Comparative sorption and desorption of benzo[a]pyrene and 3,4,3',4'-tetrachlorobiphenyl in natural lake water containing dissolved organic matter.

The sorption and desorption of two model compounds, benzo[a]pyrene (BaP) and 3,4,3',4'-tetrachlorobiphenyl (TCBP), were studied in natural lake water with high dissolved organic matter (DOM) content using the equilibrium dialysis and Tenax extraction methods. The sorption of TCBP was lower and reached steady value more slowly than did BaP. Tenax extraction revealed at least two differently desorbing fractions for both model compounds, which also supported the conclusion that DOM-HOC associations may involve several mechanisms. The rapidly desorbing fraction may be attributed to freely dissolved and loosely sorbed compound, whereas the more strongly sorbed fraction may indicate the presence of specific binding sites. The data indicated that the association between hydrophobic organic contaminants (HOC) and DOM is not simply absorption that is solely driven by the lipophilicity of the sorbates. Although contact time had a rather negligible effect on the sorption of BaP, the proportion of desorption resistant fraction increased with time, whereas the desorption of TCBP was less affected by contact time. Steric factors may be the cause of the lower sorption and smaller desorption resistant fraction of TCBP. The results indicate potential differences in the behavior of PAHs and PCBs in the aquatic environment.

Role of Fe(III), phosphate, dissolved organic matter, and nitrate during the photodegradation of domoic acid in the marine environment.

The photodegradation of domoic acid in model seawater containing varying amounts of total Fe(lll) (expressed as Fe(lll)), NO(3-), total phosphate (expressed as P04(3-)), and dissolved organic matter (DOM) is reported. A multivariate, microscale, high-throughput experimental approach is described for evaluating how these components interact to control the removal of domoic acid from natural waters. Under the nominal conditions of the study ([Fe(lll)]o 0-4 microM; [N03-]o 0-35 microM; [P04(3-)]o 0-4 microM; [DOM]o 0-10 mg/ L), it is apparent that Fe(lll) and DOM are significant promoters of domoic acid photooxidation. In contrast, P04(3-) interacts with Fe(lll) to inhibit the photooxidation of domoic acid, but P04(3-) alone does not act to slow or accelerate domoic acid photodegradation. No other variables (singly or interactively) have a statistically significant impact. At an incident light intensity of 765 W/m2 and initial domoic acid concentration of 0.96 microM, domoic acid half-lives range over 12-36 h, with half-life a function of [Fe(III)], [PO4(3-)], and dissolved organic matter loadings. An NMR based technique for measuring domoic acid-Fe(lll) binding (1.72 x 10(11)) is reported.

A chemical extraction method for mimicking bioavailability of polycyclic aromatic hydrocarbons to wheat grown in soils containing various amounts of organic matter.

In this study, we have evaluated the extent to which organic matter contents in soils influence the accumulation of PAHs by the roots of wheat plants and have developed a rapid chemical method for determining the bioavailability of PAH. Four polycyclic aromatic hydrocarbons (PAHs), naphthalene, acenaphthylene, fluorene, and phenanthrene, were added to natural soil samples with different amounts of organic matterfor pot experiments to evaluate apparent bioavailability of PAHs to wheat roots (Triticum aestivum L.). The extractabilities of PAHs in the soil were tested by a sequential extraction scheme using accelerated solvent extraction with water, n-hexane, and a mixture of dichloromethane and acetone as solvents. The water or n-hexane-extractable PAHs were positively correlated to dissolved organic matter (DOM) and negatively correlated to total organic matter (TOM), indicating mobilization and immobilization effects of DOM and TOM on soil PAHs, respectively. The apparent accumulation of PAHs by wheat roots was also positively and negatively correlated to DOM and TOM, respectively. As a result, there are positive correlations between the amounts of PAHs extracted by water or n-hexane and the quantities accumulated in plant roots, suggesting the feasibility of using water- or n-hexanes-extractable fractions as indicators of PAH availability to plants.

Aqueous oxidation of phenylurea herbicides by triplet aromatic ketones.

