Triphenyltin salicylate-antimicrobial effect and resistance--the pyrophosphatase connection.

The effect of Triphenyltin salicylate (TPS) was tested against six bacteria, Escherichia coli, Staphylococcus aureus, Shigella flexneri, Pseudomonas aeruginosa, Klebsiella pneumoniae and Salmonella typhi and five fungi, Aspergillus flavus, Aspergillus fumigatus, Aspergillus niger, Rhodotorula spp. and Saccharomyces spp. Sensitivity tests were determined with 5-500 microg/ml of TPS. All organisms were sensitive to the compound except Klebsiella pneumoniae, Pseudomonas aeruginosa, Rhodotorula spp. and Saccharomyces spp. The minimum dose of TPS that can kill 50% of the susceptible microorganisms is in the range 5-50 microg/ml. Membrane bound pyrophosphatase(s) from the organisms was non-competitively inhibited by 5 microM TPS with Ki values of 7.6, 18, 8.8 and 6.9 microM for Escherichia coli, Shigella flexneri, Aspergillus niger, and Aspergillus fumigatus, respectively. The physiological index of efficiency of the enzyme (Vmax/KM) for TPS susceptible organisms was reduced by 17-68% in the presence of 5-10 microM of the compound. In contrast the index for the non-susceptible organisms was unaffected. The mode of action of TPS is discussed.

Determination of butyltin compounds in surface sediments from the São Paulo State coast (Brazil) by gas chromatography-pulsed flame photometric detection.

Occurrence and the effects of butyltin compounds (BTs) have been studied for some years, mainly in countries of the Northern Hemisphere. Due to widespread use of tributyltin compounds (TBTs) and considering their deleterious effects, it is necessary to conduct studies on its occurrence, especially in the marine environment because of its excessive use in coatings of ship hulls to prevent fouling. Moreover, it was important to extend the evaluation to areas where there is no current information about their occurrence. The present work reports the occurrence of BTs in marine sediments of São Paulo state, Brazil. Commercial and leisure harbor sampling sites were selected because these areas are potentially exposed to BTs from antifouling paints used on ship hulls. Analytical conditions for organotin analysis in marine sediments were optimized for GC with pulsed flame photometric detection. Detection limits ranged from 8A to 66.3 ng g(-1) using a 610-nm filter, and the linearity range was 20-500 ng g(-1). Concentration levels of BTs were highest in Santos harbor (360 ng g(-1) TBT in average) and Guarujá marina (670 ng g(-1) TBT in average), which seems to be related to intensive boat traffic. Lower levels of BTs were observed in Cananéia, where only fishing boats are present (50 ng g(-1) TBT in average).

Synthesis and antimicrobial activity of ethyl N-aryl-S-(triphenylstannyl)isothiocarbamates.

Five ethyl N-aryl-S-(triphenylstannyl)isothiocarbamates were synthesized by the reaction of triphenyltin iodide with the appropriate ethyl N-arylthiocarbamate in the presence of triethylamine. The IR spectrum of each compound was obtained over the 4000--200-cm--1 range, and some bands were assigned. These new compounds were found to be generally better antifungal agents than the previously tested N-substituted N'-cyano-S-(triphenylstannyl)isothioureas. The new compounds were also investigated for antibacterial activity and were especially inhibitory toward Gram-positive species. Except for their lower activity toward Bacillus subtilis, their antibacterial activity was identical to the previously tested N-phenyl-N'-cyano-S-(triphenylstannyl)isothiourea.

DNA interaction and mutagenicity of the plastic stabilizer di-n-octyltin dichloride.

The organo-metallic compound di-n-octyltin dichloride (DOTC) is used as an additive to plastics. Since it is, therefore, present in the human environment, its long-term toxicity, particularly its genotoxic potential, is of concern. In the present studies, the agent was shown to interact with the DNA of cultured V79 Chinese hamster cells. Though the compound did not induce mutagenesis in the Salmonella-microsome assay and DNA repair in primary rat hepatocytes, it did induce mutagenesis in V79 Chinese hamster cells hypoxanthine guanine phosphoribosyl transferase (HGPRT) assay. Further studies of the long-term toxicity of DOTC, particularly its carcinogenic potential, are necessary.

Trialkyltin compounds enhance human CG secretion and aromatase activity in human placental choriocarcinoma cells.

