The muscular organization of the stomach of capybara (Hydrochoerus hydrochaeris): an architectural view.

Twenty-two stomachs from adult capybaras were used in this study, and an acid digestion mesoscopic technique was pursued using different concentrations of nitric acid to observe the muscular organization of the stomach. The capybara's stomach possessed a muscular coat composed of four layers or strata: external longitudinal, external oblique, circular and internal oblique. Also, the cardiac and pyloric sphincter muscles were comprised of three or two different layers, respectively. Furthermore, the internal oblique fibres were observed extending from the cardiac portion of the stomach to the smaller curvature, where they participated in the formation of the Ansa cardiaca together with the external longitudinal fibres. This muscular architectural arrangement was compared to that in small rodents (rat, hamster, guinea pig), as well as in rabbits and pigs. In conclusion, the stomach of the capybara has a very particular, complex and defined muscular organization that differs from that in other rodents, or domestic animals, in particular, pigs.

Triangle-programmed coulometric nanotitrations completed by continuous flow with potentiometric detection

Coulometric nanotitrations were realized in a microchannel system using a continuous-flow titration technique with a triangle current-time profile. Redox and acid-base titrations were carried out on Fe(II) and nitric acid samples, respectively, with the same nanotitrator device. A linear relation between the concentration and the coulometric current transferred to the solution was found. The advantages of this universally applicable nanotitrator are fast response, low sample volume, high sensitivity, and high reproducibility as well as the convenience of handling an automated analyzer of the flow-through type.

Development of column-pretreatment chelating resins for matrix elimination/multi-element determination by inductively coupled plasma-mass spectrometry.

Chelating resins, two kinds of iminodiacetate derivatives (IDA) of cross-linked chitosan (CCS) were synthesized and investigated for adsorption capacity, matrix elimination and collection/concentration of analytes by a column pretreatment in a multi-element ICP-MS determination method. The adsorption behavior of 54 elements at the 10 ng ml(-1) level on chitosan derivatives in a packed mini-column was systematically examined. Almost 30 kinds of metal ions were recovered quantitatively at pH 5 with CCS-HP/IDA (cross-linked chitosan possessing N-2-hydroxypropyl iminodiacetic acid groups) column. Compared with available chitosan-iminodiacetate resin, CHITOPEARL CI-03, the recovery of the metal ions such as Cu, Pb and La is satisfactory with CCS-IDA (cross-linked chitosan possessing N,N-iminodiacetic acid groups) and CCS-HP/IDA using 2 M nitric acid as an eluent, which may be attributed to the difference of cross-linking and macroporous structure. Compared with Chelex-100, the adsorption efficiency is in the order: Chelex-100 > CCS-IDA > CCS-HP/IDA, especially in the chelating ability for alkaline earth metals. The resin with a longer spacer (CCS-HP/IDA) showed higher adsorption selectivity between heavy metal ions and alkaline earth metals at pH < 7. The separation efficiency of the major matrix cations in seawater (Na. K, Mg, Ca) has also been investigated, and matrix interference was negligible even in a seawater sample at pH 5 with CCS-HP/IDA. The recoveries of Mn at pH 5 with CCS-HP/IDA or Chelex-100 were almost 100%. However, those of Mg with each resin were 4 or 98%, respectively. The adsorption capacities of synthesized CCS-HP/IDA for Cu(II), Pb(II) and La(III) were 0.90, 0.65 and 0.34 mmol g(-1), respectively. Therefore, the chelating chitosan resins developed are applicable to the pretreatment of trace amounts of elements in various kinds of water samples.

Preconcentration and determination of trace elements with 2,6-diacetylpyridine functionalized Amberlite XAD-4 by flow injection and atomic spectroscopy.

