[Ultrastructural organization of projection zones AI and AIV of the cat auditory cortex to the inferior colliculi].

By the method of anterograde degeneration, distribution of projections of functionally definite AI and AIV zones of the cat auditory cortex have been studied in the posterior calculi of the tectum opticum. It has been demonstrated electron microscopically that an essentially part of the degenerating boutons are found in the middle and caudal thirds of the colliculi, and the remaining ones terminate on the dendrites with large- and middle-size diameters and on the spines. A supposition is made that there exist two systems of the corticofugal auditory fibres. Peculiarities of the degenerative rates, specific for short- and long-axonal systems of the fibres and possible mechanisms for the influence of the auditory cortex on the neurons of the posterior colliculi of the tectum opticum are discussed.

Improvements in hemoglobin AI determination by rapid cation-exchange chromatography.

The accuracy of many convenient methods for determining the proportion of hemoglobin AI to total hemoglobin by ion-exchange chromatography is in question because those methods have not been evaluated against a reference method. In addition, several if not all of these tests are influenced by laboratory temperature. We modified the chromatographic conditions usually used, to arrive at a method that gives results that agree well with those by the Trivelli method (N. Engl. J. Med. 284, 353, 1971) and is less influenced by ambient temperature than are results obtained with the commercially available Helena and Isolab kits. We accomplished this by lowering the pH of the fast-hemoglobin eluting buffer, with a corresponding adjustment in the ionic strength. We also use a larger elution volume, which assures more nearly complete elution of the hemoglobin A1 fraction. The final method (y) agreed being y = 0.98x + 0.63 r2 = 0.887. Average results from the Helena and Isolab methods were lower than with the reference method by 1.58 and 3.03% hemoglobin A1, respectively.

Apolipoprotein AI-derived pulmonary vascular amyloid in aged dogs.

Our studies confirm the common occurrence of a unique form of apolipoprotein AI (apoAI)-derived vascular amyloidosis in dogs that appears to be unrelated to other disease conditions, but is associated with aging. Vascular amyloid deposits were most frequently located within the intima and media of medium-sized pulmonary arteries, and were not confirmed in any other tissues. Pulmonary vascular amyloid immunoreactive with antiserum to purified N-terminal (1-71) canine apoAI amyloid protein was demonstrated retrospectively in 12.8% of necropsied dogs (N = 243) 10 years of age or older. In a subsequent expanded 1-year prospective study of necropsied dogs (N = 231) of all ages, apoAI-derived pulmonary vascular amyloid deposits were demonstrated in 0.7% of dogs < 10 years of age and in 22% of dogs 10 years of age or older. The incidence of this form of amyloid in dogs 10 years of age or older was significantly associated with advancing age (P < 0.00001). However, significant differences regarding gender, breed, or the frequency of selected common disease conditions were not observed when the dogs with apoAI-derived amyloid were compared with control dogs. The possibility that this new form of senile apoAI-derived amyloidosis is a manifestation of an age-associated aberration in apoAI metabolism or is related to a mutant form of apoAI is the subject of ongoing investigations.

Activation of apolipoprotein AI gene transcription by the liver-enriched factor HNF-3.

Liver-specific expression of the apolipoprotein AI (apoA-I) gene is controlled by the coordinate action of transcription factors bound to three sites (A, B, and C) located within a powerful liver-specific enhancer which spans the -222 to -110 region upstream of the apoA-I gene transcription start site (+1). Sites A and C bind various members of the nuclear receptor superfamily including the liver-enriched factor HNF-4. In the current report, enhancer derivatives with mutagenized protein-binding sites were tested for their ability to stimulate the apoA-I basal promoter in hepatoblastoma HepG2 cells. The results revealed that occupation of both sites A and B, but not C is essential for high level expression. Electrophoretic mobility shift assays showed that in HepG2 cells site B is occupied by the liver-enriched factor HNF-3 beta. Binding of HNF-3 beta to site B transactivates the apoA-I basal promoter in hepatic and nonhepatic cells. HNF-3 beta binding and transactivation were dependent upon the close proximity of two HNF-3 beta binding motifs within site B. Furthermore, HNF-3 beta and HNF-4, bound to their cognate sites within the apoA-I enhancer exhibited strong synergy in transactivation of the apoA-I basal promoter in nonhepatic cells, highlighting the central role of HNF-3 beta in liver-specific transcription of the apoA-I gene. It is concluded that cooperative binding of HNF-3 beta to site B and synergistic interactions between HNF-4 and HNF-3 beta bound to their cognate sites in the apoA-I enhancer may play a fundamental role in apoA-I gene expression in liver.

Association of major histocompatibility complex antigens (BoLA-A) with AI bull progeny test results for mastitis, ketosis and fertility in Norwegian cattle.

