Vital fluorochromes as tracers for fungal growth studies.

Eight fluorescent dyes were tested for staining the spores or mycelia of six fungi and for their translocation into new growth when the preloaded spores or mycelia were incubated on agar coated coverslips. The dyes studied were Cellufluor, Nile red, fluorescein diacetate (FDA), carboxyfluorescein diacetate (CFDA), chloromethylfluorescein diacetate (CMFDA), aminochloromethyl coumarin (CMAC), and the carbocyamines DiIC18(3) and DiOC18(3). The fungi on which the dyes were tested included Botrytis cinerea, Fusarium oxysporum f.sp. lycopersici, Idriella bolleyi, Pythium oligandrum, Sclerotium cepivorum and Trichoderma harzianum. Most of the fluorochromes gave good initial staining of mycelia or spores; however, FDA fluorescence faded rapidly during excitation, making it impractical for use. Also, the spores and mycelia of B. cinerea and T. harzianum sometimes gave weak fluorescence with Nile red, and the spores and mycelia of I. bolleyi gave unusually weak fluorescence with Cellufluor. There were other variations of staining among the different dye/fungus combinations, but each fungus showed strong fluorescence at least one dye. Cellufluor, CMFDA, CMAC and, to a lesser extent, CFDA and Nile red, were efficiently translocated into new growth from preloaded spores or mycelia, whereas FDA, DiIC18(3) and DiOC18(3) were not. The extent of translocation ranged from 0.1 to 1.2 mm in germ tubes arising from spores, and from 0.9 to 9.2 mm in mycelia extending from dye-loaded agar blocks. The findings suggest that fluorescent dyes could be used as markers or tracers in studies of fungal growth and differentiation.

Isolation and anti-fungal activities of 2-hydroxymethyl-chroman-4-one Produced by Burkholderia sp. MSSP.

In the course of screening for biological active compounds from microbial origins, various strains were isolated from roots of Mimosa pudica using standard dilution agar plate method. One isolate was selected for further studies of bacterial anti-fungal properties. It was designated MSSP and identified as Burkholderia sp. by many biological characteristics such as carbohydrate utilization, biochemical and physiological test and 16S rDNA sequence homology search. To investigate antagonistic principle, anti-fungal compounds were extracted and fractionated by different solvent systems. An anti-fungal compound was isolated from its culture filtrate using organic solvent extraction, column chromatography and thin layer chromatography and pure solid form compound (MSSP2) was obtained. The chemical structure of anti-fungal compound was identified as 2-Hydroxymethyl-chroman-4-one based on the data obtained from spectroscopic analysis such as mass spectral analyses and NMR spectral data. The compound 2-Hydroxymethyl-chroman-4-one exhibited good activities against phytopathogen such as Pythium ultimum, Phytophthora capsici and Sclerotinia sclerotiorum.

[Purification and characterization of antifungal peptide LP-1].

An antifungal peptide LP-1 from Bacillus subtilis TG26 strain was purified by acid precipitation, acetone precipitation and Hi-pore reversed phase column chromatography. The molecular weight of LP-1 is 1057.3 D as determined by MALDI-TOF mass spectrometry, and its pI is 4.75 by PAG-IEF. It was also found to be thermostable. Its antifungal spectrum showed that LP-1 has strong inhibitory activity against many plant pathogenic fungi, such as Pythium aphanidermatum, Gibberella zeae, Alternaria longipe, Fusarium oxysporum f. lycopersici, etc.. The abnormal hyphal growth of Trichoderma viride caused by LP-1 such as swollen tips, twisted, short growth and cytoplasm condensation was also observed. Both ninhydrin reaction and peptide sequencing suggested that LP-1 is a cyclic peptide.

Contribution of phlA and some metabolites of fluorescent pseudomonads to antifungal activity.

