Sorption of tin on human teeth.

The sorption of tin ions on human teeth from water and other drinks has been studied by the radioactive tracer technique using the isotope 113Sn (T(1/2) = 115.1 d) as a tracer. The tooth holds a minimum amount of tin from coffee and a maximum amount is held from water and from tea with sugar. The same technique was also used to study the desorption of tin already taken up on the teeth. It has been found that sorbed tin is hardly desorbed from the teeth specimens. The high uptake of tin on human teeth from various drinks showed that teeth act as a natural sieve holding trace elements.

Use of radiochemical methods as tools for speciation purposes in environmental and biological sciences.

Chemical speciation for a few elements can be facilitated to a great extent by incorporating a suitable radioisotope into the system and measuring the radiation of the isolated species. This radiospiking can be applied to in vitro and in vivo labelled experiments. Radionuclides are, however, also present as an anthropogenic contaminant from various nuclear fission activities. The radiotracer should be added under such conditions that it behaves in exactly the same way as the isotopes it represents. It should possess an adequate radioactive half-life, and preferably be a gamma-emitter because of the ease of detection. Radiotracer labelling is now widely used to study speciation problems of many essential and toxic elements in body fluids and tissues. It can be used to trace the different locations where the element is metabolized and stored, and subsequently to detect the element in the isolated biocomponents. The determination of the location of a radiotracer in a cell by autoradiography proved to be impractical because of the lack of resolution. Radiochemistry is similarly very useful for investigating particular aspects of the speciation of heavy metals as they occur in the ecosystem, and to follow the fate and effects of fission nuclides in the environment as they are carried around by the water and air masses. However, in certain circumstances the behaviour of fission products appears to be different from that of their stable analogues. For the actinides they simply do not exist. Radiochemical methods are a major tool for identifying and quantifying the nuclides in the different species.

Regulation of DNA synthesis in fat cells and stromal elements from rat adipose tissue.

The incorporation of tritiated thymidine into the deoxyribonucleic acid (DNA) of adipose fat and stromal cells was followed under a variety of conditions. After in vitro incubation of adipose slices or up to 2 days after in vivo injection of the isotope, all DNA radioactivity was in the stromal cell fraction. From 2 to 15 days after thymidine injection total tissue DNA radioactivity was constant, while between 2 and 5 days after injection label in fat cell DNA increased markedly. Thus new labeled fat cells, initially collected in the stromal pool, required 2-5 days after completion of DNA synthesis to accumulate sufficient lipid to be harvested in the fat cell fraction. Fasting before thymidine injection practically abolished DNA synthesis in primordial fat cells and reduced less drastically formation of stromal elements. However fasting sufficient to deplete lipid stores by 50% neither destroyed mature fat cells nor impaired their capacity to reaccumulate fat with refeeding. Other studies evaluated the role of new fat cell formation in the process of lipid accretion accompanying refeeding. These experiments indicated that at least during the early phase of rapid weight gain, accumulation of fat was due to deposition of triglyceride in existing cells rather than to accelerated formation of new fat cells. Studies with hypophysectomized rats demonstrated that pituitary ablation variably affected stromal DNA synthesis and nearly abolished the formation and (or) maturation of primordial fat cells. In these animals growth hormone markedly enhanced thymidine incorporation into stromal DNA but had no effect on fat cell precursors. In intact animals the predominant effect of growth hormone was also on the stromal fraction, although an action of the hormone of lesser magnitude on fat cell precursors was also evident.

Relationship between sarcoplasmic reticulum volume and calcium capacity in skinned frog skeletal muscle fibres.

