GSK3 is a multifunctional regulator of Dictyostelium development.

Glycogen synthase kinase 3 (GSK3) is a central regulator of metazoan development and the Dictyostelium GSK3 homologue, GskA, also controls cellular differentiation. The originally derived gskA-null mutant exhibits a severe pattern formation defect. It forms very large numbers of pre-basal disc cells at the expense of the prespore population. This defect arises early during multicellular development, making it impossible to examine later functions of GskA. We report the analysis of a gskA-null mutant, generated in a different parental strain, that proceeds through development to form mature fruiting bodies. In this strain, Ax2/gskA-, early development is accelerated and slug migration greatly curtailed. In a monolayer assay of stalk cell formation, the Ax2/gskA- strain is hypersensitive to the stalk cell-inducing action of DIF-1 but largely refractory to the repressive effect exerted by extracellular cAMP. During normal development, apically situated prestalk cells express the ecmB gene just as they commit themselves to stalk cell differentiation. In the Ax2/gskA- mutant, ecmB is expressed throughout the prestalk region of the slug, suggesting that GskA forms part of the repressive signalling pathway that prevents premature commitment to stalk cell differentiation. GskA may also play an inductive developmental role, because microarray analysis identifies a large gene family, the 2C family, that require gskA for optimal expression. These observations show that GskA functions throughout Dictyostelium development, to regulate several key aspects of cellular patterning.

Transgenic zebrafish reveal stage-specific roles for Bmp signaling in ventral and posterior mesoderm development.

Bone morphogenetic protein (Bmp) signaling is crucial for the formation and patterning of zebrafish ventral and posterior mesoderm. Mutants defective in the Bmp pathway have expanded trunk muscle, abnormal tails and severely impaired development of ventral mesodermal derivatives such as vasculature, blood and pronephros. As Bmps continue to be expressed in the ventral and posterior mesoderm after gastrulation, it is likely that Bmp signaling continues to play an important developmental role during outgrowth of the posterior body. However, because Bmp signaling plays an essential role during the gastrula stages, it has not been possible with mutants or standard disruption techniques to determine the later functions of the Bmp pathway. To study the role of Bmp signaling in the ventral and posterior mesoderm during trunk and tail outgrowth, we generated a transgenic zebrafish line containing a heatshock-inducible dominant-negative Bmp receptor-GFP fusion. Our data show that Bmps are important for tail organizer formation and for patterning the ventral mesoderm during early gastrulation. However, from mid-gastrulation to the early somitogenesis stages, Bmp signaling is important for ventral tail fin development and for preventing secondary tail formation. We conclude that the role of Bmp signaling in the ventral and posterior mesoderm changes as gastrulation proceeds.

Cloning and developmental expression of Sna, a murine homologue of the Drosophila snail gene.

The genetic analysis of dorsoventral patterning in Drosophila has identified a zinc-finger gene, snail, that is required for mesoderm formation. The cloning and nuclease protection analysis of a Xenopus homologue of this gene has suggested a possible role in the mesoderm of vertebrates. Here, we describe the cloning of a murine homologue of snail, Sna, and in situ hybridisation studies of its developmental expression. Sequence analysis reveals substantial conservation of the second to fifth zinc fingers, but not of the first zinc finger in the Sna gene. Expression occurs in the ectoplacental cone, parietal endoderm, embryonic and extraembryonic mesoderm, in neural crest and in condensing precartilage. Based on the timing and spatial restriction of expression in embryonic mesoderm, we suggest that Sna might be required for the early development of this tissue, as is the case for its Drosophila counterpart. In addition, we propose that Sna might have an analogous role in the development of neural crest. The expression in condensing precartilage indicates that this gene also has a later function in chondrogenesis.

Genes controlling and mediating locomotion behavior of the zebrafish embryo and larva.