An on-line flow injection method for the direct determination of trace elements in environmental samples is described. A mini-column packed with 2,6-diacetylpyridine functionalized Amberlite XAD-4 was used to preconcentrate and separate 8 trace metals (Cd, Co, Cu, Mn, Ni, Pb, U and Zn) from water and extracts from solid samples. The metals were eluted with 0.1 M HNO(3) directly to the detection system (either inductively coupled plasma-mass spectrometry (ICP-MS) or flame atomic absorption spectrometry (FAAS)). As well as demonstrating that the resin could be used to preconcentrate ultra-trace analytes from natural waters, it was also shown to work well at a pH of 5.5. Therefore, after treatment of sample digests with sodium fluoride, samples that contain extremely large concentrations of iron may be analysed for trace analytes without the excess iron overloading the capacity of the resin. To this end, the analytes Cd, Co, Cu and Ni were preconcentrated from acid extracts of certified soil/sediment samples and then eluted with nitric acid to be determined on-line. Limits of detection (3sigma) of Cd = 0.33 microg l(-1), Co = 0.094 microg l(-1), Cu = 0.34 microg l(-1), Mn = 0.32 microg l(-1), Ni = 0.30 microg l(-1), Pb = 0.43 microg l(-1), U = 0.067 microg l(-1) and Zn = 0.20 microg l(-1) for the FI-ICP-MS system and Cd = 22 microg l(-1), Co = 60 microg l(-1), Cu = 10 microg l(-1) and Ni = 4.8 microg l(-1) for the FI-FAAS system were obtained. Analysis of certified reference materials showed good agreement with the certified values using the two methods.

Application of selective extraction to the study of iron species present in diet and rat gastrointestinal tract contents.

Iron speciation in rodent diet and rat gastrointestinal tract lumen during dietary digestion and absorption was investigated with a novel selective extraction technique. Five Fe fractions were identified, namely exchangeable (soluble in 1 M-magnesium chloride), carbonate-bound (soluble in mild acid), oxide-bound (soluble in hydroxylamine-acetic acid), organic-bound (soluble after treatment with peroxide in nitric acid) and residual. Fe from the pelleted diet was mobilized by rat stomach to the exchangeable fraction, then redistributed to the carbonate- and oxide-bound fractions on passage through the proximal small intestine. In vitro incubation of diet with hydrochloric acid failed to mimic the in vivo effect of the stomach. In vitro neutralization of stomach contents with bicarbonate was found to produce a similar effect on Fe speciation to that seen when diet passed the proximal small intestine in vivo. Comparison of 59Fe speciation in extrinsically labelled diet with endogenous Fe speciation showed that extrinsic labelling does not uniformly label all endogenous species. The experiments suggest that selective extraction may provide a useful approach to the study of Fe species present in diets, in vitro digestions and gastrointestinal contents.

Early alteration in glomerular reserve in humans at genetic risk of essential hypertension: mechanisms and consequences.

