In order to obtain information about the mechanism of dyskeratosis in Darier's disease, we have examined skin sections at different steps of differentiation at light and electron microscopic level in three patients. Corps ronds occurred at the basal aspect of spinous layer, while grains were abundant in the bulla and in the granular layer of the peripheral epidermis. Corps ronds contained cytoplasmic vacuoles and great number of keratinosomes. Their cytoplasm was divided by a circular ring of tonofilaments into a juxtanuclear and a peripheral compartment. More superficially corps ronds became dyskeratotic keratohyalin-containing cells by acquiring increasing amounts of keratohyalin granules; the cytoplasm turned more vacuolated and poor in keratinosomes. Grains exhibited nuclear pyknosis, shrinkage and densification of mitochondria, numerous tonofilament bundles and confluent vacuoles after disruption of their membrane. In these cells, keratohyalin granules appeared less electrondense and keratinosomes were rare or absent. Horny cells contained empty areas resulting from the persistence of vacuolar spaces. These observations suggest that corps ronds and grains represent stages in the same successive differentiation line and that grains give rise to the changed horny cells occurring in this condition.

We report a case of a neoplastic-like proliferation of epidermal cells in the limbs of an adult newt. The undifferentiated cell mass infiltrated the mesodermal tissues. Possible effects of this event on limb regeneration are discussed.

A comparative ultrastructural study study of the stage and type of differentiation of cancer cells in the primary large intestine human adenocarcinoma and its metastases in the lymph nodes, ovaries, and greater omentum is described. Metastatic cells have been found to retain the features of specific differentiation and be represented by the same groups and main ultrastructural types as in the primary tumor. The main directions of cell differentiation in metastases and in the primary tumor were similar. The role of microenvironment of different organs appeared to be insignificant in the process of cell differentiation in metastases.
A ribosomal mutant Bacillus subtilis IG1 resistant to 100 mkg/ml of streptomycin was isolated. The strA mutation is cotransduced with the cysA gene and, consequently, maps in the ribosomal cluster. The mutation does not influence cell division but does reduce a level of sporulation as well as its antibiotic and proteolytic activity. Involvement of ribosomes in the control of sporulation and secondary metabolism of spore forming bacteria is discussed.
For the determination of whether two or more stem lines with fixed of stable differentiation are present in a growing mixed adenosquamous cell carcinoma, two different mixed tumors, derived from transformed F344 rat tracheal epithelium, were dissociated and ten single cells were isolated from each tumor. Each of these single cells was allowed to grow 27 population doublings to form a clonal population. All clones were then inoculated into animals for the assessment of in vivo differentiation. All cell inocula produced carcinomas within 7-70 days. Of the ten single cell clones isolated from the first tumor, seven clones produced mixed adenosquamous carcinomas and three produced adenocarcinomas in which no squamous cell component was found. From the second tumor, eight clones produced mixed tumors and two produced squamous cell carcinomas in which no adenocarcinoma component was found. The mixed tumors derived form clones contained widely varying proportions of adeno and squamous components. Recloning of the cloned lines yielded populations that produced mixed adenosquamous cell carcinomas upon inoculation in vivo. Comparison of tumor phenotypes from clones of early and late passages further confirmed the instability of differentiation in these neoplastic epithelial cells. These studies strongly suggest that the mixture of differentiated cell types in adenosquamous cell carcinomas is not the result of a mixture of stem cells with one fixed phenotype within the tumor, but rather is the result of an instability of differentiation; the neoplastic cells can express both types of differentiation.
Acute lymphoblastic leukemia (ALL) is a heterogeneous entity that can be subcategorized on the basis of morphologic aspects, immunologic markers, membrane antigens, and enzymatic content of the blast forms. Subgroups corresponding to T-cell, pre-T-cell, pre-B-cell, and non-T-, non-B-lymphocyte derivations can be defined by immunologic markers, and the biologic differences are supported by the differential expression of membrane antigens and of nuclear, lysosomal, cytoplasmic, and membrane enzymes. These studies ease the diagnosis and characterization of ALL and enhance understanding of this disease.
