Modulation of the Rev-RRE interaction by aromatic heterocyclic compounds.

The HIV-1 Rev protein regulates the nucleocytoplasmic distribution of viral precursor RNAs that encode HIV-1 structural proteins. Rev-mediated viral RNA expression requires a sequence-specific interaction between Rev and a viral RNA sequence, the Rev responsive element (RRE). Because the Rev-RRE interaction is essential for HIV-1 replication, anti-viral agents that selectively block this interaction may be effective anti-HIV-1 therapeutics. Here, we show that certain aromatic heterocyclic compounds, in particular, a tetracationic diphenylfuran, AK.A, can block binding of Rev to its high-affinity viral RNA binding site. AK.A abolishes Rev-RRE interactions at concentrations as low as 0.1 microM. Inhibition appears to be selective and results from competitive binding of the drug to a discrete region within the Rev binding site. Interestingly, the molecular basis for the AK.A-RNA interaction, as well as the mode of RNA binding differs from previously described aminoglycoside Rev inhibitors. Analysis of a variety of aromatic heterocyclic compounds and their derivatives reveals stereo-specific features required for the inhibition. Our results further demonstrate the feasibility of identifying and designing small molecules that selectively block viral RNA-protein interactions.

[Advances in the development of new antitubercular agents from orthocondensed heterocyclic compounds. Part 3. Substances containing several types of heteroatoms in the six-membered ring].

Tuberculosis and other mycobacterial diseases are considered to be one of the most important problems of contemporary health service. Since 1985 and particularly in the 1990s and at present the search for new structures of antimycobacterial agents have ranked among the foremost areas of chemotherapeutic research. The present review paper is already the 18th communication in a group of review papers about substances with antituberculotic effects, and the third, final, communication devoted in this series about the development of new antimycobacterial agents to ortho-condensed heterocyclic compounds in recent 15 years. The classification of ortho-condensed compounds is based on six-membered heterocyclic substructural fragments and the present study deals with the substructural fragments containing a greater number of different heteroatoms. The literature search is based on the journal Chemical Abstracts, Current Awareness in Biomedicine, part Mycobacteria, and original papers. Current Awareness in Biomedicine is, however, a very imperfect literature search source, recording only a fraction of communications. Review papers about five-membered heterocyclic antituberculotic ortho-condensed compounds was published in the present journal in 1999.

Inhibition of dimethylnitrosamine metabolism by some heterocyclic compounds and by substrates and inhibitors of monoamine oxidase in the rat.

Pretreatment of rats with a number of nitrogen-containing heterocyclic compounds was found to inhibit markedly the metabolism of dimethylnitrosamine (DMN) in terms of both CO2 excretion and decline in blood DMN concentration. However, many of these compounds had either much less or no inhibitory effect on the in vivo metabolism to CO2 of a typical mixed-function oxidase substrate, aminopyrine. In addition, a number of model inhibitors of monoamine oxidase (MAO) activity also inhibited DMN metabolism in the intact animal, and a number of primary amines, known substrates of hepatic MAO, inhibited DMN metabolism but not that of aminopyrine in the isolated perfused liver system. These results, together with in vitro data and previously reported studies on the effect of MAO inhibitors and substrates on the mutagenicity of DMN, suggest that the metabolism and bioactivation of DMN may be in part mediated by a MAO type of enzyme activity.

[Synthetic studies on aromatic heterocyclic compounds].

This review covers the concise synthesis of simple coumarin with salicyl aldehydes and Wittig reagent, and a convenient synthesis of polycyclic aromatic heterocyclic compounds using the aryl-aryl coupling reaction of aryl benzamides and aryl benzoates with a Pd reagent is presented. In the first section, the reactions of salicyl aldehydes with OMe, OH, Br, CO(2)Me, and NO(2) groups with carbethoxymethylenetriphenylphophorane are described. Specifically, the reaction of 3-nitrosalicylaldehyde with amines gave benzoxazoles in moderate yield. In the second section, the syntheses of benzo[c]phenanthridine alkaloids, quinazoline alkaloids, and graphislactones in the Pd-mediated biaryl coupling reaction are described. Moreover, the synthesis of benzonaphthazepine and pyrrolophenanthridine alkaloids utilizing regioselective C-H activation with intramolecular coordination of a benzylamine to Pd is described, and the effects of oxygen substituents in the benzoyl part of benzanilides on the coupling position in its biaryl coupling reaction are presented. An effective Pd reagent for the coupling reaction of aryl tolyflate and arene was developed.

A quantitative structure-activity analysis of the mutagenic and carcinogenic action of 43 structurally related heterocyclic compounds.

Quantitative structure-activity relationships for the carcinogenicity and mutagenicity (against Salmonella typhimurium his- TA98, TA100 and TA1537 strains) of 43 structurally related heterocyclic compounds were formulated. The compounds investigated belong to the following two series of congeners : (a) benzo(thio)-pyranoquinolines and (b) benzo(thio)pyranoindoles. Their biologic activities were correlated with the following parameters : (a) the minimal topological difference (describing the fit of the considered molecules with a possible receptor, enzyme or DNA) and (b) the lipophilicity constants. The computed regression equations suggest that the structural requirements for carcinogenicity are different from those responsible for mutagenicity in the Ames test.

