Effects of chronic cochlear damage on threshold and frequency tuning of neurons in AI auditory cortex.

We describe the effects of long-term cochlear lesions on the frequency response properties of AI cortical neurons in the cat. Young animals were treated with amikacin to produce bilateral, basal to mid-turn cochlear lesions. After 12-24 months the response properties of single neurons or small unit clusters in primary auditory cortex were recorded in anesthetized animals. Responses to stimulus frequency and intensity were mapped in detail and frequency threshold curves (FTCs) and Q10dB values were derived. Subsequent to recording experiments, scanning electron microscopy of the sensory epithelium was used to characterize the degree and extent of the cochlear damage. In normal control animals, Q10dB values were, on average, lower than those derived by others from cochlear nerve fibre recordings in the same species. In amikacin-treated animals, deterioration was evident in the threshold and tuning properties of cortical neurons, particularly in those cells whose input originated in damaged cochlear regions. Often, neurons associated with 'normal' cochlear areas (as assessed by scanning microscopy) also had poor frequency tuning compared with controls. As an animal model of sensorineural hearing loss, we consider the cat with long-term cochlear lesions to be more appropriate than animals with acute or short-term pathology. We also suggest that in making physiological-psychophysical correlations, neural responses from the central auditory system (e.g. cortex) should perhaps be given more consideration than data derived at the cochlear level.

Asynchronous interleaved stimulation (AIS): a new speech coding strategy for cochlear implant.

Cochlear implants use a fixed or F0 dependent stimulation rate, whatever the sound analysed (vowel or consonant) or procedure (fixed filters or FFT). We present a procedure which varies as a function of the input signal. This method is based on FFT analysis using a variable width analysis window. Short windows are applied on the transient part of the signal, providing poor frequency resolution but good time resolution; they permit us to accurately follow the speech signal in time during its fast temporal variations. Large windows are applied on the stationary parts of the signal, providing better frequency resolution but poor time resolution. Transient parts of the speech order this window switching; they are detected using the statistical properties of the FFT; moreover, the narrow windowing is coupled to an increase of the stimulation frequency. This strategy has been implemented using the DIGISONIC cochlear implant software, and clinically assessed in 6 regular cochlear implant users, on the basis of a consonant-vowel-consonant test. This Asynchronous-Interleaved-Stimulation (AIS) strategy provides the patients with better discrimination than fixed window FFT analysis. The coding protocol will be described and results presented.

Ultrastructural study of tooth enamel with amelogenesis imperfecta in AI-nephrocalcinosis syndrome.

This paper describes the ultrastructure of the affected enamel and the clinical features in two siblings with the syndrome of nephrocalcinosis and amelogenesis imperfecta. Nephrocalcinosis was diagnosed by intravenous pyelography, and confirmed by ultrasonography and CT scan. Amelogenesis imperfecta AI was diagnosed clinically and histologically. Light microscopy showed that the affected enamel surfaces were rough and the enamel was hypoplastic and mainly positively birefringent. Scanning electron microscopy revealed a rough and extensively cracked enamel surface covered with oval shaped blister-like protrusions. TEM showed porous enamel consisting of loosely packed and randomly oriented thin ribbon-like crystals with little or no prismatic structure. Observations showed that hypoplasia together with hypocalcification and/or hypomaturation defects were present in the same tooth, indicating the possibility of an abnormality in interstitial matrix, leading to dystrophic calcification in the kidney and abnormal tooth enamel formation, or alternatively an involvement of two separate but closely linked genes.

A rapid microchromatographic method for determination of hemoglobin AI.

Hemoglobin AI (HbAI) is a minor hemoglobin fraction, which is continuously formed by a glycosylation reaction throughout the life-span of the erythrocyte. The concentration of HbAI reflects the average blood glucose level during the preceding 1 - 2 months and HbAI-determinations have been found to be of value in long-term diabetes control. A simple and reliable microchromatographic method for HbAI-determinations, suitable for routine clinical analyses, is described. In a group of diabetic patients (n = 126) the mean (+ S.D.) S.D.) HbAI--value was 9.6 + 1.8%, in the reference group (n = 33) the corresponding value was 6.0 + 0.8%. A commercial kit (Bio-Rad) based on the same principle was compared with our method. It was of equal accuracy and gave comparable results.

