An AI approach to dynamic visual field testing.

Visual field test results are crucial to the accuracy and efficiency of diagnosing blinding diseases such as glaucoma. Herein, a method of integrating self-organizing neural networks and empirical heuristics is used to perform visual field tests via a dynamic test strategy, which can lead to a reduction in the number of trials in a perimetric test. Experiments performed using clinical test records show that we are able to reduce by 20% to 30% the number of trials per test without much adverse effect on the accuracy of the tests.

Association of a polymorphism in the Apo AI gene promoter with hyperalphalipoproteinemia.

The distribution of a polymorphism due to an Adenine to Guanine transition in the ApoAI gene has been studied in 136 women and 108 men, through amplification of the promoter region of the gene and allele-specific oligonucleotide hybridization. The allelic frequencies for the A allele were 0.10, 0.14 and 0.27 in women and 0.08, 0.17 and 0.14 in men for the lowest decile, intermediate group and the highest decile of HDL-cholesterol levels, respectively. Statistical analysis showed that the A allele was associated with high HDL-cholesterol levels in women, but not in men.

WIZARD: AI in conformational analysis.

A program which utilizes the techniques of Artificial Intelligence and Expert Systems to solve problems in the area of Conformational Analysis is described. The program searches conformational space in a systematic fashion, based on the technique known as heuristic state-space search. The program proceeds by recognizing conformational units, assigning one or more conformational templates to each unit, and joining them to form conformational suggestions. These suggestions are criticized to discover logical inconsistencies, and any resulting stresses are resolved. The resulting conformational suggestions are sometimes accurate enough for immediate use, or may be further refined by a numerical program. The latter combination is shown to be quite efficient compared to purely numerical conformational search techniques.

Neuropharmacological correction of cognitive processes disrupted by extirpation of the AI-AII and Ep fields in cats.

An original neuropsychophysiological battery of tests was used to study the possibility of neuropharmacological correction of compensatory-restorative processes in cats after extirpation of a defined zone of the temporal cortex; studies were carried out in conditions of free behavior. In a variety of activation and inhibition regimes, systemic administration of cholinergic, dopaminergic, GABAergic, and serotoninergic agents could have favorable effects on restorative mechanisms, depending on cortical psycho-organic symptoms. Each cognitive activity studied (perception, gnosis, thought) had its own specific positive neuromediator "profile" in terms of compensatory outcomes.

The effect of hormone replacement therapy on the levels of serum lipids, apolipoprotein AI, apolipoprotein B and lipoprotein (a) in Turkish postmenopausal women.

OBJECTIVES: Estrogen replacement therapy alters the lipid profiles favorably for delaying atherosclerosis in postmenopausal women. The effects of estrogen plus progesterone combination therapy on lipids are controversial. This study was designed to evaluate the effect of female sex hormones on lipids and lipoproteins and to clarify the influence of progesterone on the effect of estrogen in postmenopausal women. METHODS: Of the 60 postmenopausal women admitted to our menopause clinic, 40 had intact uterus and received continuous 0.625 mg conjugated equine estrogen (CEE) plus 2.5 mg medroxyprogesterone acetate (MPA), whereas the remaining 20 were hysterectomized and received 0.625 mg CEE daily. To assess the alterations in lipids and lipoproteins during menopause, 45 healthy premenopausal women were investigated. Lipid and lipoprotein levels were assessed in each subject at baseline and at the 6th and 18th months of therapy. RESULTS: In menopause, a shift towards more atherogenic lipid and lipoprotein profiles than those of the premenopausal state was found. Following 18 months of treatment, both regimens reduced total cholesterol (TC) levels as compared with the baseline (6.4 vs. 6.9% in the CEE/MPA and CEE groups, respectively). The CEE group had a more pronounced increase in high-density lipoprotein (HDL) cholesterol than the CEE/MPA group (10.3 vs. 8.8%, respectively). Both groups displayed reduced TC, low-density lipoprotein (LDL) cholesterol and apolipoprotein-B (ApoB) concentrations, whereas triglycerides increased, with a greater tendency to increase in the CEE/MPA group at the end of the trial. Also, the lipoprotein (a) [Lp(a)] levels decreased significantly (27.6 vs. 24.5% in the CEE/MPA and CEE groups, respectively). This decrease was more pronounced in subjects with a relatively higher basal Lp(a) concentration. CONCLUSION: Both treatment regimens caused positive alterations in the lipid and lipoprotein profiles. This association might play a pivotal role in the postmenopausal increases in atherosclerotic diseases and cardioprotective effect of estrogen in postmenopausal women.

Machine learning-ready genomic biomarkers: ATF3 polymorphisms predict postoperative analgesic demand through AI-compatible phenotyping.

PURPOSE: To determine whether ATF3 polymorphisms can serve as genetic biomarkers for machine learning-based precision analgesia by establishing a genotype-phenotype association suitable for predictive modeling of postoperative opioid requirements. METHODS: In a prospective cohort of 167 adults undergoing abdominal surgery, ATF3 SNPs rs3122721 and rs3125293 were genotyped. A structured dataset architecture was developed to represent genetic profiles as input features for supervised learning models, enabling translational analysis of genotype‑dependent opioid consumption over 72 h. RESULTS: Patients with homozygous genotypes of the ATF3 SNPs had significantly higher opioid requirements than non‑carriers, despite reporting similar subjective pain scores. This consistent genotype‑dependent pattern provided a clinically relevant phenotype suitable for integration into predictive algorithms. CONCLUSION: ATF3 genotyping offers a promising biomarker for computationally informed precision analgesia. By linking genomic variability to clinically meaningful outcomes within a structured clinical and genomic framework, this approach supports the future development of risk-stratified clinical decision-support systems to optimize postoperative pain management.Trial registration ChiCTR1900021991, registered 30 April 2019. Effect of fenofibrate treatment on plasma lipoprotein lipids, high-density lipoprotein cholesterol subfractions, and apolipoproteins B, AI, AII, and E.