Excited triplet states of dissolved natural organic matter (DOM) are important players for the transformation of organic chemical contaminants in sunlit natural waters. The present study focuses on kinetics and mechanistic aspects of the transformation of phenylurea herbicides induced by well-defined excited triplet states, which have been chosen to model DOM triplet states having oxidative character. The aromatic ketones benzophenone, 3'-methoxyacetophenone, and 2-acetonaphthone were used to photogenerate their triplet states and oxidize a series of eleven substituted phenylureas. Quenching of the excited triplet states by the phenylureas was measured using laser flash photolysis in the microsecond time domain, while the oxidation kinetics of the phenylureas was followed under steady-state irradiation. Second-order rate constants for quenching and oxidation were largely identical for a given pair of ketone and phenylurea. They reached the diffusion-controlled limit (approximately 4 x 10(9) M(-1) s(-1)) and decreased with increasing free energy of electron transfer from the phenylurea to the ketone triplet. These results confirm those already obtained using phenols as the substrates to be oxidized and suggest that oxidation rates are mainly determined by the bimolecular rate constant for electron transfer, a rule that can possibly be extended to various organic contaminants. A refined estimate of the effective reduction potential of DOM excited triplet states was also obtained.

Photoinduced transformation of gamma-HCH in the presence of dissolved organic matter and enhanced photoreactive activity of humate-coated alpha-Fe2O3.

This study examined phototransformation of gamma-hexachlorocyclohexane (gamma-HCH) in different solutions. The presence of dissolved organic matter (DOM) inhibited the phototransformation of gamma-HCH. This phenomenon could be correlated to the binding interaction between gamma-HCH and DOM. Alpha-Fe2O3 promoted the transformation of gamma-HCH. The humate-coated alpha-Fe2O3 revealed a slight, however significant, favorable effect compared to the bare one. Fourier transform infrared spectroscopy (FTIR) offered the direct evidence that humate-coated alpha-Fe2O3 could form surface Fe(III)-carboxylate complexes by ligand exchange. Additional experiments demonstrated that the photocorrosion of alpha-Fe2O3 coated by DOM was much more acute than that of the bare one. These combined results suggested that the transformation of gamma-HCH on humate-coated alpha-Fe2O3 is more related to a surface complex and not to a semiconductor-assisted photoreaction. In the humate-coated alpha-Fe2O3, absorption of a photon results in an excited ligand-to-metal charge-transfer state of the complexes, and a rich variety of free radical reactions ensue, which is concurrently accompanied by the dissolution of the iron oxide. Such reactions may generate reactive transients such as superoxide and hydroxyl radicals, which may be expected to transform gamma-HCH.

Low-molecular-weight organic acids and hormone-like activity of dissolved organic matter in two forest soils in N Italy.

Concentrations of aliphatic acids, phenolic acid, and inorganic nutrients, as well as hormone-like activity, were determined in soil dissolved organic matter (DOM) from two forest sites in N Italy showing differing degrees of silver fir regeneration. In the site where silver fir recruitment was abundant, humification processes prevailed, and the soil DOM had a high content in aliphatic and phenolic acids. This enhanced the hormone-like activity in the soil, which could in turn promote growth of silver fir seedlings. In the site with poor fir recruitment, the soil DOM underwent rapid mineralization and was richer in inorganic nutrients, but had lower concentrations of aliphatic and phenolic acids, and lower hormone-like activity.

Delayed-onset muscle soreness does not reflect the magnitude of eccentric exercise-induced muscle damage.

This study investigated the relationship between delayed-onset muscle soreness and other indicators of muscle damage following eccentric exercise. Male students (n = 110) performed 12 (12ECC), 24 (24ECC), or 60 maximal eccentric actions of the elbow flexors (60ECC). Maximal isometric force, relaxed and flexed elbow joint angles, upper arm circumference, and plasma creatine kinase activity were assessed immediately before and after, and for 4 days after exercise. Muscle soreness (SOR) was evaluated by a visual analog scale (a 50-mm line, 0: no pain, 50: extremely painful) when the elbow flexors were palpated (SOR-Pal), flexed (SOR-Flx) and stretched (SOR-Ext). Although 24ECC and 60ECC resulted in significantly (P <; 0.05) larger changes in all indicators and slower recovery compared to 12ECC, no significant differences were evident for SOR-Pal and SOR-Flx between 12ECC and 24ECC, or 12ECC and 60ECC. In contrast, SOR-Ext was significantly (P <; 0.05) lower for 12ECC compared to 24ECC and 60ECC. A Pearson product-moment correlation showed SOR-Pal did not correlate significantly with any indicators, however, SOR-Ext and SOR-Flx showed weak (r <; 0.32) but significant (P <; 0.05) correlations with other indicators. Because of generally poor correlations between DOMS and other indicators, we conclude that use of DOMS is a poor reflector of eccentric exercise-induced muscle damage and inflammation, and changes in indirect markers of muscle damage and inflammation are not necessarily accompanied with DOMS.