Variances of and correlations among progeny tests for reproductive traits of cows sired by AI bulls.

Estimates of daughter fertility were computed using first artificial insemination (AI) breedings reported to the US Dairy Herd Improvement Association (DHIA) from 1995 through 1997. An animal model was used to compute estimated breeding values (EBV) of daughter groups with fixed effects of herd-year-month bred and classes of early lactation energy-corrected milk, days in milk (DIM) when bred, and parity. Standard deviations and ranges of bull EBV for daughter fertility for DIM were 9.1 and -31 to 18; standard deviations and ranges of bull EBV for daughter fertility for nonreturn were 3.8 and -11 to 10. Correlations were computed for EBV for daughter fertility with EBV for mating bull fertility and with predicted transmitting abilities (PTA) for milk, somatic cell score (SCS), and productive life for bulls (213) with minimums of 200 matings and 100 progeny with reproductive traits. None of the correlations among EBV for reproductive traits differed from 0.0. Correlations of EBV for daughter fertility with PTA for productive life were significantly positive. PTA for yield traits were not correlated with EBV for daughter differences in nonreturn or DIM. Very low correlations of EBV for daughter reproductive traits with PTA for yield indicate that, in order to improve daughter fertility, fertility must be incorporated in sire selection decisions.

AI-Driven Multi-Omics Integration of Synthetic Colon Adenocarcinoma for Cluster-Guided PROTAC Candidate Design Targeting KRASG12D.

Colorectal cancer is a leading cause of cancer death, yet its molecular heterogeneity remains poorly translated into individualized treatment. We present a reproducible artificial intelligence (AI) framework that integrates multi-omics benchmarking, sample-level drug prioritization, E3 ubiquitin ligase selection, and shape-anchored Proteolysis Targeting Chimera (PROTAC) design for KRASG12D in colon adenocarcinoma (COAD). A controlled synthetic benchmark comprising 425 tumor and 41 simulated normal profiles, parameterized to match The Cancer Genome Atlas (TCGA) distributions, was used for pipeline verification. Among sixteen methods, the Balanced Latent Integration with Stability Selection (BLISS) model achieved the highest silhouette width (0.86) and competitive agreement (Adjusted Rand Index, ARI, 0.90). The pipeline was validated on real data: a TCGA COAD cohort (186 tumors) with independent Consensus Molecular Subtype (CMS) labels and a CPTAC cohort (104 tumors). Integration modestly recovered CMS (ARI 0.28), and stage, not molecular cluster, drove survival (log-rank p = 0.005 versus 0.81). Sample-level prioritization differed from cluster-level ranking in 82.6% of profiles, below chance (p < 0.0001), without indicating efficacy. Candidate NOVEL00489 showed a good MM-GBSA estimate, matching the reference ASP3082. Compounds are computational candidates requiring experimental validation. This establishes a transparent benchmark for in silico degrader generation in precision oncology.

AI-generated familiarity estimates are a useful new source of information about word knowledge in Simplified Chinese.

This study evaluated the usefulness of AI-generated estimates of word familiarity for predicting word difficulty in Simplified Chinese, building on previous research in alphabetic languages. We found that familiarity estimates produced using large language models (LLMs) showed moderate-to-strong correlations with human familiarity ratings. These LLM estimates were the most effective predictors of both word naming and lexical decision times, surpassing traditional metrics such as word frequency and human familiarity ratings, while the latter still provided modest, non-overlapping variance. GPT-4o with English instructions produced superior results compared to the Chinese-centered models currently available. The results imply that LLM familiarity estimates are a valuable resource for Chinese psycholinguistics, supporting work across experimental design, modeling, and norming. We release familiarity estimates for 27,624 words for unrestricted research and educational use.

AI-guided analysis of human pancreatic islet sociology reveals distinct cell compositional changes in type 1 diabetes.