In this segment of a multicenter study, 36 hypercholesterolemic patients were randomly assigned to fenofibrate or placebo treatment to assess effects on plasma concentrations of lipoprotein cholesterol and triglyceride, high-density lipoprotein-cholesterol subfractions, and apolipoproteins E, B, Al, and All. All of these factors are of known or potential value in determining the patient's risk of arteriosclerosis. Observations were made during initial screening and placebo phases, a 24-week, double-blind treatment phase, and a subsequent 24-week, open-label fenofibrate phase. There were three possible expressions of fenofibrate efficacy. Changes in lipoprotein cholesterol and total triglyceride concentrations observed in these patients were very similar to those seen with the larger multicenter cohort: total triglyceride levels decreased 38 to 46 percent, low-density lipoprotein cholesterol levels decreased 13 to 20 percent, and high-density lipoprotein cholesterol levels increased 4 to 13 percent. Triglyceride concentrations were significantly reduced (p less than 0.01) in very low-density lipoprotein (50 to 56 percent, similar to those of total triglyceride and very low-density lipoprotein cholesterol), and in low-density lipoprotein cholesterol levels (17 to 21 percent). A slight but statistically insignificant decrease in high-density lipoprotein triglyceride was observed (9 to 15 percent). High-density lipoprotein2 cholesterol levels did not change significantly, whereas high-density lipoprotein3 cholesterol levels increased 8 to 16 percent, accounting for all of the increase in high-density lipoprotein cholesterol. Apoprotein All levels increased significantly (13 to 20 percent) whereas those of apolipoprotein Al did not, consistent with an increase in high-density lipoprotein3 levels, where apolipoprotein All is more abundant relative to apolipoprotein Al than in high-density lipoprotein2. Apolipoprotein B levels decreased 20 to 26 percent and those of apolipoprotein E went from 29 to 34 percent, relative to the 16 to 20 percent decreases in very low-density lipoprotein and low-density lipoprotein triglyceride and cholesterol levels. Five patients with combined elevations of triglyceride and low-density lipoprotein cholesterol treated with fenofibrate, had reductions primarily in triglyceride, total apolipoprotein E (50 percent reduction), and apolipoprotein B (18 percent) levels. High-density lipoprotein3 cholesterol levels increased 19 percent and high-density lipoprotein2 cholesterol levels were unchanged. Low-density lipoprotein cholesterol levels declined slightly in four patients and a slight rise was observed in a fifth patient.(ABSTRACT TRUNCATED AT 400 WORDS)

Electron transport between plastoquinone and chlorophyll Ai in chloroplasts. II. Reaction kinetics and the function of plastocyanin in situ.

The light-induced reaction kinetics of electron carriers between the two light reactions were studied in spinach chloroplasts. 1. The difference spectrum of the absorbance changes of plastocyanin in situ was separated from superimposing absorbance changes by flash titration described in the preceding paper (Haehnel, W. (1973) Biochim. Biophys. Acta 305, 618-631). Relative amounts of 2 : 1 electron equivalents were observed for plastocyanin and chlorophyll a1 (P-700). 2. A balance of the electron equivalents released from reduced plastoquinone and simultaneously accepted by oxidized plastocyanin, cytochrome f and chlorophyll a1 indicated a quantitative electron transfer. Additional electron carriers between plastoquinone and light reaction I can be excluded with an accuracy of about +/-0.3 electron equivalents per light reaction II. 3. The time course of the absorbance changes of plastocyanin was measured at 584 nm with negligible interference with other absorbance changes. The reduction kinetics show an initial lag followed by a rise with a half time of about 20 ms. The redox states of plastocyanin and chlorophyll a1 during this reduction via the rate-limiting step between the light reactions and during oxidation by weak far-red light suggest a true equilibrium constant of about 20. 4. The simultaneous oxidation and reduction kinetics of plastoquinone, cytochrome f, plastocyanin and chlorophyll a1 induced by two successive groups of saturating flashes after far-red illumination were measured. The oxidation kinetics of plastocyanin and the simultaneous reduction kinetics of chlorophyll a1 after the single flashes indicate a quantitative electron transfer with a half time of 200 mus. 5. The fast reduction of chlorophyll a1 by plastocyanin showed no effect of the inhibitors 3-(3',4'-dichlorophenyl)-1,1-dimethylurea and 2,5-dibromo-3-methyl-6-isopropyl-p-benzoquinone or of reduced phenazine methosulfate. But it was completed inhibited after KCN incubation. 6. The oxidation kinetics of cytochrome f were reinvestigated with high time resolution from the difference of absorbance changes at 554 minus 540 nm to minimize the disturbing interference with other absorbance changes. Absorbance changes measured 554 nm alone do not reflect kinetics of cytochrome f. The half time of the oxidation was faster than 40 microns. 7. The observed reaction kinetics gave evidence for a function of cytochrome f between plastoquinone and chlorophyll a1 in parallel to plastocyanin. In addition, they indicate that the greater portion of linear electron transport passes plastocyanin. The complex interaction between cytochrome f and chlorophyll a1 is discussed.