Zebrafish embryos and larvae have stage-specific patterns of motility or locomotion. Two embryonic structures accomplish this behavior: the central nervous system (CNS) and skeletal muscles. To identify genes that are functionally involved in mediating and controlling different patterns of embryonic and larval motility, we included a simple touch response test in our zebrafish large-scale genetic screen. In total we identified 166 mutants with specific defects in embryonic motility. These mutants fall into 14 phenotypically distinct groups comprising at least 48 genes. Here we describe the various phenotypic groups including mutants with no or reduced motility, mechanosensory defective mutants, 'spastic' mutants, circling mutants and motor circuit defective mutants. In 63 mutants, defining 18 genes, striation of somitic muscles is reduced. Phenotypic analysis provides evidence that these 18 genes have distinct and consecutive functions during somitic muscle development. The genes sloth (slo) and frozen (fro) already act during myoblast differentiation, while 13 genes appear to function later, in the formation of myofibers and the organization of sarcomeres. Mutations in four other genes result in muscle-specific degeneration. 103 mutations, defining at least 30 genes, cause no obvious defects in muscle formation and may instead affect neuronal development. Analysis of the behavioral defects suggests that these genes participate in the diverse locomotion patterns observed, such as touch response, rhythmic tail movements, equilibrium control, or that they simply confer general motility to the animal. In some of these mutants specific defects in the developing nervous system are detected. Mutations in two genes, nevermind (nev) and macho (mao), affect axonal projection in the optic tectum, whereas axon formation and elongation of motorneurons are disrupted by mutations in the diwanka (diw) and the unplugged (unp) genes.

Treatment of gonadotropin-deficient boys with recombinant human FSH: long-term observation and outcome.

BACKGROUND: Boys with prepubertal onset of hypogonadotropic hypogonadism (HH) are at a risk of poor testis growth and impaired spermatogenesis. One potential cause for this is deficient proliferation of immature Sertoli cells before and during puberty due to the absence of FSH. OBJECTIVE: To evaluate the effects of recombinant human FSH (r-hFSH) and human chorionicgonadotropin (hCG) on testicular function and pubertal development in boys with prepubertal onset of HH. DESIGN: Retrospective clinical study. SETTING: Two university central hospitals, pediatric referral endocrinology outpatient clinics. PATIENTS: Fourteen boys (aged, 9.9-17.7 years) with prepubertal (testicular volume (TV) <3 ml) onset of HH (idiopathic HH, n=2; Kallman syndrome, n=2; idiopathic panhypopituitarism, n=4; organic panhypopituitarism, n=6). INTERVENTION: Treatment with r-hFSH alone (2 mo-2.8 years) prior to induction of puberty with the combination of FSH and hCG. MAIN OUTCOME MEASURES: Progression of puberty, change in serum inhibin B, spermatogenesis. RESULTS: r-hFSH alone increased testicular volume twofold, from 0.9+/-0.6 ml (mean+/-s.d.) to 1.8 +/- 1.1 ml (P<0.005), and serum inhibin B threefold, from 27+/-14 to 80+/-57 pg/ml (P<0.01). Three boys with an apparent absence of postnatal hypothalamic-pituitary-testicular axis activation displayed attenuated inhibin B responses to long-term (>or=1 year) r-hFSH (P<0.01). Further significant increase in both TVand inhibin B occurred with induction of puberty with FSH and hCG (P<0.001). Seven boys provided semen samples: one had azoospermia, and others displayed a maximal sperm count range from 2.9 to 92 million/ml (median 8.5 million/ml). CONCLUSIONS: (i) r-hFSH induces prepubertal testis growth and increases circulating inhibin B levels, findings suggesting proliferation of immature Sertoli cells. (ii) Puberty was successfully induced with hCG and r-hFSH following r-hFSH priming. (iii) Inhibin B appears useful for monitoring spermatogenetic activity in boys treated with hCG. (iv) Despite the extremely small initial testis volume, six out of seven patients (86%) primed with r-hFSH displayed sperm in the ejaculate suggesting beneficial effect of r-hFSH priming on testicular function later in life.

Influence of meclofenamic acid on the initiation of parturition and lactation in the sow.

The effect of inhibition of prostaglandin (PG) synthesis on the initiation of parturition was studied in 4 groups of 16 sows each. Oral administration of meclofenamic acid (5.0 mg/kg body weight/day) on Days 112, 113 and 114 of pregnancy significantly delayed parturition compared to controls (P less than 0.001). Exogenous PGF-2 alpha after meclofenamic acid treatment synchronized parturition 20.4 +/- 1 h later. Functional luteolysis as indicated by a decline in maternal progesterone levels began during meclofenamic acid treatment; the fall in progesterone was then arrested in sows treated only with meclofenamic acid. Meclofenamic acid treatment in late pregnancy modified the normal initiation of lactation, as indicated by the high concentration of lactose and low concentrations of protein and Na+ in colostrum in treated sows compared to controls. We suggest that PG synthesis is required for the initiation of parturition in the sow and that luteal regression may involve two stages: the first which may be independent of prostaglandin, and the second, dependent on prostaglandin.

[Surgical procedures in patients with thyroid autoimmune disease].

