Biotypes and randomly amplified polymorphic DNA (RAPD) profiles of subgingival Candida albicans isolates in HIV infection.

A group of subgingival isolates of C. albicans recovered from Italian HIV-positive (HIV+) subjects were characterized both phenotypically and genotypically. Phenotyping of the isolates was carried out by a biotyping method based on the enzyme profiles, carbohydrate assimilation patterns and boric acid resistance of the yeasts. Genotyping was performed through randomly amplified polymorphic DNA (RAPD) analysis. Five biotypes were found among the 29 subgingival C. albicans strains examined. The predominant biotypes were A1R (55.17%), A1S (24.14%), and A2R (13.79%), while the biotypes A11R and A13R were represented by a single isolate each. RAPD profiles identified 15 genotypes among the 29 isolates. Almost every individual harboured his/her own specific isolate and in three out of the six subjects with multiple isolates (two to six each) more than one genotype (two to six) was found. The biotype distribution we found is consistent with previous reports on C. albicans isolates from other oral sources, whereas the resistance to boric acid was highly frequent in subgingival strains. RAPD analysis showed high genetic heterogeneity within subgingival isolates, also when isolates were phenotypically identical. These findings, obtained from HIV+ subjects living in Southern Italy, may be useful as baseline information on subgingival C. albicans colonization in the Mediterranean area.

[Study on PVA-alginate co-immobilization of Xanthomonas ampelina TS206].

Ice nucleation activity and anti-leaking capability are both important technical parameters in INA (Ice nucleation active bactetia) bacteria immobilization which can be adopted on freezing concentrate. Both PVA and alginate are good medium for immobilization. They can be used on co-immobilization of ice nucleation-active bacteria (Xanthomonas ampelinaTS206). The results showed that quantity of embedding affects ice nucleation activity greatly. The order of importance to comprehensive scores of technical standard should be concentration of Sodium alginate > Boric acid > PVA > CaCl2. The optimized concentration are PVA 8%, sodium alginate 1%, CaCl2 1.1% and boric acid 5%. A conclusion can be draw that ice nucleation activity increases with the number of beads and shows little pertinency with the time of immobilization, whereas anti-leaking capability can be influenced faintly by the number of beads and the time of immobilization.

Simultaneous fluorometry of urinary dopamine, norepinephrine, and epinephrine compared with liquid chromatography with electrochemical detection.

A simple fluorometric method is described for simultaneous assay of urinary norepinephrine, epinephrine, and dopamine. The catecholamines are separated by boric acid gel chromatography and subsequently determined by use of a modified trihydroxyindole reaction. By the separation with boric acid gel, dopa can be eliminated, which interferes with dopamine measurement; consequently, the reaction is specific for catecholamines. This simple, rapid method was applied to urine specimens from normal adults. Values for urinary dopamine, norepinephrine, and epinephrine agreed with those obtained by "high-performance" liquid chromatography with electrochemical detection.

Issues concerning the measurement of borate in occupational environments.

Borates are susceptible to weight change due to uptake or loss of water and this hydration instability can lead to gravimetric and interpretation errors in occupational hygiene field sampling of dust. The hydration stability for inhalable borate dust particles (mean diameter 7-22 microns) was characterized over a range of ambient temperature and relative humidity conditions simulating field sampling. Borax 10 mol (Na2O.2B2O3.10H2O), a fully hydrated borate, has a relatively high vapor pressure to water that led to rapid dehydration with significant weight change. Low hydrate borates, Neobor borax 5 mol (Na2O.2B2O3.5H2O), anhydrous boric acid (B2O3), and anhydrous borax (Na2O.2B2O3) were found to hydrate rapidly with an increase in weight. In contrast, boric acid (B[OH]3) and borax 5 mol were found to be stable to dehydration under all conditions. Boron can be measured with high analytical accuracy, but because the specific borate species or borate compounds collected in a 37-mm dust sampler cannot be accurately identified, it is argued that occupational exposure values should be revised to reflect exposure to boron and exposure values for these borates should be the same based on equivalent boron content.

Performance of gel and paste bait products for German cockroach (Dictyoptera: Blattellidae) control: laboratory and field studies.