Human choriocarcinoma cell lines have been used as placental models for the study of endocrine function, including aromatase (CYP19) activity and the secretion of human CG (hCG). In the present study, we investigated the effects of trialkyltin compounds, which are suspected endocrine disrupters, on aromatase activity and hCG secretion in human choriocarcinoma JAR, JEG-3, and BeWo cells. Protein synthesis as measured by (35)S-methionine incorporation in all cell lines was markedly decreased by treatment with both tributyltin (TBT) and triphenyltin (TPT) at concentrations above 3 x 10(-7) M, due to cytotoxicity. In JAR cells, (35)S-methionine uptake was decreased by 50% at 3 x 10(-7) M of TBT. At a TPT concentration of 1 x 10(-7) M, protein synthesis in JAR cells was not affected, whereas JEG-3 and BeWo cells demonstrated slightly decreases. In all cell lines, both TBT and TPT increased levels of hCG secretion and aromatase activity in a dose- and time-dependent fashion following exposure to nontoxic concentration ranges. In addition, these trialkyltin compounds enhanced 8-bromo-cAMP-induced hCG secretion and aromatase activity in JAR cells. TBT caused dose-related increases in steady-state mRNA levels of both hCGbeta and CYP19 in JAR cells following 24- or 48-h exposure to nontoxic concentrations of TBT. However, these mRNA changes in JAR cells were not comparable to the changes in both hCG secretion and aromatase activity. These results indicate that the observed trialkyltin-induced alterations in human choriocarcinoma cells are due to other mechanism in addition to a regulation of hCG and CYP19 mRNA levels. Our studies suggest that trialkyltin compounds are potent stimulators of human placental hCG production and aromatase activity in vitro; and the placenta represents a potential target organ for trialkyltin compounds, whose endocrine-disrupting effects might be the result of local changes in hCG and estrogen concentrations in pregnant women.

Organochlorine pesticides, polychlorinated biphenyls, and butyltin compounds in blubber and livers of stranded California sea lions, elephant seals, and harbor seals from coastal California, USA.

Concentrations of polychlorinated biphenyls (PCBs), DDTs (p,p'-DDE, p,p'-DDD, p,p'-DDT), chlordanes (CHLs; cis-chlordane, cis-nonachlor, trans-nonachlor, and oxychlordane), hexachlorocyclohexane isomers (HCHs), hexachlorobenzene (HCB), tris(4-chlorophenyl)methane (TCPMe), tris(4-chlorophenyl)methanol (TCPMOH), and mono- (MBT), di-(DBT), and tri-butyltin (TBT) were determined in blubber and livers of 15 California sea lions (Zalophus californianus), 6 northern elephant seals (Mirounga augustirostris), and 10 harbor seals (Phoca vitulina) found stranded along the coasts of California, USA, during 1991-1997. Among the organochlorines analyzed, DDTs were predominant, followed in decreasing order by PCBs, CHLs, TCPMe, TCPMOH, HCHs, and HCB. The greatest concentrations of organochlorines were found in California sea lions. The highest DDT and PCB concentrations found in the blubber of California sea lions were 2,900 and 1,300 microg/g, lipid weight, respectively. Concentrations of TCPMe and TCPMOH in California sea lions were correlated significantly with DDT concentrations. Concentration ratios of various organochlorines in harbor seal livers were different from those in California sea lions and elephant seals, which suggested that the sources of exposure of harbor seals to organochlorines were different from those in the other two species. Concentrations of butyltin compounds in livers of pinniped species ranged from 2 to 99 ng/g, wet weight, which were less than those observed in cetaceans and in California sea otters.

Steroid levels in crinoid echinoderms are altered by exposure to model endocrine disruptors.

Sexual steroids (testosterone and estradiol) were measured in the whole body of wild specimens of the crinoid Antedon mediterranea collected from the Tyrrhenian Sea (Italy). Testosterone levels (274-1,488 pg/g wet weight (w.w.)) were higher than those of estradiol (60-442 pg/g w.w.) and no significant differences between males and females were observed. No clear seasonal trend was either detected - individuals from February, June and October 2004 analyzed - apart from a peak of estradiol in males in autumn. Nonetheless, dramatic changes on tissue steroid levels were observed when individuals were exposed to model androgenic and anti-androgenic compounds for 2 and 4 weeks. The selected compounds were 17 alpha-methyltestosterone (17 alpha-MT), triphenyltin (TPT), fenarimol (FEN), cyproterone acetate (CPA), and p,p'-DDE. Endogenous testosterone levels were significantly increased after exposure to 17 alpha-MT, TPT and FEN, while different responses were observed for estradiol; 17 alpha-MT and FEN increased endogenous estradiol (up to seven-fold), and TPT lead to a significant decrease. Concerning the anti-androgenic compounds, CPA significantly reduced testosterone in a dose-dependent manner without altering estradiol levels, whereas specimens exposed to p,p'-DDE at a low dose (24 ng/L) for 4 weeks showed a four-fold increase in T levels. Overall, the data show the ability of the selected compounds to alter endogenous steroid concentrations in A. mediterranea, and suggest the existence in this echinoderm species of vertebrate-like mechanisms that can be affected by exposure to androgenic and anti-androgenic chemicals.

