Differential pulse and AC polarography of reserpine, using an aprotic organic solvent system.

Previously demonstrated advantages of aprotic organic solvent for analysis of organic pharmaceuticals have been applied to the assay of reserpine tablets. A procedure using high sensitivity electroanalytical relaxation methods was developed which is simpler and more sensitive than the USP assay. Most data were obtained with a commercial differential pulse polarograph which is widely available. However, less extensive measurements with other instruments established the general applicability of the differential pulse method, as well as various acpolarographic techniques. The results of differential pulse polarographic (DPP) and USP assays of synthetically formulated reserpine tablets compare favorably. The DPP method yields an average recovery for 0.1 mg reserpine/tablet of 100%, with a range of 97.6-102.3%. Corresponding data for 0.25 mg reserpine/tablet are 99.3%, with a range of 97.6-100.6%. Satisfactory results also were obtained in content uniformity studies, using single 0.1 and 0.25 mg tablets.

[The potentials of polarography in predicting the effect of a reconstructive operation in patients with severe ischemia of the lower extremities].

Possibilities to prognose the effect of reconstructive operation on the lower extremity vessels are shown on the basis of transcutaneous of oxygen on the dorsal surface of the foot in complex with ultrasonic Dopplerometry in patients with severe ischemia of lower extremities. Effectiveness of the test with the orthostatic growth of Po is proved. An examination of 60 patients with severe ischemia of lower extremities and 10 patients without pathology of main vessels (control group) was performed. Operations were performed on 48 patients, in 35 of them reconstructive operations were made. No correlation was found between preoperative value of Po and a result of the reconstruction. A correlation was found between the value of the orthostatic growth of Po and outcome of the reconstructive operation. The average growth of Po in the group of patients with a negative outcome of the reconstruction made up 9.93 + 2.78 mm Hg, with a positive outcome it was 14.137 + 2.30 mm Hg (p < 0.05). In patients with malleolus index of the regional systolic pressure > 0.3 with the orthostatic growth of Po > 20 mm Hg the probability of positive prognosis of the reconstruction made up 88.8%.

[Oxygen consumption by the yeast-like and filamentous forms of Sporothrix schenckii as measured by polarography].

The study of the oxygen uptake by cultures of Sporothrix schenckii as measured with the Clark electrode has shown that when the fungus was grown in a liquid medium, the atmospheric oxygen went into solution very slowly even when the liquid was rapidly stirred. The partial oxygen pressure was very small after some days of culture (no more than 2 or 3% expressed as the saturated value). Hence, it is postulated that the linear part of the growth curve is due to the dissolved oxygen acting as a limiting factor. When the oxygen uptake by filaments, conidia, or yeasts isplotted against the time the curve variations follow the transformations of the fungus.
