Pi (antitrypsin) typing methods. A comparison of acid starch-gel electrophoresis and isoelectric focusing.

The Pi typing methods acid starch-gel electrophoresis (ASGE) and isoelectric focusing (IEF) have been compared by three reference laboratories: 564 samples of phenotypes Pi M, MS and MZ were tested in each of the three laboratories with a 96% agreement on initial typing. The discrepancies are recorded and reasons for disagreement discussed. IEF is a reliable method for Pi typing and gives results comparable to those obtained by ASGE.

Comparison of methods to detect enteroviral genome in frozen and fixed myocardium by polymerase chain reaction.

BACKGROUND: Understanding the pathogenesis of viral myocarditis is linked to the availability of sensitive assays to detect viral genome in clinical material. Recent advances in molecular techniques permit detection of viral-specific RNA in tissue samples. We describe here a comparison of different methods for RNA extraction, reverse transcription, and gene amplification of Coxsackievirus B3 virus in fixed and frozen mouse myocardium. EXPERIMENTAL DESIGN: Homogenized Coxsackie B3 virus-infected myocardium was assayed for virus titer and formalin fixed, paraffin-embedded sections from the same heart were subjected to RNA extraction by 3 different methods. Optimal conditions were determined for a one-step assay combining reverse transcription and the polymerase chain reaction to detect enteroviral genome in RNA extracted by these different methods. The presence of amplifiable cDNA was confirmed by amplification of porphobilinogen deaminase mRNA as a positive control. RESULTS: Extraction of RNA from paraffin-embedded myocardium after overnight digestion with proteinase K (200 micrograms/ml) and 0.5% sodium dodecyl sulfate was the most efficient of the three methods compared. With our optimized polymerase chain reaction assay, in which the cDNA synthesis and amplification steps are combined, we detected as little as 10 TCID50 of virus from frozen viral stocks and tissue homogenates and 1.0 TCID50 of virus from fixed tissue. CONCLUSIONS: This sensitive polymerase chain reaction assay will facilitate examination of archival clinical samples to establish a retrospective diagnosis of enterovirus infection.

Comparison of methods for estimating carbonaceous BOD parameters.

The performance of seven different methods (Differential, Fujimoto, Thomas, Graphical, Integral, Log-Difference, and Nonlinear Regression) for estimating first-stage, carbonaceous biochemical oxygen demand (CBOD), curve parameters, namely k and L0, were compared using synthetic data generated by Monte Carlo simulation technique. The comparison of the methods was made based on their efficiency in retrieving the original values of k and L0, which were selected to generate the synthetic data. In the first part of the study, five sets of "true" data (without error substitution) with different k and L0 value pairs, (k (d(-1))-L0 (mg l(-1)): 0.23-10,000; 0.23-250; 0.23-50; 0.10-250; and 0.50-250) were used to obtain information about the effect of different k-L0 combinations and of using 5-day and 20-day CBOD data on the performance of the methods. In the second part, the same methods were used to calculate k and L0 for ten sets of synthetic data with log-normally distributed random errors at the coefficient of variation (COV) levels of 0.1, 0.2, and 0.3 for a single k-L0 value pair, (0.23 d(-1); 250 mg l(-1)). The results indicated that: (1) different combinations of k-L0 values had no significant effect on the performance of CBOD curve parameter estimation methods with the "true" data; (2) use of CBOD20 data, i.e., CBOD data collected for 20 days, provided better estimates for k and L0; (3) the Integral and Nonlinear Regression techniques were found to be the most reliable methods for the estimation of CBOD curve parameters among the other methods considered in this study.

Comparison of methods to determine glucose kinetics and measurement of recycling and lactose formation in lactating cows.

Several investigators have suggested that the conventional multicompartmental exponential analysis of in vivo glucose metabolism is arbitrary and possibly not the most accurate description of glucose kinetics, especially in the large animal. In support of that hypothesis, we found that in a systematic comparison of 3 methods, blood-specific radioactivity data in single-injection studies of glucose metabolism in lactating cows was better described graphically, or by a hybrid polynomial-biexponential curve fit, than by an exclusively exponential curve fit. We hypothesized that this finding was attributable to partial failure of linearity and steady-state assumptions that underlie the exponential model. Second, using both an irreversible tracer (3H-labeled glucose) and reversible tracer (14C-labeled glucose), we found that glucose carbon recycling had no effect during the first 2 hours, but became significant in lactating cows 7 hours after injection. Finally, we determined that approximately 52 to 55% of the glucose replacement rate was being used to generate lactose.