Triorganotin compounds as antimicrobial agents.

Six triorganotin derivatives of thiolupinine(1-mercaptolupinane), 2-mercaptobenzoxazole and 2-mercaptobenzothiazole were prepared and tested against several bacteria, fungi and protozoa. Most compounds exhibited high activity against the tested microorganisms and particularly worth noting was the activity of triethyltin lupinylsulfide on Gram-negative strains. Triethylgermanium lupinylsulfide was also prepared but was devoid of action on the whole set of tested microorganisms.

Diorganotin(IV) complexes of pyridoxal thiosemicarbazone: synthesis, spectroscopic properties and biological activity.

The complexes [SnR2(L)] (R = Me, Et, Bu, Ph; H2L = pyridoxal thiosemicarbazone) have been prepared and characterized. In the light of the spectral properties of the complexes in the solid state (IR, mass, Mössbauer) the bideprotonated thiosemicarbazonato anion is O(phenolic)-, N(3)-, S-bonded to the tin atom which probably has trigonal bipyramidal coordination with N(3) atom and R groups occupying equatorial positions. NMR ( 1H, 13C and 119Sn) data in CDCl3 or DMSO-d6 suggest that this coordinative picture remains in these solutions. The ethyl, butyl and phenyl derivatives suppress proliferation of Friend erithroleukaemia cells (FLC). Of the pyridoxal thiosemicarbazone complexes so far evaluated. [SnBu2(L)] and [SnPh2(L)] showed the lowest thresholds for inhibition of FLC proliferation. The effects of these compounds on DMSO-induced differentiation of FLC, DNA synthesis and reverse transcriptase were also assayed.

The relationship between the length of the alkyl chain of dialkyltin compounds and their effects on thymus and bile ducts in mice.

The effects on thymus and bile ducts have been studied in male mice 4 days after intravenous administration of 15 and 20 mumol/kg of di-n-butyltin dichloride, di-n-pentylin dichloride, di-n-hexyltin dichloride, di-n-heptyltin dichloride and di-n-octyltin dichloride. The degree of the reduction of relative thymus weight and thymocyte count is not dependent on the length of the alkyl chain of the di-n-alkyltin dichlorides (4 to 8 C atoms). Signs of bile duct lesions (increase of the serum alkaline phosphatase activity and bilirubin concentration, enlargement of bile duct diameter, histopathological findings) were induced only after administration of di-n-alkyltin dichlorides with alkyl chains of 4 to 6 C atoms. The toxic effects of the di-n-alkyltin compounds on thymus and bile ducts seem to be independent of each other. The investigated dialkyltin derivates exert cytotoxic effects on macrophages from spleen fragments of guinea pigs in concentrations of 10(-6) to 10(-4) M. There is a tendency toward a decrease of the cytotoxicity with the increasing length of the alkyl chains.

The effect of steric constraints on the adsorption of phenyltin onto the dipalmitoylphosphatidylcholine bilayer.

In this paper, we present studies concerning phenyltin adsorption onto the dipalmitoylphosphatidylcholine bilayer. Phenyltin compounds are known to be biologically active, and their molecular geometry makes it possible to study the effect of steric constraints on their ability to penetrate the model lipid membrane. Using a fluorescence probe as a reporter of the amount of adsorbed compound, we evaluated their affinity to the membrane as a function of the membrane state. The amount of the adsorbed compound was found to depend on the adsorbing molecule's geometry and lipid bilayer organization. The fluorescence measurements were supported by the density functional theory (DFT) method of quantum mechanical computations. The penetrant location was correlated with the possible relative positions of its polar and hydrophobic moieties to determine if it could adopt structural requirements of the local membrane environment. Molecules were deformed by a model force, mimicking interactions within the membrane interfacial region. Computations show that the diphenyltin molecule can be deformed to such an extent that it can adopt an amphiphilic conformation. Triphenyltin is different, as its bending requires more energy. Born repulsion energies from hydrophobic fluid into water for phenyltins were also computed in an isodensity-polarized continua model of DFT computation. Our results indicate that the phenyltin compounds incorporate into the interface of the lipid membrane, although diphenyltin integrates more deeply than triphenyltin, which locates on the double layer's surface, and this is due to the fact that the main role is played by steric and not electrostatic interactions.

