Skin sensitization with the new reagents NOPYE (4-nitro-1-cyclohexyl-3-ethoxy-2-oxo-3-pyrroline) and NOPY-L-phenylalanine.

The aim of this study was to investigate the compound 4-nitro-1-cyclohexyl-3-ethoxy-2-oxo-3-pyrroline (NOPYE) and some related compounds for skin sensitization in guinea pigs, as the first step in a search for more effective skin sensitizers for immunotherapy of cutaneous tumors. In guinea pigs, NOPYE and NOPYE-L-alanine produce far milder delayed hypersensitivity reactions than DNCB. Both NOPYE and DNCB fail to act as adjuvants for skin sensitization to tuberculin purified protein derivative (PPD) and ovalbumin (OV). This suggests an explanation for the lack of effectiveness of DNCB in immunotherapy of metastases: DNCB may be relatively ineffective as an adjuvant for production of specific antitumor immunity. Such adjuvant activity may be essential if the action of the immunotherapeutic reagent is not to be confined to its site of application but is to be effective at the site of distant metastases.

High-pressure ion source combined with an in-axis ion trap mass spectrometer. 1. Instrumentation and applications

A new combination of a dual EI/CI ion source with a quadrupole ion trap mass spectrometer has been realized in order to efficiently produce negative ions in the reaction cell. Analysis of volatile compounds was performed under negative ion chemical ionization (NICI) during a reaction period where selected reactant negative ions, previously produced in the external ion source, were allowed to interact with molecules, introduced by hyphenated techniques such as gas chromatography. The O2*-, CH3O-, and Cl- reactant ions were used in this study to ensure specific ion/molecule interactions such as proton transfer, nucleophilic displacement, or charge exchange processes, respectively leading to even-electron species, i.e., deprotonated [M - H]- molecules, diagnostic [M - R]- ions, or odd-electron M*- molecular species. The reaction orientation depends on the thermochemistry of reactions within kinetic controls. First analytical results are presented here for the trace-level detection of several contaminants under NICI/Cl- conditions. Phosphorus-containing compounds (malathion, ethyl parathion, and methyl parathion as representative for pesticides) and nitro-containing compounds (2,4,6-trinitrotoluene for explosive material) have been chosen in order to explore the analytical ability of this promising instrumental coupling.

Comparative metabolism of nitroaromatic compounds in freshwater, brackish water and marine decapod crustaceans.

1. The metabolic pathways of p-nitroanisole, 4-nitro-m-cresol and methyl parathion in Malaysian prawns (Macrobrachium rosenbergii), ridgeback prawns (Sicyonia ingentis) and crayfish (Procambarus clarkii) were compared. 2. The Malaysian prawns and ridgeback prawns were shown to O-demethylate 29% and 11%, respectively, of an accumulated level of p-nitroanisole, while crayfish were able to O-demethylate 98% of the accumulated level of nitroanisole. 4-Nitro-m-cresol oxidation was not detected in either prawn species. In contrast, 5-hydroxy-2-nitrobenzaldehyde was the major metabolite formed from nitrocresol metabolism by crayfish. 3. Both prawn species readily dearylated methyl parathion to form p-nitrophenol and p-nitrophenyl conjugates. Crayfish displayed a similar trend in methyl parathion metabolism. 4. Nitroreduction was observed in the metabolism of 4-nitro-m-cresol by ridgeback prawns, which excreted 4-nitroso-m-cresol as a minor product. Reduction products were not observed in the metabolism of the three substrates by Malaysian prawns or crayfish. 5. Conjugation was overall the dominant detoxication pathway observed in the decapods. Malaysian prawns conjugated p-nitrophenol and 4-nitro-m-cresol to form the corresponding beta-D-glucosides and sulphate monoesters. Ridgeback prawns formed beta-D-glucosides in small quantities, preferring conjugation of p-nitrophenol and 4-nitro-m-cresol to form sulphates and unknown conjugates through a unique conjugation pathway. Crayfish conjugated the phenolic substrates to form exclusively the beta-glucosides. 6. The unknown conjugates formed by ridgeback prawns had chromatographic properties similar to the corresponding beta-D-glucosides but were refractory to the deconjugation by alpha- or beta-glucosidase, beta-galactosidase, aryl sulphatase and beta-glucuronidase. Phenolic conjugation ability appeared to follow the order of ridgeback prawns greater than Malaysian prawns greater than crayfish.

The induction of cytochrome P4502E1 by nitrogen- and sulfur-containing heterocycles: expression and molecular regulation.

