The use of progestins in regimens for fixed-time artificial insemination in beef cattle.

Four experiments were conducted to investigate modifications to gonadotropin releasing hormone (GnRH)-based fixed-time Al protocols in beef cattle. In Experiment 1, the effect of reducing the interval from GnRH treatment to prostaglandin (PGF) was examined. Lactating beef cows (n = 111) were given 100 mg gonadorelin (GnRH) on Day 0 (start of treatment) and either 500 microg cloprostenol (PGF) on Day 6 with Al and 100 microg GnRH 60 h later, or PGF on Day 7 with Al and GnRH 48 h later (6- or 7-day Co-Synch regimens). Pregnancy rates were 32/61 (53.3%) versus 26/50 (52.0%), respectively (P = 0.96). In Experiment 2. cattle (n = 196) were synchronized with a 7-day Co-Synch regimen and received either no further treatment or a CIDR-B device (Days 0-7). Pregnancy rates were 32/71 (45.1%) versus 33/77 (42.9%) in cows (P < 0.8), and 9/23 (39.1 %) versus 17/25 (68.0%) in heifers (P < 0.05). In Experiment 3, 49 beef heifers were randomly assigned to receive 12.5 mg pLH on Day 0, PGF on Day 7 and 12.5 mg of pLH on Day 9 with Al 12 h later (pLH Ovsynch), or similar treatment plus a CIDR-B device from Days 0 to 7 (pLH Ovsynch + CIDR-B), or 1 mg estradiol benzoate (EB) and 100 mg progesterone on Day 0, a CIDR-B device from Days 0 to 7 (EB/ P4 + CIDR-B), PGF on Day 7 (at the time of CIDR-B removal) and 1 mg i.m. EB on Day 8 with AI on Day 9 (52 h after PGF). Pregnancy rate in the EB/P4 + CIDR-B group (75.0%) was higher (P < 0.04) than in the pLH Ovsynch group (37.5%): the pLH Ovsynch + CIDR-B group was intermediate (64.7%). In Experiment 4, 266 non-lactating cows were allocated to a 7-day Co-Synch protocol (Co-Synch), a 7-day Co-Synch plus 0.6 mg per head per day melengestrol acetate (MGA) from Days 0 to 6 inclusive (Co-Synch + MGA) or MGA (Days 0-6) plus 2 mg EB and 50 mg progesterone on Day 0. 500 microg PGF on Day 7, 1 mg EB on Day 8 and fixed-time Al 28 h later (EB/ P4 + MGA). Pregnancy rates (P < 0.25) were 44.8% (39/87: Co-Synch), 47.8% (43/90; Co-Synch + MGA), and 60.7% (54/89: EB/P4 + MGA). In conclusion, a 6- or 7-day interval from GnRH to PGF in a Co-Synch regimen resulted in similar pregnancy rates in cows. The addition of a progestin to a Co-Synch or Ovsynch regimen significantly improved pregnancy rates in heifers but not in cows. Progestin-based regimens that included EB consistently resulted in high pregnancy rates to fixed-time Al.

Repeat breeding in dairy heifers: follicular dynamics and estrous cycle characteristics in relation to sexual hormone patterns.

Repeat breeding occurs at an incidence of 10% in the Swedish dairy cow population. Evidence is available for a hormonal asynchrony around estrus in repeat-breeder heifers (RBH). This asynchrony seems to be the underlying cause for a series of dysfunctions such as prolonged standing estrus and delayed ovulation, leading to fertilization failure. For further determinations of repeat-breeder estrous cycle characteristics, seven strictly selected RBH and six virgin heifers (VH) were studied during 3-7 consecutive cycles, with particular attention paid to the estrous period. Follicular dynamics were studied by ultrasonography and related to estrous behavior and pattern of sexual hormones (progesterone, estradiol-17beta, and LH) in peripheral circulation. Mean group data were compared and a classification model was designed. The most prominent findings for RBH were prolonged duration of estrus, delayed LH peak, prolonged lifespan of the preovulatory follicle, and a late postovulatory rise in plasma progesterone. There was also a strong tendency for peri-ovulatory suprabasal progesterone levels in RBH. It is suspected that these deviations cause changes in the microenvironment of the preovulatory follicle, negatively affecting the final maturation of the oocyte. The heterogeneity of the RBH group underlines the multifactorial cause of the repeat-breeder syndrome. The VH formed a homogenous group with data varying within physiological limits. A classification model based on three characteristic variables managed to identify 81% of the VH and 79% of the RBH correctly. Results from this study propose that some heifers have general, consistent problems in synchronizing estrous events, displayed as varying symptoms in the course of consecutive estrous cycles. These subfertile animals could be classified as repeat-breeders.