Synthetic, spectral, thermal and antimicrobial studies on some mixed 1,3-dithia-2-stannacyclopentane derivatives with dialkyldithiocarbamates.

1,3-dithia-2-stannacyclopentane derivatives with dialkyldithiocarbamates of the types SCH(2)CH(2)SSn[S(2)CNR(2)]Cl (I) and SCH(2)CH(2)SSn[S(2)CNR(2)](2) (II) (where R = CH(3), C(2)H(5) and -CH(2)-CH(2)-) have been synthesized by the reaction of 2,2-dichloro-1,3-dithia-2-stannacyclopentane and sodium/ammonium salts of dialkyldithiocarbamates in 1:1 and 1:2 molar ratios, respectively, in anhydrous benzene. These newly synthesized derivatives have been characterized by elemental analyses (C, H, N, S and Sn), thermal [thermogravimetry (TG) and differential thermal analyses (DTA)] as well as spectral [UV, IR and multinuclear NMR ((1)H, (13)C and (119)Sn)] studies. The monodentate behaviour of the dialkyldithiocarbamate ligands was confirmed by IR and (119)Sn NMR spectral data and distorted tetrahedral structures have been suggested for both type (I) and (II) compounds. The free ligands and their tin complexes have also been screened for their antibacterial and antifungal activities. These results made it desirable to delineate a comparison between free ligands and their tin complexes. These exhibit higher antibacterial effect than some of the previously investigated antibiotics.

Effects of butyltin compounds on mitochondrial respiration and its relation to hepatotoxicity in mice and Guinea pigs.

Because the mechanisms responsible for the difference in toxicity between different experimental animal species remain unclear, the effects of tributyltin chloride (TBTC) and dibutyltin dichloride (DBTC) on mitochondrial respiration were compared among the livers of mice and guinea pigs in vitro and in vivo. Further, the levels of these butyltin compounds and their derivatives in the mitochondrial fractions of the hepatocytes were investigated in these animal species. Administration of TBTC and DBTC to mice resulted in the obvious elevation of serum enzymatic activities, as well as the inhibition of succinate-linked State 3 respiration in hepatic mitochondria at 24 h after administration. On the other hand, these metal compounds failed to induce such hepatotoxicity or to inhibit mitochondrial respiration in guinea pigs. There was no significant difference between mice and guinea pigs in the IC50 (metal concentration observed in 50% inhibition of mitochondrial respiration) of TBTC and DBTC against the succinate-linked State 3 respiration of hepatic mitochondria in vitro, although the mitochondrial respiration of succinate-linked State 3 was inhibited in the liver of mice treated with the metals in vivo. The levels of total butyltin compounds in the mitochondrial fractions of hepatocytes were higher in the mice than in the guinea pigs, and the main butyltin compound in the mitochondrial fractions was DBTC in both species at 24 h after TBTC or DBTC administration. The amount of sulfhydryl groups, which were capable of binding with DBTC, in mice hepatic mitochondria was twice as large as that in guinea pigs, and the affinity of DBTC for the isolated hepatic mitochondria was higher in mice than in guinea pigs in vitro. These results suggested that the induction of hepatotoxicity by TBTC and DBTC in vivo was closely associated with the depression of mitochondrial respiration and that the difference in susceptibility to the metal-induced mitochondrial damages between mice and guinea pigs might result from the high affinity of butyltin compounds, in particular DBTC, for hepatic mitochondria in mice containing higher levels of sulfhydryl groups, compared with guinea pigs.

Synthesis, characterization and biological activity of triorganotin 2-phenyl-1,2,3-triazole-4-carboxylates.

The triorganotin 2-phenyl-1,2,3-triazole-4-carboxylates, 2-PhC2N3CO2SnR3 (R=C6H5, 1; c-C6H11, 2; C6H5C(CH3)2CH2, 3), have been prepared and characterized by means of elemental analysis, IR and NMR (1H, 13C and 119Sn) spectroscopy. The crystal structures of 1 and 3 have been determined. Compound 1 is polymeric in nature with a trigonal bipyramidal configuration, and compound 3 shows a tetrahedral geometry. Bioassay results have shown that these compounds have good antibacterial and antitumor activity. The activity against three human tumor cell lines (HeLa, CoLo205 and MCF-7) decreased in the order 1>2>3.

