Phase II study of oral doxifluridine in elderly patients with advanced non-small-cell lung cancer.

Elderly patients with advanced non-small-cell lung cancer (NSCLC) are usually excluded from most clinical trials because of the toxicity associated with chemotherapy. About 50% of the new cases of lung cancer occur in patients older than 65 years. Doxifluridine is a fluoropyrimidine derivate which can be administered orally with very low toxicities. This phase II study evaluates the toxicity and activity of a home therapy with oral doxifluridine in elderly advanced NSCLC patients. Thirty-three advanced NSCLC patients, aged 70 years or more, entered the study; median ECOG performance status was 1 (0-2) and 22 patients (66.6%) had metastatic disease. Doxifluridine was given orally in three divided doses, for a total daily dose of 2,250 mg, for 4 consecutive days every week. The treatment was well tolerated; five patients (15%) experienced a grade 3 diarrhea which required doxifluridine dose reduction to 1,500 mg daily. Thirty-one patients are evaluable for response; four partial responses (12.9%) have been observed (95% confidence limit interval 3.6-29.8%); 17 patients (54.8%) had a stabilization of the disease. This study demonstrates that a home therapy with oral doxifluridine in elderly NSCLC patients is feasible and well tolerated and should encourage further studies.

A new Myxococcus xanthus gene cluster for the biosynthesis of the antibiotic saframycin Mx1 encoding a peptide synthetase.

The gene cluster for the biosynthesis of the heterocyclic quinone antibiotic saframycin Mx1 of Myxococcus xanthus DM504/15 was inactivated and tagged by Tn5 insertions. The tagged genes were cloned in Escherichia coli and used to select overlapping cosmid clones spanning 58 kb of the M. xanthus genome. Gene disruption experiments defined a > or = 18 kb contiguous DNA region involved in saframycin biosynthesis. Sequencing of part of this region revealed a large ORF containing two 600-amino-acid domains with similarity to peptide synthetase amino-acid-activating sequences, suggesting that saframycin Mx1 is synthesized by a nonribosomal multienzyme complex, similar to other bioactive peptides.

Mobilization of photosystem II induced by intense red light in the Cyanobacterium Synechococcus sp PCC7942.

We use confocal fluorescence microscopy and fluorescence recovery after photobleaching to show that a specific light signal controls the diffusion of a protein complex in thylakoid membranes of the cyanobacterium Synechococcus sp PCC7942 in vivo. In low light, photosystem II appears completely immobile in the membrane. However, exposure to intense red light triggers rapid diffusion of up to approximately 50% of photosystem II reaction centers. Particularly intense or prolonged red light exposure also leads to the redistribution of photosystem II to specific zones within the thylakoid membranes. The mobilization does not result from photodamage but is triggered by a specific red light signal. We show that mobilization of photosystem II is required for the rapid initiation of recovery from photoinhibition. Thus, intense red light triggers a switch from a static to a dynamic configuration of thylakoid membrane protein complexes, and this facilitates the rapid turnover and repair of the complexes. The localized concentrations of photosystem II seen after red light treatment may correspond to specific zones where the repair cycle is active.