Human pancreatic islets exhibit greater anatomic and cellular heterogeneity than previously appreciated, raising fundamental questions about how their composition varies with age, sex, region, and islet size and how type 1 diabetes (T1D) alters these relationships. Yet these questions remained largely unresolved due to the bottleneck of manual tissue inspection. Here, we developed an integrated artificial intelligence (AI)-guided imaging, processing, and statistical pipeline enabling unbiased, high-throughput analysis of more than 2 million candidate islets from 106 non-diabetic (ND) and T1D donors. We identified age-, region-, sex-, and islet size-dependent differences in islet distribution and composition between ND and T1D donors. Profound β-cell loss in T1D was accompanied by reciprocal α-cell expansion, whereas δ-cells and pancreatic polypeptide cells were largely resilient. Cell area and pseudotime analyses uncovered regional and age-dependent trajectories of islet remodeling across T1D progression, along with distinct patterns of cytoarchitectural reorganization of the endocrine pancreas.

[Parameters of lipid metabolism and polymorphism of apolipoprotein aI and angiotensin-converting enzyme genes in patients with endometrial carcinoma].

Data on lipid metabolism in 109 cases of endometrial carcinoma and 33 patients in control are presented. Relevant disorders were detected in 72.5% of the study group [stage I obesity--23 (21.1%); II--29 (26.6%); III--25 (22.9%); IV--2 (1.9%)] which was 1.5 times the mean level in controls (51.5%). The abdominal pattern of obesity was predominant (77.7%). Symptoms of cardiosclerosis were identified in 93 patients with endometrial tumors (85.3%), 93.5% of the latter group presenting with concomitant hyperglyceridemia. In 68.8% of endometrial carcinoma patients, incidence of arterial hypertension was higher than in controls (54.5%). Lipoproteins played a major role in dyslipoproteid pathogenesis involved in obesity and high blood pressure. A study of the insertion-deletion (I/D) polymorphism of apolipoprotein AI genes showed two deletion alleles (DD--6%) and one heterozygote (ID--3%) in control group; no deletion alleles were identified in endometrial tumor patients (0.1 (p(0.05). An investigation of the I/D polymorphism of angiotensin-converting enzyme genes identified deletion homozygotes in 30, insertion homozygotes (II)--27, and heterozygotes--41% (control). In endometrial cancer group, the deletion allele distribution was: DD--29; II--28 and ID--45%. Deletion allele frequency in control was 0.485 while in endometrial carcinoma--0.484 (p(0.05), i.e. with out significant difference.

[Introduction of wide spectrum rice bacterial blight resistance gene Xa21 into two-line genic male sterile rice variety Pei'ai 64S].

Agrobacterium-mediated transformation of two-line genic male sterile Indica rice variety Pei'ai 64S was conducted using a cloned gene, Xa21, as the foreign gene and mature embryo calli as the recipients. A total of 46 transgenic plants had been obtained. The PCR analysis and Southern blotting showed the integration of Xa21 gene into the genome the transgenic plants. Results of inoculation with philippine race 6 of Xanthomonas oryzae pv. oryzae indicated that most of transgenic plants obtained high resistance to rice bacterial blight disease (Xoo). Analyses of T1 plants of the tested transgenic lines showed that integrated Xa21 gene could be steadily inherited and segregated in a 3:1 ratio.

[Clone and tissue expression of tree shrew apolipoprotein AI cDNA].

OBJECTIVES: To clone apolipoprotein AI (apoAI) gene from tree shrew (TS) which is an animal insusceptible to atherosclerosis and to analyze the characteristics of the gene. METHODS: A cDNA library of the liver tissue was constructed by using the isolated mRNA as the template during reverse transcription. The positive clones of apoAI cDNA were identified in the library with an anti-serum to TS apoAI. Sequencing, analyzing and expressing of the clones were finished. RESULTS: A complete cDNA sequence of TS apoAI was determined as 900 nucleotides. It contained 28 bp in 5' untranslated region, and 795 bp in an open reading frame encoding a 265-amino acid apoAI precursor (an 18-amino acid signal peptide, a 6-amino acid propeptide and a 241-amino acid mature protein), two stop codens (TGA, TAA) and following 72 bp in 3' untranslated region. The novel gene was submitted to GenBank. TS apoAI deduced from the cDNA sequence was more hydrophobic than that of human. The results of Northern blot indicated the plentiful presence of apoAI mRNA in TS liver and intestine. CONCLUSIONS: A new gene was cloned from TS, one animal insusceptible to atherosclerosis. TS apoAI may have a stronger activity of binding cholesterol and cholesterol ester than that of human apoAI in reverse cholesterol transport. A good foundation was laid for studying the molecular structure and function of TS apoA I.