Genome-wide scan for loci of Asperger syndrome.

Asperger syndrome (AS), characterised by inadequate social interaction, lack of empathy and a dependence of routines and rituals, is classified as belonging to the autism spectrum disorders (DSM-IV and ICD-10). Although the prevalence of AS has been estimated to range from 0.3 up to 48.4 per 10 000, the phenotype still remains relatively unrecognised by clinicians. Several reports, including the original description by Hans Asperger (1944), have suggested that AS has a strong genetic component. Here, we have performed a genome-wide scan on Finnish families ascertained for AS with a strictly defined phenotype. In the initial scan, Z(max)>1.5 was observed on nine chromosomal regions, 1q21-22, 3p14-24, 3q25-27, 4p14, 4q32, 6p25, 6q16, 13q31-33 and 18p11. In the fine mapping stage, the highest two-point LOD scores were observed on chromosomes 1q21-22 (D1S484, Z(max dom)=3.58), 3p14-24 (D3S2432, Z(max dom)=2.50) and 13q31-33 (D13S793, Z(max dom)=1.59). The loci on 1q21-22 and 3p14-24 overlap with previously published autism susceptibility loci, and the loci on 1q21-22 and 13q31-33 overlap with the reported schizophrenia susceptibility loci. The present study is the first genome-wide screen in AS and therefore replication data sets are needed to evaluate further the significance of the AS-loci identified here.

The importance of having your SOX on: role of SOX10 in the development of neural crest-derived melanocytes and glia.

SOX10 is a member of the high-mobility group-domain SOX family of transcription factors, which are ubiquitously found in the animal kingdom. Disruption of neural crest development in the Dominant megacolon (Dom) mice is associated with a Sox10 mutation. Mutations in human Sox10 gene have also been linked with the occurrence of neurocristopathies in the Waardenburg-Shah syndrome type IV (WS-IV), for which the Sox10(Dom) mice serve as a murine model. The neural crest disorders in the Sox10(Dom) mice and WS-IV patients consist of hypopigmentation, cochlear neurosensory deafness, and enteric aganglionosis. Consistent with these observations, a critical role for SOX10 in the proper differentiation of neural crest-derived melanocytes and glia has been demonstrated. Emerging data also show an important role for SOX10 in promoting the survival of neural crest precursor cells prior to lineage commitment. Several genes whose regulation is dependent on SOX10 function have been identified in the peripheral nervous system and in melanocytes, helping to begin the identification of the multiple pathways that appear to be modulated by SOX10 activity. In this review, we will discuss the biological relevance of these target genes to neural crest development and the properties of Sox10 as a transcription factor.

Catecholamines and pituitary function. VII: Effects of acute and chronic dopamine-receptor blockade on pituitary response to TRH-GNRH in normal women and in patients with hyperprolactinemic amenorrhea.

To investigate whether an enhanced dopamine (DA) inhibition on pituitary thyrotrophs and gonadotrophs may account for the abnormal TSH and LH dynamics in pathological hyperprolactinemia, we examined the effect of an acute lysis of the putative DA overinhibition, as obtained with continuous domperidone (DOM) infusion, on both basal and TRH-GnRH stimulated PRL, TSH and LH release in both normal cycling women and patients with pathological hyperprolactinemia. The effect of TRH-GnRH administration was also examined in women with DA-antagonist induced hyperprolactinemia, in order to evaluate the effect of a chronic lack of the physiological DA inhibition on pituitary hormone dynamics. Patients with both pathological and DA-antagonist induced hyperprolactinemia displayed an evident TSH and LH hyper-responsiveness to TRH-GnRH. The PRL response was reduced in the former but enhanced in the latter group. Domperidone infusion resulted in a marked increase in serum PRL levels in normal cycling women, but not in patients with pathological hyperprolactinemia. The abolition of the putative DA-overinhibition at the pituitary level with DOM infusion in patients with pathological hyperprolactinemia was followed by a slight increase in basal TSH output but did not modify the TSH and LH hyperresponsiveness to TRH-GnRH. The similarities in TSH and LH dynamics between patients with pathological and DA-antagonist induced hyperprolactinemia and the ineffectiveness of DOM infusion in modifying the TSH and LH hyper-responses to TRH-GnRH in the former group, seem to exclude the widely accepted idea that endogenous DA overactivity is responsible for the abnormal thyrotroph and lactotroph dynamics in women with hyperprolactinemic amenorrhea.