A novel superfamily of transporters for allantoin and other oxo derivatives of nitrogen heterocyclic compounds in Arabidopsis.

A wide spectrum of soil heterocyclic nitrogen compounds are potential nutrients for plants. Here, it is shown that Arabidopsis plants are able to use allantoin as sole nitrogen source. By functional complementation of a yeast mutant defective in allantoin uptake, an Arabidopsis transporter, AtUPS1 (Arabidopsis thaliana ureide permease 1), was identified. AtUPS1 belongs to a novel superfamily of plant membrane proteins with five open reading frames in Arabidopsis (identity, 64 to 82%). UPS proteins have 10 putative transmembrane domains with a large cytosolic central domain containing a "Walker A" motif. Transport of (14)C-labeled allantoin by AtUPS1 in yeast exhibited saturation kinetics (K(m) approximately 52 microM), was dependent on Glc and a proton gradient, and was stimulated by acidic pH. AtUPS1 transports uric acid and xanthine, besides allantoin, but not adenine. Protons are cosubstrates in allantoin transport by AtUPS1, as demonstrated by expression in Xenopus laevis oocytes. In plants, AtUPS1 gene expression was dependent on the nitrogen source. Therefore, AtUPS1 presumably is involved in the uptake of allantoin and other purine degradation products when primary sources are limiting.

Effect of adroxazine, a heterocyclic compound of the adrenals, on the rate of replication of lymphoblastoid cells.

It has been found that the addition of 10(-2)-10(-8) microL/mL of adroxazine, heterocyclic compound of the adrenals, to the tissue culture medium increases the rate at which three strains of lymphoblastoid cells replicate. For example, the addition of 10(-5) micrograms/mL of adroxazine to cultured L5178Y cells decreases their doubling time from approximately 11 to 8 h and addition of 10(-3) micrograms/mL of adroxazine to the tissue culture medium of Huly-16 or Huly-29 cells decreases their doubling time from approximately 43 to 34 and 36 h, respectively. Other concentrations of adroxazine, from 10(-2) to 10(-8) micrograms/mL, cause a less but measurable decrease in doubling time. The effect of adroxazine on the rate of incorporation of [3H]thymidine into acid-precipitable material by lymphoblastoid cells was found to parallel the effect of adroxazine on doubling time. The effect of adroxazine on the length of DNA synthesis and mitosis was calculated from the percent of cells that are labeled with [3H]thymidine during a 20-min interval and the percent of cells in mitosis, respectively. Results showed that adroxazine does not decrease the length of DNA synthesis or mitosis, but markedly decreases the sum of pre- and post-mitotic resting times (TG2 + TG1). Addition of 10(-3) micrograms/mL of adroxazine to the medium decreases the resting times of Huly-16 cells from approximately 20 to 10 h and Huly-29 cells from approximately 12 to 5 h. Addition of 10(-5) micrograms/mL of adroxazine to the medium decreases the resting time of L5178Y cells from 3.3 to 0.1 h.(ABSTRACT TRUNCATED AT 250 WORDS)

Artificial neural network prediction of retention factors of some benzene derivatives and heterocyclic compounds in micellar electrokinetic chromatography.

A 5-4-1 artificial neural network (ANN) was constructed and trained for prediction of the retention factors of some benzene derivatives and heterocyclic compounds in micellar electrokinetic chromatography (MEKC) based on quantitative structure-property relationship (QSPR). The inputs of this network are theoretically derived descriptors that were chosen by the stepwise variable selection techniques. These descriptors are: molecular surface area, maximum value of electron density on atom in molecule, path four connectivity index, average molecular weight, and sum of atomic polarizability which were selected by using stepwise multiple linear regression as a feature selection technique. The standard errors of training, test, and validation sets for the ANN model are 0.091, 0.119, and 0.114, respectively. Results obtained showed that nonlinear model can simulate the relationship between the structural descriptors and the retention factors of the molecules in data set accurately. Also the appearance of these descriptors in QSPR models reveals the role of electronic and steric interactions in solute retention in MEKC.

Fast HPLC for the analysis of oxygen heterocyclic compounds of citrus essential oils.

A fast HPLC method for the determination of the oxygen heterocyclic compounds of citrus essential oils was developed. Five different oils were analyzed under identical conditions, by reversed-phase HPLC with photodiode array detector, for a direct comparison of the composition of their oxygen heterocyclic fraction. Analysis time was 7 min. The oils analyzed were lemon, bergamot, mandarin, sweet orange, and bitter orange. The method developed is good for rapid screening or fingerprinting of these essential oils; a slightly slower method is recommended for higher resolution and better quantitative results.
