Pulmonary vascular amyloidosis in aged dogs. A new form of spontaneously occurring amyloidosis derived from apolipoprotein AI.

The N-terminus of a mutant form of apolipoprotein AI [apoAI] has previously been shown to be the subunit protein of amyloid fibrils in a human kindred with a form of familial amyloid polyneuropathy (FAP, type III) and in a recently reported kindred with a form of non-neuropathic hereditary amyloidosis. In this study, we demonstrate by amino-acid sequence analysis, that a form of vascular amyloidosis occurring in the lungs of aged dogs is derived from a N-terminal fragment of apoAI and that no amino acid substitution is present in this confirmed sequence. This represents the first documentation of apoAI as a precursor for a form of amyloidosis in animals, and provides the first documentation of apoAI as a precursor for amyloid fibrils in a form of age-associated ("senile") amyloidosis. Secondary structure prediction analysis of the N-terminal regions of normal human and dog apoAI indicated a propensity for beta-pleated sheet conformation, and thus amyloidogenesis, in 40 and 45% of the respective sequences. These results suggest that apoAI (like transthyretin) may serve as an amyloid precursor protein for both familial and senile forms of amyloidosis. ApoAI should, therefore, be considered as a potential amyloid precursor when forms of human senile amyloidosis of unknown origin are evaluated.

Segmental evaluation of the surface and radiological deformity after Cotrel-Dubousset (CD) instrumentation for King type II and III. adolescent idiopathic scoliosis (AIS): surgical and etiological implications.

In this paper we report the segmental effects of CD in each of the three planes of deformity (frontal, transverse and sagittal) in 35 children with AIS (King types II and III) treated by Cotrel-Dubousset instrumentation. Patients were examined preoperatively and in follow-up at 8 weeks and one year, and 15 patients were also examined at two years. Back shape was appraised segmentally as angle of trunk inclination (ATI) at each of ten levels by ISIS and the Scoliometer. The radiographs were measured nonsegmentally for Cobb angle, apical vertebral rotation and spinal balance, and segmentally for both vertebral rotation and tilt (frontal and sagittal plane), and in the rib cage for rib-vertebra angles (RVAs).

[Transferring of multiple insect-resisting genes into two-line genetic male sterile rice Pei'ai 64S by particle bombardment].

Transformation of two-line genetic male sterile indica rice variety Pei'ai 64S was conducted by particle bombardment. Three insect-resisting genes ligased into plasmid vector pKC-3 was introduced into calli derived from mature embryo and a total of 33 transgenic plants had been obtained. The PCR and Southern blot analysis of each different gene in R0 plants and also PCR analysis of each different gene in R1 plants showed the integration of three insect-resisting genes into the genome of transgenic plants which could stably pass down.

Estimation of Hardy-Weinberg and pairwise disequilibrium in the apolipoprotein AI-CIII-AIV gene cluster.

Departures from Hardy-Weinberg (HW) equilibria and pairwise disequilibria were estimated in a sample of unrelated healthy individuals typed for six RFLPs in the apo AI-CIII-AIV gene region. The sample was composed of males and females, selected for health, from two populations, those of exclusively French-Canadian (FC) and those of some non-French-Canadian (NFC) ancestry. An approach suggested by Weir and Cockerham, which includes estimates of nonrandom association (disequilibria) between three and four alleles at two loci as well as the traditional associations between two alleles, at two loci was used. The pattern of departures from HW equilibria suggested that the genetic structures of the FC and NFC are different. Departure from HW equilibrium at an RFLP locus could not be predicted from information about other loci in the same gene region. Nonrandom associations were also evident from the pairwise analyses. Two pairs of loci had significant diallelic disequilibria, while two other pairs had significant triallelic disequilibria. All of the RFLP pairs had at least one measure of disequilibrium at its maximum value determined by allele frequencies. Inferences about pairwise disequilibria depended on the statistical approach used. Sizes of the pairwise disequilibria were not correlated with the physical distance between loci. The impact of these disequilibria on RFLP-phenotype association studies is discussed.

Associations between quality-of-life and internal or external spinal deformity measurements in adolescent with idiopathic scoliosis (AIS).

In theory, quality-of-life measures should correlate with impairments such as spinal deformity. The goal was to determine the ability of the SRS-22 and surface topography measures to predict internal deformity. Data from 227 females with AIS were collected. Correlations and regression were used to predict Cobb angle or category. In subjects treated conservatively, the Cobb angle correlated with pain, self-image, satisfaction and total score. Only self-image correlated with surface topography. In subjects having had surgery, Cobb angle correlated with self-image, mental health, satisfaction and total scores. The cosmetic score was the only external deformity measure to correlate with the SRS-22. Function, self-image and trunk-twist predicted subjects within 3 categories (Cobb <30 degrees , 30-50 degrees , >50 degrees) with 57% accuracy compared to 53% when using self-image only. Accuracy in predicting subjects with curves smaller or larger than 50 degrees was 79% (using self-image, trunk-twist) or 72% (self-image only). Correlations between quality-of-life and deformity measures were low. However, it was possible to predict subjects within clinically meaningful categories of internal deformity using SRS-22 scores.