The use of plasma progesterone profiles to predict the reproductive status of anestrous gilts and sows.

A plasma progesterone profile obtained from three consecutive blood samples with an interval of 7 days was evaluated for usefulness as the basis for the diagnosis and treatment of anestrous gilts and sows. Four reproductive statuses were categorized based on the plasma progesterone levels and pathological examination of the reproductive organs from 25 gilts and 12 sows with anestrus. Category 1: fluctuating (at least one sample <2.5 ng/ml and one >10 ng/ml) with normal ovary; Category 2: sustained low (<2.5 ng/ml) with inactive ovary; Category 3: persistent high (>5 ng/ml) with normal sized or cystic corpora lutea; and Category 4: animals not included in the categories mentioned, such as pigs with luteinized cysts and follicular cysts. Using the plasma progesterone profiles and this categorization, the reproductive status of 54 gilts and 38 sows with anestrus was predicted. Hormonal treatments were performed with moderate to high success. Results from this study indicate that plasma progesterone profiles can be useful for the determination of estrus status, for the diagnosis of the causes of anestrus, and for the prediction of the next estrus for an appropriate hormonal treatment in anestrous gilts and sows.

Effect of probe location on changes in vaginal electrical impedance during the porcine estrous cycle.

The impedance technique is one of many methods that can be used for noninvasive monitoring of reproductive events occurring in cyclic animals. The influence of the depth of probe insertion on changes in vaginal impedance in sows during the estrous cycle was examined. Sows were checked twice a day for estrus via exposure to a sexually mature boar. The criterion for confirmation of ovulation was an increase in plasma progesterone levels above 4.0 ng/ml 8 and 12 days after the beginning of estrus. The impedance measurements were carried out using a four-terminal method at a distance of 8, 10, 12, 14, 16 and 18 cm from the vulva. In all six locations of the vagina the mean impedance values decreased gradually after weaning (P<0.01), achieved a nadir 1-2 days before estrus and increased during estrus (P<0.01). It was found that the probe location within the vagina and the parity of sows significantly affected the impedance measured by means of a four-terminal method. The study suggests that the causes of impedance fluctuation are not only technical but also include a number of poorly understood biological causes.

Effect of resynchronization with GnRH on day 21 after artificial insemination on pregnancy rate and pregnancy loss in lactating dairy cows.

The objectives of the present study were to determine the effects of resynchronization with GnRH on Day 21 after artificial insemination (AI) on pregnancy rate and losses of pregnancy in lactating dairy cows. Holstein cows (n=585) on two dairy farms were assigned to one of two treatments in a randomized complete block design. On Day 21 after a pre-enrollment AI, animals assigned to the resynchronization (RES) group received 100 microg of GnRH i.m., whereas animals in the control (CON) group received no treatment. All animals were examined ultrasonographically on Days 21 and 28 after AI, and blood samples were taken for progesterone measurement on Day 21. Pregnancy was diagnosed on Day 28 and reconfirmed 14 days later. Nonpregnant cows on Day 28 were inseminated using timed AI after the completion of the Ovsynch protocol 10 and 17 days after enrollment in the study for RES and CON groups, respectively. Progesterone concentration > or =2.35 ng/ml was used as an indicator of pregnancy on Day 21. For RES and CON cows, pregnancy rate at Days 21 (70.9% versus 73.0%, P<0.56), 28 (33.1% versus 33.6%; P<0.80) and 42 (27.0% versus 26.8%; P<0.98) after the pre-enrollment AI did not differ. Administration of GnRH on Day 21 after AI had no effect on pregnancy loss in RES and CON groups from days 21 to 28 (53.2% versus 53.5%; P<0.94) and days 28 to 42 (17.9%; P<0.74) after AI. Pregnancy rate after the resynchronization period was similar for both treatment groups. Resynchronization with GnRH given on Day 21 after AI for initiation of a timed AI protocol prior to pregnancy diagnosis does not affect pregnancy rate and pregnancy loss in lactating dairy cows.