Autoimmune thyroid disease is a serious medical problem in which various operative procedures are performed. The objective of the study is to explore the type of applied surgical procedures in autoimmune thyroid disease, advantages and disadvantages of various procedures, and criteria they have to meet. This is retrospective clinical study on 1478 patients, operated for Graves' toxic goiter (117 males and 795 females mean age 37.7) and Hashimoto thyroiditis (27 males and 539 females mean age 50.6) from 1995 to April 2005. Cancer in Graves' disease was found in 61 patients (6.7%), papillary in 60 (occult in 53 or 6.6%) and metastatic in 1, Hashimoto thyroiditis and thyroid cancer was found in 141 patients (24.9%), papillary in 116 or 20.5% (occult in 55 or 9.7%), follicular in 2 (0.3%), Hurthle in 11 (1.9%), medullary in 8 (1.4%), anaplastic in 2 (0.3%) and lymphoma in 3 (0.5%). We performed subtotal bilateral lobectomy in 344 (312 in Graves and 32 in Hashimoto), total lobectomy on one side with subtotal on the oposite in 307 (228 in Graves and 79 in Hashimoto); out of them, in 59 patients, the remnant was left in the region of the upper pole which we called atypic lobectomy. The most common procedure, total or near by total thyroidectomy, performed in 719 (371 in Graves and 349 in Hashimoto). One side lobectomy was performed in 103 patients with Hashimoto thyroiditis. Lymph node dissection was performed in 21 (1 in Graves and 20 in Hashimoto), in all central, in 10 lateral functional and in 6 mediastinal, in 15 patients with cancer and in 6 patients with benign disease. There was no operative mortality. In Graves' disease, there was postoperative bleeding in 4 (0.4%), wound infection in 2 (0.2%) recurrent pulsy in 18 (2%) and permanent hypoparathyroidism in 13 (1.4%). In Hashimoto thyroiditis, there was postoperative bleeding in 2 (0.4%), recurrent nerve pulsy in 11 (1.9%) and permanent hypoparathyroidism in 6 (1.1%). The most common surgical procedure in autoimmune thyroid disease is total thyroidectomy which is followed by low complication rate in specialised centers. Cancer is more frequent in Hashimoto than in Graves' disease.

Sex differences and handedness in hemispheric lateralization of tactile-spatial functions.

Sex differences in spatial processing and handedness were studied with a tactile-spatial task in 27 male and 29 female right-handed undergraduate students in psychology. Subjects were asked to identify amorphous shapes to investigate possible male right-hemisphere specialization for spatial functions and bilateral representation among the women. The Annett handedness questionnaire estimated extent of right-handedness. Subjects were classified by major, and women by phase of menstrual cycle. Analysis shows significantly more right-handedness in women and ambidexterity in men. Over-all, men do not perform significantly better than women, although men outperform women with their left hands when handedness is covaried. Within sex, no difference is seen between left and right hand scores for men, but women perform significantly better with right than left hands. Further analyses suggest men appear right-hemisphere dominant for this task whereas women show left-hemisphere dominance. Analyses of hormonal data support recent research, in that for women on the pill there is a trend to perform worse than all other groups. Engineering students perform significantly better than all other majors.

Hemisphere specialization of the animal brain for information processing principles.

The experiments were performed on white rats. Different techniques for forming conditioned reflexes were used. Hemisphere cortex inactivation was carried out by means of spreading depression. The right hemisphere was shown to dominate for space perception, simultaneous information processing, concrete characteristics perception, and deductive processing; the left hemisphere was shown to dominate for time perception, successive information processing, abstract characteristics perception, and inductive processing. A hypothesis of induction and deduction being the main factors determining function lateralization in the hemispheres of the animal brain was put forward.

EEG time and frequency domain measures of cerebral specialization.

Electroencephalographic (EEG) studies of lateralized cognitive functions have primarily relied upon measures of averaged evoked potentials (AEP) and power spectra (PS) to demonstrate hemispheric differences. The nature of the tasks appropriate to each of these methods of signal analysis generally require different cognitive activities and suggest different assumptions about cerebral specialization. This study investigated whether the tasks associated with either method were more likely to produce EEG asymmetries. Eighteen right-handed subjects participated in a three phase experiment which varied the amount of subject involvement in task performance. Phase I entailed passive listening to speech and nonspeech sounds. Phase II required active attention to speech and nonspeech sounds. Phase III involved a verbal matching task, a spatial rotations task, and a noncognitive control task. Auditory AEPs were measured in Phases I and II, and PS were measured in Phase II and III. EEG recordings were obtained from frontal and parietal areas of both hemispheres (F3, F4, P3, P4) referred to linked ears. Power spectra were derived from the 4-8, 8-13, and 13-20 Hz frequency bands at each lead. Despite statistically significant task and/or stimuli differences found in all three phases of study, essentially no task by lead interactions were obtained. That is, interhemispheric activity did not covary with differing stimuli and tasks. The absence of such interactions calls into question uncritical acceptance of a simple direct relation between task performance, EEG activity, and hemisphere specialization.