[Effect of chang'ai kangfu decoction on immunity in postoperational patients with large intestine cancer].

OBJECTIVE: To explore the effect of Chinese drug Chang'ai Kangfu decoction (CAKF) on immunity in post-operational patients with large intestine cancer (LIC). METHODS: Forty-eight patients with LIC in Dukes' B, C stage after operation were randomly assigned to 3 groups, the CAKF group (16 cases), chemotherapy group (17 cases) and combination therapy (CAKF plus chemotherapy) group (15 cases). 5-FU and mitomycin C were given to the chemotherapy group. The dynamic changes of T-lymphocyte subsets, NK cells and immunoglobulins were investigated. RESULTS: Before operation, the CD3+, CD4+, CD4+/CD8+ and the activity of NK cells in LIC patients were lower, but CD8+ was higher than that of normal level (P < 0.01), which indicated that cellular immunity in LIC was in immunosuppressive state, they all further reduced 1 week after operation, particularly CD3+ cell counts, but CD3+, CD4+ and the activity of NK cells normalized 1 month after operation in CAKF group, and 2 months were needed to normalize in combination therapy group. Both groups recovered to a certain extent in comparing with before treatment, but the chemotherapy group recovered slower. The similar results appeared in humoral immunity. CONCLUSION: CAKF could obviously increase the immunity in LIC patients after operation.

[A case control study on the distribution of apolipoprotein AI gene polymorphisms in the survivors of atherosclerosis cerebral infarction].

OBJECTIVE: To investigate the frequency of the restrict fragment length polimorphisms (RFLP) at 2 MspI sites in the 5' - end of apolipoprotein AI (ApoAI) gene and its potential interaction to atherosclerotic cerebral infarction (ACI) in Chinese Han population. METHOD: Polymerase chain reaction (PCR) technique was used in a sample of 199 cases with documented ACI and 204 healthy matched by age and sex individuals selected from Chinese Han nationality in Beijing. The studied loci include promoter region (-75 bp) and the intron - 1 (+83 bp). RESULTS: (1) Both ACI and control groups M1 + + and M1 + - were major frequent genotypes in Beijng area, M2 + + were major frequent genotypes in intron - 1; (2) M1 + - genotype appeared more common among patients than in controls (0.487 vs 0.368, P < 0.05, OR = 1.64); (3) Individuals with M1 + -/M2 + + haplotype were significantly increased in patients compared with controls (0.437 vs 0.314, P < 0.05, OR = 1.70). CONCLUSION: It is suggested that both M1 + - genotype and M1 + -/M2 + + haplotype of ApoAI gene might associate with ACI in our study.

[Clinical analysis of urolithiasis in Poh Ai Hospital of I-Lan, Taiwan, R.O.C.--a comparative study with urolithiasis in Japan].

Between August 1987 and December 1990, 546 patients were admitted to the department of Urology at the Poh Ai Hospital of I-Lan, Taiwan, R.O.C. for the treatment of urinary stones. These urinary stone cases accounted for 50 to 60% of all urology patients admitted. The incidence of urolithiasis in I-Lan was estimated at 147/100,000 population in 1990. There were 402 male patients and 144 female patients, The male to female ratio was 2.8: 1. There were 450 upper urinary tract stones (kidney, ureter) in 314 males and 136 females, and 79 lower urinary tract stones (bladder, urethra) in 72 males and 7 females. The ratio of upper to lower urinary tract stones was 6:1. Endourological treatments such as percutaneous nephrolithotripsy and transurethral ureterolithotripsy have increased rapidly in recent years. A summary of the present analysis for composition of 365 stones follows. The most frequent type was calcium-containing stone (92.3%), followed by infection stone (4.7%), then uric acid (UA) stone (3.0%). There were no UA stones found in the female patients. According to urinalysis criteria of more than 10 WBC/HPF (x 400), pyuria was found in 67 cases of 334 metabolic stones (20.1%), and 11 cases of 17 infection stones (67.7%). There were neither pediatric case of stone formation nor cystine stones.