Semitendinosus fibres from Rana pipiens were examined in the electron microscope. When the aqueous solutions in which the fibres were prepared contained no sucrose, the sarcoplasmic reticulum was markedly swollen and possibly fragmented. When as little as 50 mM sucrose was included in the aqueous solution, the sarcoplasmic reticulum retained the flattened appearance characteristic of intact fibres. The degree of flattening was not substantially different with concentrations of sucrose ranging from 50 to 300 mM, suggesting that the flattened volume may be determined by structural elements. In all the fibre segments, the regularity of the hexagonal filament lattice was disrupted and the disruption was not affected by sucrose. Radioactive calcium uptake was measured in the presence and absence of 200 mM sucrose. The capacity of the sarcoplasmic reticulum (SR) was depressed by about 30% by the sucrose but the initial uptake was not significantly affected. This finding suggests that most of the calcium within the SR is bound and not free in solution.

Oral absorption of lead and iron.

Oral lead and iron absorptions have been measured by the use of radioactive tracer techniques in ten subjects, five of whom had low iron stores as shown by low serum ferritin. The absorption of both elements was abnormally high in these five and also in one subject with normal serum ferritin. The other 4 subjects, who were iron-replete, had absorption values in the normal range. The statistically significant positive correlation between oral lead and iron absorptions suggests that the sizeable proportion of the population "overabsorbing" iron may absorb two to three times more lead than the generally accepted value of 10% of dietary input.

Synthesis of peptidoglycan and membrane during the division cycle of rod-shaped, gram-negative bacteria.

A modified procedure for determining the pattern of peptidoglycan synthesis during the division cycle has allowed the measurement of the rate of side wall synthesis during the division cycle without the contribution due to pole formation. As predicted by a model proposing that the surface growth of the cell is regulated by mass increase, we find a decrease in side wall synthesis in the latter half of the division cycle. This supports the proposal that, upon invagination, pole growth accommodates a significant proportion of the increasing cell mass and that residual side wall growth occurs in response to the residual mass increase not accommodated by pole volume. The observed side wall synthesis patterns support the proposal that mass increase is a major, and possibly sole, regulator of bacterial surface increase. Membrane synthesis during the division cycle of the gram-negative, rod-shaped bacteria Escherichia coli and Salmonella typhimurium has also been measured with similar methods. The rate of membrane synthesis--measured by incorporation of radioactive glycerol or palmitate relative to simultaneous labeling with radioactive leucine--exhibits the same pattern as peptidoglycan synthesis. The results are compatible with a model of cell surface growth containing the following elements. (i) During the period of the division cycle prior to invagination, growth of the cell occurs predominantly in the side wall and the cell grows only in length. (ii) When invagination begins, pole growth accommodates some cytoplasmic increase, leading to a concomitant decrease in side wall synthesis. (iii) Surface synthesis increases relative to mass synthesis during the last part of the division cycle because of pole formation. It is proposed here that membrane synthesis passively follows the pattern of peptidoglycan synthesis during the division cycle.

[A case-control study on the risk factors of leukemia in mining areas of rare-earth in South Jiangxi].

OBJECTIVE: In order to explore the correlation on radioactive contamination of lanthanon to leukemia, and provide clues for the causes and prevention of leukemia in mining areas of rare-earth elements. METHODS: 1:1 matched case-control study was used. A total of 51 clinically confirmed leukemia cases, individually matched with controls from general population, were interviewed in mining areas of rare-earth in South Jiangxi from November to December, 2001. Data were analyzed, using conditional logistic regression. RESULTS: The main risk factors would include frequently drinking water from river (OR = 5.543), distance from residence to rare-earth mine and years for living in the area (OR = 3.308), exposure to organophosphorus pesticide (OR = 3.014). Tea drinking habit appeared to be a protective factor. CONCLUSIONS: Leukemia seemed to be related to environmental pollution with rare-earth elements around the residential areas and organophosphorus pesticide exposure. The protective factor of tea drinking habit seemed to be unique in this study, which called for further studies.

Spallation processes and nuclear interaction products of cosmic rays.