Different effects of di- and triphenyltin compounds on lipid bilayer dithionite permeabilization.

Phenyltins are chemicals widely used in industry, hence their occurrence in the human environment is frequent and widespread. Such compounds include hydrophobic phenyl rings bonded to positively charged tin. This molecular structure makes them capable of adsorbing onto and penetrating through biological membranes, hence they are potentially hazardous. Two such compounds, diphenyltin and triphenyltin, show different steric constraints when interacting with the lipid bilayer. It has been demonstrated that these compounds are positioned at different locations within model lipid bilayers, causing dissimilarity in their ability to affect membrane properties. In this paper we present a study regarding the ability of these two phenyltins to facilitate the transport of S2O4(-2) ions across the lipid bilayer, evaluated by a fluorescence quenching assay. In concentration range of up-to 60 microM those compounds do not affect lipid bilayer topology, when evaluated by vesicle size distribution. Both phenyltins facilitate the transfer of S2O4(-2) across the model lipid bilayer, but the dependence of dithionite transport on phenyltin concentration is different for both. In principle, above 20 microM triphenyltin is more efficient in transferring ions across the lipid bilayer than diphenyltin.

Simultaneous determination of mono-, di- and tributyltin in environmental samples using isotope dilution gas chromatography mass spectrometry.

The development of a rapid, precise and accurate speciation method for the simultaneous determination of mono-, di- and tributyltin in environmental samples is described. The method is based on using isotope dilution gas chromatography/mass spectrometry (GC/MS) with electron ionization, a widely used technique in routine testing laboratories. A mixed spike containing (119)Sn-enriched monobutyltin (MBT), dibutyltin (DBT) and tributyltin (TBT) was used for the isotope dilution of the samples. Five molecular ions were monitored for each analyte, corresponding to the (116)Sn, (117)Sn, (118)Sn, (119)Sn and (120)Sn isotopes. The detection at masses corresponding to (116)Sn and (117)Sn were used to correct for m + 1 and m + 2 contributions of (13)C from the organic groups attached to the tin atom on the (118)Sn, (119)Sn and (120)Sn masses with simple mathematical equations and the concentrations of the butyltin compounds were calculated based on the corrected (118)Sn/(119)Sn and (120)Sn/(119)Sn isotope ratios. The (119)Sn-enriched multispecies spike was applied with satisfactory results to the simultaneous determination of MBT, DBT and TBT in three certified reference materials: two sediments, PACS-2 and BCR 646, and the mussel tissue CRM 477. The method was compared with a previously published GC/inductively coupled plasma MS isotope dilution procedure, developed in our laboratory, by injecting the same samples into both instruments. Comparable analytical results in terms of precision and accuracy are demonstrated for both atomic and molecular mass spectrometric detectors. Thus, reliable quantitative organotin speciation analysis can be achieved using the more widespread and inexpensive GC/MS instrument.

Diorganotin(IV) and triorganotin(IV) complexes of meso-tetra(4-sulfonatophenyl)porphine induce apoptosis in A375 human melanoma cells.

The cytotoxic effect of several diorganotin(IV) and triorganotin(IV)-meso-tetra(4-sulfonatophenyl)porphine derivatives was tested and only the (Bu(2)Sn)(2)TPPS and the (Bu(3)Sn)(4)TPPS showed cytotoxicity on A375 human melanoma cells. To examine the pathway of (Bu(2)Sn)(2)TPPS or (Bu(3)Sn)(4)TPPS induced A375 cell death, DNA fragmentation analysis, Annexin V binding and PI uptake as well as caspases activation analysis by Western blot were carried out. A375 cells treated exhibited several typical characteristics of apoptosis. Both the (Bu(2)Sn)(2)TPPS and the (Bu(3)Sn)(4)TPPS compounds activate caspase-8 and caspase-9 leading to caspase-3 activation. Thus, we propose that these two porphirin derivatives lead to the apoptosis of human melanoma cells via both death receptor-mediated and mitochondrial apoptotic pathways.

Seven new synthetical (Z)-1-[2-(Triarylstannyl) vinyl]-1-cycloheptanols organic tin compounds were studied by X-ray photoelectron spectroscopy and mass spectrum method. The influence of different replacement radical in compounds on the d orbit of tin atom was discussed. The coordinate substance had an effect on the chemical displacement of Ols binding energy. The O-->Sn bond was affected by the different replacement radical. The results of X-ray photoelectron spectroscopy and mass spectrum show that Cl, Br and I had the important action.