Essential hypertension has a familial predisposition, but the phenotype of elevated blood pressure has delayed penetrance. Because the kidney is a crucial determinant of blood pressure homeostasis, we studied early glomerular alterations in still-normotensive young subjects at genetic risk of hypertension. Thirty-nine normotensive adults (mean age 29 to 31 years), stratified by genetic risk (parental family history [FH]) of hypertension (26 with positive FH [FH+], 13 with negative FH [FH-]), underwent intravenous infusion of mixed amino acids. Before and during amino acid administration, we measured glomerular filtration rate (GFR), putative second messengers of amino acids (nitric oxide [NO.] metabolites and cGMP), serum insulin and amino acid concentrations, and the FE(Li)+ as an index of renal proximal tubular reabsorption. The FH+ group had a blunted GFR rise in response to amino acids (2.43+/-8.16% versus 31.0+/-13.4% rise, P:=0.0126). The amino acid-induced change in GFR correlated (r=0.786, P:<0.01) with the change in urinary NO. metabolite excretion; a diminished rise in urinary NO. metabolite excretion in the FH+ group (P:=0.0105) suggested a biochemical mechanism for the different GFR responses between FH groups: a relative inability to convert arginine to NO. The FH+ group had a far lower initial cGMP excretion at baseline (261+/-21.1 versus 579+/-84.9 nmol. h(-1)/1.73 m(2), P:=0.001), although cGMP did not change during the amino acid infusion (P:=0.703). FH status, baseline GFR, and baseline serum insulin jointly predicted GFR response to amino acids (P:=0.0013), accounting for approximately 45% of the variance in GFR response. Decline in FE(Li)+, an inverse index of proximal tubular reabsorption, paralleled increase in GFR (r=-0.506, P:=0.01), suggesting differences in proximal tubular reabsorption during amino acids between the FH groups. GFR response to amino acid infusion was blunted in the FH+ group despite significantly higher serum concentrations of 6 amino acids (arginine, isoleucine, leucine, methionine, phenylalanine, and valine) in the FH+ group, suggesting a novel form of insulin resistance (to the amino acid-translocating action of insulin) in FH+ subjects. We conclude that blunted glomerular filtration reserve in response to amino acids is an early-penetrance phenotype seen even in still-normotensive subjects at genetic risk of hypertension and is linked to impaired formation of NO. in the kidney. Corresponding changes in GFR and fractional excretion of Li(+) suggest that altered proximal tubular reabsorption after amino acids is an early pathophysiologic mechanism. Resistance to the amino acid-translocating actions of insulin may play a role in the biological response to amino acids in this setting. This glomerular reserve phenotype may be useful in genetic studies of renal traits preceding or predisposing to hypertension.

Observations on dental structures when placed in contact with acids: experimental studies to aid identification processes.

In a "mafia" crime case, a magistrate asked us whether it is possible to destroy a cadaver by immersing it in acids, and would it be possible to identify any residues. The aim of this study was to observe the behavior of teeth exposed to four kinds of acid solutions. The teeth were placed in plastic containers with 25 mL of acid and observed. The experiences showed that teeth are completely dissolved after 14 h of immersion in 37% solution of hydrochloric acid, while at 90h in 96% sulfuric acid, the destruction of the samples is still incomplete. In nitric acid the teeth undergo a complete dissolution in 12 h, and in 17 h in aqua regia (chloroazotic acid-hydrochloric/nitric acid 1:3). It was possible to recognize the characteristic morphological features of dental tissues and structures up until the advanced stages of degradation.

Nicotinamide and dexamethasone inhibit interleukin-1-induced nitric oxide production by RINm5F cells without decreasing messenger ribonucleic acid expression for nitric oxide synthase.

Nitric oxide (NO) generation may be a final common pathway for beta-cell damage in early insulin-dependent diabetes mellitus. Insulin-producing cells express an inducible form of NO synthase (iNOS), which is similar to that observed in activated macrophages. Induction of iNOS mRNA in these cells depends on protein synthesis. To further characterize the regulation of iNOS induction in insulin-producing cells, RINm5F cells (RIN cells) were exposed for 6 h to human recombinant interleukin-1 beta (rIL-1 beta; 1 ng/ml) alone or in combination with either nicotinamide (10, 20, or 50 mM) or dexamethasone (1 or 5 microM). These agents have been previously shown to prevent activation of iNOS in macrophages, fibroblasts, and hepatocytes. rIL-1 beta induced the expression of iNOS mRNA in RIN cells and a 12- to 13-fold increase in medium nitrite accumulation, the latter indicating NO production. Nicotinamide decreased nitrite production in a dose-dependent way. Thus, 10 mM nicotinamide decreased rIL-1 beta-induced nitrite formation by 30%, 20 mM by 60%, and 50 mM by 90%. The highest concentration of nicotinamide also prevented rIL-1 beta-induced iNOS mRNA, an effect associated with inhibition of total protein biosynthesis. However, 10 or 20 mM nicotinamide did not modify rIL-1 beta-induced iNOS mRNA expression or inhibit protein biosynthesis. Dexamethasone also decreased rIL-1 beta-induced nitrite production without affecting iNOS mRNA expression. As a whole, these data suggest that both nicotinamide and dexamethasone may prevent NO accumulation in insulin-producing cells by posttranscriptional mechanisms. It is also possible that these drugs induce direct inhibition of iNOS enzymatic activity and/or scavenge NO. Higher concentrations of nicotinamide might also inhibit iNOS mRNA expression, possibly by blocking protein biosynthesis.

