Effect of free and bovine fibrinogen-bound methotrexate and dibromoaminopterin on the dissemination of solid Gardner lymphosarcoma.

Solid Gardner lymphosarcoma transplanted subcutaneously to mice of strain C3H/Sumice does not grow solely at the site of inoculation, but disseminates also to remote tissues. An i. v. administration of fibrinogen-Methotrexate derivative or Methotrexate (8-15 mg antimetabolite per 1 kg) to mice with an early tumor, reduced the weight of the local tumor, prolonged their survival time and also the posttransplantation period during which the dissemination of malignant cells could not be detected. When the above substances were given to mice with advanced tumors whose dissemination could be reliably established, Methotrexate proved more effective than its fibrinogen derivative. 3.5'-dibromoaminopterin administered alone or bound to fibrinogen (14.6 mg of antifolate per kg) showed no effect both on survival rate and on tumor dissemination.

Chemotherapeutic responses in dogs with lymphosarcoma and hypercalcemia.

Twelve dogs with lymphosarcoma and hypercalcemia were treated over a period of 36 months. Signs and laboratory findings were referable to hypercalcemia and azotemia. All dogs were staged, classified histologically, and given cytoreductive chemotherapy, using 5 drugs (vincristine sulfate, cytosine arabinoside, cyclophosphamide, L-asparaginase and prednisone). For azotemia, symptomatic therapy (0.9% NaCl solution and furosemide) was given. Seven dogs responded completely, with marked reduction of lymphadenopathy and return of serum calcium concentration to normal. Median duration of remission in this group was 48 days (range, 14 to 93), and median survival time was 112 days (range, 85 to 153). Five nonresponding dogs had less than 50% reduction in measurable tumor mass, although serum calcium concentration returned to normal. The median survival time for this group was 34 days (range, 23 to 68). Two of the nonresponders died from sepsis and another from disseminated intravascular coagulation. Response to therapy did not appear to be influenced by age, breed, sex, initial calcium concentration, degree of azotemia, or histologic classification.

Lymphosarcoma associated with virus-like intranuclear inclusions in a California king snake (Colubridae: Lampropeltis).

A laboratory-bred and laboratory-reared male California king snake was submitted for clinical evaluation, and a mass was palpated caudal to the stomach and within the abdominal cavity. The snake was anesthetized and the mass was excised. A constricted atonic section of adjacent small intestine was removed and the ends were subsequently reanastomosed. The snake died 2 days post surgery; after histopathologic examination of all major organs (except muscle and brain), lymphosarcoma was diagnosed. Lymphocytes, approximately 70% of which contained small to large eosinophilic intranuclear inclusions, had infiltrated all major organs. By electron microscopic examination these inclusions were found to be large electron-dense granular structures. Aggregates of circular structures that measured 50 nm were seen in the cytoplasm of several cells.

Presence of DNA polymerase in lymphosarcoma in northern pike (Esox lucius).

Northern poke lymphosarcoma DNA polymerase was partially purified from particulate fractions banding at 1.15 to 1.16 g/ml from homogenates prepared from frozen necropsies of tumor-bearing pike. The enzyme behaves as a typical reverse transcriptase, in that it prefers ribotemplates to deoxytemplates. The isoelectric point (pl 5.5) is similar to that of avian myeloblastosis virus polymerase. The pike enzyme elutes from a phosphocellulose column at 0.22 M potassium phosphate, the same as avian myeloblastosis virus DNA polymerase. The enzyme activity is inhibited by pyran, a specific inhibitor of viral DNA polymerases. The most striking difference between the pike lymphoma polymerase and the other viral DNA polymerases tested is the low maximum temperature of 20 degrees, compared to 30 degrees for Rauscher leukemia virus polymerase and 38 degrees for avian myeloblastosis virus and Rous sarcoma virus.

Causes of death and sex differences in survival times of pigs with untreated hereditary lymphosarcoma (leukemia).

The survival times from birth of 39 entire male Large White pigs with hereditary lymphosarcoma (L/S) were significantly shorter than those of 76 affected females. The survival times of 19 male castrates were for the most part intermediate between those for entire males and females. Analysis of the cause of death revealed that pigs with L/S (L/S-P) were more likely than pigs without L/S to die from colibacillosis, but this disease showed to sex predisposition. In uncomplicated deaths from L/S, weights of splanchnic lymph nodes, spleen, and thymus were greatest in females. The cell-mediated and humoral antibody responses of L/S-P were significantly poorer against 2,4-dinitrofluorobenzene (DNFB) and sheep red blood cells, poorer against skin homografts, but better against bovine serum albumin than those of healthy littermates. Healthy females had stronger DNFB reactions than did healthy males or castrates. Castrated L/S-P responded to DNFB better than did females and significantly better than did entire males.

