An industrial approach to evaluation of pyrolysis and combustion hazards.

In addition to the usual toxicology studies necessary for the safe manufacture and use of polymers at room temperature, special studies are needed for polymers which will be used at elevated temperatures. This paper discusses various areas to be investigated and principles for deciding on test materials, tests, and test conditions, polytetrafluoroethylene (PTFE) and fluorinated polyethylene-propylene (PFEP) pyrolysis studies being used as an illustrative case history. Some limitations of animal testing also are mentioned. A toxicological spectrum relating toxicological determinants to PTFE temperature is developed.

Formation of brominated dibenzofurans from pyrolysis of the polybrominated biphenyl fire retardant, firemaster FF-1.

The polybrominated biphenyl (PBB) fire retardant, FireMaster FF-1, was pyrolyzed for 20 min at 380-400 degrees C in open glass tubes and in tubes sealed after nitrogen flushing. The pyrolyzed residue was extracted with benzene, and extracts were cleaned up on columns of graphite (Carbopack A) and alumina. Analysis was carried out by low resolution direct probe mass spectrometry (MS). Spectra from extracts of the open tube pyrolyzed material had a series of ions characteristic of tetra- and pentabrominated dibenzofurans as evidenced by comparison with spectra from 2,3,7,8-tetrabromodibenzofuran (TBDF). Confirmatory evidence for the brominated dibenzofurans was obtained by high resolution MS dual ion analysis of certain fragment and molecular ions. Recovery values of TBDF through the cleanup procedure averaged 50% and, using this recovery value and TBDF as an external standard, dual ion analyses indicated that 40 ppm tetra- and 4 ppm pentabrominanted dibenzofuran were produced based on the PBB level used in the pyrolysis experiments. Additional analysis of the open tube pyrolyzed material by gas chromatography/mass spectrometry provided evidence that there was one tetrabromodibenzofuran compound with a retention time equal to that of TBDF. Trace levels (less than 1 ppm) of the molecular ion of tetrabrominated dibenzofuran were found after analysis by low resolution MS of the PBB pyrolyzed under nitrogen in sealed tubes. The experimental evidence is consistent with a mechanism for brominated dibenzofuran formation involving attack of oxygen on PBB compounds.

Analysis of lipid components in zooplankter individuals by reactive pyrolysis-gas chromatography in the presence of organic alkali.

It has been eagerly requested to develop a highly-sensitive method to characterize extremely minute amounts of natural organic materials occluded in meteorites and/or space dusts in order to confirm the existence of life in the extraterrestrial space. In this article, the reactive pyrolysis-gas chromatography (Py-GC) applied to the analysis of the lipid components contained in every zooplankter individual is introduced for the sake of its potential extension to the characterization of trace amounts of the extraterrestrial organic materials. Here, Py-GC was applied to 1) the discriminative analysis among zooplankter individuals cultured in different food concentrations, and 2) the correlation analysis between the distributions of fatty acid components in the lipids of zooplankter individuals and ingested algae phytoplankton.

The value of pyrolysis mass spectrometry to investigate nosocomial outbreaks caused by Serratia marcescens.

Simultaneous outbreaks of S. marcescens infection going on in the Neonatal Intensive Care Unit and the Surgical Department of the same hospital were investigated by pyrolysis mass spectrometry (PyMS). The PyMS analysis of the strains clearly demonstrated that the two outbreaks were caused by different strains. The 14 S. marcescens isolates from the first outbreak were closely related, with the exception of one environmental isolate, which did not harbour the ESBL plasmid, which was present in all other isolates. However, the phage type of all 14 isolates was the same. Among the 9 S. marcescens isolates from the second outbreak, PyMS clearly distinguished 3 that exhibited gentamicin resistance from the remaining 6 gentamicin-susceptible isolates. Phage typing was unhelpful in this case, as none of the isolates were typable. The PyMS typing of nosocomial outbreak strains can reach the level of discrimination approaching that achieved by molecular genetic analysis.

Pyrolysis-gas chromatographic analysis of black paints.

Samples of 79 black paints from 27 manufacturers in the United States were examined to determine the extent to which paints can be distinguished by pyrolysis-gas chromatography. Evaluation by a Carbowax column showed some differences among paints from different manufacturers and among batches of paints from the same manufacturer. However, it was not possible to conclude that paints with identical chromatograms had a common origin.