Effects of Alfaprostol and uterine palpation on postpartum interval and pregnancy rate to embryo transfer in Brahman influenced beef cows.

Two trials were conducted to determine the effects of Alfaprostol, a prostaglandin F2 alpha analogue, and uterine palpation on reproductive performance in postpartum beef cows. In Trial J, 143 multiparous and 132 primiparous Brahman crossbred embryo recipient cows were assigned to receive one 5 mg injection of Alfaprostol between 29 and 65 d postpartum or to serve as controls. Alfaprostol treatment between 29 and 42 d postpartum decreased (P<0.05) postpartum interval in multiparous but not primiparous cows. Alfaprostol treatment increased pregnancy rate to embryo transfer. Trial II utilized 369 multiparous Brangus cows, 17 to 49 d postpartum, that were assigned to one of three groups: 1) Alfaprostol plus uterine palpation (APP), 2) Uterine palpation only (UP) or 3) Control. Completion of uterine involution was determined at treatment. Alfaprostol plus uterine palpation decreased postpartum interval regardless of uterine involution, while UP had a greater effect if uterine involution was not complete. Both APP and UP increased the number of cows which were detected in estrus by 80 d postpartum. Alfaprostol and uterine palpation, alone or in combination, can increase postpartum reproductive performance in Brahman crossbred and Brangus beef cows.

Ovarian follicular dynamics in Nelore breed (Bos indicus) cattle.

The most common beef cattle raised in Brazil is the Nelore breed (Bos indicus). Information obtained by ultrasonography on follicular growth in Bos taurus cattle has been accumulating rapidly. However, there are few publications to date on follicular development in Bos indicus breeds. The follicular dynamics in Nelore heifers and cows during natural or prostaglandin (PG)-induced estrous cycle were studied. From the detection of estrus onward, all animals were examined daily by ultrasonography for one (n = 35) or two (n = 10) consecutive estrous cycles. The follicular dynamic in Nelore cattle was characterized by the predominance of 2 follicular waves in the cows (83.3%, n = 18, P < 0.05) and 3 waves in the heifers (64.7%, n = 16, P < 0.05). Most of the cattle observed over 2 consecutive estrous cycles presented the same pattern of follicular waves in the first and second cycle, and only 30% showed variation in the number of waves from one cycle to the other. Most of the follicular parameters analyzed were not affected by PG treatment or age but were altered by follicular waves. Consequently, data on cows and heifers were combined according to the number of follicular waves. The ovulatory follicle was larger than the other dominant follicles (P < 0.05), and the ovulatory wave was shorter than the preceding waves (P < 0.05). The interovulatory interval was longer in animals showing 3 waves than those exhibiting 2 waves (P < 0.05). Maximum diameter of the dominant follicle (around 11 mm) and of the corpus luteum (CL, approximately 17 mm) were smaller than those reported for European breeds. In conclusion, the results demonstrate that although the dominant follicle and corpus luteum are smaller than in European breeds, the follicular dynamics in Nelore cattle were similar to those observed in European breeds and were characterized by 2 or 3 follicular waves for cows and heifers, respectively, during the natural or prostaglandin-induced estrous cycle.

Effect of injection of beta-carotene or vitamin E and selenium on fertility of lactating dairy cows.