Subliminal words activate semantic categories (not automated motor responses).

Semantic priming by visually masked, unidentifiable ("subliminal") words occurs robustly when the words appearing as masked primes have been classified earlier in practice as visible targets. It has been argued (Damian, 2001) that practice enables robust subliminal priming by automatizing learned associations between words and the specific motor responses used to classify them. Two experiments demonstrate that, instead, the associations formed in practice that underlie subliminal priming are between words and semantic categories. Visible words classified as pleasant or unpleasant in practice with one set of response key assignments functioned later as subliminal primes with appropriate valence, even when associations of keys with valences were reversed before the test. This result shows that subliminal priming involves unconscious categorization of the prime, rather than just the automatic activation of a practiced stimulus-response mapping.

The surgical treatment of overcorrected esotropia.

The medical records of 33 consecutive patients who developed secondary exotropia following surgery for esotropia were reviewed. Only six patients (18%) had normal medial rectus function. Lateral rectus recession proved to be an excellent procudure for correcting the deviation in these patients, provided the surgery was limited to the eye with the narrower palpebral fissure. Twenty-seven patients (82%) exhibited limitation of adduction secondary to medial rectus underaction. Of these patients, 75% were successfully corrected following a 12 mm advancement and resection of the underacting medial rectus muscle. The details of the surgical technique performed are discussed.

Interplay between TCR affinity and necessity of coreceptor ligation: high-affinity peptide-MHC/TCR interaction overcomes lack of CD8 engagement.

CD8 engagement is believed to be a critical event in the activation of naive T cells. In this communication, we address the effects of peptide-MHC (pMHC)/TCR affinity on the necessity of CD8 engagement in T cell activation of primary naive cells. Using two peptides with different measured avidities for the same pMHC-TCR complex, we compared biochemical affinity of pMHC/TCR and the cell surface binding avidity of pMHC/TCR with and without CD8 engagement. We compared early signaling events and later functional activity of naive T cells in the same manner. Although early signaling events are altered, we find that high-affinity pMHC/TCR interactions can overcome the need for CD8 engagement for proliferation and CTL function. An integrated signal over time allows T cell activation with a high-affinity ligand in the absence of CD8 engagement.

Renal substrate uptake, oxygen consumption and gluconeogenesis at low temperature.

One of the important needs in the surgery of renal transplantation is the long-term preservation of the removed kidney. Up to now, preservation time has been limited to about 72 hours. The viability of the stored organ decreases sharply with prolongation of the preservation over 24 hours. At present, two methods of preservation are widely used: the storage of the organ in a cold electrolyte solution, and the continuous, hypothermic perfusion with albumin or cryoprecipitated plasma [1]. Both methods require a low temperature to reduce the endogenous renal metabolism. Besides the quality of the removed kidney, the type of perfusate and the duration of perfusion, two other factors might influence the viability and later function of the transplanted kidney: the absolute temperature during preservation, and the supply of energy - yielding substrates to the isolated kidney [2,3]. Up to now, the influence of the latter factor on the metabolic performance of the kidney tissue has been studied only in a few investigations [4,5,6,7]. In the following study, we determined the uptake of various energy - yielding substrates, oxygen consumption and gluconeogenesis at various low temperatures in isolated rat kidney tubules.

Lobar decrease in 99mTc-GSA accumulation in hilar cholangiocarcinoma.

