Effect of fat substitutes, sucrose polyester and tricarballylate triester, on digitoxin absorption in the rat.

The effect of non-absorbable fat substitutes (sucrose polyester (SPE) and tricarballylate triester (TCTE)) on [3H]digitoxin intestinal absorption was studied in the rat using a small intestine in-situ perfusion technique. The effect of SPE and TCTE was compared with that of sunflower oil, oleic acid, and saline. After 120 min perfusion, 5% SPE emulsion significantly reduced (P < 0.001) digitoxin absorption compared with all other treated groups. Five per cent TCTE emulsion had a less marked effect than SPE (P = 0.0002) and did not differ from sunflower oil. No difference was found between saline and 5% oleate emulsion, which did not reduce digitoxin absorption compared with other treated groups (P < 0.02). When taurocholic acid and lipase were added, results for the saline-, TCTE-, and SPE-treated groups were similar to those above, but the sunflower oil-treated group showed significantly enhanced (P < 0.01) digitoxin absorption. Thin-layer chromatography of the lipid phases showed hydrolysis of sunflower oil in the presence of taurocholic acid and lipase, but not of TCTE or SPE. The inhibitory effect of the non-absorbable fat substitutes on digitoxin absorption could be related to drug sequestration by the persistent oil phase constituted by the undigested and then unabsorbed fat substitutes. That part of digitoxin dissolved in the undigested oil phase is consequently unavailable for intestinal absorption.

Ribosome-catalyzed polyester formation.

Deamination of phenylalanyl-transfer RNA with nitrous acid yields the alpha-hydroxyacyl analog, phenyllactyl-transfer RNA. When this is incubated in a protein-synthesizing system directed by polyuridylic acid, it yields an acid-precipitable, alkali-labile polyester of phenyllactic acid.

Elucidation of the chain conformation in a glassy polyester, PET, by two-dimensional NMR

The chain conformation of glassy poly(ethylene terephthalate) (PET) was characterized by two-dimensional double-quantum nuclear magnetic resonance (NMR). In amorphous carbon-13-labeled PET, the statistics of the O-13CH2-13CH2-O torsion angle were determined, on the basis of the distinct shapes of the two-dimensional NMR patterns of trans and gauche conformations. In crystalline PET, the trans content is 100 percent, but in the amorphous PET it is only 14 percent (+/-5 percent). An average gauche torsion angle of 70 degrees (+/-9 degrees) was obtained. Implications for materials properties of polyesters are discussed.

Formation of Polyesters by Pseudomonas oleovorans: Effect of Substrates on Formation and Composition of Poly-(R)-3-Hydroxyalkanoates and Poly-(R)-3-Hydroxyalkenoates.

Pseudomonas oleovorans grows on C(6) to C(12)n-alkanes and 1-alkenes. These substrates are oxidized to the corresponding fatty acids, which are oxidized further via the beta-oxidation pathway, yielding shorter fatty acids which have lost one or more C(2) units. P. oleovorans normally utilizes beta-oxidation pathway intermediates for growth, but in this paper we show that the intermediate 3-hydroxy fatty acids can also be polymerized to intracellular poly-(R)-3-hydroxyalkanoates (PHAs) when the medium contains limiting amounts of essential elements, such as nitrogen. The monomer composition of these polyesters is a reflection of the substrates used for growth of P. oleovorans. The largest monomer found in PHAs always contained as many C atoms as did the n-alkane used as a substrate. Monomers which were shorter by one or more C(2) units were also observed. Thus, for C-even substrates, only C-even monomers were found, the smallest being (R)-3-hydroxyhexanoate. For C-odd substrates, only C-odd monomers were found, with (R)-3-hydroxyheptanoate as the smallest monomer. 1-Alkenes were also incorporated into PHAs, albeit less efficiently and with lower yields than n-alkanes. These PHAs contained both saturated and unsaturated monomers, apparently because the 1-alkene substrates could be oxidized to carboxylic acids at either the saturated or the unsaturated ends. Up to 55% of the PHA monomers contained terminal double bonds when P. oleovorans was grown on 1-alkenes. The degree of unsaturation of PHAs could be modulated by varying the ratio of alkenes to alkanes in the growth medium. Since 1-alkenes were also shortened before being polymerized, as was the case for n-alkanes, copolymers which varied with respect to both monomer chain length and the percentage of terminal double bonds were formed during nitrogen-limited growth of P. oleovorans on 1-alkenes. Such polymers are expected to be useful for future chemical modifications.