[The distribution of apolipoproteins AI, CII and CIII mRNA in rat tissues].

It was demonstrated that apo AI, CII and CIII mRNA were distributed in rat (Sprague Dawley) liver, intestine, brain and kidney by RNA-dot blot analysis with apoAI, CII and CIII cDNA probes. The relative abundances of apoAI in liver, intestine, brain and kidney tissues are 100%, 212%, 170% and 224%, respectively, those of apoCII are 100%, 61%, 163% and 214%; and those of apoCIII 100%, 65%, 70% and 73%, respectively. The experiment was repeated for three times. The results were discussed in this paper.

[An electron microscopic study of the origins of the projections of the first auditory area of the cortex (AI) to the medial geniculate body in cats].

An electron microscope study of retrogradely labelled neurons in layer VI of the primary auditory cortex (AI) after injection of the horseradish peroxidase to the medial geniculate body was carried out in cats. Three-eight synapses (4.6 +/- 0.6 at an average) were revealed on the somata profiles of these retrogradely labelled cortico-geniculate neurons. Synapses occupied 10.8 +/- 1.0% of the somatic profile of cortico-geniculate neurons. Almost all (98.7%) of these axosomatic synapses had symmetrical contacts and were formed by axonal terminals with small elongated synaptic vesicles. HRP retrogradely labelled axonal terminals of geniculo-cortical fibres were revealed in neuropil of layer VI. They contained large round synaptic vesicles and formed asymmetrical synapses, mainly on spines. The role of axo-somatic synapses in regulation of the activity of cortico-geniculate neurons was discussed.

Rate immunonephelometry and radial immunodiffusion compared for apolipoproteins AI and B assay.

We measured apolipoproteins (apo) AI and B in fresh plasma samples and serum control pools by using two rate immunonephelometric assay (INA) methods (Beckman "Array" and Behring) and radial immunodiffusion (RID). Both INA methods on average gave apoAI values similar to those by RID in fresh plasma samples. The coefficients of correlation for the two INA-RID method pairs were as follows: Beckman INA vs RID, n = 94, r = 0.73; and Behring INA vs RID, n = 112, r = 0.66. The bias between the INA and RID methods was reflected reasonably well by either lyophilized or frozen control pools. For apoB, the Beckman INA measurements in fresh plasma averaged 40% lower than RID values, due almost entirely to the values assigned to calibration sera, but results by the two methods were highly correlated (n = 94, r = 0.93). The Behring INA values for fresh plasma averaged only 13% lower than RID values, but the two methods were less well correlated (n = 112, r = 0.74). Frozen control pools were most suitable for apoB analysis.

Cloning of apoprotein AI gene and observation on DNA polymorphism of apoprotein AI/CIII gene-loci in genomic library from a liver of Chinese.

For the research on the structure and function of the HDL apoproteins, we have successfully constructed a complete genomic library, in self-prepared EMBL3 lambda vector and the packaging extracts of lysogenic strains BHB2688, BHB2690, from a Chinese fetal liver, which will be widely used in the research on the structure and function of apoproteins. In this study, a DNA sequence polymorphism, revealed by digestion of human DNA with the restriction endonuclease Sst1 and hybridization with an apoprotein AI cDNA probe, has been shown to be located at apoAI/CIII gene-loci. Three gene related fragments (5.7 Kb, 4.0 Kb and 3.0 Kb) were detected. The 5.7 Kb fragment is common and the polymorphism is demonstrated by the presence of either a 4.0 Kb fragment (S1 allele) or the 3.0 Kb fragment (S2 allele). Individuals were genotyped S1S1 or S1S2. Our result showed that the frequency of genotype S1S2 was higher in hypertriglyceridemic subjects than that in normolipidemic subjects.

The detection of linkage disequilibrium between closely linked markers: RFLPs at the AI-CIII apolipoprotein genes.

Study of very closely linked DNA variants at various loci has frequently shown linkage disequilibrium. We studied three closely linked RFLPs at the apolipoprotein AI-CIII locus. Two variants detected by MspI and SstI were in strong linkage disequilibrium; but when conventional statistical tests were used, a third variant (PstI), located between the MspI and SstI markers, appeared to be in linkage equilibrium with these two "outside" markers. Similar discrepancies from the expected monotone relationship between physical distance and linkage disequilibrium have been reported by others. To investigate these discrepancies, the power to detect linkage disequilibrium was calculated. It could be shown that, for the gene frequencies encountered, very large sample sizes would be required to demonstrate negative (i.e., repulsion-phase) linkage disequilibrium. Such numbers are usually very difficult to attain in human studies. Failure to demonstrate linkage disequilibrium by conventional methods therefore does not imply its absence. Appropriate nomograms and tables are provided.

Apolipoprotein B and apolipoprotein AI as predictors of coronary artery disease.