Patterns of association between genetic variability in apolipoprotein (apo) B, apo AI-CIII-AIV, and cholesterol ester transfer protein gene regions and quantitative variation in lipid and lipoprotein traits: influence of gender and exogenous hormones.

Patterns of RFLP association were studied, to identify gene regions influencing quantitative variation in lipid and lipoprotein traits (coronary artery disease [CAD] risk factors or metabolically related traits). Subjects (118 female and 229 male; age 20-59 years) were selected for health. Multiple RFLPs were used to sample variability in regions around genes for apolipoprotein (apo) B (restriction enzymes HincII, PvuII, EcoRI, and XbaI), apo AI-CIII-AIV (BamHI, XmnI, TaqI, PstI, SstI, and PvuII) and cholesterol ester transfer protein (TaqI). Separate analyses were done by gender. The sample was truncated at mean +/- 4 SD, to remove extreme outliers. There was no significant gender difference in RFLP genotype frequency distribution. After trait-level adjustment to maximize removal of concomitant variability, analysis of variance was used to estimate the percentage trait phenotypic variance explained by measured variability in the gene regions studied. Fewer gene regions were involved in men, with less influence on quantitative trait variation than in women, in whom hormone use affected association patterns. Gender differences imply that pooling genders or adjusting data for gender effects removes genetic information and should be avoided. The association patterns show that variability around the candidate genes modulates trait levels: the genes are contributors to the genetics of CAD risk variables in a healthy sample.

Expression of low density lipoprotein receptor, apolipoprotein AI, AII and AIV in various rat organs utilizing an efficient and rapid method for RNA isolation.

Intact RNA from various rat organs was isolated by an efficient and rapid method. This method of RNA isolation is a modification of an earlier method that uses guanidinium isothiocynate followed by extraction in the presence of sarcosyl, acetate and phenol. The RNA obtained by the method reported here was comparable with the RNA prepared by the CsCl2 ultracentrifugation method and the commercially available kit based on published methods. The quality of RNA was found suitable for Northern blotting analysis, RNase protection assays and reverse transcriptase-polymerase chain reaction (RT-PCR). Since reverse transcriptase is active in the buffer used for Taq DNA polymerase, only one reaction needs to be set up. We also found that the use of aurintricarboxylic acid in the RNA preparation prevents the degradation of RNA during storage. Expression of low density lipoprotein (LDL) receptor, apolipoprotein (apo) AI, AII and AIV mRNAs were quantified in various rat organs. Our results indicated that rat LDL receptor mRNA is expressed in several organs whereas apoAI and AIV mRNAs were expressed mainly in the liver and intestine. However, apo AII mRNA is expressed mainly in the liver. Unlike mice and some species of monkeys, in the rat apoAI mRNA is expressed at 5-6 times higher levels in the intestine compared to liver. Apo AIV mRNA abundance was also found to be several fold higher in intestine compared to hepatic tissues. We present here, for the first time, data on the absolute amounts of LDL receptor, apoAI, AII and AIV mRNA in various rat organs which were quantified by a novel RNase protection/solution hybridization assay.

Apolipoprotein AI expression and high density lipoprotein distribution in transgenic mice during development.

The developmental changes in apoAI expression in human apolipoprotein AI (apoAI) transgenic and nontransgenic control mice and the effect of these changes on high density lipoprotein (HDL) sizes were investigated. Results demonstrated that both human and mouse apoAI mRNA levels sharply increased following delivery and then decreased prior to weaning. Unlike the changes in apoAI mRNA, plasma apoAI concentration and HDL mass increased following delivery and remained elevated as the animals matured. In these animals, parallel increases in plasma apoAI and HDL mass with development were accompanied by increases in HDL particle sizes. Control mice demonstrated a monodisperse population of particles that increased in particle size from 7.5 nm (at 2 days prior to birth) to 9.8 nm (at 13 days after birth). The latter is similar to the adult HDL size and distribution. Transgenic mice, unlike controls, exhibited several distinct HDL populations, which also increased in size as animals matured. These results suggest that post-transcriptional factors are important in regulating apoAI plasma levels and that developmental changes in plasma apoAI concentrations are associated with changes in HDL size distribution.