Gel and paste bait formulations containing boric acid or hydramethylnon were evaluated in field and laboratory experiments with the German cockroach, Blattella germanica (L.). Bait formulations contained 14-62% water by weight and lost the most water after 1 d of desiccation. Bait age and water content did not alter the toxicity (LT50) of boric acid-based baits. Toxicity of hydramethylnon (Maxforce) bait, however, increased with bait age. In continuous exposure tests without harborage, boric acid formulations had LT50s of 8.5 and 10.1 d for Blue Diamond and IT WORKS, respectively. Maxforce bait had an LT50 of 4.2 d. LT50s determined with cardboard harborage or in Ebeling choice boxes were 1.5 to 2.2 times lower than those from continuous exposure tests without harborage. Relative repellency, measured as the mean percent of live cockroaches in the light side of the choice box, was greatest for IT WORKS (21.4%) and Maxforce (25.4%) formulations, followed by Blue Diamond (10.9%). A performance index that combined choice box repellency and mortality data indicated that Maxforce had the greatest potential for field effectiveness. Blue Diamond and Maxforce significantly reduced German cockroach populations in the field. Maxforce performed best in field trials when applied at 10 or 20 placements per apartment. Apartments treated with IT WORKS had significantly greater cockroach trap catches. Factors that may affect the predictive power of the performance index are discussed.

HPLC determination of chloramphenicol degradation in eye drops.

An HPLC-method is described for the simultaneous determination of chloramphenicol and its most important degradation products. A 15 cm long C-18 reversed phase column was used with a mobile phase consisting of boric acid solution-acetonitrile (60 + 45) adjusted to a pH of 3. The amount of degradation of chloramphenicol eye drop solutions containing boric acid and borax, at pH 4.7 and 7.2 respectively, is determined at 4, 21, 100 and 120 degrees C. The solution of pH 7.2 is more stable than that of pH 4.7 at 4 and 21 degrees C, but is less stable at 100 and 120 degrees C. Preparation has to be accomplished in subdued light. The solutions have to be protected from light during storage.

Scanning electron microscopy of Thebesian ostia (microdissection by ultrasonication: enzymatic digestion).

Thebesian vasculature provides for communication between the coronary system and the chambers of the heart. Anatomic, embryologic, physiologic, and therapeutic investigations have involved this component of cardiac anatomy from the early 18th century to the present time. The scanning electron microscope (SEM) now affords an innovative approach to the study of the ostia of these veins as they open into the chambers of the heart. The surface of the intact endocardium is continuous, whether it is treated with boric acid or not, as long as it remains intact. Enzymatic microdissection of tissues with trypsin, hyaluronidase and pronase, followed by similar treatment with boric acid, reveals continuity of successive component layers of the endocardium extending into Thebesian substructure. Thebesian tributaries are easily visualized from the ostia but the deeper capillary network of the Thebesian system is not demonstrable by this approach. Valvular structures such as might prevent retroflow during the cardiac cycle are not present. Our observations with SEM support anatomic relationships indicated by previously published work.

Nonamphoteric isoelectric focusing: II. Stability of borate-glycerol pH gradients in recycling isoelectric focusing.

By complexing polyols with borate in recycling isoelectric focusing and by varying the ratio of polyol to borate over the useful pH range of 4.0-6.0, it is possible to control pH. Twelve solutions of 0.1 M boric acid and varying glycerol concentration were used to vary pH in a twelve-compartment commercial recycling isoelectric focusing (RIEF) system. Various concentrations of boric acid were tested as anolyte, and various Tris(hydroxymethylamino)methane-borate buffer systems were tested as catholyte. Electroosmosis, hydrogen ion flow, and fluid balancing were characterized in two glycerol gradients; one was maintained at 0.06 pH/fraction and the other at 0.12 pH/fraction. In the latter case, ovalbumin (pI4.70) migrated to the pH 4.61 and 4.72 compartments. It is concluded that the borate-glycerol system can be adequately stabilized in RIEF for isoelectric purification of certain proteins.

Treatment of aquarium water by denitrifying photosynthetic bacteria using immobilized polyvinyl alcohol beads.

During the purification of an aquarium for carp breeding, a relatively high level of chemical oxygen demand (COD) was removed by filtration systems packed with both alginate- and polyvinyl alcohol (PVA)-immobilized gel beads of Rhodobacter sphaeroides S. Low nitrate accumulation was observed in the alginate gel beads packed system due to denitrification, but high levels of nitrate and nitrite accumulation were observed in the PVA gel beads packed system. This phenomenon was caused by the inhibitory effect of PVA on nitrite reductase. The boric acid used for hardening gel beads of PVA slightly inhibited nitrate reductase. On the other hand, during the denitrifying growth experiments for this strain, boric acid inhibited cell growth, but PVA only partially inhibited cell growth. Based on electron equivalent (Y(eq.)), growth yields using various kinds of concentrations of PVA were almost identical. It was suggested that PVA might only limit the growth rate of this strain by the inhibition of nitrite reductase.