Washing as a remediation technology applicable in soils heavily polluted by mining-metallurgical activities.

A permanent solution in order to remediate a heavily contaminated soil is washing it utilizing the appropriate reagents. The study presented in this paper deals with the washing of a soil highly polluted by mining and metallurgical activities. Pollution consists of slags, sulphur compound waste and various low grade lead condensates, all rich in heavy metals and metalloids (Pb, As, Cu, Zn, Mn and Fe). Soil was washed with deionized water, 6, 3, 2 and 1 M HCl, 6 M H(2)SO(4), 6 M HNO(3) and 0.1 M Na(2)EDTA. High extraction efficiency was achieved with respect to hydrochloric acid even at the lowest concentration; the solubility of pollutants in nitric acid was low, while sulphuric acid was proven to be problematic with respect to lead. Mobilization of metals and metalloids under DI water effect was minimal indicating that the elements fraction that is weakly bound to soil particles is insignificant. Furthermore, a kinetic study was undertaken, utilizing 1 M HCl and 0.1 M Na(2)EDTA. Results illustrated that 1 M HCl was more effective for intermediate mixing times (up to 4 h) while 0.1 M Na(2)EDTA was better applicable to short mixing times (<1 h) and low pH values (pH < 7.1). 1 M HCl was proven to be very effective for the removal of pollutants from contaminated soil but the high metal content of the resulting solution necessitates further treatment for the retention of metals.

Adhesion and activation of platelets and polymorphonuclear granulocyte cells at TiO2 surfaces.

The initial reactions of two TiO2 surfaces with blood were investigated by short-time exposure to capillary blood and analysis of surface-adsorbed plasma proteins and surface-adhering cells by using immunofluorescence techniques. Antibodies directed against platelet membrane antigen and P-selectin were used to visualize platelet adhesion and activation. Acridine orange and anti-CD11b were used to detect adhesion and activation of polymorphonuclear granulocytes (PMNs). Antibodies against thrombospondin were used as markers for platelet alpha-granules. The fluorescence intensity was quantitated by computer-aided image analysis. Commercially pure, polished sheet titanium was oxidized in two different ways: (1) the natural oxide was dissolved with hydrofluoric acid and a new oxide layer was grown by oxidation in nitric acid, or (2) annealing was performed at 700 degrees C in air. Auger electron spectroscopy and x-ray photoelectron spectroscopy showed that both surfaces had similar composition consisting of TiO2 covered by a carbonaceous surface contamination layer. The thickness of the oxide layer was 4 nm on the acid-oxidized surface and 39 nm on the annealed surface. Optical profilometry and scanning electron microscopy showed that the acid-oxidized surface was rough and the annealed surface was smooth. The fibrinogen/prothrombin-thrombin ratio in the initial protein film differed between the surfaces. The number of adhering platelets was larger at the surface with a high surface concentration of adsorbed fibrinogen. Platelet activation (CD62) and priming of PMNs (CD 11b) were also significantly higher on the acid-oxidized surface. The results indicate that non-self recognition of biomaterials is an array of transient reactions comprising protein-material, protein-cell, and cell-cell interactions.

Adsorption of anionic chloro complexes of 59Fe and 195Au on non-ionic resins of macro-reticular type.

Adsorption behavior of 59Fe and 195Au on the non-ionic macro-reticular resin, Amberlite XAD-7 is studied. Distribution coefficients (Kd) for both nuclides in hydrochloric acid or lithium chloride solutions above 6M are particularly high. In nitric acid solution below 2M, 195Au is highly adsorbed on the resin but 59Fe is negligible over any concentrations. This resin did not adsorb other important radionuclides such as 54Mn, 60Co, 65Zn, 90Sr, 106Ru, 137Cs and 144Ce at all. Based on such peculiar adsorption behavior, application to selective separation and determination of radio and stable iron in sea water was also studied.