UNLABELLED: Hilar cholangiocarcinoma can obstruct hepatic ducts and involve the portal veins. Both biliary stasis and decrease in portal venous flow are known to reduce 99mTc-diethylenetriamine pentaacetic acid-galactosyl human serum albumin (GSA) accumulation. The specific relationship between these pathological conditions due to hilar cholangiocarcinomas and 99mTc-GSA accumulation has never been clarified. METHODS: Sixteen patients with hilar cholangiocarcinomas who underwent 99mTc-GSA liver scintigraphy were reviewed. The relationship between significant decrease in 99mTc-GSA accumulation and lobar biliary stasis, or decrease in the portal venous flow, was evaluated. Average counts of region of interest placed in both right and left lobes were compared in the same transaxial SPECT section. Count ratios of right and left lobes were calculated. RESULTS: Significant lobar decrease in 99mTc-GSA accumulation was observed in 6 of the 16 patients. Ipsilateral portal venous stenosis or obstruction was seen in all these 6 patients, whereas ipsilateral portal venous stenosis or obstruction was seen in only 1 of the other 10 patients. Symmetric bile duct dilatation was seen in 13 patients, and asymmetric bile duct dilatation was seen in 3. Lobar decrease in 99mTc-GSA accumulation correlated well with decrease in ipsilateral portal venous flow (P < 0.0005). The count ratio was significantly reduced when unilateral portal venous flow decreased (P < 0.05). CONCLUSION: Using 99mTc-GSA liver scintigraphy, we can predict lobar decrease in ipsilateral portal venous flow and monitor hepatic functional lateralities in patients with hilar cholangiocarcinomas.

Progression of meiotic DNA replication is modulated by interchromosomal interaction proteins, negatively by Spo11p and positively by Rec8p.

Spo11p is a key mediator of interhomolog interactions during meiosis. Deletion of the SPO11 gene decreases the length of S phase by approximately 25%. Rec8p is a key coordinator of meiotic interhomolog and intersister interactions. Deletion of the REC8 gene increases S-phase length, by approximately 10% in wild-type and approximately 30% in a spo11Delta background. Thus, the progression of DNA replication is modulated by interchromosomal interaction proteins. The spo11-Y135F DSB (double strand break) catalysis-defective mutant is normal for S-phase modulation and DSB-independent homolog pairing but is defective for later events, formation of DSBs, and synaptonemal complexes. Thus, earlier and later functions of Spo11 are defined. We propose that meiotic S-phase progression is linked directly to development of specific chromosomal features required for meiotic interhomolog interactions and that this feedback process is built upon a more fundamental mechanism, common to all cell types, by which S-phase progression is coupled to development of nascent intersister connections and/or related aspects of chromosome morphogenesis. Roles for Rec8 and/or Spo11 in progression through other stages of meiosis are also revealed.

The diaphragmatic compound muscle action potentials (DCMAPs) at different lung volumes--a prospective electrophysiological study of the respiratory system.

Phrenic nerve conduction study and measurements of static lung volumes such as functional residual capacity (FRC), total lung capacity (TLC) and residual volumes (RV) using body plethysmography were carried out in 31 normal healthy male subjects (31 +/- 8 years). The objective was to correlate changes in latency, amplitude, duration and area of diaphragmatic compound muscle action potential (DCMAPs) with lung volumes and changes in them with changes in postures. The mean phrenic nerve latency did not show any significant change with lung volumes or postures, but the latency and height of the subject had a significant correlationship (r = 0.68). The peak to peak amplitude of DCMAP showed a significant relationship with the quantitative lung volumes (r = 0.65). The amplitude was significantly higher and duration reduced (p < 0.001) at TLC as compared with those at measured FRC and RV in both sitting and supine postures. The area under the curve did not change significantly. Similarly, amplitudes and latency did not show any significant relation with other anthropometric parameters. The study evaluates a new quantitative relationship between DCMAPs amplitude and lung volumes. This may be carefully used along with other clinical parameters in critically ill patients for an early weaning from ventilator. Posture exerts minimal influence on DCMAPs amplitude; since DCMAPs amplitudes depends on the position of the diaphragm which in turn depends on dipolic potential, its electromagnetic fields and its moving angle subtends at the recording electrode, one can confidently use it to predict lung volume in respiratory failure due to neuromuscular diseases where subjective lung function assessment is impossible. However, a further study is in process in the critical care unit to confirm its utility.

[Reconstruction of the long bones and mandible with vascularized fibular grafts].

In two prospective studies at the Clinic for Plastic Surgery and Burns and Clinic for Maxillofacial Surgery of the Military Medical Academy in the period from 1991 to 1996 we performed 47 transplantations of the vascularized fibular graft in order to compensate bone defects of the long bones and mandibular inflicted by war wounding. The importance of the length of the vascular pedicle preparation for the total success was pointed out as well as the immobilization of the elbow region for the compensation of the defects of the upper arm and forearm and total healing of the mandibular for the later functional rehabilitation. The realized primary efficiency of 70.2% and total successfulness of 93.6% indicated that at the compensation on of bone defects the etiology of war wound could be compared to the etiology of malignant tumors resection and trauma.