[The effect of compression treatment on RDF pyrolysis].

The micropore surface area and micropore structure of refuse derived fuel (RDF) was changed by compression treatment. Thermogravimetric curves can be obtained at different pyrolysis temperatures, to estimate some of the kinetic parameters of different samples. A number of data collected and analysis was presented in the paper, together with the rules of kinetic parameters and discussion on the influence of the character of RDF's micropore structure. The reaction rates of the compressed samples were much lower than those of the uncompressed samples. And the optimal reaction temperature was 550 degrees C-650 degrees C. There was a compensation effect between activation energy and the Arrhenius parameters of some compressed samples. The activation energy decreases and the Arrhenius parameters are ten times less than uncompressed samples.

[Investigation of a gas chromatographic column system for the on-line analysis of gaseous components in de-propane tower of pyrolysis equipment].

Multi-dimensional gas chromatograph has become an important process analyzer due to the advantages of high resolution and fast speed. According to the production requirement, a gas chromatographic column switching system has been investigated for the on-line analysis of gaseous components from high-pressure and lower-pressure de-propane towers of pyrolysis equipment. By using two different injection times on three injectors, and fore-flush and back-flush techniques, C2-hydrocarbons, propane, propene, methylacetylene, propadiene and C4-hydrocarbons can be separated on 7 columns in 7 minutes. The practical application showed the developed column system is suitable for the on-line monitoring of the production process.

Elevation of levels of carcinogenic tryptophan pyrolysis products in plasma and red blood cells of patients with uremia.

The carcinogenic tryptophan pyrolysis products, 3-amino-1,4-dimethyl-5H-pyrido[4,3-b]indole (Trp-P-1) and 3-amino-1-methyl-5H-pyrido[4,3-b]indole (Trp-P-2), have been measured in plasma and red blood cells (RBC) of patients with uremia and normal subjects by using a high-performance liquid chromatography (HPLC) method. In both uremic patients and normal subjects, these carcinogens have been detected in RBC as well as plasma. Trp-P-1 and Trp-P-2 levels in plasma of uremic patients were significantly higher than those of normal subjects. Moreover, these carcinogen levels in RBC (per hemoglobin) were significantly elevated in uremic patients in spite of the presence of severe anemia. These results suggest that patients with uremia are continuously exposed to higher levels of these carcinogens as compared with normal subjects. Our data also support the idea that these carcinogens in plasma and RBC are suitable for monitoring exposure levels in humans.

[Determination of mercury in Chinese medicinal material and biological samples using pyrolysis atomic absorption spectrometry].

In this paper a rapid and simple method using pyrolysis coupled with atomic absorption spectrometry for the analysis of total mercury in Chinese medicinal material and biological samples is presented. No sample digestion was needed and this greatly simplifies the analytical procedure and minimizes potential sources of contamination. Under optimum conditions, the reproducibility of the method was 2.1% for peak area and 9.1% for peak height. The detection limit (3sigma) was 6.3 ng x g(-1), and the recovery was within the range of 95%-105%. Several standard reference materials were analyzed and the results were obtained with satisfaction. The performance of the method was compared with inductively coupled plasma-mass spectrometry (ICP-MS), and excellent agreements were observed between these two methods.

Analysis and pyrolysis of some N-nitrosamino acids in tobacco and tobacco smoke.

A new tobacco-specific nitrosamine, 4-(N-nitrosomethylamino)-4-(3-pyridyl)butyric acid (iso-NNAC), has been identified in tobacco, and its structure was confirmed by gas chromatography-mass spectrometry following enrichment of a tobacco extract. The levels of iso-NNAC ranged from 0.01 to 0.95 ppm. It does not induce DNA repair in primary rat hepatocytes and is inactive as a tumorigenic agent in strain A mice. In order to study the fate of nitrosamino acids during smoking, we spiked cigarettes with the following N-nitrosamino acids: iso-NNAC, 3-(nitrosomethylamino)propionic acid (NMPA), 4-(nitrosomethylamino)butyric acid (NMBA), N-nitrososarcosine (NSAR) and N-nitrosoproline (NPRO). NMPA and NMBA were partially transferred, unchanged, during smoking and partially formed the corresponding methyl esters, while pyrolysis of NSAR and NPRO resulted mainly in their decarboxylating products. This is the first time that the pyrosynthesis of methyl esters has been observed during smoking.

