[The effect of ultrasonic condensation on gold condenser fillings and on the abutting dental hard tissue].

A prototype ultrasonic condensation device was applied to condense two class V mat-gold fillings in each of 10 extracted molars (group A). For 10 other molars a conventional electric device was used (group B). In five molars of each group the nib face of the condenser was not brought into direct contact with the cavity margins (AI/BI); in the other five molars of the sample this was done intentionally (AII/BII). Only one of the two fillings in each tooth was polished. Replicas of the fillings were examined for marginal adaptation by means of scanning electron microscopy (SEM). After dye penetration for 24 h, ground sections of the teeth were examined by light microscopy and SEM. Enamel cracks and cementum injuries were predominantly observed in unpolished fillings of group AII and BII. The marginal adaptation in group AI was significantly better than in group AII, BI, BII, respectively. The polished fillings demonstrated a significantly lower incidence of dye penetration than unpolished restorations. Our results indicate that only direct treatment of the cavity margins with the ultrasonic device causes enamel cracking and cementum injuries. Avoidance of direct treatment of the cavity margins seems to result in better marginal adaptation. In addition to that, polishing seems to lead to lower rates of microleakage.

The superstructure of chromatin and its condensation mechanism. V. Effect of linker length, condensation by multivalent cations, solubility and electric dichroism properties.

Comparison between the internucleosomal distance found by X-ray solution scattering for chicken erythrocyte (23 nm) and sea urchin (30 nm) chromatin indicates that this distance is proportional to the linker length. The diameter of the condensed sea urchin chromatin fibers is about 45 nm which is significantly larger than in chicken erythrocyte chromatin (35 nm). Trivalent cations (Gd, Tb, Cr) and polyamines spermine and spermidine were found to induce compaction at much lower concentrations than the divalent cations but Gd, Tb, Cr induce aggregation before full compaction of the fibers. The influence of hydrogen bonding is illustrated by comparison of the effects of NaCl, ammonium chlorides on condensation. Solubility experiments indicate that there is a nearly linear dependence of the Mg++ concentration at which precipitation occurs on chromatin concentration and confirm the differences between cations observed by X-ray scattering. The chicken erythrocyte chromatin samples were further characterized by their reduced electric dichroism. The values found are consistent with the model derived from X-ray scattering and are compared with those reported in the literature.

Mean evaporation and condensation coefficients based on energy dependent condensation probability.

A generalization of the classical Hertz-Knudsen and Schrage laws for the evaporation mass and energy fluxes at a liquid-vapor interface is derived from kinetic theory and a simple model for a velocity dependent condensation coefficient. These expressions, as well as the classical laws and simple phenomenological expressions, are then considered for the simulation of recent experiments [Phys. Rev. E 59, 419 (1999)]]. It is shown that mean condensation and evaporation coefficients in the mass flow influence the results only if they are small compared to unity and that the expression for evaporation mass flow determines the temperature of the liquid. Moreover, it is shown that the expression for evaporation energy flow plays the leading role in determining the interface temperature jump, which can be obtained in good agreement with the experiment from the generalized kinetic theory model and phenomenological approaches, but not from the classical kinetic-theory-based Hertz-Knudsen and Schrage laws. Analytical estimates show that the interface temperature jump depends strongly on the temperature gradient of the vapor just in front of the interface, which explains why much larger temperature jumps are observed in spherical geometry and the experiments as compared to planar settings.

Condensed matter. Quantum condensate gets a fresh squeeze.

Cooled to a few billionths of a degree above absolute zero, atoms in a Bose-Einstein condensate represent an extreme state of matter. But physicists have now subjected the atoms in condensates to even more outrageous ordeals, squeezing them into one-dimensional lines and two-dimensional planes. The experiments, reported in the 24 September Physical Review Letters, open the door to investigating a new regime of physics in which the rules are easier to understand.

Studies on condensed-heterocyclic azolium cephalosporins. II. Synthesis and antibacterial activity of 7 beta-[2-(2-aminothiazol-4-yl)-2(Z)-alkoxyiminoacetamido]-3-(condensed- heterocyclic azolium)methyl-3-cephem-4-carboxylates.

