Variation in nitrogen supply changes water-use efficiency of Pseudotsuga menziesii and Populus x euroamericana; a comparison of three approaches to determine water-use efficiency.

We studied the effects of three nitrogen (N) supply rates (low, intermediate and high) on Douglas-fir (Pseudotsuga menziesii (Mirb.) Franco) seedlings and poplar clone "I-214" (Populus x euroamericana (Dole) Guinier) cuttings growing in mini-stands. Our specific objectives were to: (1) evaluate the effects of N supply on water-use efficiency (WUE) and biomass production; (2) determine if N affects WUE through control of carbon assimilation rates or through stomatal control of water loss; and (3) compare three methods of estimating WUE: one short-term method (WUE(i), based on gas exchange measurements) and two long-term methods (WUE(T), based on the ratio between biomass production and transpired water, and Delta, based on leaf carbon isotope discrimination tested as a proxy of WUE). In both species, biomass production, WUE(i) and WUE(T) increased with increasing N supply, but there was no effect of N supply on either transpiration or stomatal conductance and Delta was negatively related to leaf N concentration. Plots of Delta versus both WUE(i) and WUE(T) revealed negative trends, but the regression between WUE(i) and Delta was significant only for Douglas-fir, and the regression between WUE(T) and Delta was significant only for poplar. Thus, the mechanisms underlying the response of WUE to N supply were mainly related to a positive effect of N supply on photosynthetic rates. The data confirm that carbon isotope discrimination may be a useful proxy of WUE. The finding that N availability enhances both biomass production and WUE may have practical implications in regions where these factors impose constraints on forest productivity.

High efficiency gene transfer is an efficient way of defining therapeutic targets: a functional genomics approach.

BACKGROUND: Dendritic cells are the most potent antigen presenting cells and key to many aspects of the immune function. Studying the intracellular signalling mechanism used by dendritic cells would provide an insight into the functioning of these cells and give clues to strategies for immunomodulation. METHOD: Highly efficient adenoviral infection of dendritic cells for the delivery of transgenes was obtained. These viral vectors were used to introduce IkappaB alpha into dendritic cells for the inhibition of NF-kappaB. This was used to investigate the role of NF-kappaB in dendritic cell function. RESULTS: By blocking the NF-kappaB function a potent inhibition of the expression of costimulating molecules by dendritic cells with the concomitant loss of T cell stimulating function was demonstrated. CONCLUSION: The use of adenoviral vectors may be a useful way of studying the role of genes in dendritic cell function.

Highly efficient, 0.84 slope efficiency, 901 nm, quasi-two-level laser emission of Nd in strontium lanthanum aluminate.

The possibility of using direct pumping into the emitting level of the Nd3+ ion in magnesium-compensated strontium lanthanum aluminate (Sr(1-x)La(x-y)Nd(y)Mg(x)Al(12-x)O19) to improve 900 nm 4F(3/2) --> 4I(9/2) laser emission is discussed. Selection of the composition parameters x and y for optimization of laser emission and reduction of heat generation is based on the spectroscopic and crystal growth characteristics. Pumping in the 865.5 nm absorption band 4I(9/2)(Z1) --> 4F(3/2)(R1) transforms the laser process into a quasi-two-level scheme of very low (below 4%) quantum defects. A very high slope efficiency (over 84%) for 901 nm continuous-wave laser emission is demonstrated with Ti:sapphire laser pumping in this band for a crystal with x = 0.4 and y = 0.05.

Phosphine-triggered complete chemo-switch: from efficient aldehyde-alkyne-amine coupling to efficient aldehyde-alkyne coupling in water.

[reaction: see text] A phosphine ligand served as a remarkable chemo-switch for the silver-catalyzed reaction of alkynes with aldehydes in the presence of amines in water. Exclusive aldehyde-alkyne-amine coupling product was observed in the absence of phosphine, whereas in the presence of a phosphine ligand, exclusive aldehyde-alkyne coupling product was obtained.