[Tests on ionic release from glass-ionomer cements].

BACKGROUND: Dissolution process in oral liquids by the presence of glass-ionomer systems (due to surface corrosion, to diffusion through solutions and through mass) make an ionic release (particularly F, Al, Pb, As) which is a non secondary problem, due to the usual utilization of these materials in pedodontic and restorative dentistry. METHODS: In this work, considering the high toxicity of low quantity of Arsenic ion, a comparative research has been made in order to determine, by using high level liquid Cromatography (HPCL), the quantity in ppm of As hydro- and acid soluble given by five ionomeric products, in water and in nitric acid concentrated solution. RESULTS AND CONCLUSIONS: The results show that in some products arsenical concentrations are higher then the quantity accepted by ISO-FDI; therefore, a better control in the production of these products is needed as well as a limited use in dentistry. It is suggested to use glass-ionomer systems in patients with dental dike and varnish on the surfaces that are in contact with oral liquids action.

Effect of Acids on Water Vapor Uptake by Pyrogenic Silica

Effect of gaseous HCl and HNO3 on the water vapor uptake by pyrogenic silica was studied at different relative humidities (RH) for pure water and different compositions of binary and ternary vapor mixtures. Experiments showed that the ability of silica to uptake water strongly depends on RH and on the type of acids and their concentration in the vapor mixtures. At low acid concentration in the binary mixtures the influence of acids is probably small. Water uptake by silica does not change monotonically with acid concentration: at first it decreases and then starts to grow. However, the presence of acids promotes water uptake, and the effect is very significant at low RH. HCl seems to be more effective acid to enhance water uptake than HNO3 . In the case of ternary mixtures the adsorbed weight of water is a bit larger than that adsorbed from the binary mixtures. Acids are accumulated by silica surface, and the accumulation is larger for nitric acid.

Characterization and CO2 Adsorptivity of Acid-Washed and Cation-Exchanged Natural Mordenites

A Japanese natural mordenite was modified by acid washing as well as cation-exchange. Crystal structure, porosity, and active sites of the modified natural mordenites were characterized, and their CO2 adsorptivity were examined. Contraction as well as expansion of the mordenitic unit cell in the natural zeolite and distortion of zeolite structure by acid washing are plausibly shown by XRD. Long-term washing by nitric acid induces enlargement of micropore volume, increasing the micropore dimension but decreasing the large pore dimension over the mesoporous range. The degree of dehydration has an important role in CO2 adsorption. Metal ionic sites affect both the irreversible CO2 adsorption and the CO2 equilibrium adsorption at low concentration. Li+ ion-exchange brings about a great CO2 adsorption but reduces the CO2-surface interaction due to molecular size effect. Ca2+ ion-exchange has its main effect in the enhancement of CO2 desorption energy. Copyright 1997 Academic Press. Copyright 1997Academic Press

Application of capillary electrophoresis combined with a modified BCR sequential extraction for estimating of distribution of selected trace metals in PM2.5 fractions of urban airborne particulate matter.

The capillary electrophoresis (CE) method combined with a sequential extraction was applied to determine the distribution of Fe, Zn, Cu, Mn and Cd in urban ambient air PM2.5 samples. PM2.5 was collected on Teflon filters with dichotomous sampler, and the modified extraction procedure following the BCR leaching procedure was used to chemically fractionate metals into "easily exchangeable" with water, "acid extractable" with 0.11 mol/l acetic acid, "reducible" with 0.1 mol/l hydroxylamine hydrochloride acidified to pH 2.0 with nitric acid, and "oxidisable" with oxidation by 8.8 mol/l hydrogen peroxide (H2O2) followed by extraction with 1.0 mol/l ammonium acetate. Based on the obtained results it was concluded that the application of the studied methodology provides chemical fractionation data that reflect the general sources and potential health hazards of the studied metals.