From our series of studies on cephalosporins bearing condensed-heterocyclic azolium methyl groups at the 3 position in the cephalosporin nucleus, we describe here the synthesis and antibacterial activity of 7 beta-[2-(2-aminothiazol-4-yl)-2(Z)-alkoxyiminoacetamido]ceph alosporins containing imidazo[1,5-a]pyridinium, imidazo[1,2-b]pyridazinium, imidazo[1,2-a]pyrimidinium, imidazo[1,2-c]pyrimidinium, and pyrazolo[1,5-a]pyridinium methyl groups at the 3 position. Among the cephalosporins tested, 7 beta-[2-(2-aminothiazol-4-yl)-2(Z)-methoxyiminoacetamido]-3- (imidazo[1,5-a]pyridinium-2-yl) (1), (imidazo[1,2-b]pyridazinium-1-yl) (2), and (pyrazolo[1,5-a]-pyridinium-1-yl) (3)methyl-3-cephem-4-carboxylates showed potent antibacterial activity and broad antibacterial spectrum. The antibacterial activity of these cephalosporins (1 approximately 3) was superior to that of ceftazidime (CAZ). These results imply that the delocalization of the positive charge of the imidazo[1,5-a]pyridinium, pyrazolo[1,5-a]pyridinium and imidazo[1,2-b]pyridazinium groups leads to an expanded antibacterial spectrum and increased activity and that these condensed-heterocyclic compounds as well as imidazo[1,2-a]pyridine are effective moieties for improving antibacterial activity and spectrum.

Studies on condensed-heterocyclic azolium cephalosporins. IV. Synthesis and antibacterial activity of 7 beta-[2-(5-amino-1,2,4-thiadiazol-3-yl)-2(Z)- alkoxyiminoacetamido]-3-(condensed-heterocyclic azolium)methyl cephalosporins including SCE-2787.

The synthesis and in vitro antibacterial activity of 7 beta-[2-(5-amino-1,2,4-thiadiazol-3-yl)-2(Z)-alkoxyiminoacetami do] cephalosporins bearing various condensed-heterocyclic azolium groups at the 3 position in the cephalosporin nucleus are described. The thiadiazolyl cephalosporins showed good antibacterial activity against both Gram-positive and Gram-negative bacteria and the MICs of the thiadiazolyl cephalosporins against Pseudomonas aeruginosa was more potent than that of the corresponding 7 beta-[2-(2-aminothiazol-4-yl)-2(Z)-alkoxyiminoacetamido]-3- (condensed-heterocyclic azolium)methyl cephalosporins. Also, the thiadiazolyl cephalosporins bearing (imidazo[1,2-b]-pyridazinium-1-yl)methyl groups at the 3 position showed antibacterial activity against methicillin-resistant Staphylococcus aureus (MRSA). Among the cephalosporins tested, 7 beta-[2-(5- amino-1,2,4-thiadiazol-3-yl)-2(Z)-methoxyiminoacetamido]-3-(imidaz o[1,2- b]pyridazinium-1-yl)methyl-3- cephem-4-carboxylate (4, SCE-2787) which exhibited the most potent antibacterial activity and the broadest antibacterial spectrum was selected as a parenteral cephalosporin candidate for further biological evaluation.

Requirement of protamine for maintaining nuclear condensation of medaka (Oryzias latipes) spermatozoa shed into water but not for promoting nuclear condensation during spermatogenesis.

Protamine is an arginine-rich basic protein found in the sperm nuclei of many vertebrates, but its actual roles in spermatozoa remain to be elucidated. In this study, we investigated the physiological roles of protamine by examining protamine-less spermatozoa produced in vitro in the presence of the transcriptional inhibitor actinomycin D. Even under inhibited transcription, medaka spermatocytes underwent meiosis and differentiated into spermatozoa with a condensed nucleus and an elongated flagellum. Using a newly produced anti-medaka protamine antibody, we confirmed the absence of protamine protein in the spermatozoa differentiated in the presence of actinomycin D. These findings clearly indicate that sperm nuclear condensation in medaka is independent of protamine. Since medaka spermatozoa are shed into water upon natural fertilization, we also investigated the roles of protamine by comparing the differences between the nuclear morphology of protamine-equipped and protamine-less spermatozoa immersed in water. The nuclei without protamine more rapidly swelled than did those with protamine and completely broke down within 10 min, whereas more than 80% of the sperm nuclei with protamine resisted the disruption under similar conditions. These findings strongly suggest that a physiological role of protamine in medaka spermatozoa is to protect the ejaculated spermatozoa against the disruption by low osmotic pressure until arrival at the eggs for successful fertilization.

[The balance of harmful trace contaminants between the air humidity condensate and air in a simulator of the Mir orbit station moisture condensation unit].

Subject of the investigation was the balance of harmful trace contaminants (HTC) between the air moisture condensate and air in a simulator of the MIR moisture condensation unit. Experiments involved various classes of water-solvent compounds including alcohols (C1-C4), ketons (C1-C2), aldehydes (C1-C2), fatty acids (C2-C4), esters (acetates C4-C6), and ammonium. For most of the compounds, removal efficiency correlates with air humidity and virtually does not depend on the HTC concentration within the range of 0.25 to 59.1 mg/m3.

A new model of counterion condensation in polyelectrolyte solutions. III. Theoretical predictions of competitive condensation between counterions of different valences.

