Bi-enzyme reactor for electrochemical detection of low concentrations of uric acid and glucose.

An enzyme-based flow-injection amperometric analysis system (FIA) for monitoring of uric acid and glucose is described. The oxidase and peroxidase enzymes are physically coimmobilised in a sandwich-type reactor and ferrocene serves as a mediator. The assays are based on the measurement of a reduction current resulting from the enzymatic reactions, at a glassy carbon electrode held at 0.00 mV (vs. Ag/AgCl). The high selectivity (ascorbic acid did not interfere) is coupled to high sensitivity (a detection limit of 30 and 60 nmol/l for uric acid and glucose, respectively; signal/noise = 3) and good stability (the enzymes remained active for more than 6 weeks at 30 degrees C). The usefulness of the assay in clinical chemistry is illustrated by the measurement of human serum uric acid and glucose concentration. The results obtained were in fairly good agreement with those obtained using conventional hospital laboratory methods.

Routine analysis with immobilized enzyme nylon tube reactors.

The basic strategy involved in the design, development, and application of immobilized enzyme nylon tube reactors for routine analysis is described in this chapter, touching on some of the attractive features of these methods. Extensive data (Tables I and II) and the references provide details which may be needed based on specific methods.

[Morphologic, biochemical and functional properties of the mitochondria of the rabbit gastric mucosa].

Purified fractions of the rabbit gastric mucosa and liver mitochondria were isolated. Electrone microscopy and functional studies of mitochondria showed the most intact mitochondria of gastric mucosa to be localized in gradient fractions of p=1.16-1.18, and those of the liver in fractions with p=1.19-1.20. The gastric mitochondria differed from the liver mitochondria by specific localization in oxyntic cells, abundance of crists, small matrix volume, presence of high respiratory control at lesser values of pH, high respiratory activity, great amount of cytochromes of electron transport chain in inner mitochondria membrane, two-fold excess of cytochroma a in relation to b and c, absence of b5 and by presence of great amount of phosphathydilaethanolamine and unsaturated fatty acids.

Highly sensitive flow injection analysis of glucose and uric acid in serum using an immobilized enzyme column and chemiluminescence.

A method for the flow injection analysis of glucose and uric acid in serum using immobilized enzymes in column form and chemiluminescence detection is described. The method is based on the determination of chemiluminescence formed by the reaction of a luminol-ferricyanide mixture with hydrogen peroxide which is produced by the action of the respective oxidases on glucose and uric acid. Glucose or uric acid in serum were determined with 1 microliter of the sample at a speed of 120 samples/h without carryover and at an assay time of approximately 10 s. The immobilized glucose oxidase column measured only 1.0 X 5 mm, and the immobilized uricase column 1.0 X 20 mm. The present method gave perfect linearity of the data up to 4.0 g glucose per liter or 0.10 g uric acid per liter with satisfactory precision, reproducibility, and accurate reaction recoveries. Furthermore, the present method was hardly affected by ascorbic acid, while the peroxidase-linked colorimetric method is usually influenced significantly by ascorbic acid. Both column reactors showed good operational stability for a 2-month period, during which time they were repeatedly used for analyses over 2000 times. The results on glucose and uric acid correlated satisfactorily with those obtained by other well-established methods.

Peroxisomal oxidases: cytochemical localization and biological relevance.

(1) alpha-HAOX has a broad substrate specificity. In rat kidney, the enzyme reacts with aliphatic and aromatic alpha-hydroxy acids, in rat liver, however, only with aliphatic ones. (2) The best substrate for the demonstration of alpha-HAOX activity in rat and human liver is glycolate. (3) alpha-hydroxy butyric acid is the best substrate in the luminometric assay for the demonstration of alpha-HAOX activity in the rat kidney, whereas glycolate is not catalysed by the enzyme. (4) In the proximal tubulus epithelial cells of the rat kidney alpha-HAOX is concentrated in the peripheral matrix of the peroxisomes.

Deactivation of bilirubin oxidase by a product of the reaction of urate and O2.