Excretion of 3-methylhistidine and hydroxyproline following acute weight-training exercise.

Changes in urine excretion of 3-methylhistidine (3MH) and hydroxyproline (OHP) were investigated following a single session of weight training that produced delayed onset muscle soreness (DOMS). The subjects (n = 18) maintained meat-free diets for 6 days and collected 24-h urine volume on day 4 (PRE), day 5 (POST1), and day 6 (POST2) for analysis of 3MH, OHP, and creatinine (CRE). At the end of PRE, nine subjects (EX) performed intensive weight-training exercises. The remaining subjects (NE) served as a nonexercised group. All subjects were measured for fat free weight (FFW) and rated DOMS at the end of each urine collection period, i.e., PRE, POST1, and POST2. The results were significant DOMS ratings on POST1 and POST2 compared with PRE2 for EX only. Excretion of 3MH (x +/- SE in mumol/day) by group were, for EX, PRE 271 +/- 18, POST1 243 +/- 16, POST2 259 +/- 23; for NE, PRE 203 +/- 23, POST1 209 +/- 27, POST2 225 +/- 32. Excretion of OHP (x +/- SE in mg/day) were, for EX, PRE 30.0 +/- 5.4, POST1 32.8 +/- 6.4, POST2 29.6 +/- 6.1; for NE, PRE 22.5 +/- 5.2, POST1 21.6 +/- 6.0, POST2 22.7 +/- 6.0. Changes in urine 3MH and OHP were not statistically significant, whether analyzed in absolute terms or relative to CRE and FFW. It was concluded that the acute session of weight training which produced muscle soreness did not effect excretion of 3MH or OHP.

Differential white cell count after two bouts of downhill running.

The purpose of this study was to compare blood markers associated with eccentrically biased exercise and muscle damage, after two bouts of downhill running. Nine active, untrained males performed 2 x 45 min bouts of downhill running (-0.16 radians), at a speed that would elicit 70% of each subjects VO2max, on a level grade; runs were spaced 14d apart (RUN1, RUN2). Blood samples were obtained before, after, and every hour for 12 h after exercise, as well as every 24 h for 5 d, to assess numbers of circulating neutrophils, monocytes, and lymphocytes, serum cortisol, creatine kinase (CK); subjective sensations of delayed onset muscle soreness (DOMS) were monitored. To control for diurnal variation, two weeks prior to the RUN1, subjects had blood draws performed at the same time as would occur after exercise, but did no exercise (CONTROL). During the 5 d after exercise, DOMS and CK were significantly greater (p < 0.05) after RUN1 compared to RUN2 and CONTROL. During the 12 h after RUN1 and RUN2, neutrophils showed similar responses compared to CONTROL. However, neutrophils were significantly elevated at 96 h after RUN1 and 24 h after RUN2. Monocytes were significantly elevated during 5-11 h after RUN1 and RUN2, compared to CONTROL. Cortisol showed a similar significant diurnal decrease for all three conditions during the 12 h following exercise. The significantly lower levels of CK and DOMS seen after RUN2, compared with the initial run is consistent with the literature. The similar changes in neutrophils and monocytes during the 12 h following RUN1 and RUN2, followed by disparate responses over the subsequent 5 d, requires further investigation.

Effects of alkali treatment of wheat straw on intake and microbial protein synthesis in cattle.