Comparison of methods for transcatheter fragmentation of gallstones.

Alternative methods have been considered for treating cholelithiasis. Compared to extracorporeal shockwave lithotripsy (ESWL), a percutaneous endoscopic approach would be more invasive, but would offer the advantage of immediate stone removal without the need for subsequent drug therapy. We performed an in vitro comparison of three methods of transcatheter cholecystolithotripsy with regard to effectiveness of stone fragmentation, damage to the gallbladder mucosa, and compatibility with percutaneous delivery systems. The three devices used for cholecystolithotripsy were the ultrasonic lithotriptor (UL), the electrohydraulic lithotriptor (EHL), and the thulium-holmium-chromium: YAG laser (THC:YAG). The UL effectively fragmented all types of stones studied, although it is necessary to hold the stone against the tip of the probe. The EHL quickly fragmented noncalcified and pigment stones simply by placing the tip in the vicinity of the stone, but calcified stones had to be held in position near the electrode. The THC:YAG was effective at fragmenting each type of stone, but the number of pulses required was quite large, corresponding to 7 min for some stones. The EHL had the most capacity for mucosal damage, followed by the THC:YAG laser. The UL produced no mucosal damage at the exposure times tested. The UL is not compatible with flexible endoscopes while the EHL and the THC:YAG are. Because of the specific advantages and disadvantages of each device, a combination of devices may be required for successful clinical cholecystolithotripsy.

Comparison of six respirator fit-test methods with an actual measurement of exposure in a simulated health care environment: Part I--Protocol development.

Quantitative fit tests (QNFT) have been assumed to be predictive of the protection respirators would provide to a wearer in the workplace. Workplace studies have consistently found no correlation between quantitative fit factors and workplace protection factors. This article is the first in a series of three describing a study designed to compare the fit factors from six QNFT methods against the actual dose of 1,1,2 trichloro-1,2,2 trifluoroethane (Freon-113) received under the same laboratory conditions. Five preliminary studies conducted to develop the protocol to assess the respirator wearer's dose through end-exhaled air analysis are described in this article: (1) chamber characterization, (2) end-exhaled air sampling, (3) skin absorption testing, (4) pharmacokinetic modeling, and (5) subject characterization. It was established that the concentration of corn oil aerosol and Freon-113 could be generated simultaneously in the chamber. It was ascertained that the optimum time to sample the exhaled breath was 30 minutes after the subject exited the chamber. It was also found that in a chamber concentration of 500 ppm, without any respiratory exposure, Freon-113 was still present in the end-exhaled air. This was attributed to skin absorption. The end-exhaled air of subjects exposed to 0.5, 3, 5, 25, 50, and 100 ppm (30 minute time-weighted average) of Freon-113 was evaluated at 30 minutes postexposure. This characterization was then used to predict the actual dose of Freon-113 received during the method comparison and validation testing to be described in subsequent articles.

HLA polymorphism in Bulgarians defined by high-resolution typing methods in comparison with other populations.

In the present study we analyzed for the first time HLA class I and class II polymorphisms defined by high-resolution typing methods in the Bulgarian population. Comparisons with other populations of common historical background were performed. Most HLA-A, -B, -DRB alleles and haplotypes observed in the Bulgarian population are also common in Europe. Alleles and haplotypes considered as Mediterranean are relatively frequent in the Bulgarian population. Observation of Oriental alleles confirms the contribution of Asians to the genetic diversity of Bulgarians. The use of high-resolution typing methods allowed to identify allele variants rare for Europeans that were correlated to specific population groups. Phylogenetic and correspondence analyses showed that Bulgarians are more closely related to Macedonians, Greeks, and Romanians than to other European populations and Middle Eastern people living near the Mediterranean. The HLA-A,-B,-DRB1 allele and haplotype diversity defined by high-resolution DNA methods confirm that the Bulgarian population is characterized by features of southern European anthropological type with some influence of additional ethnic groups. Implementation of high-resolution typing methods allows a significantly wider spectrum of HLA variation to be detected, including rare alleles and haplotypes, and further clarifies the origin of Bulgarians.

Cerium-based demonstration of phosphatase activity in plastic-embedded sections: a comparison with conventional methods.

Cerium-based methods have been used for the demonstration of several phosphatases at alkaline, acid and neutral pH in low temperature acetone-fixed, plastic-embedded sections. At alkaline pH calcium is used as capturing agent and the precipitated calcium phosphate converted to cerium phosphate. At neutral and acid pH cerium is used directly as capturing agent. Cerium phosphate is subsequently visualized using the H2O2-DAB method. A comparison has been made with conventional calcium-cobalt and lead methods. It appeared that calcium-cobalt methods are more susceptible to improvement than lead methods.