Pseudomonas oleovorans as a Source of Poly(beta-Hydroxyalkanoates) for Potential Applications as Biodegradable Polyesters.

Pseudomonas oleovorans was grown in homogeneous media containing n-alkanoic acids, from formate to decanoate, as the sole carbon sources. Formation of intracellular poly(beta-hydroxyalkanoates) was observed only for hexanoate and the higher n-alkanoic acids. The maximum isolated polymer yields were approximately 30% of the cellular dry weight with growth on either octanoate or nonanoate. In most cases, the major repeating unit in the polymer had the same chain length as the n-alkanoic acid used for growth, but units with two carbon atoms less or more than the acid used as a carbon source were also generally present in the polyesters formed. Indeed, copolymers containing as many as six different types of beta-hydroxyalkanoate units were formed. The weight average molecular weights of the poly(beta-hydroxyalkanoate) copolymers produced by P. oleovorans ranged from 90,000 to 370,000. In spite of the higher cell yields obtained with octanoate and nonanoate, the use of hexanoate and heptanoate yielded higher-molecular-weight polymers. These copolyesters represent an entirely new class of biodegradable thermoplastics.

Survival of microorganisms in laundered polyester-cotton sheeting.

The effects of wash-water temperature, cold-water or regular detergent, wash-cycle design, drying, and drying temperature on survival of four microorganisms on polyester-cotton sheeting were examined. Escherichia coli T3 bacteriophage survived washing at 24, 35, 46, and 57 C, but not at 68 C. Serratia marcescens survived only the lowest three wash temperatures. Levels of residual Staphylococcus aureus were diminished at the highest two wash temperatures, but survival was substantial even at 68 C. Counts of Bacillus stearothermophilus spores were not altered appreciably by wash temperature. Type of detergent had no practical effect on observed counts. The regular wash cycle was significantly more efficient in removal of microorganisms than the permanent-press cycle. Counts, especially of the bacteriophage and the gramnegative bacterium, were decreased by drying; after drying, the effects of wash-water temperature on S. aureus and B. stearothermophilus were not significantly different. Microorganisms were transferred from inoculated to sterilized sheeting during laundering. The public health significance of these observations is discussed.

Molecular dynamics of hyperbranched polyesters in the confinement of thin films.

Broadband Dielectric Spectroscopy is employed to investigate the molecular dynamics in thin films of hyperbranched polyesters (type AB(1)B(2), with -OH and -OCOCH(3) as terminal groups). Three relaxation processes are detected: alpha, beta and gamma. While the latter two are not influenced by the confinement, a pronounced effect is observed on the alpha relaxation: with decreasing film thickness the slower relaxation modes of the dynamic glass transition are gradually suppressed, resulting in an increase of the average relaxation rate and in a linear decrease of the dielectric strength. This is attributed to an immobilization in confinement of the polymeric segments located at the periphery of the hyperbranched macromolecular structures.

Nonabsorbable fat (sucrose polyester) and the regulation of energy intake and body weight.

A substantial number of people have difficulties in controlling their body weight and energy/fat intake. Fat substitutes may be helpful in solving these problems. We here report the first study on the longer-term effects of the nonabsorbable fat sucrose polyester (SPE) on energy intake in normal-weight subjects. We studied the longer-term (12 days) effects of the consumption of warm meals with normal dietary fat (5.0 MJ, 1,195 kcal) or meals in which 52 g of fat were replaced by the fat replacer SPE (3.1 MJ, 740 kcal) on spontaneous food intake (measured by dietary records), body weight, and gastrointestinal complaints in healthy volunteers in two studies. In the first study the 48 subjects were unaware of the treatment, whereas in the second study the 47 subjects were informed of what they received. In both studies men and women consumed less energy per day during the SPE (11.2 MJ) compared with the fat treatment (12.7 MJ) (P values < 0.0001) for the whole 12-day period. This was accompanied by equivalent changes in body weight. The percentage of energy from fat decreased from 43% during the fat treatment to 32% during the SPE treatment. During the SPE treatment, more gastrointestinal complaints and a higher frequency of defecation were reported. Information about the treatment did not affect. the outcomes. It is concluded that the use of a fat replacer such as SPE helps in reducing fat and energy intake in normal-weight people.