Analytical potential of 6-oxy-(N-succinimidyl acetate)-9-(2'-methoxycarbonyl) fluorescein for the determination of amino compounds by capillary electrophoresis with laser-induced fluorescence detection.

The analytical potential of a fluorescein analogue, 6-oxy-(N-succinimidyl acetate)-9-(2'-methoxycarbonyl) fluorescein (SAMF), for the first time synthesized in our laboratory, as a labeling reagent for the labeling and determination of amino compounds by capillary electrophoresis (CE) with laser-induced fluorescence (LIF) detection was investigated. Biogenic monoamines and amino acids were chosen as model analytes to evaluate the analytical possibilities of this approach. The derivatization conditions and separation parameters for the biogenic amines were optimized in detail. The derivatization was performed at 30 degrees C for 6 min in boric acid buffer (pH 8.0). The derivatives were baseline-separated in 15 min with 25 mM boric acid running buffer (pH 9.0), containing 24 mM SDS and 12.5% v/v acetonitrile. The concentration detection limit for biogenic amines reaches 8 x 10(-11) mol.L(-1) (signal-to-noise ratio = 3). The application of CE in the analysis of the SAMF-derivatized amino acids was also exploited. The optimal running buffer for amino acids suggested that weak acidic background electrolyte offered better separation than the basic one. The proposed method was applied to the determination of biogenic amines in three different beer samples with satisfying recoveries varying from 92.8% to 104.8%. Finally, comparison of several fluorescein-based probes for amino compounds was discussed. With good labeling reaction, excellent photostability, pH-independent fluorescence (pH 4-9), and the resultant widely suited running buffer pH, SAMF has a great prospect in the determination of amino compounds in CE.

[Toxicological analyses over the past five years at a single institution].

We investigated 352 patients with acute poisoning, 145 males and 207 females, who were sent to the Department of Emergency and Critical Care Center between June 1999 and March 2004. The insurance application rate averaged 72.5%. The causative agents included 98 prescribed drugs, 23 pesticides and herbicides, cyanide, arsenic and boric acid. Hypnotic and psychotropic drugs accounted for 192 cases (47.5%); antipyretics and analgesics, 93 (23.0%); pesticides and herbicides, 65 (16.1%); amphetamine-like drugs, 6 (1.5%); boric acid, 4 (1.0%); cyanide, 3 (0.7%); arsenic, 1 (0.2%); and other agents, 40 (9.9%). In young people, the most prevalent cause of poisoning was hypnotic,or psychotropic drugs prescribed in psychiatry clinics. In people older than 30, poisoning due to pesticides and herbicides was increased, reaching a peak among those in their 40s. Of 23 patients who died, 20 took Paraquat and Diquat, 2 took organophosphates and 1 took an overdose of antidepressants. The other 329 were transferred to different medical departments or were discharged from our hospital after recovery.

Loss and Gain of Aqp10 Paralogs With Broad Solute Selectivity in Anguillid Eels.

Aquaporin (Aqp) 10 is a member of the aquaglyceriporin family, which transports small, uncharged solutes in addition to water. Although the solute selectivity of aquaglyceroporins varies, the mechanism of solute selectivity has not yet been fully elucidated. The common ancestor of ray-finned fish possessed two paralogous genes for aquaporin 10, aqp10.1 and aqp10.2, which produce Aqps with different solute selectivities. Most teleosts possess one or more ohnologs derived from aqp10.1 and aqp10.2; however, the common ancestor of Anguilliformes species lost all aqp10.1-derived ohnologs. Anguilliformes species, except Anguilla species, have one aqp10.2b, but recent tandem duplications in the European eel have generated three aqp10.2b paralogs (aqp10.2b1-aqp10.2b3), whose activities remain ambiguous. In this study, we found that the four sites forming the aromatic/arginine (ar/R) selectivity filter in European eel Aqp10.2b1 were identical to those in Aqp10.2b of other species. However, the Y residue at position 3 was replaced with G in the ar/R selectivity filter of Aqp10.2b2 and b3. When expressed in Xenopus oocytes, Aqp10.2b2 and b3 showed higher permeability to urea and boric acid than Aqp10.2b1, indicating that Aqp10.2b2 and b3 acquired broad solute selectivity similar to that of Aqp10.1, which was lost in the ancestral Anguilliformes species. Urea and boric acid permeabilities of Aqp10.2b1 increased when the Y residue at position 3 of the ar/R selectivity filter was replaced with G. Overall, our results outline the history of the loss and gain of Aqp10 paralogs with broad solute selectivity in anguillid eels.

Control of American cockroaches (Dictyoptera: Blattidae) in sewers.