[Estimate of digestibility in dairy cows using an indicator method in comparison to the Hohenheimer feed value test].

In order to measure the digestibility of feed with an indicator method two trials were realized on the dairy research farm "Karkendamm". The first trial was to test the suitability of TiO2 as an indicator to determine the digestibility in cows. The rate of recovery of the indicator was 98.8% (95.5%-101.9%). The concentration of TiO2 of the rectal samples in the morning (daily or every second day) corresponded very well with the values from total faeces collection. The samples taken in the evening showed lower concentrations. The estimation of digestibility of organic matter led to the same values with the total collection or the indicator method with morning samples. The digestibility derived from evening samples were 1.7-2.3%-units digestibility derived from evening samples were 1.7-2.3%-units lower in comparison to total collection. In the second trial the digestibility derived from the in vitro method "Hohenheimer-Futterwert-Test" and from a digestion trial with 30 cows using the indicator method were compared. The digestibility calculated from the date of the in vivo trial was 9.9%-units lower than the one from the "Hohenheimer-Futterwert-Test". The depression of digestibility was due to the variation in concentrate feeding between cows. If the cows got less than 8 kg concentrates the depression in digestibility was 1.9% when increasing the level of feeding one unit of maintenance requirement. If the cows got more than 8 kg concentrates the depression was 3.1%-units. It could be concluded that the "Hohenheimer-Futterwert-Test" underestimates the digestibility if the level of feeding is increased to a certain extent.

Evaluation of Aution Max AX-4280 automated urine test-strip analyser.

Aution Max AX-4280, an automated urine test-strip analyser, was evaluated in three centres. Method comparison, imprecision, carry-over, linearity, detection limit and drift studies were performed for glucose, protein, blood and leukocytes using Uriflet S 9UB strips. These strips enable measurement of pH, glucose, protein, blood, leukocytes, ketones, bilirubin, urobilinogen and nitrite. Specific gravity is determined by the refractive index method. Within-run and between-day imprecision, assessed using pooled urines and quality control materials, were good. No drift over 24 h or sample carry-over was observed. Method comparison with quantitative methods for glucose, protein and specific gravity yielded good correlations. Ascorbate negatively interfered with haemoglobin, glucose and nitrite measurements. Acetylsalicylic acid lowered pH, the effect being greatest when protein was absent. During the assessment period no malfunction or breakdown was reported. The Aution Max is easy to use and needs minimal maintenance.

The effect of administration method: a comparison of the Rapaport and Exner Rorschach systems.

This study compared two Rorschach administration methods across a number of test variables. With a within-subjects, counter-balanced design, 20 female subjects randomly received either an initial Rapaport (Rapaport, Gill, & Schafer, 1968) or an Exner (Exner, 1974, 1986) Rorschach administration. Results showed that the Exner administration produced significantly more Color (C), Shading (Shd), and Blend (B) responses than did the Rapaport administration. Intersystem differences were most prominent on the first presentation of the two administrations. The first Exner administration produced significantly more C, Shd, and Blend responses than did the first Rapaport administration. Findings are discussed in light of their clinical implications, limitations in the experimental design, and suggestions for improving future research.

Comparison of methods for the analysis of 226Ra in water samples.

226Ra is one of the key nuclides among the natural radionuclides from the point of view of radiation protection. For monitoring the radiation exposure risk along the water pathway it has to be measured routinely with low detection limits. Because of the properties of 226Ra and its progenies, a number of quite different methods are possible for this purpose. This paper presents a comparison of routine techniques for the determination of 226Ra in water samples as applied by the authors.

[Comparison of methods for computer and visual assessment of stenosing lesions of the coronary arteries. 1].

A system of computer analysis of coronary arteries stenotic lesions developed in the USSR Cardiology Research Centre, Academy of Medical Sciences of the USSR, is described. Reliability of the computer analysis data was assessed, and computer and visual methods were compared on the basis of phantom study. The study was performed in 5 models of stenotic coronary arteries, the main parameters of which were measured mechanically as well as by computer and visual analysis of their images. Both methods proved to give highly reproducible intraobserver and interobserver results. Comparison to mechanically obtained data showed that the results of computer analysis are highly significant while visual evaluation allows no accurate evaluation of the stenosis degree (it is overestimated).

Knee arthroscopy with different anesthesia methods: a comparison of efficacy and cost.