Effects of sodium hydroxide treatment on intake and digestion of straw organic matter (OM), and microbial protein synthesis were examined in comparison with untreated and treated straws. Intakes (kg dry matter (DM)/d) of untreated, NaOH- and sodium carbonate-treated straws were 5.21, 7.04 and 6.48 respectively and intakes (kg/d) of digestible organic matter (DOM) for the respective diets were 2.57, 3.73 and 2.98 respectively. NaOH-treated straw ranked higher than untreated straw in intakes of DM and DOM (P less than 0.01). The proportion of DOM intake that was apparently digested in the rumen was 0.74, 0.77 and 0.66 on untreated, NaOH- and Na2CO3-treated straws respectively. Although the amount of bacterial N leaving the abomasum was higher with NaOH-treated than with untreated straw (74.6 v. 56.8 g/d, P less than 0.01), efficiencies of bacterial N synthesis were not different between these treatments (28.6 v. 31.5 g bacterial N/kg OM apparently digested, P greater than 0.05). The live-weight gain which the NaOH-treated straw could support was predicted to be 350 g/d and it was concluded that a protein supplement would have provided further increase in production in diets based on NaOH-treated straw.

The effects of static stretching and warm-up on prevention of delayed-onset muscle soreness.

It has been suggested in the lay literature that static stretching and/or warm-up will prevent the occurrence of Delayed-Onset Muscle Soreness (DOMS). The primary purpose of this study was to determine the effects of static stretching and/or warm-up on the level of pain associated with DOMS. Sixty-two healthy male and female volunteers were randomly assigned to four groups: (a) subjects who statically stretched the quadriceps muscle group before a step, (b) subjects who only performed a stepping warm-up, (c) subjects who both stretched and performed a stepping warm-up prior to a step test, and (d) subjects who only performed a step test. The step test (Asmussen, 1956) required subjects to do concentric work with their right leg and eccentric work with their left leg to voluntary exhaustion. Subjects rated their muscle soreness on a ratio scale from zero to six at 24-hour intervals for 5 days following the step test. A 4x2x2 ANOVA with repeated measures on legs and Duncan's New Multiple Range post-hoc test found no difference in peak muscle soreness among the groups doing the step test or for gender (p greater than .05). There was the expected significant difference in peak muscle soreness between eccentrically and concentrically worked legs, with the eccentrically worked leg experiencing greater muscle soreness. We concluded that static stretching and/or warm-up does not prevent DOMS resulting from exhaustive exercise.

The characteristics of residual motion perception in the hemifield contralateral to lateral occipital lesions in humans.

Unilateral damage to the lateral occipital region in humans can give rise to impaired motion perception in the contralateral visual field [Plant et al. (1993), Brain, 116, 1303-1335]. We report the following characteristics of the residual vision. (i) Spatial acuity and spatial frequency discrimination are not affected. (ii) Contrast thresholds for direction-of-motion (DOM) discrimination of luminance modulated (LMod) sine-wave gratings is unaffected regardless of drift temporal frequency and the effect of spatial and temporal frequency on drifting/counterphase sensitivity ratios is normal (providing further evidence that cortical directionally selective mechanisms are intact). (iii) Contrast thresholds for DOM discrimination of contrast modulated (CMod) gratings are elevated by a log unit across a range of drift velocities. (iv) The residual motion perception shows neither a directional nor a naso-temporal asymmetry. (v) Weber fractions for velocity discrimination are shown in one patient in whom this measurement was carried out, to be elevated by around a factor of three but the functions relating velocity discrimination to stimulus contrast and to the velocity of the standard are parallel in the affected and unaffected hemifields. (vi) Weber fractions for temporal frequency discrimination using counterphase modulated gratings are also elevated. We conclude that the degraded motion perception is mediated by mechanisms which have similar contrast and temporal properties to those subserving normal motion perception. Mechanisms subserving DOM discrimination of LMod gratings may be spared because they are more widely distributed in extra-striate cortex than mechanisms subserving non-Fourier (second-order) motion perception or velocity discrimination. The anomaly resembles that described in some recent animal studies of impaired motion perception after extra-striate cortical damage.

Peripheral opioid analgesia in experimental human pain models.