Localized chemotherapy of Gardner's lymphosarcoma of C3H mice using combinations of carrier-sorbed antifolics and detoxicating tetrahydrofolates.

The effect of aminohexyl-2-hydroxyethylmethacrylate polymer (HEMA-Hex) with sorbed methotrexate (MTX) or 3',5'-dibromoaminopterin (BrAP) on the survival of C3H mice with Gardner lymphosarcoma was studied. The measured bits of HEMA-Hex-MTX or HEMA-Hex-BrAP were implanted into the solid tumor growing 4 to 8 8 days. The doses of sorbed antimetabolites amounting in MTX 4.3 to 13.5 mg.kg-1 and in BrAP 5.1 and 12.6 mg.kg-1 were calculated from the area of the carrier and mean weight of animals. Following implantation i. p. injections of leucovorin or anhydroleucovorin were applied. The treatment of early tumors showed better results than that of advanced ones if evaluated either as prolonged survival or as a number of mice surviving the observation period. The 18-hr. interval between implantation of HEMA-Hex-MTX and anhydroleucovorin injection was optimum if considered both the protection of mice from lethal MTX toxicity and therapeutic effect measured as prolonged survival. Doses of leucovorin or anhydroleucovorin close to MTX doses in term mg.kg-1 resulted in best results. In case leucovorin was applied to tumor-bearing mice pretreated with HEMA-Hex with nonlethal dose of MTX, the survival of mice was shortened. Folic acid did not show this effect. Intra-tumorous implantation of HEMA-Hex-BrAP toxicity even if applied into an advanced tumor. The therapeutic effect of the sorbed BrAP seemed to decrease with tumor progression at a lower rate than of that the sorbed MTX. The application of anhydroleucovorin after implantation of HEMA-Hex-BrAP shortened the survival of tumor-bearing mice.

Ataxia associated with lymphosarcoma in a dog.

A 4-year-old English Springer Spaniel with ataxia was examined because of progression of neurologic signs. Complete physical examination and results of CBC and serum biochemical analysis revealed no other abnormalities. The dog developed respiratory arrest and was euthanatized during recovery from general anesthesia, which had been performed to obtain a CSF sample. Results of CSF analysis were within reference limits. Necropsy revealed a single, well-circumscribed mass in the cerebellum, which was diagnosed as lymphosarcoma. The dog had no peripheral lymphadenopathy or organomegaly suggestive of the generalized form of the disease.

Are matrix-immobilized neoglycoproteins, plant and human lectins and carbohydrate--binding antibodies from human serum mediators of adhesion in vitro for carcinoma and lymphosarcoma cells?

Mediation of cell adhesion by defined molecules can be studied by their immobilization onto a nitrocellulose matrix and incubation with cells. In order to infer the capacity of deliberately selected protein-carbohydrate interactions to establish sugar-inhibitable cell adhesion, a panel of immobilized neoglycoproteins was employed for the murine lymphosarcoma lines RAW-117 with low (P) and high (H10) metastatic capacity, a human mammary carcinoma line (DU4475) and three human colon carcinoma lines (C205, SW480, SW620). Exhibiting an otherwise rather similar behavior relative to the line with low metastatic potential, the murine line RAW117-H10 bound strongly to the matrix with carboxyl group-bearing N-acetylneuraminic acid and glucuronic acid as well as rhamnose. Whereas the analysis of carbohydrate-mediated adhesion yielded comparable results for the three colon carcinoma lines, a markedly reduced number of adherent cells was counted for matrix-attached alpha- and beta-galactosyl, alpha-mannosyl and alpha-glucosyl moieties in the case of the mammary carcinoma line, raising evidence for cell lineage-dependent alterations of this property. From the carbohydrate-binding proteins, the plant lectin, concanavalin A and Viscum album agglutinin almost invariably served well as cell adhesion molecules. Appropriate cell surface sugar receptors, probed with neoglycoproteins, and glycoconjugates, probed with lectins, thus can contribute to adhesion in this model system. The immobilized human beta-galactoside-binding lectin (Mr 14kDa) caused adhesion of the murine lines and one colon carcinoma line (SW480). Neither C-reactive protein under conditions that induce its activity as lectin nor serum amyloid P component nor a lactose-binding immunoglobulin G fraction from human serum were reactive. However, cell adhesion to the alpha-galactoside-binding immunoglobulin G fraction of human serum was seen with the murine line of low metastatic capacity and the mammary carcinoma line. Cells of this line adhered also to the mannan-binding protein from human serum, supporting the view for its potential role in host defence against aberrantly glycosylated tumor cells.