Experiments tested whether supplemental antioxidants improved fertility. To test effects of beta-carotene, cows in a hot environment were injected with prostaglandin F2 alpha (PGF2 alpha) and were given 3 injections, i.m., of 800 mg beta-carotene or saline at Days -6 and -3 before the anticipated date of insemination and at insemination (n = 37-41 inseminated cows/group). There was no effect of beta-carotene on the proportion of cows detected in estrus following PGF2 alpha, timing of estrus after PGF2 alpha injection or pregnancy rate in inseminated cows. In a second trial, cows in a temperate climate received intramuscular injections of vitamin E (500 mg) and selenium (50 mg) at 30 d post partum (n = 97) or were untreated controls (n = 89). Treatment did not affect interval from calving to first insemination or the proportion of cows pregnant at first service, but it increased the pregnancy rate at second service (69.8 vs 52.1%; P = 0.07) and reduced services per conception (1.7 vs 2.0; P < 0.05) and interval from calving to conception (84.6 vs 98.1; P < 0.05). Thus, injection of vitamin E and selenium increased fertility in cattle that did not become pregnant at first service.

Effect of a deep uterine insemination on spermatozoal accessibility to the ovum in cattle: a competitive insemination study.

A competitive insemination study was conducted to determine the effect of a deep uterine insemination on accessory sperm number per embryo in cattle. Cryopreserved semen of a fertile bull characterized by spermatozoa with a semi-flattened region of the anterior sperm head (marked bull) was matched with cryopreserved semen from an unmarked bull having spermatozoa with a conventional head shape. Using 0.25-mL French straws and a side delivery embryo transfer device, deep uterine insemination (0.125 mL deposited in each horn) was performed 2 cm from the uterotubal junction. Immediately after, the uterine body was artificially inseminated using semen (0.25 mL) from an alternate bull and a conventional insemination device. The complete dose (both inseminations) was 50x10(6) total sperm cells consisting of an equal number of spermatozoa from each bull. Single ovulating cows (n = 95) were inseminated at random with either the unmarked semen in the uterine body and marked semen in the uterine horn, or the unmarked semen in the uterine horn and marked semen in the uterine body. Sixty-one embryos(ova) were recovered nonsurgically 6 d post insemination, of which 40 were fertilized and contained accessory spermatozoa. The ratio and total number of accessory spermatozoa recovered was different among treatments: 62:38 (326) for the unmarked semen in the uterine body and marked semen in the uterine horn, and 72:28 (454) for the unmarked semen in the uterine horn and marked semen in the uterine body (P<0.05). Deep uterine insemination using this semen in a split dose and a side delivery device favors accessibility of spermatozoa to the ovum compared with conventional uterine body insemination.

An ultrasonographic study of luteal function in breeds of sheep with different ovulation rates.

Development and demise of luteal structures were monitored using daily transrectal ultrasonography in 2 breeds of sheep differing in ovulation rates (nonprolific Western white-faced cross-bred, n = 12 and prolific pure-bred Finn sheep, n = 7), during 1 estrous cycle in the mid-breeding season. Jugular blood samples were collected once a day for radioimmunoassay (RIA) of progesterone. The mean diameter of ovulatory follicles was higher in Western white-faced than in Finn ewes (6.4 +/- 0.2 and 5.3 +/- 0.2 mm, respectively; P < 0.001). The mean volume of luteal structures was higher (P < 0.05) in Western white-faced compared with Finn sheep from Days 5 to 15 of the cycle (Day 0 = day of ovulation). This accounted for the higher (P < 0.05) total luteal volumes recorded in Western white-faced ewes on Day 7 and from Days 11 to 15, despite the higher ovulation rate in Finn ewes (2.7 +/- 0.3 and 1.7 +/- 0.2, respectively; P < 0.05). Mean serum progesterone concentrations were higher (P < 0.05) in Western white-faced than in Finn ewes from Days 4 to 14. Daily total luteal volumes were positively correlated with daily serum progesterone concentrations throughout the cycle in Finn sheep (r > or = 0.40, P < 0.02), and during luteal growth and regression (r > 0.60, P < or = 0.00001) but not during mid-cycle in white-faced ewes (r = 0.16; P = 0.22). During the growth of the corpora lutea (CL), luteal tissue volume increased faster (P < 0.05) than serum progesterone concentrations in both breeds of sheep. During luteolysis, the decrease in luteal volumes parallelled that in serum progesterone concentrations in Finn (P = 0.11) but not in Western white-faced ewes, where luteal volumes decreased more slowly (P = 0.02) in relation to progesterone secretion. Increased ovulation rate in prolific Finn ewes resulted in more but smaller CL, and lower serum progesterone levels compared with nonprolific Western white-faced ewes. We conclude that breed-specific mechanisms exist to control the formation of luteal tissue and progesterone secretion in cyclic ewes differing in prolificacy. The mechanisms may involve ovulation of Graafian follicles at different sizes and inhibitory paracrine effects of CL on co-existing CL.