Fluorescent Pseudomonas species are an important group of PGPR that suppress fungal root and seedling disease by production of antifungal metabolites such as 2,4-diacetylphloroglucinol (2,4-DAPG), pyoluteorin, pyrolinitrin, siderophores and HCN. The compound 2,4-DAPG is a major determinant in biocontrol of plant pathogens. A 7.2 kbp chromosomal DNA region, carrying DAPG biosynthetic genes (phlA, phlC, phlB, phlD, phIE and phlF). Detecting the ph1 genes make them an ideal marker gene for 2,4-DAPG-producing fluorescent pseudomonad's. In this study we detected ph1A gene (that convert MAPG to 2,4-DAPG) using PCR assay with primers phlA-1r and phlA- f that enabled amplification of phlA sequences from fluorescent pseudomonad's from ARDRA group 1 and 3. We could detect phlA gene in P. fluorescens strains CHAO, Pf-44, Pf-1, Pf-2, Pf-3, Pf-17, Pf-62 and Pf-64, native isolates of Iran. The efficacy of this method for rapid assay characterizing rhizosphere population of 2,4-DAPG producing bacteria from soil of different area of Iran is in progress. We used a collection of 48 fluorescent pseudomonas strains in vitro, with known biological control activity against some soil born phytopathogenic fungi such as, Macrophomina phaseoli, Rhizoctonia solani Vericillium dahlia, Phytophthora nicotiana, Pythium spp. and Fusarium spp. and the potential to produce known secondary metabolites such as protease. Strains Pf-1, Pf-2, Pf-3, Pf-17, Pf-33 and Pf-44 showed the best antifungal activity against all fungi used in this study. Thirty-eight of 48 strains produced protease. The ability to rapidly characterize populations of 2,4-DAPG producers will greatly enhance our understanding of their role in the suppression of root disease.

Biological protection of the main cereals against fungal specific diseases.

Our field experiments showed that the use of Czech biopreparations (Supresivit, Ibefungin and Polyversum) based on the following microorganisms: Trichoderma harzianum, Bacillus subtilts and Pythium oligandrum applied as the seed treatment, the spray on the plants and like the mixture with mineral fertilizers (NPK, ammonium sulphate) lead namely after the seed-treatment and after the application as the mixture with mineral fertilizers to the increasing of the yield about 3-5 % (spring barley, winter wheat). This increasing was given by depression of soil-borne phytopathogenic fungi of the genera: Fusarium, Drechslera (Helminthosporium), Pseudocercosporella (Tapesia), Gaeumannomyces, and partially Rhynchosporium. The number of the fungi on plant rests were also influenced. No effect was observed on smuts and rusts. The smuts needed the seed-treatment with Vitavax (carboxim, thiram, imazalil) which didn't influence the biological treatment with biopreparations. The doses of biopreparations were following: the mixture with mineral fertilizer - 0.1 g of biopreparation/ kg of mineral fertilizer, the seed treatment - 0.1 g of biopreparation/1 kg of the seed and in the spray 0.1 g of biopreparation/1 litre of distilled water. Biological biopreparations will be usefull probably also in the future because they don't create the harmfull residues. The impact of the biopreparations under different ways of soil tillage (conventional variant with ploughing, variants with different plant residues and variants without ploughing) was studied as well. The influence of different soil preparation on composition of the soil mycoflora and the influence on quantity of the pathogenic genera Fusarium, Drechslera and others was observed. Quantity of named genera was negatively influenced while yield and health status of the plants were influenced positively.

Biological studies on 2,1-benzisothiazole derivatives. I. Evaluation of antibacterial, antifungal and genotoxic activities of 1-acyl-3-acylimino-2,1-benzisothiazolines.

The in vitro evaluation of antibacterial, antifungal and genotoxic activities of some 1-acyl-3-acylimino-2,1-benzisothiazolines is described. Compounds 55, 58, 60 and 62 exhibited a selective antibacterial activity. Compounds 1, 2, 30, 59 and 63 were active against Botrytis cinerea, compounds 6, 20, 39 and 54 against Pythium irregulare. DNA-damaging activity in the Bacillus subtilis rec-assay was demonstrated for compounds 45, 46, 54, 55, 57 and 61. By the Salmonella-microsome assay, genotoxic activity was confirmed and demonstrated also for compounds 10, 28, 29, 40, 44, 47-53, 56, 58-60, 62 and 63. Relationships between structure and genotoxic activity showed that all compounds containing an aromatic -NO2 group, except compound 27, were active. Among derivatives containing a -CH2Cl group in the acylic chain, only compound 40 showed mutagenic activity.

Studies of aquatic fungi. XX. Seasonal changes of mycoflora in the pool in the palace park.