[Functional status of mast cells in subcutaneous connective tissue of rats with lymphosarcoma of Pliss].

The functional status of a population of mast cells taken from the subcutaneous connective tissue of rats with lymphosarcoma of Pliss and those suffering from aseptic inflammation was evaluated. The experiment established such manifestations of remote influence on the mast cell population, at early stages of tumorigenesis, as faster rates of tumor growth and mast cells maturation and enhanced degranulation. Initial signs of functional exhaustion of the mast cells population was seen as a paraneoplastic syndrome.

[The morphoimmunological correlates in the leukemic degeneration of prolymphocytic lymphosarcoma].

Morphocytochemical and immunologic features of the bone marrow and peripheral blood lymphoid elements were studied in 40 patients with B-prolymphocytic lymphosarcoma in the stage of leukemic degeneration. Four types of pathologic lymphoid cells were distinguished: large prolymphocytes expressing pan-B CD19 and HLA-Dr antigens, medium-sized prolymphocytes with round nuclei, characterized by activation CD38 and CD45Ra antigens, prolymphocytes with split nuclei containing light and heavy immunoglobulin chains, and leukemic lymphocytes containing, besides pan-B antigens and immunoglobulins, antigens of the early hemopoietic (CD34) and early B-(CD10) precursors. The content of the RAS-positive substance in the prolymphocytes was inversely proportional to the parameters of activation antigens expression. Acid phosphatase activity correlated with the expression of CD22 and CD45Ra antigens.

[Cytokinetic indices and their role in formulating the prognosis and therapy of lymphosarcomas].

The paper presents a retrospective analysis of clinical evidence elucidating correlations between the disease cytomorphology, proliferative activity, clinical stages, hemopoiesis and survival of the patients. A combined approach oriented on individual measurement of the disease proliferative parameters is recommended to increase lymphosarcoma prognosis reliability and administration of adequate therapy.

[Effect of heparin on the permeability of skin vessels in rats with lymphosarcoma of Pliss].

The effects of heparin on the permeability of skin vessels were investigated in intact rats with lymphosarcoma of Pliss. It has been ascertained that heparin treatment diminishes skin vessels permeability in both intact rats and those with blastoma. More pronounced effect was observed in the experiments involving chronic heparinization of animals, the permeability of vessels in tumor-bearing animals being lower than in intact ones.

Specific expression of cellular oncogenes c-myc and c-myb in T-cell lines established from three types of bovine lymphosarcomas.

Expression of cellular oncogenes in 3 lymphoid cell lines, BTL-PC3 (BoCD2-, BoCD4-, BoCD8-, BoWC1+), BLS1 (BoCD2+, BoCD4-, BoCD8-, BoWC1+) and BLT2 (BoCD2-, BoCD4-, BoCD8-, BoWC1-), which have been established from calf, skin, and thymic types of lymphosarcomas, respectively, were analyzed by DNA-RNA (northern blot) hybridization. To determine specific expression of oncogenes involved in malignant transformation of the lymphoid cells, cellular RNA was isolated from bovine tumor cell lines, BTL-PC3, BLS1, and BLT2, and from Madin Darby bovine kidney cells used as a control for bovine cell lines. The RNA was hybridized against 5 viral oncogene probes (v-jun, v-myc, v-erbB, v-erbA and v-fes), 6 human cellular oncogene probes (N-ras, c-Blym-1 c-erbB-2, c-fos, c-myb and c-abl), human p53 tumor suppressor gene, and bovine LDH-A gene probes. Line BTL-PC3 expressed 2.4-kilobase (kb) c-myc and 4.0- and 3.6-kb c-myb transcripts, and line BLT2 expressed a 3.8-kb c-myb transcript, but line BLS1 expressed no message for the oncogenes tested. Specific transcripts of p53 were found in BTL-PC3 and BLT2 lines, but not in BLS1. Madin Darby bovine kidney cell line expressed multiple cellular oncogenes, c-jun, c-myc, and c-fos, and p53 genes. Southern blot hybridization did not reveal abnormal DNA rearrangements associated with the expressed oncogenes (c-myc and c-myb) in the 3 bovine tumor lines. (ABSTRACT TRUNCATED AT 250 WORDS)

Confirmation of a pulmonary component of multicentric lymphosarcoma with bronchoalveolar lavage in two dogs.