To evaluate the cost and time effectiveness for different anesthesia methods when performing knee arthroscopy, this study compared three different anesthesia methods. Four hundred healthy patients scheduled for knee arthroscopy were randomized to either local anesthesia (LA) (n=200), spinal anesthesia (SA) (n=100) or general anesthesia (GA) (n=100). The LA arthroscopies were performed in a facility set up in our outpatient department without anesthesia service. The SA and GA arthroscopies were performed in our central operating department with full anesthesia service. Comparisons were made between total hospital stay, anesthesia time, surgery time, recovery time, pharmaceutical and total cost. The total hospital time was 130.4 min (SD 35.14, range 63-383) in the LA group compared to 280.4 min (SD 79.29, range 155-589) in the GA group (p<0.001). The total hospital time in the GA group was also shorter than that of 350.3 min (SD 65.37, range 198-502) in the SA group (p<0.001). The time from start of anesthesia to start of surgery was significantly longer in the LA group, 39.2 min (SD 13.13, range 17-87), compared to 20.1 min (SD 4.93, range 11-35) in the SA group and to 17.6 min (SD 4.64, range 9-44) in the GA group. There were no differences in the surgery time for the three groups. The use of LA was shown to save SEK 1011 (Swedish Crowns) per patient compared to SA and GA.

Landscape evaluation: comparison of evaluation methods in a region of Spain.

Landscape evaluation is now widely recognised as a powerful, interdisciplinary, environmental research method. The aim of the present work was to compare two landscape evaluation methods as part of a physical planning process: the cartographic assessment method and the in situ assessment method. This comparison was performed using an area of the Montes de Toledo, in the centre of Spain. Both methods provided similar results for the majority of the landscape units studied. This shows that the cartographic method can be successfully used in landscape evaluation, allowing important savings in terms of fieldwork. However, this is only the case when the information provided by the available maps is sufficiently accurate to allow the correct assignment of coefficients by a panel of experts.

[Comparison of 4 methods of bacterial count determination in burn wounds].

The present study includes twelve patients with second and third degree fresh burns involving 10-70 per cent total body surface area. Standardized surface swab, gaze pad contact, Rodac plates, and burn wound biopsy cultures were obtained twice per week. The various techniques were compared by 48 independent measurements. Confluent growth occur in more than 70 per cent of the Rodac agar plates. There was a significant (alpha less than 1%) correlation between the results of the surface swab and the gaze pad method. A comparison of the number of viable bacteria detected with the burn wound biopsy technique and the surface methods (gaze pad and surface swab) showed no significant correlation. In addition, the number of colony forming units in 8 parallel cultures obtained from different sites of the burn wound was determined. The standard error of mean (SEM) varied between 1.01 log (biopsy), 0.91 log (gaze pad) and 0.61 log (surface swab). Several case reports emphasize that the quantitative biopsy culture seems to give most reliable data in predicting burn wound sepsis.

Comparison of methods eliciting cerebral ischaemic pressor response in the cat.

Three methods of intercepting the blood supply to the brain were tested by means of X-ray angiography and by monitoring the pressor response following cerebral ischaemia. The methods were: (1) occlusion of carotid and basilar artery; (2) occlusion of carotids and vertebral arteries in the cervical canal of the third vertebra; (3) occlusion of carotid and subclavian arteries. The X-ray angiographs showed that in most cases we could close the accessory sources of the cerebral circulation and drastically reduce the blood supply to the brain of the cat. With all three methods the cerebral ischaemia evoked strong blood pressor elevation, which was weaker however when the carotid and basilar artery were clamped, in comparison with the other two methods. This may be explained by the existence of small arteries supplying the lower brain stem and originating intracranially from the vertebral artery near to the junction of the vertebral and anterior spinal artery.

Comparison of methods for separating polycyclic aromatic hydrocarbons by high-performance thin-layer chromatography.

A comparison is made of different stationary phases (silica gel, cellulose, acetylated cellulose, and 3-aminopropyl-, ethyl-, octyl-, octadecyl-, and diphenylsilanized silica gel), mobile phases, and development techniques (conventional, continuous, and multiple) for the separation of environmentally important polycyclic aromatic hydrocarbons by high-performance thin-layer chromatography. Reversed-phase separations using octadecylsilanized silica gel and methanol-water or methanol-water-based ternary solvent systems were found to give the best separations. A new development technique, multiple development with application of the mobile phase further up the plate at each successive development, provided superior resolution of the polycyclic aromatic hydrocarbons compared with conventional multiple development.