[Classification of Chinese traditional medicine, moutan (Paeonia suffruticosa Andr.) cortex by means of pyrolysis-high resolution gas chromatography/pattern recognition].

Thirty-eight samples of Moutan (Paeonia suffruticosa Andr.) Cortex obtained from three different regions (Southwest, East and Middle China) and two samples of unknown region were subjected to analysis with pyrolysis-high resolution gas chromatography (Py-HRGC). Each sample was thus characterized by the peak area of 41 peaks in each Py-HRGC profile. Discriminant analysis (DA), PRIMA and SIMCA pattern recognition were used to recognize the 40 x 41 data matrix. These data analysis gave satisfactory results (DA, 100% correct; PRIMA, 100% correct, 92.2% unique; SIMCA, 92.2% correct, 79% unique). The correct classification of Moutan Cortex for the unknown territory was obtained by three pattern recognition methods. The results showed that Py-HRGC/pattern recognition technique might be a potential tool for the identification of Chinese traditional medicine.

Sonolysis of concentrated aqueous solutions of nonvolatile solutes: spin-trapping evidence for free radicals formed by pyrolysis.

The sonolysis of argon-saturated aqueous solutions of sodium acetate, sodium propionate, amino acids, and sugars was investigated by ESR and spin trapping over a large range of concentrations. The spin trap 3,5-dibromo-2,6-dideutero-4-nitrosobenzene sulfonate was used to examine the possibility of detecting new radicals specifically generated in the high temperature zones surrounding the collapsing cavitation bubbles. At lower concentrations of these solutes, no evidence for specific new radicals formed in the high temperature regions induced by cavitation could be found, and only radicals formed by hydroxyl radical and hydrogen atom abstraction reactions could be detected. These were identified by comparison with the radicals produced by uv photolysis in the presence of hydrogen peroxide. However, at higher concentrations, new radicals (typically methyl radicals) formed in the high temperature interfacial regions induced by cavitation were found for sodium acetate, sodium propionate, amino acids, and sugars (D-mannose, D-glucose, 2-deoxy-D-ribose). These results indicate that pyrolysis radicals can be detected when the nonvolatile solutes are present at high concentrations in the interfacial regions of the cavitation bubbles.

[Chronic obstructive airway diseases caused by long-term occupational exposure to asphalt pyrolysis products].

The paper reports on 6 roofers who have been specialized on production of damming up roofs and insulation of wet rooms for 20 years. There was chronic obstructive bronchitis with functional impairment of different degree in 4 of them. These diseases were acknowledged as occupational diseases by special committee decision (BK SE). The pyrolysis products developing while heating the bituminous masses used for insulation purposes were recognized as causal agents.

Induction of gamma-glutamyl transpeptidase positive foci in rat liver by pyrolysis products of amino acids.

The activities of pyrolysis products for initiating or promoting preneoplastic lesions were tested. In experimental series I, N-2-fluorenylacetamide (2-FAA) was used as an initiator or a promoter and 3-amino-1,4-dimethyl-5H-pyrido-[4, 3-b] indole (Trp-P-1), 3-amino-1-methyl-5H-pyrido [4, 3-b] indole (Try-P-2), 2-amino-6-methyldipyrido [1,2-a:3', 2'-d] imidazole (Glu-P-1) or 2-amino-dipyrido [1, 2-a:3', 2'] imidazole (Glu-P-2) was administered at a fixed dose and for the same period in the initiation stage or the promotion stage. Animals were subjected to partial hepatectomy or treated with carbon tetrachloride (CCl4) to increase the induction of gamma-glutamyl transpeptidase (gamma-GT) positive foci. Trp-P-1 and Glu-P-2 administered in either the initiation or promotion stage significantly increased the induction of gamma-GT positive foci (p less than 0.001). Glu-P-1 increased the induction of gamma-GT positive foci (P less than 0.001) when administered in the initiation stage but it was not effective when administered during the promotion stage. In experimental series II, animals were given a single dose of these chemicals at one of three different dose levels with partial hepatectomy and then were fed with 2-FAA and given a single dose of CCl4. Trp-P-1, Glu-P-1 and Glu-P-2 caused dose-dependent inductions of gamma-GT positive foci.

