway lining fluid.

[Study of the possibility of utilizing the transpired mositure condensate from sweet potato for growing plants in biological life support systems].

The effect of nonpurified condensate obtained during prolonged cultivation of batata in a sealed chamber upon batata cuttings and seedlings of garden cress, radish and Chinese cabbage was studied. It was shown that nonpurified condensate produced an inhibitory effect on the formation of roots in batata cuttings and on the growth of previously developed roots of batata cuttings and seedlings. The studies which used a chemical model of 3,4-dihydroxy phenylalanine indicated that the condensate contained biologically active substance of organic origin. However, only experiments with the real continuous culture of batata, using real dilutions of the condensate that depend on the size of the greenhouse and the amount of the nutrient solution would clarify wheather condensate of transpiration water of batata plants can be repeatedly utilized in life support systems.

Pneumatic versus hand condensation of amalgam: effect on microleakage.

This study compared two techniques of amalgam condensation for effect on early microleakage. Class V preparations were restored with a high-copper amalgam using pneumatic condensation or conventional hand condensation. Significantly less microleakage was observed when pneumatic condensation was used. Further, pneumatic condensation proved equally effective in reducing early microleakage whether or not a dentinal sealant was used as an intermediary.

[Exhaled breath condensate and its analysis--a new method in pulmonology].

In the middle of the nineties a new, non-invasive method for investigation of the lung aroused the interest of many researchers: the exhaled breath condensate. It shows the extent of the interest that in the last five years more than 80 original articles have been published in this theme. Many substances are found in the expired breath which are detectable in the liquid that we obtain by cooling (= condensing) the exhaled breath. The advantages of this method are that it is non-invasive, convenient, it could be performed with mechanically ventilated patients as well as with children. The most studied substance is the hydrogen-peroxide, which is the marker of oxidative stress, and its level in condensate is elevated in numerous inflammatory diseases. 8-isoprostane was also studied a lot, which is another marker of oxidative stress. Numerous substances could be even measured in condensate, so the decay-product of nitric-oxide (nitrite, nitrate, nitrotyrosine), further nitrosothiol, adenosine, ammonia, different ions, leukotrienes, cytokines; recently even other feature of condensate is examined, such as its pH. The different mediators could help us to know better the diseases, support the diagnosis, follow the treatment or the disease. In this study the authors attempt to present the most important knowledge till now.

Outcome of endodontic treatment of teeth filled using lateral condensation versus vertical compaction (Schilder's technique).

The purpose of this prospective clinical and radiographic investigation was to assess the treatment results following endodontic therapy of teeth filled with lateral condensation versus teeth filled with vertical compaction of warm gutta-percha. A total of 290 patients were treated using the standardized step-back technique for canal preparation, which were filled with either lateral condensation or vertical compaction in one single session. Five years later, the treatment results were assessed clinically and radiographically and related to the type of the obturation technique using Chi-square analysis. Of the 340 teeth that were reexamined, 160 teeth were filled with lateral condensation, and 180 teeth were filled with vertical condensation. The results showed a significantly higher success rate for the vertical compaction versus the lateral condensation technique of teeth presented with preoperative periapical lesions P<0.04. Regardless of the preoperative periapical status of the teeth, no statistically significant difference was found between the two techniques. The overall success rate of both filling techniques was 80.3%.

Condensed images for evaluating gastric motility patterns.

A condensed imaging technique was applied to gastric emptying studies to investigate (a) whether different types of motility disorders may be distinguished by characteristic image patterns and (b) whether the findings obtained provide additional information compared to standard quantitative measurements. Condensed images and quantitative data of gastric emptying were evaluated in 75 consecutive patients with normal function (n = 16) and various disorders (n = 59) such as peptic ulcer, postvagotomy, pyloric obstruction, dumping syndrome, gastroparesis etc. Condensed images were generated from a gastric region of interest. They display the distribution and behaviour of a radioactive test meal in a space-time matrix, whose horizontal and vertical dimensions are temporal and spatial, respectively. As shown in a series of representative examples condensed images disclose a variety of well-defined image patterns reflecting different pathophysiological mechanisms. This qualitative characterization of gastric emptying patterns provided in 34 of the 75 patients (45%) important new information compared to quantitative data. The application of condensed imaging techniques to gastric emptying studies (complementary to quantitative measurements) may, therefore, enhance the diagnostic value of scintigraphic techniques.

Preparation and monitoring of marijuana smoke condensate samples.

The objective of this study was the production of smoke condensate from the marijuana cigarettes on schedule for the bioassay study and involved quality control on each batch of smoke condensate. Examination of the condensate production data (tables 1 and 2) reveal that it is not possible to strictly control the butt length of cigarettes when smoked on a robot smoking machine thereby accounting for some variation in the yield of the dry particulate matter and increment9-tetrahydrocannabinol. No increment8-tetrahydrocannabinol was detected in any batch of the smoke condensate. Recently, Hoffman and co-workers (13) have reported that marijuana that exhibited carcinogenic potential similar to tobacco cigarette smoke condensate in the dermal assay in mice.

[Clinically controlled study of success of lateral and thermo-mechanical Gutta-percha condensation].

76 root canal treated teeth were followed up within 2 years after treatment with the help of the periapical index PAI and the root canal filling index WKF-I. The root canals were obturated with gutta-percha and Sealer according to lateral or thermom chanical condensation. Treatment was successful in 26 of the lateral condensated and in 25 of the thermomechanical condensated cases. The thermomechanical condensation method represents an alternative method for filling root canals for those endodontists who are well practiced in this method and ensures a high success rate at exact indication positions. Intense practice on extracted teeth prior to clinical application is a strong prerequisite. The lateal condensation method is the root canal filling technique of choice for the general dental practice.

Endogenous polyamines associate with DNA during its condensation in mammalian tissue. A fluorescence cytochemical and immunocytochemical study of polyamines in fetal rat liver.

The polyamines spermidine and spermine are essential for cell proliferation and differentiation. By two independent fluorescence cytochemical methods as well as by immunocytochemistry, we have studied the distribution of these molecules in fetal rat liver. Strong reactions for polyamines were found in highly condensed chromatin, present in chromosomes in mitotic cells, and in condensed nuclei in late erythropoietic cells. Moreover, polyamines were so closely associated with DNA in condensed chromatin that DNase pretreatment was necessary for making them available for reaction with antibodies. In other cells, polyamines were mainly localized to the cytoplasm. Studies of cells at different stages in erythropoiesis revealed that polyamines become associated with DNA during its condensation and inactivation. Our data strongly indicate that polyamines participate in the condensation of DNA.