Action of sucrose polyesters on lipid metabolism in rats.

The action of sucrose polyesters (SPE) as undigestible fat-like compounds on lipid metabolism of growing rats was studied. Over a period of 4 weeks the animals were fed a semi-synthetic diet containing 6% of SPE with or without reduction of dietary fat. The substitution of dietary fats by SPE or the additional supply of SPE led to an increase of fecal lipid and cholesterol excretion, which was accompanied by a significant stimulation of in vitro cholesterol synthesis of the liver and by a significant reduction of liver cholesterol. The serum cholesterol level, however, was not affected, whereas the level of triglycerides both in serum and liver was significantly reduced to about 40% of that found in control animals.

Two new sesquiterpene polyesters from the seeds of Euonymus nanoides LOES.

Two new beta-dihydroagarofuran sesquiterpene polyesters, 1S,13-diacetyloxy-4S-hydroxy-6R-(3-)furancarbonyloxy-9S-cinnamoyloxy-beta-dihydroagarofuran (1) and 1S,6R-di(2-)methylbutanoyloxy-4S-hydroxy-8S-benzoyloxy-9R-(3-)furancarbonyloxy-13-acetyloxy-beta-dihydroagarofuran (2), together with one known compound (3) were isolated from the seeds of Euonymus nanoides LOES. Their structures were elucidated by spectroscopic data interpretation. Compound 2 showed moderate antitumor activity against BEL-7402, P-388, HL-60 and A-549.

The ABC carbon and polyester prosthetic ligament for ACL-deficient knees. Early results in 31 cases.

We treated 39 knees with chronic deficiency of the anterior cruciate ligament by reconstruction using the ABC carbon and polyester prosthetic ligament; 31 (79.5%) were reviewed at an average follow-up of 34 months. There had been four complete failures requiring revision. The remaining 27 were studied in detail. On the Lysholm rating, only 11 knees (41%) had good results with a score of over 76. The mean anterior drawer movement was reduced from 7.6 mm before operation to 5.8 mm at review. The mean difference from the opposite uninjured knee was 3.9 mm before operation, 1 mm (in 21 patients) at mean follow-up of 7.4 months and 2.5 mm (in 27 patients) at 34 months, indicating progressive loss of effect. In our opinion the results are unsatisfactory: we do not recommend the use of this prosthetic ligament.

Polarization holographic and surface-relief gratings at 257 nm in an amorphous azobenzene polyester.

Polarization holographic and surface-relief gratings have been recorded in an amorphous azobenzene polyester by use of a frequency-doubled argon-ion laser beam at 257 nm. Higher excited states of azobenzene in the trans and cis configurations contribute to the formation of a diffraction grating in this experiment. A combination of right and left circularly polarized writing beams has been found to give the highest diffraction efficiency. The contributions to the total phase difference that arise from anisotropy and surface relief have been separated experimentally, and it is shown that the surface-relief grating contributes a larger phase difference than that which is due to anisotropy.

Evanescent polarization holographic recording of sub-200-nm gratings in an azobenzene polyester.

I demonstrate high-resolution polarization holographic recording with evanescent waves in a thin film of an azobenzene polyester deposited directly on the hypotenuse of a highly refractive prism. A spatial frequency greater than 7000 lines/mm and diffraction efficiency greater than 1% have been achieved. It was found that diffraction efficiency increases in the dark after the writing beams have been switched off. The biphotonic effect found in other azobenzene polymers, which converts cis states of the azobenzene to trans states followed by an ordering process that is due to aggregation, is proposed as the reason for this increase in diffraction efficiency.

Adsorption of a cholesteric liquid crystal polyester on silver nanoparticles studied by surface-enhanced Raman scattering and micro Raman spectroscopy.