Our model has been extended for theoretical estimation of competitive condensation of counterions of different valences onto polyelectrolytes in solution. The estimations are compared with those obtained from Manning theory and with experimental data on counterion activity coefficients. The agreement with the data for sodium polystyrenesulfonate/MgCl2, CaCl2 is satisfactory.

Morphology and histogenesis of squamous cell metaplasia of the rat lung after chronic exposure to a pyrolized pitch condensate and/or carbon black, or to combinations of pyrolized pitch condensate, carbon black and irritant gases.

Female Wistar rats were exposed to different concentrations of a pyrolized pitch condensate and/or carbon black particles and/or a combination of irritant gases for 18 hours/day, 5 days/week for 10 months, followed by a clean air period of up to 20 months. Bronchiolo-alveolar hyperplasia and squamous metaplasia were important components of the resulting lesions. Squamous metaplasia and associated hyperplasia was investigated by routine histology, scanning and transmission electron microscopy, and by immunohistochemical detection of various cytokeratins (CKs). Intensely CK positive squamous metaplasia lacking a distinct stratum spinosum was distinguishable from squamous metaplasia with a distinct stratum spinosum that reacted weakly CK positive or CK negative. The CK positive type was histologically characterized by narrow intercellular spaces, the weakly CK positive or CK negative type had markedly enlarged intercellular spaces. Differentiated hyperplastic epithelium and the normal lung parenchyma reacted CK negative. In poorly differentiated hyperplasia of the alveolar type associated with squamous metaplasia scattered cells with characteristics of squamous differentiation were detected. Ultrastructurally these cells showed increased amounts of filament bundles and immunohistochemically a positive reaction with the CK antibody. These cells were regarded as precursor stages of squamous metaplasia of the lung periphery in rats.

Condensed thienopyrimidines. II. Synthesis and gastric antisecretory activity of thiazole and polymethylene condensed thienopyrimidine derivatives.

Thiazolo[3,2-a]thieno[3,2-d]-, [3,4-d]- and [2,3-d]pyrimidin-5-one derivatives (6, 11 and 16), and polymethylene condensed thieno[3,2-d]-, [3,4-d]- and [2,3-d]pyrimidin-5-one derivatives (19-21), in which the oxygen atom of the oxazolidine moiety in 3 was replaced by a sulfur atom or methylene groups, were synthesized and evaluated for gastric antisecretory activity in pylorus-ligated rats. The structure-activity relationships of these compounds are discussed.

Studies on condensed-heterocyclic azolium cephalosporins. III. Synthesis and antibacterial activity of 7 beta-[2-(2-amino-5-substituted-thiazol-4-yl)-2 (Z)-alkoxyiminoacetamido]-3-(condensed-heterocyclic azolium)methyl-3-cephem-4- carboxylates.

As a part of our research on the synthesis of cephalosporins bearing condensed-heterocyclic azolium groups at the 3 position in the cephalosporin nucleus, we describe herein the synthesis of 7 beta-[2-(2-amino-5-halogeno-, methylthio-, methylsulfinyl-, methylsulfonyl- and sulfothiazol-4-yl)-2(Z)-alkoxyiminoacetamido] cephalosporins and their antibacterial activity. Among the compounds prepared, 7 beta-[2-(2-amino-5-chlorothiazol-4-yl)-2(Z)- methoxyiminoacetamido]-3-(imidazo[1,5-a]-pyridinium-1-yl)methyl-3-cephem -4-carboxylate (14) showed good antibacterial activity against both Staphylococcus aureus including methicillin-resistant Staphylococcus aureus (MRSA) and Pseudomonas aeruginosa, whereas the antibacterial activity against other Gram-negative bacteria was a slightly lower than that of 7 beta-[2-(2-aminothiazol-4-yl)-2(Z)-methoxyiminoacetamido]-3-(im ida zo [1,2-a]pyridinium (I-1) and imidazo[1,5-a]pyridinium (I-4)-1-yl)methyl-3-cephem-4-carboxylates.

[Studies on prematurely condensed chromosomes and induced condensed chromosomes with SEM].

The ultrastructure of prematurely condensed chromosomes (PCC) in BK cells and the CHO metaphase chromosomes (the PCC inducer) were studied with cell fusion technique and SEM. The methods for preparing complete specimens for SEM, were used to eliminate artifact as complete as possible, so the natural states of PCC ultrastructure, especially the ultrastructure of the early, mid, and late S-PCC, were observed for the first time. In the three phases of S-PCC multiple double strand helical structure were revealed, on the chromosome segments of BK cells. On the surface of CHO metaphase chromosomes which may induce PCC may regularly arrayed short hair-like loop-protrusions consisting of helical chromatin fibers were observed.
