Effects of some sugars, amino acids, glycerol and alpha-glycerophosphate on oxygen uptake of some digenetic trematodes.

Effect of various substrates on the oxygen uptake of some digenetic trematodes -- Srivastavaia indica, Gastrothylax crumenifer, Gigantocotyle explanatum from the buffalo, Bubalus bubalis and Isoparorchis hypselobagri from the fish Wallago attu was studied. In all the four species, the effect of various substrates except maltose was found to be stimulatory. The inhibitory or stimulatory effect of these substrates on oxygen uptake reflects the importance of these compounds in the energy metabolism. In spite of the fact that these trematodes were subjected to similar concentration of substrates, the percentage change in oxygen uptake is different in each of the trematodes, the differential utilization is probably related to the different ecophysiological conditions of the habitat of these trematodes. Such studies can be useful in devising better in vitro culture media for these trematodes.

Heat stability studies at the alpha-glycerophosphate dehydrogenase locus in populations of Drosophila melanogaster.

The level of hidden variation in populations of Drosophila melanogaster at the Gpdh+ locus was determined by thermal stability studies of the protein. The results indicate a lack of variation using these methods both in and between the two common electrophoretic variants. It is suggested that alpha-GPDH is conserved in primary structure, which may be related to its critical role in flight muscle metabolism.

Induction of enzymes of the glycerophosphate pathway in leu-5 mutants of Neurospora crassa.

The inducible cytosolic glycerokinase and mitochondrial glycerol-3-phosphate dehydrogenase have been examined during the glycerol-specific induction in Neurospora crassa. Although both the fully induced levels and the respective rates of synthesis of these two enzymes were less than observed with wild-type cells, there were no major differences in the relative rates of induction of the glycerol-3-phosphate dehydrogenase at either permissive or restrictive temperatures. These results indicate that the processes involved in the assembly of this enzyme into the mitochondrial inner membrane are normal in a mutant lacking the mitochondrial leucyl tRNA synthetase and suggest that the functions of the mitochondrial synthetase may be replaced by those of the cytosolic leucyl tRNA synthetase.

Variants of alpha-glycerophosphate dehydrogenase in diploids and triploids of Drosophila melanogaster.

On the basis of band staining intensities in electrophoretic runs of single flies homozygous and heterozygous for two alleles at the autosomal locus for GPDH, F allele activity is believed to be 8% lower than S allele activity. Indeed, the intensity distribution in the patterns of FSS and FFS triploid females shows that both are not equally expressed. On a per fly or live weight basis, females with two and three doses of the Gpdh gene show bands with equal staining intensity, thus exhibiting a dosage effect when GPDH activity is estimated on a per cell basis.