Altogether 424 Norwegian AI bulls, progeny tested for clinical mastitis, ketosis and fertility (recorded as nonreturn percentage), were typed by Edinburgh and Oslo allo-antisera to bovine lymphocyte antigens (BoLA-A) over a 7-year period. Significant effects of BoLA-A on disease were revealed. A2 was associated with relative resistance to mastitis, a positive influence on fertility, and a possible relative resistance to ketosis, while A13 was associated with relative resistance to ketosis. The previously reported associations of A11 and w16 with relative susceptibility to mastitis were not confirmed in the present material.

Transcriptional repression of apolipoprotein AI gene expression by orphan receptor ARP-1.

Expression of the apolipoprotein AI (apoAI) gene in the liver is controlled by a liver-specific enhancer. The function of this enhancer depends on synergistic interactions between transcription factors bound to at least three sites (designated A, B, and C) located within this enhancer. We have previously shown that an apoAI gene reporter construct containing the entire enhancer is expressed efficiently in a hepatoma cell line and that its activity is repressed by the orphan receptor ARP-1. Moreover, repression by ARP-1 is overcome by the retinoid X receptor RXR alpha in the presence of retinoic acid. In this study, we show that ARP-1 represses the apoAI promoter by binding to site A of the apoAI liver-specific enhancer, the repression being a promoter context-specific event. Mapping analysis of ARP-1 indicated that its DNA binding domain is essential but not sufficient for repression. Two separate repression domains located at the amino- and carboxyl-terminal halves of ARP-1 were found to individually complement the DNA binding domain for efficient repression. We also demonstrate the reversibility of ARP-1 repression by transcription factors C/EBP and Egr-1, which might also be involved in apoAI gene expression. Significantly, repression by ARP-1 was found to be a prerequisite for C/EBP-mediated transactivation. We interpret our results in terms of a model in which ARP-1 repression via its interaction with site A is an obligatory intermediate step in switching from one activated state of the apoAI gene to another.

Familial nephropathic systemic amyloidosis caused by apolipoprotein AI variant Arg26.

A point mutation in the apolipoprotein AI (apoAI) gene causing autosomal dominant non-neuropathic systemic amyloidosis is described in a previously unreported Canadian family of British origin with five affected individuals in three generations. Amyloid deposits in the renal biopsy from the proband, a 31-year-old female presenting with hypertension and renal failure, stained immunospecifically with antiserum to apoAI. The plasma of all family members with amyloidosis contained both wild-type apoAI and a variant bearing one additional positive charge. Sequencing of the apoAI gene demonstrated that the proband was a heterozygote for a single base substitution in exon 3, changing codon 26 from GGC(Gly) to CGC(Arg). Concordance of the mutant allele with the presence of variant plasma apoAI and clinical features of amyloidosis was demonstrated. This is the third family in which this amyloidotic mutation has been described, but the distribution of amyloid deposits and their clinical effects are clearly determined by other genetic and/or environmental factors.

Coronary artery disease risk predicted by plasma concentrations of high-density lipoprotein cholesterol, apolipoprotein AI, apolipoprotein B, and lipoprotein(a) in a general Chinese population.

We measured lipid and lipoprotein concentrations in blood samples from control subjects and patients with coronary artery disease (CAD) in Taiwan. We found significant differences (P < 0.01) in the concentrations of high-density lipoprotein cholesterol (HDLC), apolipoprotein AI (ApoAI), apolipoprotein B (ApoB), and lipoprotein(a) [Lp(a)]. Concentrations of HDLC < 350 mg/L, ApoAI < 900 mg/L, ApoB > 800 mg/L, and Lp(a) > 200 mg/L occurred, respectively, 2.8, 5.2, 1.7, and 2.3 times more frequently in the patients than in the control group. If one considers HDLC at < 350 mg/L, ApoAI at < 900 mg/L, ApoB at > 800 mg/L, and Lp(a) at > 200 mg/L as separate risk factors for CAD, the ratio of individual patients to control subjects having 4, 3, 2, 1, or 0 risk factors was [symbol: see text] 9.4, 2.1, 0.2, 0.2, respectively. Individuals displaying three or more risk factors were found 15 times more frequently in the CAD group than in the control group. These risk factors may be used clinically for the prediction and prevention of CAD in the general population.

[DNA polymorphisms of the apolipoprotein AI-CIII-AIV gene cluster in coronary artery disease].

Two polymorphic sites of the apolipoprotein AI (ApoAI) gene, PstI and SstI, were examined in a sample of 43 unrelated Chinese patients with coronary artery disease (CAD) and 60 unrelated healthy individuals. Allele frequencies were compared between patients and controls and their impact on lipid and oxygen free radicals metabolism were also studied. The frequencies of the uncommon P2 and S2 alleles in CAD cases were found as 0.14 and 0.22 respectively, which were significantly higher than 0.05 and 0.13 in controls. The more the number of vessel disease in CAD cases, the higher the frequencies of S2 alleles which were the highest in the group of 3 vessel diseases. Some serum lipid and oxygen free radicals levels differed between CAD cases of different genotypes. These data suggest that genetic variation in P2 and S2 alleles of APOAI gene may contribute to the development and extent of CAD in Chinese subjects.