Metabolic activation of mutagenic tryptophan pyrolysis products by rat liver microsomes.

In an attempt to determine whether cytochrome P-450 metabolizes tryptophan pyrolysis products, 3-amino-1,4-dimethyl-5H-pyrido[4,3-b]indole (Trp-P-1) and 3-amino-1-methyl-5H-pyrido[4,3-b]indole (Trp-P-2) to active forms, studies were done using microsomes and the 9000 x g supernatant (S-9) from polychlorinated biphenyl mixture-treated rat livers together with Salmonella typhimurium TA 98 as a tester strain. The number of revertants increased with increments in the amount of S-9 fraction added. The highest mutation was seen with an amount of S-9 that is equivalent to 2.5 mg of liver tissue; however, further increase in the S-9 fraction resulted in a sharp decline in the number of revertants. There was a similar biphasic response when a relatively large amount of S-9 fraction was incubated for an increasing length of time. Microsomes showed parallel biphasic responses, and, in addition, the 105,000 x g supernatant fraction was ineffective in increasing the number of revertants. These results suggested that most if not all of the ability of the S-9 fraction to convert Trp-1 and Trp-P-2 to active forms resided in the microsomes. The involvement of microsomal cytochrome P-450 in this process was further confirmed by the following evidence. The treatment of rats with polychlorinated biphenyl mixture and 3-methylcholanthrene resulted in a marked increase in the ability of microsomes to activate Trp-P-1 and Trp-P-2, and the activation reaction required reduced nicotinamide adenine dinucleotide phosphate as a cofactor and was inhibited by carbon monoxide, 7,8-benzoflavone, n-octylamine, and 2-diethylaminoethyl-2,2-diphenylvalerate. The mutagenic metabolite formed as a result of microsomal metabolism of Trp-P-2 was fairly stable and survived heating at 90 degrees for 7 min. With a high-performance liquid chromatography, an active metabolite of Trp-P-2 was purified. A preliminary analysis showed this molecule to be an N-hydroxylated derivative of Trp-P-2.

Pyrolysis gas chromatography of Streptococcus faecalis: effect of cultural conditions on pyrochromatograms.

Streptococcus faecalis 251 was cultured under a variety of different growth conditions, i.e. incubation for 24 or 70 h; at 22 degrees, 37 degrees or 45 degrees C; on blood agar or on MacConkey agar plates; aerobically or anaerobically. Replicate cultures were analysed by pyrolysis-gas liquid chromatography on columns of 7% Carbowax 2 M, TPA on Chromosorb G (AW-DMCS, 80-100 mesh) programmed from 40 degrees C up to 170 degrees C. Culture grown under identical conditions resulted in reproducible pyrochromatograms which were only slightly modified by change in temperature of growth from 37 degrees to 45 degrees C, or length of growth from 24 to 70 h, or growth on MacConkey agar instead of blood agar. Growth under anaerobic conditions resulted in a modified pyrochromatogram; while growth at only 22 degrees C resulted in a major change in pyrochromatogram.

[Effect of pyrolysis products of polypropylene plastic on the defence mechanisms of the respiratory system (author's transl)].

Since the use of plastic materials, a change in the pathology of fire victims has been observed. We studied the effects of a single short-term inhalation (30 min) of a sub-lethal dose of polypropylene pyrolysis products (one LD-0). Including control and test animals, 66 rats and 112 mice were used. The exposure provoked disturbances in the antixenic defense mechanisms of the respiratory system, chiefly in tracheo-bronchial defenses, since we observed a lowering of ciliary activity of 35 to 78% in test animals exposed a few hours before, compared with the controls. These changes provoked a significant increase in death-rate of test animals, following experimental airborne infection by Klebsiella pneumoniae. The combustion products of polypropylene plastic materials did not contain hydrocyanic acid nor hydrochloric acid, and neither the temperature of the inhaled air, nor the concentration of carbon monoxide could explain these effects. On the contrary, we can suspect the well known irritative properties of aldehyde compounds formed during smoldering combustion of polypropylene.