Histologic examination of lung tissue has been the only definitive diagnostic procedure used to confirm pulmonary involvement with lymphosarcoma (LSA) in dogs. Lung involvement with LSA was diagnosed by cytologic examination of bronchoalveolar lavage fluid in 2 dogs with multicentric LSA. Both dogs had cough or dyspnea, in addition to peripheral lymphadenopathy or visceral organomegaly. Both dogs had nonspecific, abnormal thoracic radiographic findings, including diffuse pulmonary interstitial pattern, hydrothorax, and mediastinal and retrosternal lymphadenopathy. In these 2 dogs, postmortem lung histologic examination, performed immediately after bronchoalveolar lavage, confirmed the cytologic diagnosis of pulmonary infiltration with LSA.

Identification of tumor-associated antigen that is expressed on bovine leukemia virus-induced lymphosarcoma cells and expression of its human homologue in human T-cell lymphotrophic virus I-infected cell lines.

By using the monoclonal antibody (MAb) c143 against tumor-associated antigen that is expressed in tumor cells of cattle with bovine leukemia virus (BLV)-induced enzootic bovine leukosis (EBL), we found a novel bovine MHC class II-related antigen which consists of alpha chain (36-37 kDa) and beta chain (32 and 34 kDa). The nature of the c143 antigen was different from previously identified class II antigens, such as DR and DQ, as indicated by test for reactivities with mouse L cell transfectants expressing human class II antigens, sequential immunoprecipitation and tryptic peptide mapping. With the progression of EBL, the number of cells carrying the c143 antigen increased, and the beta chain was specifically phosphorylated at serine residue(s) in lymphosarcoma cells and the cell lines derived from them. A marked difference between expression patterns of the c143 antigen and the class II antigens was observed in lymph nodes from BLV-free and -infected animals. Although bovine mixed lymphocyte reaction (MLR) was inhibited by the addition of MAbs against class II antigens, the c143 MAb did not inhibit a lymphoproliferative response of T cells in the MLR, suggesting that the function of this antigen is distinct from those of classical class II molecules. Significantly, although the human homologue of the c143 antigen is expressed little if at all in human T-cell lines, such as CEM, Molt-4, Jurkat and HPB-ALL, its expression become apparent in T-cell lines infected with human T-cell lymphotrophic virus I (HTLV-1).

Specific inhibition of pre-ribosomal RNA processing in extracts from the lymphosarcoma cells treated with 5-fluorouracil.

To elucidate the molecular mechanism by which the potent anticancer drug, 5-fluorouracil (5-FUra), inhibits cell proliferation, the effect of its metabolite, 5-fluorouridine triphosphate, on transcription of rat rRNA gene and processing of pre-rRNA was investigated in S-100 extract from the mouse lymphosarcoma cells. The in vitro processing of pre-rRNA substrate synthesized from the T3 promoter occurred at the correct primary processing site. Replacement of UMP with 5-fluorouridine monophosphate in the rRNA substrate did not affect the pre-rRNA processing. Similar result was obtained when coupled transcription-processing was studied. When the coupled reaction was examined using extracts from the cells treated with 5-FUra, rRNA processing was abolished whereas transcription of rRNA gene was unaffected. Treatment of cells with thymidine along with 5-FUra did not reverse the inhibitory effect of the drug on rRNA processing. In contrast to the effect on rRNA processing, treatment of cells with 5-FUra did not impede the 3' end processing of pre-mRNA. These data show that inhibition of pre-rRNA processing is a major mechanism of action of 5-FUra and suggest that the activity and/or synthesis of a trans-acting factor(s) involved in this reaction is altered by the anticancer drug.

[Lymphosarcoma of the lacrimal gland].

The malignant lymphoreticular tumours of the lacrimal gland are quite rare. A female patient 65 years old presents a neoformation at the level of the lacrimal gland of the left eye, adhering to the profound planes. The extirpation of the lacrimal gland shows a diffuse infiltrate with atypical cells, shape, volume and tinctorial inequalities with frequent atypical mitosis, having an aspect of lymphosarcoma. The lymphoreticular infiltrate exceeds the own capsule of the gland, so the patient must follow a roentgen-therapeutical treatment.