Assessing an AI knowledge-base for asymptomatic liver diseases.

Discovering not yet seen knowledge from clinical data is of importance in the field of asymptomatic liver diseases. Avoidance of liver biopsy which is used as the ultimate confirmation of diagnosis by making the decision based on relevant laboratory findings only, would be considered an essential support. The system based on Quinlan's ID3 algorithm was simple and efficient in extracting the sought knowledge. Basic principles of applying the AI systems are therefore described and complemented with medical evaluation. Some of the diagnostic rules were found to be useful as decision algorithms i.e. they could be directly applied in clinical work and made a part of the knowledge-base of the Liver Guide, an automated decision support system.

From prediction to mechanism: Explainable AI uncovers plasma and CSF proteomic signatures of Alzheimer's disease.

Alzheimer's disease (AD) plasma and cerebrospinal fluid (CSF) proteomics can distinguish AD from cognitively normal controls, but the generalizability of machine learning performance and the recurrence of biological signals across datasets require cautious interpretation. We developed an explainable artificial intelligence framework spanning two fluids and four ADNI proteomic datasets, covering 2082 modality specific samples, all analysed internally within ADNI. Phase 1 analysed plasma using a 119 analyte NULISA and targeted UPENN panel (n = 727; 216 CE, 511 controls). Phase 2 extended the analysis to CSF using SOMAscan7k, TMT-MS and targeted SET2, with Elecsys Aβ42, Aβ40, total tau and p-tau181 as anchor biomarkers. Only SOMAscan was subject-independent relative to Phase 1 plasma; TMT-MS and SET2 overlapped with Phase 1 for 96.0% and 97.7% of subjects and therefore are not independent replication cohorts. Under subject-level splits with fold internal preprocessing, we compared Elastic Net, Explainable Boosting Machines and gradient boosted trees with SHAP-based explanations. Among the candidate pipelines, we selected the pipeline with the highest held-out test ROC AUC for each platform; the selected values were 0.927 in plasma and 0.954-0.973 across the three CSF datasets. Because the same held out test performance was used for pipeline selection and headline reporting, these are optimistically selected single-holdout estimates, not unbiased estimates of generalizable or clinical performance. Explanations identified five recurring biological axes within ADNI: cholinergic (ACHE), tau/14-3-3 (YWHAG, YWHAZ, YWHAB, YWHAE), neuro-axonal (NEFL, NEFH), microglial/complement (CHIT1, SMOC1, CHI3L1, C7, CFH) and synaptic (NPTXR, NPTX2, DLG4, SYT5, VSNL1, ELAVL2). CSF analyses showed synaptic vesicle-cycle enrichment (q = 2 × 10-6), and CSF YWHAG correlated strongly with total tau (ρ = 0.87). Cross-fluid directional concordance was modest overall (54-57%) but increased to 73-80% among mapped analyte/protein rows reaching q < 0.05 in CSF. These findings provide hypothesis-generating, internally supported evidence within ADNI. Independent external cohorts with locked pipelines are required to evaluate generalizable performance and biological reproducibility; the overlapping TMT-MS and SET2 analyses should not be interpreted as independent replication.

Correlates of cigarette smoking among selected Southwest and Northern plains tribal groups: the AI-SUPERPFP Study.

OBJECTIVES: We describe the prevalence and correlates of cigarette smoking in 2 American Indian tribal groups. METHODS: We performed multinomial logistic regression on epidemiological data from a population-based, cross-sectional study of Southwest and Northern Plains American Indians aged 15 to 54 years. RESULTS: We found that 19% of Southwest men, 10% of Southwest women, 49% of Northern Plains men, and 51% of Northern Plains women were current smokers. Male gender and younger age were associated with higher odds of smoking in the Southwest tribe, whereas current or former marriage and having spent less time on a reservation were associated with higher odds of smoking in the Northern Plains population. Alcohol consumption was strongly associated with higher odds of smoking in both groups. CONCLUSIONS: Cigarette smoking is a major public health concern among American Indians. Because correlates and smoking patterns vary among different tribal groups, each group's unique characteristics should be considered when designing and implementing comprehensive, culturally appropriate interventions in American Indian communities.

Malignant neoplasms in cyclosporin A-induced autoimmunity (CyA-AI).