Uptake of small oxygenated organic molecules onto ammonium nitrate under upper tropospheric conditions.

The uptake of formic (C1), propanoic (C3), butanoic (C4), and pentanoic (C5) acids onto ammonium nitrate (AN) has been investigated as a function of temperature and relative humidity using a Knudsen cell flow reactor coupled with FTIR-reflection absorption spectroscopy (FTIR-RAS). The uptake of acetone and methanol onto AN was also briefly studied. Initial uptake coefficients (gamma) were determined over the temperature range 200-240 K. Formic, propanoic, and butanoic acids exhibited efficient but temperature-dependent uptake on AN, with larger uptake coefficients observed at lower temperatures. Pentanoic acid was not taken up by AN under any of the conditions studied. Uptake of acetone and methanol onto AN was observed, but in insignificant amounts under atmospherically relevant conditions. Infrared spectra revealed that propanoic and butanoic acids ionized on the surface, despite the fact that the AN films were effloresced. Formic acid reacted with the AN film to produce ammonium formate and ionized nitric acid. Adding small amounts of water vapor (4% RH) to the chamber resulted in dramatically increased gamma values for all of the acids. Furthermore, the IR spectra showed the formation of a liquid layer when propanoic and butanoic acids adsorbed on the surface at RH = 20% and greater. Liquid water features were not observed at a similar relative humidity in the absence of the acids. These results show that small organic acids can be efficiently scavenged by AN and lead to enhanced water uptake under upper tropospheric conditions.

Selective precipitation separation and inductively coupled plasma mass spectrometric determination of trace metal impurities in high purity silver.

A simple and rapid method for the determination of some trace element impurities in high purity silver, combining the isolation of analytes from the silver matrix with selective precipitation followed by ICP-MS determination was developed. On the basis of an extreme difference in the solubilities of the chlorides of silver and the other accompanying trace elements, silver can be separated completely through the addition of hydrochloric acid. The sample of silver was at first dissolved in 7 M nitric acid followed by addition of hydrochloric acid to remove the silver matrix by formation of a silver chloride precipitate, while leaving the trace element impurities in the solution, which was subsequently analysed by ICP-MS. Eleven elements (Al, Au, Cd, Co, Cu, Fe, Mg, Mn, Ni, Pb and Sn) were determined with good accuracy and precision. The limits of detection (based on the 3 sigma criterion) of these elements were 10(-1)-10(-3) ng g-1. The proposed method was successfully applied to the determination of metal impurities in high-purity silver samples (EM9465 and EM 9343) and validated by the analysis of NIST SRM 8171 (Fine Silver FS 14).

[Changes in microstructure of ceramic alloys after heat and electrochemical acid treatment].

The technique of electrochemical etching was developed by Dr. Tegart for application in both research and industry in 1959. There is considerable evidence showing that in conjunction with chemical bonding, surface roughness of etched casting restorations can apparently improve the mechanical adherence between the porcelain and the metal. It is obvious that surface roughness produced by sandblasting or electrochemical etching of the metal substrate not only provides mechanical interlocking but also increases the surface area for porcelain attachment. It was observed in this study that a properly heat-treated surface was significantly rougher than a surface without appropriate treatment after electrochemical acid etching. The roughness of base metals also treated with 0.5N nitric acid appeared greater obvious than those treated with 10% sulfuric acid. The etching pattern can be evaluated by selective removal of the alloy phases present between the dendritic arms in the base metal alloy. The technique of electrochemical acid etching is limited to alloys that solidify with a multiphasic structure- that is, a base metal alloy. And there are no known noble alloys suitable for this technique in this experiment. Electrolytic etching is most conveniently evaluated under a light microscope at about 30-80 magnifications to inspect surface relief and etching patterns. It remains to be determined by further research what is the exact cause of the increased roughness and how it improves the metal for ceramic mechanical bonding.
