Hepatic accumulation of glutamic acid branched neogalactosyllipid modified liposomes.

We synthesized branched type galactosyllipid derivatives for liposome modification for the targeting of asialoglycoprotein receptors on the surface of liver cells. Galactose was coupled to the alpha- and gamma-carboxyl groups of glutamic acid via a triethyleneglycol spacer, then this glutamic moiety was bound to the lipid anchor. Ricinus communis agglutinin (RCA120) induced the agglutination of liposomes modified with mono-, bi- and tri-antennary neogalactosyllipid. With the bi- or tri-antennary derivatives, agglutination was observed at fewer galactosyl residues on the liposomes. We examined the effect of the branching structure in vivo. The difference in accumulation of liposomes between non-branched type neogalactosyllipid and branched type neogalactosyllipid was not large. Liver accumulation of liposomes depended on the galactosyl residues. The number of galactosyl residues was more effective for accumulation in the liver than for branching. We studied the effect of asialofetuin preinjection on the hepatic accumulation of neogalactosyllipid modified liposomes. Hepatic accumulation of liposomes was inhibited by preinjection of asialofetuin. The effect of preinjection was almost equal among the ligands. These results show that the saccharide density on the liposome surface seemed to be a more important factor than the branching structure of the ligand for liver targeting.

Azoospermia in bilharziasis and the presence of sperm antibodies.

Sperm agglutinating, haemagglutinating and immobilizing antibodies were detected more frequently and in higher titres in the sera of azoospermic patients with urinogenital bilharziasis than in the sera of unaffected males. There is evidence that bilharziasis enhances the autoimmune response of the individual.

[Seroepidemiologic survey of leptospirosis among environmental sanitation workers in an urban locality in the southern region of Brazil].

Sera from 386 environmental sanitation workers, concerned with water supply, drains and drainage galleries, sewers, garbage collection and road sweepers, were examined for leptospiral agglutinins by the microscopic agglutination test. Altogether 40 of the 386 workers (10.4%) were positive to one or more serovars; however, the difference in seropositivity between the professional categories was not significant (p < 0.05). Twelve serovars were recorded among the seropositive workers with predominance of L. castelonis and L. australis; but the difference between the serovars was not statistically significant (p > 0.05). Of the seropositive workers, 86.9% had agglutination titres > or = 100 and < or = 400; the rates for titres 100 and 400 were higher than 800, 1,600 and 3,200 (p < 0.05).

[Obtaining specific non-adsorbed sera to Haemophilus influenza for the agglutination test on glass].

Type-specific agglutinating sera to capsular antigenas of H. influenzae were recovered (from 1:3200 to 1:25600). With the given immunization scheme there formed mostly IgG immunoglobulins, and only insignificant amount of IgM (separation of G-200 Sephadex gel). IgG were found to be responsible for the type-specificity of the sera to bacteria, whereas the cross reactions within the H. influenzae genus were connected connected with the IgM. Use of the sera heated at various temperature regimens offered a possibility of obtaining highly specific agglutinating sera with the aid of which express serodiagnosis of the isolated H. influenzae strains was possible.

Observations on the serologic changes in the recombinants in Shigella flexneri Serotype 4a.

Studies on the antigenic composition of 84 diagnostic strains of S. flexneri serotype 4a and their Lac+ recombinants from a cross with E. coli HfrC were carried out. Considerable differentiation of agglutination reaction was found both in original material and in recombinants. An interesting group included recombinants which completely lost their capability to agglutinate with sera specific for type and group antigens and preserved only the capability to agglutinate in the polyvalent serum of S. flexneri subgroup. One of them (recombinant 140 Lac+) had been subject to detailed serological analysis and was shown to acquire as a result of recombination a new antigen appearing neither in any flexneri subgroup serotype, nor in dysenteriae and boydii subgroups. This antigen is of thermostable character.

[Intrageneric O-antigenic relations of Providencia bacteria].

The authors present literature and personal data on the intragenera O-antigenic relations of the strains of a collection of bacteria belonging to Providencia genus, including standard strains of 53 serological O-groups whose antigenic interrelations were studied serologically in the tests of agglutination and adsorption of agglutinins. Experimental results confirmed the presence of unilateral relations by the O-antigen between the serological groups 01-021, 03-021 and bilateral relations between the serological groups 01-03; there were also revealed relations by the O-antigens between the serological groups 09-036, 014-023, 018-035, 039-053, and 039-027 of a unilateral character. The data obtained added information to the antigenic structure of Providencia and were of significance for the preparation of diagnostic O-sera of Providencia.