Baits, dusts, and aqueous sprays were applied to sewer shafts to control American cockroaches, Periplaneta americana (L). Boric acid and hydramethylnon baits failed to provide consistent control because of problems with mold on the baits and because large populations of cockroaches depleted the bait. Bendiocarb, boric acid powder, and a powder mixture of silica gel + synergized pyrethrins provided excellent control for at least 1 mo, but cockroaches reinfested treated shafts within 3-6 mo. Treatments made in the fall when adult cockroach populations were lowest (about 26%) provided best results. Aqueous sprays of wettable powder chlorpyrifos and microencapsulated diazinon provided outstanding control for at least 9 mo. Microencapsulated and emulsifiable concentrate chlorpyrifos sprays provided greater than 94% reductions for a year.

Monitoring of the conversion from triptolide to tripchlorolide in Triptergium wilfordii by micellar electrokinetic caillary chromatography.

A novel concocting method to convert Triptolide (T) into Tripchlorolide (T(4)) in the traditional Chinese herb Tripterygium wilfordii Hook F. and a micellar electrokinetic capillary chromatographic (MEKC) approach by which the conversion of Triptolide (T) and Tripchlorolide (T(4)) was identified and determined had been established. Investigations of the influence of different pH values of boric acid and borax buffer and of sodium dodecyl sulfate (SDS) and organic additive concentrations had been carried out, and the optimum separation for T and T(4) was achieved using boric acid and borax of pH 7.0 with 30 mM SDS and 20% (volume ratio) methanol as the running buffer. It was found that MEKC exhibited good accuracy, precision and repeatability and the content of T(4) was greatly increased in the herb that was treated by the new concocting method.

Analysis of destruxins produced from Metarhizium anisopliae by capillary electrophoresis.

Destruxins are insecticidal metabolites of a fungus, Metarhizium anisopliae. These metabolites are usually secreted into the culture medium during growth. The structure of destruxins is classified as being a cyclic hexadepsipeptide. More than 35 different destruxins have been characterized with a wide range of insecticidal activities. In this report, the destruxins are extracted by acetonitrile and crystallization by lyophilization. The final crystal is subjected for capillary electrophoresis analysis. Because destruxins are relatively hydrophobic compounds, micellar electrokinetic capillary chromatography is used in this series of experiments. The borate-based running buffer is optimized according to (1) boric acid concentration, (2) sodium dodecyl sulfate (SDS) concentration, (3) acetonitrile concentration, and (4) the pH of the running buffer. Optimization is based on resolution and running speed. The results indicate that 20 mM boric acid with 40 mM SDS plus 10% acetonitrile with pH 9.24 is the best set of conditions for both resolution and running speed.

Preventing ascorbate interference in ion-chromatographic determinations of urinary oxalate: four methods compared.

In an attempt to decrease ascorbate interference on the ion-chromatographic determination of urinary oxalate, we compared the effectiveness of four different methods for ascorbate elimination by analyzing a representative urine pool supplemented with successive ascorbate additions. Two of the methods--treatment with ferric ions or boric acid--have been described elsewhere; treatments with nitrites or ascorbate oxidase (EC 1.10.3.3) are investigated here as possible alternatives. Consideration of the main features, advantages, and drawbacks of the four procedures leads us to conclude that boric acid dilution is a good routine method and that pre-incubation with ascorbate oxidase reliably prevents ascorbate interference in assays of urinary oxalate.

The effect of vehicle on corneal penetration of triturated ketoconazole and itraconazole.

BACKGROUND AND OBJECTIVES: Triturated (crushed and suspended) ketoconazole has been recommended for the treatment of fungal keratitis when commercial antifungal eyedrops are unobtainable. The authors evaluated the in vivo corneal stromal concentration with different vehicles in the eyes of adult rabbits. MATERIALS AND METHODS: Ketoconazole and itraconazole tablets were triturated to 20 mg/ml in four vehicles: polyvinyl alcohol (PVA), boric acid, olive oil, and balanced salt solution (BSS). Six eyes (deepithelialized for better penetration) received one drop every 15 minutes for 2 hours. A yeast overlay bioassay of extracts determined the stromal concentration. RESULTS: Itraconazole in BSS, olive oil, PVA, and boric acid produced inhibition zones of 17.3, 15.6, 15.4, and 13.2 mm, respectively. Ketoconazole produced inhibition zones of 35.9, 39.4, 41.8, and 44.7 mm, respectively. From a standard curve, the concentrations of ketoconazole in tissue were 512, 773, 1221, and 1492 micrograms/g, respectively. CONCLUSION: The vehicle that is used to triturate antifungals affects the tissue concentration. This may have an impact on fungal keratitis therapy.
