[The effect of rolitetracycline on the level of nonesterified fatty acids, phospholipids, and triacylglycerols in ischemic damage to the myocardium].

Myocardial ischemia developed in rabbits after ligation of coronary artery within 60 min. Single dose of rolytetracycline 40 mg/kg was administered intravenously before or after ligation. Alterations in content of unesterified fatty acids and phospholipids were studied in blood serum as well as of unesterified fatty acids, phospholipids and triacyl glycerols--in the myocardium. After ligation of coronary artery content of unesterified fatty acids and phospholipids was increased 2-fold in blood serum as compared with control values, while in the ischemic myocardium these substances were increased more than 2-fold and triacyl glycerol--about 7-fold. Administration of rolytetracycline before ligation of the artery led to a decrease in content of unesterified fatty acids and phospholipids in blood serum down to control values; the ligation-induced high content of triacyl glycerols and phospholipids in the myocardium was also decreased, while alterations in unesterified fatty acids content were not similar. Rolytetracycline administered after the artery ligation caused a decrease in content of unesterified fatty acids but triacyl glycerols were markedly increased. The antibiotic did not affect the lipid content in nonischemized tissues of myocardium.

Improvement of chemical analysis of antibiotics. X. Determination of eight tetracyclines using thin-layer and high-performance liquid chromatography.

Analytical methods for eight tetracyclines (TCs) were established using silica gel high-performance thin-layer chromatography (HPTLC), reversed-phase thin-layer chromatography (RP-TLC) and high-performance liquid chromatography (HPLC). Good separations of eight TCs were obtained using chloroform-methanol-5% disodium ethylenediaminetetraacetate solution (65:20:5) (lower layer) and methanol acetonitrile 0.5 M oxalic acid solution (1:1:4) (pH 3.0) on silica gel HPTLC and C8 TLC plates, respectively. A combination of HPTLC and RP-TLC made possible the identification of the eight TCs. Each calibration graph was linear between 0.1 and 1.0 microgram using UV densitometry except for rolitetracycline. For detection reagents, the diazonium salts including Fast Violet B gave variously coloured spots with the eight TCs and good sensitivities were obtained except with minocycline. In HPLC, the simultaneous analysis of the eight TCs on a C8 column was possible using methanol-acetonitrile-0.01 M oxalic acid solution (1:1.5:7) adjusted to pH 3.0 as the mobile phase. A linear relationship was obtained between 1.0 and 10 ng using the usual sample preparation except for rolitetracycline. The direct determination of rolitetracycline was possible using tetrahydrofuran, dimethyl sulphoxide and the mobile phase as solvents for preparation of the sample. For the determination of residual rolitetracycline, it was effective to measure the amount of rolitetracycline as tetracycline by HPLC, HPTLC and RP-TLC after conversion of rolitetracycline to tetracycline by incubating for 5 min in methanol at 50 degrees C.

[The behaviour of creatine phosphokinase in serum after the intramuscular injection of a Tetracyclin preparation (author's transl)].

The single intramuscular injection of 275 mg Rolitetracyclin (Reverin) led to a rise in serum creatine phosphokinase in 11 out of 20 heart healthy patients, 7 cases with values over 100 mU/ml. In sane cases the initial values had still not been reached 72 hours after injection. With Rolitetracyclin given intravenously the creatine phosphokinase values do not alter, as with an isotonic Na-Cl solution given intramuscularly. A rise in the serum creatine phosphokinase was seen in 2 out of 6 cases after an intramuscular injection of Oxytetracyclin (Terramycin -Depot). One is not dealing with a reaction which is typical only to Rolitetracyclin. The cause is thought to be the setting free of enzymes through the musclelesions. The results underline the sensitivity of and problems involved with creatine phosphokinase in the diagnosis of heart-infarction.

Comparative trial of sclerotherapy for hydroceles.

We present a prospective single-blind trial of sclerotherapy for hydroceles with tetradecyl sulphate and rolitetracycline as sclerosants. Twenty-seven hydroceles were treated with tetradecyl sulphate and 28 with rolitetracycline. The median follow-up was 13 months. There were no statistically significant differences between the 2 drugs in respect of cure rate and complications. The overall cure rate for both drugs was 96%. Complications were rare and common to both drugs. Tetradecyl sulphate and rolitetracycline were found to be equally effective as sclerosants.

Effect of tetracyclines and 4-epiderivatives on the ureter.

1. The effect of tetracyclines on the isolated dog ureter is dependent on: (a) the tetracyclines used-mepicycline and doxycycline antagonizing, and tetracycline and rolitetracycline increasing the contractor action of barium chloride; (b) the percentage of 4-epiderivatives in the tetracyclines used-the higher the epiderivative concentration, the smaller the effect of mepicycline or doxycycline, and the greater the action of tetracycline or rolitetracycline.2. In vivo the addition of the antibiotics into the renal pelvis shows no significant differences between the various tetracyclines or different 4-epiderivative concentrations on the intra-ureteral flow of the dog or guinea-pig.3. Intravenous injection of mepicycline or doxycycline does not induce a significant change in the intra-ureteral flow, while intravenous administration of tetracycline or rolitetracycline produces a triphasic response: (a) a marked decrease of the intra-ureteral flow for a few minutes; (b) a return to the control condition for 30-60 min; and (c) a lesser but persistent decrease in flow for 60-120 minutes. In the first phase the ureteral smooth muscle is directly affected by the antibiotics circulating in the blood, while in the third phase the tetracyclines act via the intra-ureteral mucosa.4. Neurogenic effects on the ureter-bladder junction in vivo are not affected by the tetracyclines tested.

[Antibacterial substances in tamponades after tympanoplasties - an "in-vitro"-study (author's transl)].

This "in-vitro"-study is concerned with the efficiency of antibacterial agents used in packings after tympanoplasties. For this purpose, gel-foam-material was impregnated either with rolitetracyclin (mostly used in German ENT-clinics), or comparatively with a combination of bacitracin and neomycin or with antiseptics like phenylmercuriborate and povidon-iodine. It is tested, how these impregnated "packings" behave in the presence of pathogenic germs (Staph. aureus, Ps. aeruginosa). Antibacterial effects and alterations of the material were judged. It is shown, that in some regards the rolitetracyclin - very successful in clinical use - is superior to the other substances tested.