The "wired" bilirubin oxidase (BOD) bioelectrocatalyst is superior to pure platinum as an electrocatalyst of the four-electron electroreduction of O(2) to water. Not only is its overpotential for O(2) reduction lower, but unlike platinum, it is not affected by organic compounds like glucose. The "wired" BOD-coated carbon cathode operates for >1 week at 37 degrees C in a glucose-containing physiological buffer solution. One of its key applications would be in a glucose-O(2) biofuel cell, which would operate in living tissues. The cathode is, however, short-lived in serum, losing its electrocatalytic activity in a few hours. Here we show that the damaging serum component is a product of the reaction of urate and dissolved oxygen. Exclusion of urate, by application of Nafion film on the cathode, improves the stability in serum.

Effect of dietary n-3 and n-6 polyunsaturated fatty acids on lipid-metabolizing enzymes in obese rat liver.

This study was designed to examine whether n-3 and n-6 polyunsaturated fatty acids at a very low dietary level (about 0.2%) would alter liver activities in respect to fatty acid oxidation. Obese Zucker rats were used because of their low level of fatty acid oxidation, which would make increases easier to detect. Zucker rats were fed diets containing different oil mixtures (5%, w/w) with the same ratio of n-6/n-3 fatty acids supplied either as fish oil or arachidonic acid concentrate. Decreased hepatic triacylglycerol levels were observed only with the diet containing fish oil. In mitochondrial outer membranes, which support carnitine palmitoyltransferase I activity, cholesterol content was similar for all diets, while the percentage of 22:6n-3 and 20:4n-6 in phospholipids was enhanced about by 6 and 3% with the diets containing fish oil and arachidonic acid, respectively. With the fish oil diet, the only difference found in activities related to fatty acid oxidation was the lower sensitivity of carnitine palmitoyltransferase I to malonyl-CoA inhibition. With the diet containing arachidonic acid, peroxisomal fatty acid oxidation and carnitine palmitoyltransferase I activity were markedly depressed. Compared with the control diet, the diets enriched in fish oil and in arachidonic acid gave rise to a higher specific activity of aryl-ester hydrolase in microsomal fractions. We suggest that slight changes in composition of n-3 or n-6 polyunsaturated fatty acids in mitochondrial outer membranes may alter carnitine palmitoyltransferase I activity.

Kinetic flow-injection determination of hydrogen peroxide by use of iron(III)-catalyzed coloration and its application to the determination of biological substances.

A kinetic flow-injection (FI) method is described for the determination of hydrogen peroxide. This method is based on an iron(III)-catalyzed oxidative coupling of 4-aminoantipyrine with N,N-dimethylaniline by hydrogen peroxide. By measuring the change in the absorbance of the dye formed at 560 nm, 1 x 10(-6) - 6 x 10(-4) M hydrogen peroxide could be determined with a sampling rate of 15 h(-1). The relative standard deviation (n = 30) was 0.8% for 5 x 10(-5) M hydrogen peroxide. There was little interference of the co-existing ions and compounds. After introducing some immobilized enzyme reactors to the FI system, the proposed method allowed the determination of glucose and uric acid ranging from 1 x 10(-6) to 6 x 10(-4) M with relative standard deviations of below 2%. The applicability of the method was demonstrated by determining these substances in serum samples.

[Cold adaptation and changes in peroxisome enzyme in various organs of the rat].

In the rat brown fat peroxisomes - thermogenetic organules - an peroxisomal enzyme activities undergo remarkable changes during the adaptation to cold of the animals (see 3). In this paper was show that changes of peroxisomal enzyme activities occur also in liver and kidney during cold-adaptation. Catalase, L-hydroxyacid oxidase, uricase and D-aminoacid oxidase (DAO) were assayed as in (6). During cold-adaptation, the activity of the former three enzymes (Table 2) increases with the weight of the organs (Table 1) whereas that of DAO exhibits a much larger increase (Table 3). Results are discussed with regard to the contribution of the liver to non-shivering thermogenesis.
