[Study on reversed-phase ion-pair chromatographic determination of thiosulfate and iodide anions].

This paper describes an indirect reversed-phase ion-pair chromatographic separation and UV indirect photometric detection of and S2O3(2-) and I- by pre-column derivatization with Hg-EDTA to form their corresponding ternary complexes EDTA-Hg-S2O3 and EDTA-Hg-I for the first time. On a Zorbax ODS column with a mobile phase of methanol-water we have studied the influences of the ion-pair TBA+ concentration, pH value, the addition of EDTA, the concentration of organic modifier and ionic strength. In our experiments, the addition of EDTA had a notable effect on retention and peak shape. So the best chromatographic conditions are: methanol:water = 23:77 (V/V), the ion-pair reagent TBA+ 3.0 mmol/L, EDTA 1.0 mmol/L, NaNO3 2.0 mmol/L and buffer solution KH2PO4-Na2HPO4 (pH 7.0). The eluate is monitored by a UV detector operated at 254 nm and the detection limits of S2O3(2-) and I- are 61.1 ng and 37.6 ng respectively. Good linear relationships between the peak area (Y) and the amount of anion ions (X) with r = 0.9851 and 0.9892, respectively, are obtained. Interference of some ions is also studied. The method is both convenient and selective. It has already been used for the determination of I- in kelp and the results are satisfactory. The amount of I- in kelp is 0.0183%.

[Pharmacokinetics following intraperitoneal infusion of cisplatinum with and without sodium thiosulfate rescue in carcinomatous peritonitis in advanced gastric cancers].

The pharmacokinetics of ip cisplatinum (100 mg/body) was studied in 10 patients with advanced gastric cancer. Five patients were administered ip-cisplatinum with STS and another 5 patients were given the same without STS. As a result, 1) the level of free-CDDP in ascites fluid was significantly elevated after administration, showing that free-CDDP possessing anti-tumor activity persisted in the ascites fluid for a long time and reached the peak concentration 30 minutes after administration. 2) The free-CDDP transferred into the blood after administration also persisted for a long time in parallel with changes in the free-CDDP level in the ascites fluid. 3) The AUC, MRT and VRT of the free-CDDP concentration in the peritoneal cavity after ip administration of CDDP were high, clearly indicating the direct effect on peritoneal concentration. 4) The AUC, MRT and VRT of free-CDDP in the venous phase after ip administration of CDDP were higher than after its iv administration, suggesting that effects equivalent to or greater than those of the iv route drug can be expected in the venous phase. 5) The frequency of side effects induced by the combination of STS was lower than that of side effects induced by the administration without combined STS. Much larger doses of CDDP may be given ip by combining with STS.

Acute renal failure associated with intraperitoneal cisplatin chemotherapy with systemic thiosulfate protection in ovarian cancer patients.

Acute renal failure associated with intraperitoneal cisplatin chemotherapy occurred in 4 courses of treatment each given to one patient, and comprised 3.4% of the total number of intraperitoneal treatment courses. The 4 patients recovered completely, 2 of them after dialysis. Several possible reasons for this complication, which occurred in spite of thiosulphate systemic protection, are considered. Strict adherence to the treatment protocol is mandatory in order to avoid severe nephrotoxicity.

[The diminishing oocyte toxicity following intraperitoneal infusion of cisplatin (CDDP) combined with subcutaneous injection of sodium thiosulfate (STS) in syngeneic young mice].

Small oocytes in 6-week-old C57BL/6 mice were destroyed in a dose-dependent fashion following intraperitoneal infusion of CDDP. ED65, the effective dose of which theoretically produced 65% destruction of small oocytes according to Probit analysis, was determined as 0.2mg/mouse. In this study, the oocyte toxicity induced with intraperitoneal treatment with CDDP (0.2mg) was offset by subcutaneous treatment with STS (40mg, 400 x molar ratio compared to that of CDDP). STS was subcutaneously injected into the back skin of mice at 60, 30, 15 minutes before, at the same time or at 15, 30, 60 minutes after the CDDP injection. STS markedly diminished the oocyte toxicity and reduced oocyte destruction about 45% when it was injected at 30, 15 minutes before or at the same time as CDDP infusion. On the other hand STS failed to reduce oocyte destruction when it was injected 15, 30 or 60 minutes later. STS might neutralize CDDP in the circulating blood to decrease the amount of active CDDP, but STS might not have any effect on CDDP once it is incorporated and conjugated with the DNA in the oocytes.
