[Characteristics of the terminal segments of the lymphatic system of microcirculation according to the data of scanning electron microscopy].

Lymphatic capillaries and postcapillaries in the system of the connective tissue were studied by the method of scanning (raster) electron microscopy. It was shown that these capillaries consisted of chains of ballon-shaped structures-sinuses. Lymphatic capillaries represent an open system which is communicated with the intercellular matter of the connective tissue by means of openings ensuring filtration of the tissue liquid. Impairment of the absorbing function in the fibrous process in the lungs led to deposition in connective tissue cells of cellular detritus and foreign particles with formation around the walls of lymphatic capillaries and postcapillaries of residue-collector zones. Overgrowth of a rough fibrous tissue around these zones brought about atrophy and obliteration of vessels, gradual attenuation of the drenage function in the given region of microcirculation.

[Realities and uncertainties in normal and pathological physiology of the lymphatic system of the extremities].

In the special literature, values of pressures in the lymphatics trunks vary within very large limits, frequently in a contradictory manner, a fact that may generate confusion in the case of interpretation of pathological data. Following an analysis of numerous lymphographic aspects an explanation can be provided, for the radiological modifications of lymph vessels in various pathological circumstances, starting from the most plausible data concerning the lymph circulation in the limbs.

Effect of bile on absorption of mepitiostane by the lymphatic system in rats.

The effects of bile and site of gastrointestinal absorption on the lymphatic absorption of the highly lipophilic drug, mepitiostane were examined using thoracic duct-cannulated rats. The lymphatic absorption from the small intestine was very small in the absence of bile compared with that when bile was present. The lymphatic absorption was greatest when drug was administered to the upper small intestine with bile, was smaller for the lower regions of the small intestine, and was negligible for the stomach and the large intestine. A correlation was observed between the extent of lymphatic absorption and the secretion of chylomicron and very low density lipoproteins after administration to various regions with or without bile. The portal absorption data of mepitiostane confirmed that site specificity occurs in the partition of drug between blood and lymph.

Immunological detection of the oestradiol receptor protein in cell lines derived from the lymphatic system and the haematopoietic system: variability of specific hormone binding in vitro.

Extracts of human MCF 7 mammary carcinoma cells, the human lymphoblastoid cell lines AEH 1 and IM 9, T-cell derived CCRF cells, HL 60 myeloic leukaemia cells and murine myeloma cells SP 0 and NS I were analysed for immunoreactivity with polyclonal goat antibodies raised against homogeneous preparations of C-terminal fragments (32 kDa) of porcine uterine oestradiol receptor (ER). Whole cells and low speed cytosols were analysed for specific oestradiol-binding activity. ERs were enriched from cell extracts by either fractionated ethanol precipitation (0-25% (v/v) ethanol) and/or microscale-immunoaffinity chromatography. Immunoreactive proteins of identical molecular weight (approximately 65 kDa) were detected in all cell lines examined. Whole cell binding assays showed specific oestradiol-binding activity in MCF 7, IM 9 and CCRF cells. Borderline binding was found in HL 60 myeloid cells. No specific binding could be detected in AEH 1, NS I and SP 0 cells. Identical results were obtained using agar-electrophoresis after dextran-coated charcoal treatment. Immunoaffinity purified ERs from MCF 7, AEH 1 and HL 60 cells were subjected to limited proteolysis, where identical tryptic fragments were generated. In conclusion, we have confirmed by immunological methods that ERs are expressed in a variety of cell lines derived from the immune system and the haematopoietic system. The lack of specific hormone binding or very low-affinity hormone binding in some of the cells examined may be due to post-translational events or point mutations.

[Changes in the lymphatic system in the crush syndrome].

In vivo indirect electrolymphoroentgenography was conducted to study the reaction of lymphatics during prolonged compression syndrome (PCS). The studies were made on white rats, Wistar-line, males. It was found out that the early period of clinical course of PCS was characterized by lymphostasis. The intermediate period of PCS corresponds to the stage of the collateral lymphokinesis. At the edge of intermediate and late period of PCS the electrolymphoroentgenogram shows the removal of lymphostasis and recovery of main lymphokinesis.

[The lymphatic system of the larynx. A lymphangioscopic and ultrastructural study (author's transl)].

From post mortem and in vivo tissues, the lymphatic drainage of the human larynx has been evaluated by means of lymphangioscopy. The fine structure of lymphatic capillaries and collecting vessels was examined in an electron microscope study. At the ultrastructural level, the structural components of the laryngeal lymphatic vascular wall showed only minor differences when compared to other lymphatic tissues. These differences concerned the valvular morphology of the lymphatic collecting vessels. There was no evidence of specific lymphatic drainage for distinct laryngeal areas, with the exception of the lymphatic capillaries of the free margin of the vocal cords. On account of anastomoses of lymphatic capillaries and collecting vessels lymphatic drainage could cross over both the laryngeal midline and other laryngeal levels. In pathological conditions with impediment of lymphatic flow, these collateral communications are of clinical significance.

Effect of oily vehicles on absorption of mepitiostane by the lymphatic system in rats.

[14C]Mepitiostane in various vehicles was administered to the small intestine of anaesthetized rats with cannulated thoracic ducts, and the effect of lipids on lymphatic absorption was examined. The extent of lymphatic absorption was greatest when administered in triolein and sesame oil, which are triglycerides of long-chain fatty acids. Absorption in the presence of other vehicles was in the order of 10% Tween 80 aqueous solution greater than monolein greater than oleic acid approximately oleic acid/monolein (2:1 mol/mol) greater than aqueous suspension. Differences between the extents of lymphatic absorption of mepitiostane in the various formulations were not due to variation in the lymph flow but to the increased secretion of chylomicron and very low density lipoproteins. During absorption of mepitiostane from the small intestine, oil affected not only the penetration into epithelium cells and the metabolism in them, but also the partition between blood and lymph.