Lethally X-irradiated LEW rats reconstituted with syngeneic bone marrow and given low-dose Cyclosporine A (CyA) for 5 weeks develop, after withdrawal of CyA, symptoms of disease resembling graft-vs.-host disease (GVHD) as seen after allogeneic bone-marrow transplantation. Symptoms of disease may include acute dermatitis and chronic disease resembling scleroderma. Since anti-class II MHC cytotoxic lymphocytes are generated in this model, it has been proposed as an anti-tumor regimen in humans. We now report that LEW rats treated according to this protocol may, after cessation of CyA administration, paradoxically develop malignant neoplasms. Of 48 experimental animals, 31 developed rapidly progressive subcutaneous and/or intracutaneous tumors commencing at 6 weeks, 13 weeks and 6 months after cessation of CyA. Tumors were of mesenchymal origin, usually high-grade sarcoma, adenocarcinoma or both mesenchymal and epithelial tumors. Such tumor incidence exceeded the incidence of tumor growth in X-irradiated controls, and in rats subjected to thymectomy prior to X-irradiation and CyA administration. CyA by itself induced no tumors. Our results show that total body X-irradiation is required for tumor development but that the presence of CyA-induced autoimmune disease increases the incidence significantly.

Reactivity of various four-coordinate aluminum alkyls towards dioxygen: evidence for spatial requirements in the insertion of an oxygen molecule into the AI-C bond

The interaction of dioxygen with various tetrahedral aluminum alkyls, (tBu)3Al.OEt2 (1), tBu2Al(mu-OtBu)2AltBu2 (6), (tBu)2Al(mesal) (2) [mesal=methyl salicylate anion], R2Al(mu-pz)2AlR2 [pz=deprotonated pyrazole, R= Me (3a), Et (3b), and tBu (3c)], R2Al(mu-3,5-Me2pz)2AIR2[3,5-Me2pz = deprotonated 3,5-dimethylpyrazole, R= Me (4a), and Et (4b)], and Et2B(mu-pz)2AlEt2 (5), has been investigated. We were particularly interested in the effect of steric hindrances both caused by the metal-bonded substituents and those that result from the nature of the bifunctional ligand used in the oxygenation reaction. In the reaction of 1 with O2, only the formation of the monoalkoxide compound6 was observed. The latter di-tert-butyl compound as well as all planar aluminapyrazoles, that is, the tert-butyl derivative 3c and lower alkylaluminum derivatives with the more demanding 3,5-dimethylpyrazoyl ligands 4a and 4b, are stable under an atmosphere of dry oxygen and ambient conditions. Inspection of the space-filling representation of these compounds has undoubtedly shown that the bulky tert-butyl groups or pyrazolyles ligands, respectively, provide steric protection for the metal center from the dioxygen attack. In contrast, the dialkylaluminum derivatives of pyrazole, 3a and 3b, and the diethylaluminum bis(1-pyrazolyl)borate complex 5, all with the metal center eclipsed with respect to the plane defined by the four nitrogen atoms, react smoothly with O2 to form the alkyl(alkoxy)aluminum complexes. In the reaction of 5 with O2 for example, the Et-B bonds remained intact, and the dimeric five-coordinate compound [Et2B(mu-pz)2 Al(mu-OEt)Et]2 (9) was isolated in good yield. The interaction of mononuclear di-tert-butyl chelate complex 2 with O2 at -15 degrees C gives (tBuOO)(tBuO)Al(mu-OtBu)2Al(mesal)2 (7) in high yield, and the presence of the alkylperoxo moiety is a particularly significant point in the resulting product. All the compounds have been characterized spectroscopically, and the structures of 3c, 4a, 6, 7, and 9 have been confirmed by X-ray crystallography. Structural features of 1-6 are discussed and are considered in relation to the possible approach pathways of the O2 molecule to the four-coordinate metal center. This analysis and the observed apparent dissimilarity in the reactions of model four-coordinate aluminum alkyls with O2 clearly show that the stereoelectronic prerequisites are responsible for the fundamentally different reactivity.

Relative Lewis basicities of six AI(ORF)4- superweak anions and the structures of LiA.

The relative Lewis basicities of six Al(ORF)4- ions, Al[OC(CH3)(CF3)2]4-, Al(OC(CF3)3]4-, Al(OCPh(CF3)2]4-, Al[OC[4-C6H4(tBu)](CF3)2]4-, Al(OC(Cy)(CF3)2]4-, and Al(OCPh2(CF3)]4-, have been determined by measuring their relative coordinating abilities towards Li+ in dichloromethane. The relative Li- Lewis basicities of the Al(ORF)4- ions are linearly related to the aqueous pKa values of the corresponding parent HORF fluoroalcohols. The Lewis basicity of Al[OCH(CF3)2]4- could not be measured because two of these anions can coordinate to one Li+ cation. The structures of LiAl[OCH(CF3)2]4 and [1-Et-3-Me-1,3-C3H3N2][Li[Al[OCH(CF3)2)4]2] were determined.