Studies of aquatic fungi were carried out in the annual cycle in the years 1987-1989 against a background of hydrochemical relations in the pool in the park of the Medical Academy in Białystok. 59 species of aquatic fungi were found, of which a prevailing number were reported many times in various water reservoirs of Poland. Such species, however, as Diplophlyctis complicata, Nowakowskiella atkinsii, Allomyces moniliformis, Pythium pulchrum, Zoophage phanera, Lemonniera filiforme and Tricladium patulum are new to Polish hydromycoflora. In summer months (July-August) the mycoflora of the pool was poor; the occurrence of various species of aquatic fungi increased in autumn and spring, and less so in winter.

The interactive effect of some molluscicides on the inermediate host of human bilharziasis and their associated zoosporic fungi in the river Nile, Egypt.

Biomphalaria alexandrina and Bulinus truncatus are the main vectors of human Bilharziasis in Egypt. The mycotic inhabitants of both control and treated snails were surveyed at 7 and 15 days intervals. The two molluscicides affected more on the mortality of Biomphalaria alexandrina than Bulinus truncatus. The highest population of zoosporic fungi was collected from Biomphalaria alexandrina, the lowest from Bulinus truncatus. Copper sulphate was more effective than Bayluscide on the fungal species of both snails. Achlya, Dictyuchus and Saprolegnia were the common genera while Leptolegnia caudata and Pythium ultimum completely missed from treated snails. The total colonies of zoosporic fungi were lowered after. 15 days of treatment. The two molluscicides inhibited the cell division of both tested snails.

The occurrence of fungi along the Red Sea coast and variability among isolates of Fusarium as revealed by isozyme analysis.

Nineteen identified species which belong to nine fungal genera were recovered from 14 samples collected from different sites of the Red Sea governorate. The aquatic fungal genera were Allomyces, Dictyuchus, Saprolegnia and Pythium while, the terrestrial fungal genera were Aspergillus, Penicillium, Fusarium, Neurospora and Rhizopus. Aspergillus was the most frequent genus, represented by seven species, of which A. niger, A. flavus and A. ustus were the most common. Penicillium was of occurred less frequently and was represented by two species, while Fusarium was isolated unfrequently and contributed four species. The remaining genera were unfrequent or rare and were each represented by one species. In addition, two electrophoretic isozyme patterns, esterase and glutamate oxalate transaminase (GOT), were determined to measure variability among 10 isolates of Fusarium. The results revealed that the tested fungi differed from each other in one or more esterase bands, except that F. moniliforme isolated from Safaga and from 40 Kilometers south of El-Kaussier yielded similar banding pattern. The activity of GOT was observed in the samples of F. solani and F. oxysporum and not detected in other isolates of Fusarium. The results indicated that F. solani differed from F. oxysporum in the isozymes of GOT, while no differences were observed between the isolated of the same species.

Immunodiffusion test for diagnosing human pythiosis.

An immunodiffusion test was developed for diagnosing subcutaneous and systemic pythiosis in humans. When culture filtrate antigen (CFA) from P. insidiosum was reacted against patient and rabbit antisera, 1-5 precipitin bands occurred both in patient and rabbit antisera, and a line of identity also occurred between patient and rabbit sera. When control P. insidiosum CFA was reacted with 30 apparently normal persons, 20 Thalassemia patients, 2 candidosis and 5 aspergillosis patients, no precipitin bands were found. P. insidiosum CFA also tested with rabbit antibodies to B. dermatitidis, C. immitis, H. capsulatum, P. brasiliensis, C. albicans, M. furfur and A. fumigatus revealed no cross reactions. This test is practical, sensitive and specific.

Rhamnose lipids--biosynthesis, microbial production and application potential.

Biosurfactants containing rhamnose and beta-hydroxydecanoic acid and called rhamnolipids are reviewed with respect to microbial producers, their physiological role, biosynthesis and genetics, and especially their microbial overproduction, physicochemical properties and potential applications. With Pseudomonas species, more than 100 g l-1 rhamnolipids were produced from 160 g l-1 soybean oil at a volumetric productivity of 0.4 g l-1 h-1. The individual rhamnolipids are able to lower the surface tension of water from 72 mN m-1 to 25-30 mN m-1 at concentrations of 10-200 mg l-1. After initial testing, rhamnolipids seem to have potential applications in combating marine oil pollution, removing oil from sand and in combating zoosporic phytopathogens. Rhamnolipids are also a source of L-rhamnose, which is already used for the industrial production of high-quality flavor components.