Endocrine function of neuropeptide Y knockout mice.

Among its many proposed functions, neuropeptide Y (NPY) is thought to modulate the hypothalamic-pituitary axis. Specifically, increased hypothalamic NPY signaling may be critical in mediating the neuroendocrine response to fasting. To determine the consequences of NPY deficiency on endocrine physiology, multiple hormones were quantitated in wildtype and NPY-knockout mice under fed and fasted conditions. Serum concentrations of leptin, corticosterone, thyroxine, and testosterone were normal in NPY-knockout males fed ad libitum. A 48-hour fast resulted in a 50% reduction in leptin, a 60% reduction in thyroxine, a 75% reduction in testosterone, and a 12-fold increase in corticosterone in both wildtype and NPY-knockout mice. Fasting also increased the estrous cycle length by 3 days in both wildtype and NPY-deficient female mice. We conclude that NPY is not essential for appropriate function of the gonadotropic, thyrotropic, or corticotropic axes under ad lib fed conditions or in response to acute fasting.

Use of urea-molasses-multinutrient block and urea-treated rice straw for improving dairy cattle productivity in Vietnam.

After conducting a preliminary survey, a feeding trial was carried out to determine the effect of urea-molasses-multinutrient block (UMMB) and urea-treated rice straw (UTRS) as a feed supplement on the productivity of dairy cows. Sixty Holstein-Friesian crossbred cows on 11 smallholder farms were divided equally into control, UMMB and UTRS supplementation groups. Milk yield and feed intake were recorded daily. Milk fat content, body weight and body condition score (BSC) of each cow were determined at two week intervals. Milk samples for progesterone analysis were collected once a week commencing one month after parturition. Data were recorded for date of onset of ovarian activity, estrus, insemination, and conception rate. Milk production increased by 10.3-11.9% and milk fat content increased by 3-5%, therefore, profit for farmers increased by US $0.55-0.73 per cow per day (exchange rate US $1 = VN $11,000). The intervals from calving to onset of ovarian activity (91-94 days), to estrus (110-114 days), to conception (121-122 days) and the calving interval (13.4-13.6 months) in the trial groups were significantly shorter than those in the control group (112, 135, 152 days and 14.4 months, respectively.

Oviductal isthmic motility patterns as monitored by polyview in unrestrained sows around ovulation.

A method for monitoring oviductal isthmic motility in sows incorporating a computer programme (Polyview) was developed. This method was found to be reliable and easy for recording and analysing data. Isthmic motility patterns were monitored from 11 h prior to and up to 36 h after ovulation in 13 unrestrained multiparous sows during their second oestrus after weaning. The amplitudes and frequencies of phasic pressure fluctuations in relation to the hormonal profiles were also calculated. The isthmic motility patterns were regular before ovulation changing to wave patterns during the peri-ovulatory period and eventually to irregular patterns after ovulation. The amplitudes and frequencies of phasic pressure fluctuations were significantly higher (p<0.05) prior to and soon after ovulation than afterwards. Plasma oestradiol-17beta levels significantly (p<0.05) decreased before ovulation while plasma progesterone levels increased significantly (p<0.05) after ovulation. Despite a significant decrease in the plasma levels of oestradiol-17beta prior to ovulation, the amplitudes and frequencies of phasic pressure fluctuations remained high until shortly after ovulation. This could have been due to the endogenous levels of oestradiol-17beta bound to the nuclear oestradiol-17beta receptors that might still have been present in the isthmus. Conversely, the irregular isthmic motility patterns, the decline in the frequencies of phasic pressure fluctuations and amplitudes seen after ovulation may have been due to the rising plasma levels of progesterone. The amplitudes and frequencies of phasic pressure fluctuations were highest at the time when oestradiol-17beta levels were highest and when progesterone levels were low. It can be concluded that the changes in the isthmic motility patterns, amplitudes and frequencies of phasic pressure fluctuations in relation to the changes in the plasma levels of oestradiol-17beta and progesterone seen in the present study prior to and after ovulation indicate a possible role of the oviduct in regulating gamete transport.