A Raman study of the adsorption of thermotropic cholesteric liquid crystal polyester PTOBDME ([C(34)H(36)O(8)](n)) on Ag surfaces is presented in this work. The affinity and adsorption mechanism of this polymer was tested on Ag metal colloids and on Ag films obtained by direct immobilization of the colloidal nanoparticles. We have first studied the structure of PTOBDME suspended in several solvents in order to identify the Raman bands used as structural markers. The adsorption of the polymer leads to a deep conformational change involving both the main chain and the aliphatic side chain. The interaction of polymers like PTOBDME with metals could be interesting in the formation of functionalized surfaces, providing them with specific physicochemical properties with possible applications in recognition phenomena, which can be easily characterized by Raman spectroscopy.

DOT-ELISA for detection of anti-Cysticercus cellulosae antibodies in human cerebrospinal fluid using a new solid-phase (resin-treated polyester fabrics). Preliminary report.

A dot enzyme-linked immunosorbent assay (DOT-ELISA) was developed to detect specific antibodies in cerebrospinal fluid (CSF) for human neurocysticercosis immunodiagnosis, with Cysticercus cellulosae antigen dotted on a new solid-phase. This was represented by sheets of a synthetic polyester fabric impregnated with a polymerized resin (N-methylol-acrylamide). A very stable preparation was thus obtained, the antigen being covalently bound by cross-linking with free N-methylol groups on the resin. Since robust, no special care was necessary for handling the solid-phase. The test could be performed at room-temperature. From 30 CSF samples assayed, 14 were positive, from a group of 15 cases of neurocysticercosis, with titers from 1 to 128; 15 other samples, from normals or other neurological diseases, were all negative. Test characteristics seem to indicate it as adequate for epidemiological surveys. A more detailed study on sensitivity, specificity, reproducibility and the use in serum samples is being conducted.

Dot-ELISA-IgM in saliva for the diagnosis of human leptospirosis using polyester fabric-resin as support (preliminary report).

In order to improve the diagnosis of human leptospirosis, we standardized the dot-ELISA for the search of specific IgM antibodies in saliva. Saliva and serum samples were collected simultaneously from 20 patients with the icterohemorrhagic form of the disease, from 10 patients with other pathologies and from 5 negative controls. Leptospires of serovars icterohaemorrhagiae, canicola, hebdomadis, brasiliensis and cynopteri grown in EMJH medium and mixed together in equal volumes, were used as antigen at individual protein concentration of 0.2 micrograms/microliters. In the solid phase of the test we used polyester fabric impregnated with N-methylolacrylamide resin. The antigen volume for each test was 1 microliter, the saliva volume was 8 microliters, and the volume of peroxidase-labelled anti-human IgM conjugate was 30 microliters. A visual reading was taken after development in freshly prepared chromogen solution. In contrast to the classic nitrocellulose membrane support, the fabric support is easy to obtain and to handle. Saliva can be collected directly onto the support, a fact that facilitates the method and reduces the expenses and risks related to blood processing.

Dot-ELISA for the detection of anti-Cysticercus cellulosae antibodies in cerebrospinal fluid using a new solid phase (resin-treated polyester fabric) and Cysticercus longicollis antigens.

A dot-ELISA was developed for the detection of antibodies in CSF in the immunologic diagnosis of human neurocysticercosis, using antigen extracts of the membrane and scolex of Cysticercus cellulosae (M+S-Cc) and, alternately, membrane (M) and vesicular fluid (VF) of Cysticercus longicollis (Cl) covalently bound to a new solid phase consisting of polyester fabric treated with N-methylol-acrylamide resin (dot-RT). The test was performed at room temperature, with reduced incubation times and with no need for special care in the manipulation of the support. The sensitivity rates obtained were 95.1% for antigen Cc and 97.6% for antigen Cl. Specificity was 90.6% when Cc was used, and 96.9% and 100% when M-Cl and VF-Cl were used, respectively. No significant differences in titer were observed between tests carried out with homologous and heterologous antigens. The low cost and easy execution of the dot-RT test using antigen extracts of Cysticercus longicollis indicate the test for use in the immunodiagnosis of human neurocysticercosis.

Noise problem in a polyester fiber plant in Pakistan.

Noise study has been undertaken in the three units of a polyester fiber plant. The level and frequency characteristics of the prevailing noise have been studied and speech interference levels evaluated. The results are discussed with reference to the maximum permissible occupational noise exposure limits as allowed by the ISO and other national standards. Some recommendations have also been made to provide safety measures to the workers against high level noise in these units.