Most cosmic-ray nuclei heavier than helium have suffered nuclear collisions in the interstellar gas, with transformation of nuclear composition. The isotopic and elemental composition at the sources has to be inferred from the observed composition near the Earth. The source composition permits tests of current ideas on sites of origin, nucleosynthesis in stars, evolution of stars, the mixing and composition of the interstellar medium and injection processes prior to acceleration. The effects of nuclear spallation, production of radioactive nuclides and the time dependence of their decay provide valuable information on the acceleration and propagation of cosmic rays, their nuclear transformations, and their confinement time in the Galaxy. The formation of spallation products that only decay by electron capture and are relatively long-lived permits an investigation of the nature and density fluctuations (like clouds) of the interstellar medium. Since nuclear collisions yield positrons, antiprotons, gamma rays and neutrinos, we shall discuss these topics briefly.

The role of radioactive iodine in the treatment of well-differentiated thyroid cancer.

The prognosis in differentiated thyroid cancer is excellent. The completeness of thyroidectomy and removal of involved regional metastases are the most important elements of management. Iodine 131 has a role in ablating residual thyroid and treating regional and distant metastases. The effectiveness of the treatment is confirmed by negative follow-up scans and low or undetectable Tg. This therapy is relatively specific, but other tissues can receive meaningful doses of radiation and acute and long-term complications can occur. Therefore, whether the patient will have a better prognosis after treatment with 131I and whether the benefits of treatment are greater than the side effects must be determined for every patient.

Leaching behavior and characteristics of glass components and surrogate nuclides in radioactive vitrified waste forms.

Several vitrified waste forms were fabricated and characterized, which contain simulated radioactive waste incineration ash, and a long-term leaching test was conducted by an ISO method for 820 days to assess the chemical durability of vitrified waste forms. Two semi-empirical mechanism models were applied to find out the dominant leaching mechanism of glass elements. For glass elements, dissolution associated with diffusion was the dominant leaching mechanism and leaching characteristics also depend upon solubilities of components. A type of prediction model was applied to observe the long-term leaching behavior of major glass elements and surrogates. Diffusion coefficients and dissolution rate constants, the main parameters in the long-term prediction model, were obtained for glass elements and surrogate nuclides using experimental data for short and long-term periods. The model could be used to predict long-term behavior of such elements to observe and assess the stability of vitrified waste forms.

Dietary modulation of small intestinal glutamine transport in intestinal brush border membrane vesicles of rats.

The effects of a glutamine-enriched diet on the transport of glutamine across brush border membrane vesicles (BBMV) from the rat jejunum were studied to gain further insight into the effects of diet on regulating gut glutamine utilization. Following fasting, rats were randomized to one of three nutritionally complete elemental diets supplemented with glutamine, glutamate, or glycine (control). Brush border membrane vesicles were prepared by a Mg2+ aggregation/differential centrifugation technique and uptake of radioactive [3H]glutamine by the BBMV was studied using a rapid mixing/filtration technique. BBMVs from all test diet groups were enriched in alkaline phosphatase 14-fold. [3H]Glutamine uptake courses for all groups demonstrated sodium dependency, overshoots, and similar 2-hr equilibrium values. Vesicles from animals fed the glutamine-enriched diet had a 75% increase in glutamine uptake compared to those of the control diet and a 250% increase compared to those of the glutamate-enriched diet (P less than 0.05). alpha-Methylamino isobutyric acid and glycine did not significantly inhibit total [3H]glutamine uptake, whereas asparagine and glutamine inhibited total [3H]glutamine uptake compared to the mannitol control. The brush border appears to possess the glutamine selective System N transporter, the activity of which can be stimulated by providing dietary glutamine.

Radionuclide applications in laboratory studies of environmental surface reactions.

The advantages of using radionuclides for laboratory studies of environmental processes include the wide range of element concentrations that can be studied, the capability to simultaneously study several isotopes in a single experiment, the direct applicability to the behaviour of radioactive waste or fallout, and the ability to study the mechanisms, reversibility and kinetics of environmental reactions under controlled conditions. These attributes are demonstrated using specific examples drawn from case studies in Australia, including radionuclide fallout onto tropical soils, the association of trace metals with harbour sediments and the behaviour of uranium in natural and contaminated systems.