This placebo-controlled, double-blind crossover study assessed whether exclusive activation of peripheral opioid receptors results in significant pain reduction. To achieve opioid activity restricted to the periphery, we used a short-term (2 h) low dose infusion of morphine-6-beta-glucuronide (M6G) because M6G does not pass the blood-brain barrier during this time in amounts sufficient to induce CNS effects. The lack of central opioid effects of M6G was confirmed by a lack of change of the pupil size and absence of other opioid-related CNS effects. As a positive control, morphine was infused at a dosage that definitely produced CNS effects. This was evident by a rapid decrease of the pupil size and by other typical opioid-related side effects including nausea, vomiting, itchiness, hiccup and sedation. Three different pain models were employed to evaluate the analgesic effects: (i) cutaneous inflammatory hyperalgesia induced by briefly freezing a small skin area to -30 degrees C ('freeze lesion'); (ii) muscle hyperalgesia induced by a series of concentric and eccentric muscle contractions (DOMS model; delayed onset of muscle soreness); and (iii) pain induced by electrical current (5 Hz sinus stimuli of 0-10 mA). M6G significantly reduced cutaneous hyperalgesia in the 'freeze lesion' model as assessed with von Frey hairs. It also reduced muscle hyperalgesia in the DOMS model. Electrical pain, however, was not affected by M6G. Morphine was significantly more active in the 'freeze lesion' and DOMS model, and also significantly increased the electrical pain threshold and tolerance. Subcutaneous tissue concentrations of M6G and morphine as assessed with microdialysis were about half those of the respective plasma concentrations. The results of the study indicate that M6G has antihyperalgesic effects in inflammatory pain through activation of peripheral opioid receptors. Since this occurs at concentrations that do not cause central opioid effects, M6G might be useful as a peripheral opioid analgesic.

Activity of the sex-determining gene tra-2 is modulated to allow spermatogenesis in the C. elegans hermaphrodite.

In the nematode C. elegans, there are two sexes, the self-fertilizing hermaphrodite (XX) and the male (XO). The hermaphrodite is essentially a female that makes sperm for a brief period before oogenesis. Sex determination in C. elegans is controlled by a pathway of autosomal regulatory genes, the state of which is determined by the X:A ratio. One of these genes, tra-2, is required for hermaphrodite development, but not for male development, because null mutations in tra-2 masculinize XX animals but have no effect on XO males. Dominant, gain-of-function tra-2 mutations have now been isolated that completely feminize the germline of XX animals so that they make only oocytes and no sperm and, thus, are female. Most of the tra-2(dom) mutations do not correspondingly feminize XO animals, so they do not appear to interfere with control by her-1, a gene thought to negatively regulate tra-2 in XO animals. Thus, these mutations appear to cause gain of tra-2 function in the XX animal only. Dosage studies indicate that 5 of 7 tra-2(dom) alleles are hypomorphic, so they do not simply elevate XX tra-2 activity overall. These properties suggest that in the wild type, tra-2 activity is under two types of control: (1) in males, it is inactivated by her-1 to allow male development to occur, and (2) in hermaphrodites, tra-2 is active but transiently inactivated by another, unknown, regulator to allow hermaphrodite spermatogenesis; this mode of regulation is hindered by the tra-2(dom) mutations, thereby resulting in XX females.

Safety and feasibility of a health-related fitness test battery for adults.

BACKGROUND AND PURPOSE: Health-related fitness (HRFI) assessment may be useful in promoting physical activity. Health-related fitness refers to those components of fitness that are related to health status. The safety and feasibility of a test battery designed for the assessment of HRFI were evaluated. SUBJECTS AND METHODS: Middle-aged men (n = 246) and women (n = 254), evenly selected from five age cohorts of a random sample (N = 826), were tested. The subjects had a mean age of 47.0 years (SD = 7.9, range = 37-57). Screening to identify subjects with health limitations was conducted by fitness testers who had master's degrees in sport or health sciences. Safety was assessed in terms of acute complications, delayed-onset muscle soreness (DOMS), and heart rate after each test. Subject exclusion and time costs were evaluated for feasibility. RESULTS: No acute complications occurred. The leg function test caused severe DOMS among inactive women. The overall exclusion rate increased with age. Up to 27% of subjects aged 52 and 57 years were excluded from muscle endurance tests, mainly due to self-reported heart disease or elevated blood pressures. Over 90% of the subjects, however, qualified for balance, flexibility, muscle force, and walk tests. CONCLUSION AND DISCUSSION: The test battery offers a safe and feasible method for the assessment of HRFI in working-aged adults, with the limitation that the one-leg squat function test may cause DOMS, particularly in inactive women.