The sow endometrium at different stages of the oestrous cycle: studies on morphological changes and infiltration by cells of the immune system.

The aim of this study was to investigate the distribution of leukocytes and the morphological changes of the sow endometrium throughout the oestrous cycle. Fifteen crossbred multiparous sows (Swedish Landrace x Swedish Yorkshire), with an average parity number of 3.4 +/- 0.7 (mean +/- S.D.) were used. Blood samples were collected from the jugular vein 1h before slaughter for analyses of oestradiol-17beta and progesterone levels. Uterine samples from the mesometrial side of both horns, taken immediately after slaughter at late dioestrus, prooestrus, oestrus, early dioestrus and dioestrus, were fixed, embedded in plastic resin and stained with toluidine blue. The surface and glandular epithelium as well as subepithelial and glandular connective tissue layers were examined by light microscopy. The significantly highest surface and the glandular epithelium were observed at oestrus and dioestrus, respectively. The largest number of capillaries (underneath the surface epithelium) was found at oestrus. In the surface epithelium, the largest number of intraepithelial lymphocytes (IELs, round nucleus) was found at early dioestrus. The largest number of lymphocytes and macrophages within the glandular epithelium were found at early dioestrus and oestrus, respectively. In the subepithelial connective tissue layer, the most common type of leukocytes during all stages was the lymphocyte. The largest numbers of lymphocytes and neutrophils were found at oestrus while the largest number of eosinophils was found at dioestrus. The dominating cells of the immune system in the connective tissue of the glandular layer were lymphocytes and macrophages. The significantly largest numbers of lymphocytes and plasma cells were found at early dioestrus and dioestrus, respectively. The number of lymphocytes in the connective tissue of the glandular layer and the number of plasma cells in the subepithelial layer were positively correlated with the plasma level of progesterone (P < or = 0.05). The numbers of capillaries and neutrophils in the subepithelial layer underneath the surface epithelium as well as the number of macrophages in both surface and glandular epithelium were positively correlated with the plasma level of oestradiol-17beta (P < or = 0.05). In conclusion, the present study showed a variation in the infiltration and distribution of lymphocytes, neutrophils, eosinophils, macrophages, mast cells and plasma cells in the sow endometrium during different stages of the oestrous cycle. Also morphological parameters (e.g. height of surface and glandular epithelium, capillaries density and degree of oedema) varied throughout the oestrous cycle.

Pregnancy diagnosis in dairy goats using progesterone assay kits and oestrous observation.

Two qualitative on-farm milk progesterone test kits were used for early pregnancy diagnosis in goats. One kit was based on latex agglutination (LA) and the other on enzyme-linked immunosorbent assay (ELISA). The accuracy of early pregnancy diagnosis by these two methods was compared with the accuracy of oestrous observation (OeO) and the level of plasma progesterone measured by radioimmunoassay (RIA). Dairy goats (n = 73) from a single herd were used for collection of milk and blood samples 20 days after breeding. Ultrasound examination 50 days after mating found that 49 (67%) goats were pregnant, and 24 (33%) were not pregnant. Using ultrasound as a reference method, the accuracy of early pregnancy diagnosis by RIA and OeO was 92% and 86%, respectively, and for non-pregnancy 100% for both RIA and OeO. The accuracy of early pregnancy diagnosis by ELISA and LA was 82% and 79% and for non-pregnancy, 88% and 100%, respectively. The kappa-statistic for RIA, OeO, ELISA and LA was 0.93, 0.84, 0.77 and 0.73, respectively. It was concluded that LA and ELISA tests can be used for early pregnancy diagnosis in dairy goats. However, in the herd studied, early pregnancy diagnosis by OeO was as good as that achieved with progesterone determination using the kits.