The evidence that plasma levels of apolipoprotein B and apolipoprotein AI are important determinants of the risk of premature coronary artery disease (CAD) are reviewed and a metabolic framework for these relations presented. ApoB and apoAI are compared to total cholesterol and to lipoprotein lipids as predictors of CAD. The emphasis is on apoB as this is the particular interest of this laboratory and because a structural basis has been elucidated that may relate the epidemiologic observations about apoB to the metabolic characteristics of low density lipoprotein. The measurement of apoproteins has revealed much that was previously obscure about normal and abnormal lipoprotein metabolism. However, one of the major challenges is the development of an accurate, widely available commercial assay system for apoproteins.

Aminodiphosphonate or AI preincubation inhibits calcification of aortic homografts in the rat subdermal model.

Calcification is frequently noted in valved and non-valved aortic homografts, predominantly in the aortic wall portion. This study was designed to assess the individual anticalcification effects of the trivalent cation, aluminum (AI ), and the compound aminopropanehydroxydiphosphonate (APDP) when used to pretreat aortic wall homograft tissue before subdermal implantation in the rat model. Thoracic aortas of male rats (CD, Sprague-Dawley, 350 to 400 g) were harvested distal to the aortic valve and incubated in the following solutions: 1) APDP (0.004 M), 2) APDP (0.004 M) + glutaraldehyde (GLUT) (0.02 M), 3) GLUT (0.02 M), 4) ALCL3 (0.01 M), 5) HCL (0.001 M), and 6) HEPES (0.05 M). The homograft tissue was then implanted subcutaneously for 21 days in weanling male rats (50 to 60 gm) of the same strain. Explant results showed that calcification (CALC) was markedly inhibited in the APDP and 0.01 M ALCL3 groups, compared to the control and GLUT pretreated groups. No diphosphonate or aluminum related side effects were seen.

[Modification of serum lipids, lipoproteins and apoproteins AI and B in patients with hyperlipidemia Type IIa and IIb using polyenylphosphatidylcholine].

In a double blind study 27 patients with type II hyperlipidemia (8 IIa and 19 IIb) were treated as follows: 13 received placebo and 14 sn-polyenylphosphatidylcholin (PPC) (P 0206/1/01, Nattermann GmbH, Cologne) in a dose of three times 450 mg b.i.d. In all patients, and also in the two subclasses of patients with type IIa and type IIb hyperlipidemia, total cholesterol and LDL cholesterol were lowered significantly by PPC. The other parameters showed only minor variation. There was a downward trend in apoprotein B, triglycerides and VLDL cholesterol, and an upward trend in apoprotein AI, with virtually unchanged HDL cholesterol. None of these variations was significant compared with placebo. The fall in LDL cholesterol with unchanged HDL cholesterol caused a statistically significant decrease in the LDL cholesterol/HDL cholesterol ratio, thus supporting the hypothesis of an antiatherogenic property of PPC, as demonstrated experimentally in various animals.

[Cholinergic structures in the auditory area of the cerebral cortex in the cat (zone AI)].

Histochemical reaction to AChE was used to reveal the network of cholinergic varicose fibres in the neuropile of different cortical layers. Its density was maximal in layer I and deeper, and minimal in layers II and III. Among neurons of zone AI few stellate neurons located in layers II-VI are cholinergic. Axons of some stellate neurons terminate on adjacent neurons, axons of a part of layer VI neurons travel to the subcortical layer of the associative arcuate fibres. Cholinergic terminals are located on the soma and proximal parts of dendrites of neurons most of which contain no acetylcholinesterase. Such cholinoceptive neurons of various size and shape are found in all cortical layers.

Immunonephelometric assay of human apolipoprotein AI.

We report an immunonephelometric assay for apolipoprotein AI, which involves use of a laser nephelometer and an antiserum prepared in our laboratory. Coefficients of variation were 2.5% within-run, 6.3% between-run. Several types of manipulation of the samples (heating, delipidation, dissociation with urea), reportedly necessary for accurate determinations by radioimmunoassay or gel diffusion immunoassay, did not substantially affect results obtained with our technique. The assay is sensitive, reproducible, and simple, and appears suitable for routine use.

Relation between hemoglobin AI and determinations of glucose in diabetics treated with and without insulin.

The concentration in blood of hemoglobin AI is known to correlate well with glucose regulation if blood and urinary glucose values are carefully documented over several weeks. A slight increase of HbAI with age was found in 94 healthy subjects. In 41 adult diabetics treated with sulfonylurea drugs or diet alone, good correlations were obtained between HbAI and single random measurements of morning blood glucose (r = 0.69) and log urinary glucose/24h (r = 0.79) whereas in 47 adult patients with insulin-treated diabetes the corresponding correlation coefficients were only 0.42 and 0.49, respectively. In 29 insulin-treated children the HbAI-values were not correlated with single morning blood glucose determinations (r = 0.16 n.s.) but a correlation coefficient of 0.83 was found between HbAI and an index for regular testing of urinary glucose (Clinitest) at home. For the long-term monitoring of insulin-treated diabetics, single random determinations of blood and urinary glucose may often provide misleading information and particularly in these patients, routine determinations of HbAI should be of value.
