On the interaction of bare and hydrated aluminum ion with nucleic acid bases (U, T, C, A, G) and monophosphate nucleotides (UMP, dTMP, dCMP, dAMP, dGMP).

The B3LYP/6-311+G(2df,2p) density functional approach was used to study the interaction that aluminum trication, in the bare and hydrated forms, establishes with the nucleic acid bases and the corresponding monophosphate nucleotides. In this investigation, we determine equilibrium geometry of all possible complexes resulting from the attachment of the ion on the different binding sites selected on each ligand. The relative energies of complexes and metal ion affinities are also given. The most meaningful aspect was found to lie in the energetics of this interaction that underlines a very high affinity of aluminum ion for the examined biological systems.

Clock mutation lengthens the circadian period without damping rhythms in individual SCN neurons.

Spontaneous discharges of individual neurons in the suprachiasmatic nucleus (SCN) of Clock mutant mice were recorded for over 5 days in organotypic slice cultures and dispersed cell cultures using a multi-electrode dish. Circadian rhythms with periods of about 27 hours were detected in 77% of slice cultures and 15% of dispersed cell cultures derived from Clock/Clock homozygotes. These findings indicate that the Clock mutation lengthens the circadian period but does not abolish the circadian oscillation, and suggest an important role of intercellular communication in the expression of circadian rhythm in the SCN.

Control of the vibration damping through surface functionalization of a shear force probe.

Shear force mapping on thiolipid Langmuir-Blodgett monolayers probed with Al-coated tips leads to a contrast highly dependent on the monolayer molecular organization, which does not correspond to the topographical relief of the sample. The use of functionalized surface probes offers the possibility to better control the probe-to-sample interaction. In addition, hydrophilic surface probes are totally insensitive to alkyl chain arrangements in the monolayer. Hydrophobic probes, instead, can be chosen to map shear force on soft samples in liquid environment, since their mechanical properties are not influenced by the surrounding liquid.

Induction of systemic resistance to Pythium damping-off in cucumber plants by benzothiadiazole: ultrastructure and cytochemistry of the host response.

Benzo (1,2,3) thiadiazole-7-carbothioic acid S-methyl ester (BTH, CGA 245704), a non-toxic, synthetic chemical, was applied as a foliar spray to cucumber plants and evaluated for its potential to induce defense mechanisms in root tissues infected by the soilborne pathogen, Pythium ultimum Trow. In non-treated cucumber plants, fungal colonization was intense and paralleled marked host tissue damage, whereas in BTH-treated plants, pathogen ingress towards the vascular stele was apparently halted by the massive deposition of a phenolic-enriched material which occluded a large number of cortical and vascular parenchyma cells. This considerable increase in the accumulation of phenolics was accompanied by cytological disorders of the invading pathogen at a time when the wall-bound cellulose component was preserved. In addition to phenolic compounds, the occluding material contained large amounts of beta-glucoside residues. These residues gradually decreased in the areas neighboring fungal cells whereas phenolic deposition appeared to be more uniformly distributed throughout the occluded host cells. Pathogen penetration in non-occluded cucumber root cells coincided with other changes, mainly characterized by both the deposition onto the inner surface of the cell walls of some heterogeneous wall appositions and the coating of some intercellular spaces with an electron-opaque material. Evidence is provided in this study that BTH has the ability to induce SAR in cucumber. Exogenous, foliar applications of the chemical sensitize susceptible cucumber plants to react more rapidly and more efficiently to P. ultimum attack, mainly through the massive accumulation of phenolic compounds at sites of attempted pathogen penetration.

Application of the Wolf damped Coulomb method to simulations of SiC.

A multibody interatomic potential is developed for bulk SiC using a modification of the Wolf et al. summation technique [D. Wolf, P. Keblinski, S. R. Phillpot, and J. Eggebrecht, J. Chem. Phys. 110, 8254 (1999)] for the electrostatic interaction. The technique is modified to account for the short-range nonpoint charge effect. The nonelectrostatic interaction is modeled by a simple Morse-stretch term. This potential is then applied to beta-SiC to calculate various bulk properties using molecular dynamics simulations. The simulated x-ray diffraction pattern, radial distribution functions, lattice constant, elastic constants, and defect energy agree well with experimental data.

Suppression of damping-off in maize seedlings by Pseudomonas corrugata.

Two strains of Pseudomonas corrugata, (1 and 7), isolated from subtropical and temperate soils in Sikkim Himalaya, respectively, were subjected to Petri-dish as well as plant-based bioassay to examine their potential for disease suppression against three major pathogens of maize. A mixture of Pythium ultimum, P. arrhenomanes and Fusarium graminearum was introduced in the soil; maize seed inoculated with one of the two strains of Pseudomonas corrugata (1 or 7) were sown in pots containing such soil. The bacterial inoculation resulted in significant disease suppression as well as growth promotion of seedlings. The bacterial strains were also evaluated for their intrinsic antibiotic resistance against a range of concentrations of ten antibiotics. While the bacteria were found to be sensitive to gentamycin and rifampicin, they exhibited resistance against ampicillin, carbenicillin and penicillin, even at high concentrations.