Several structurally related sulfur- and nitrogen-containing heterocycles including thiazole, pyrazine, pyridazine, pyrimidine, thiophene, and triazole, which are present in tobacco, tobacco smoke, and certain foods, have been employed with the goal of characterizing the effects of these agents on the inhibition and expression of P4502E1 in hepatic tissue and on the molecular level regulatory events governing enhanced expression. The results of this study reveal that whereas the binding constants of these compounds to 2E1 moderately correlated with the percentage inhibition of metabolic activity in vitro (r = 0.66), neither inhibition of metabolic activity nor binding to P4502E1 correlated with relative induction of P4502E1 levels (r = 0.07 and 0.03, respectively). Thiazole, which produced the greatest inhibition of metabolic activity (88%) and exhibited the highest binding affinity for P4502E1 (35 microM), induced P4502E1 approximately fourfold. In contrast, pyrazine and pyridazine, which only marginally inhibited metabolic activity (54 and 41%, respectively), and weakly bound 2E1 (73 and 384 microM, respectively), increased P4502E1 levels approximately four- and fivefold, respectively. A common feature associated with these inducers, however, was the substantial decrease in hepatic P4502E1 poly(A)+ RNA levels in treated animals relative to untreated animals. Slot and Northern blot hybridization analyses revealed an approximately 80% decrease in P4502E1 poly(A)+ RNA levels at 48 hr following treatment of rats with thiazole, and at 24 hr following treatment of animals with either pyrazine or pyridazine, relative to controls. P4502E1 poly(A)+ RNA levels appeared to increase gradually, returning to levels which approximated 60% of the P4502E1 poly(A)+ RNA levels present in untreated animals at 48 and 72 hr following treatment with pyrazine or pyridazine, respectively. The results of these experiments show that thiazole, pyrazine, and pyridazine induce P4502E1 in rats, that the induction of 2E1 is associated with a concomitant decrease in 2E1 poly(A)+ RNA levels, and that these agents differentially affect the expression of P4502E1.

Quantum chemical, ballistic and explosivity calculations on 2,4,6,8-tetranitro-1,3,5,7-tetraaza cyclooctatetraene: a new high energy molecule.

Ab initio molecular orbital calculations have been carried out on 2,4,6,8-tetranitro-1,3,5,7-tetraazacyclooctatetraene, the tetramer of the series (NO(2)CN)(n) where n=1-4, using the Hartree-Fock theory with the 6-31 G(d) basis set. These calculations yield three conformers for the tetramer with D(4h), C(4h) and C(2) symmetries. The nonplanar conformer with the C(2) symmetry turns out to be 99.0 and 164.4kJmol(-1), respectively, lower in energy than the C(4h) and D(4h) conformers. The electron density topography - the density at the bond critical point - has been used as a measure of the CNO(2) strengths. Based on these bond strengths, heats of formation [obtained from the parametric model 3 (PM3) method] and specific decomposition energies, it may be concluded that (NO(2)CN)(4) is a promising candidate in the class of high energy molecules. Theoretically computed explosive (velocity of detonation, detonation pressure, etc.) and ballistic (characteristic velocity, specific impulse, etc.) parameters support these conclusions.

Prediction of shock sensitivity of explosives based on small-scale gap test.

A new method is described to predict shock sensitivity of C(a)H(b)N(c)O(d) explosives without using any experimental data. It can determine shock sensitivity based on small-scale gap test as the pressure required to initiate material pressed to 90%, 95% and 98% of theoretical maximum density. Three essential parameters would be needed in the new scheme which contain a+b/2-d, the existence of alpha-C-H linkage in nitroaromatic compounds or NNO(2) functional group and difference of the number of amino and nitro groups attached to aromatic ring. Predicted shock sensitivities in some well-known explosives have a root mean square (rms) deviation of 3.97, 4.02 and 5.89kbar of experiment to initiate pressure of material pressed to 90%, 95% and 98% of theoretical maximum density, respectively.

Regioselective nucleophilic addition of triphenylphosphine to the nitrosylruthenium alkynyl complexes having a hydrotris(pyrazol-1-yl)borate: formation of phosphonio-alkenyl, alkynyl, and allenyl species.

A nitrosylruthenium alkynyl complex of TpRuCl(C[triple bond]CPh)(NO)(1a) was reacted with PPh3 in the presence of HBF4.Et2O at room temperature to give a beta-phosphonio-alkenyl complex (E)-[TpRuCl{CH=C(PPh3)Ph}(NO)]BF4(2.BF4). On the other hand, for gamma-hydroxyalkynyl complexes TpRuCl{C[triple bond]CC(R)2OH}(NO)(R = Me (1b), Ph (1c), H (1d)), similar treatments with PPh3 were found to give gamma-phosphonio-alkynyl [TpRuCl{C[triple bond]CC(Me)2PPh3}(NO)]BF4(3.BF4),alpha-phosphonio-allenyl [TpRuCl{C(PPh3)=C=CPh2}(NO)]BF4(4.BF4), and a novel product of gamma-hydroxy-beta-phosphonio-alkenyl (E)-[TpRuCl{CH=C(PPh3)CH2OH}(NO)]BF4(5.BF4), respectively. Dominant factors for the selectivity in affording 3-5 were associated with the steric congestion and electronic properties at the gamma-carbons, along with those around the metal fragment. From the bis(alkynyl) complex TpRu(C[triple bond]CPh)2(NO)6, a bis(beta-phosphonio-alkenyl)(E,E)-[TpRu{CH=C(PPh3)Ph}2(NO)](BF4)2{7.(BF4)2} was produced at room temperature. However, similar reactions at 0 degrees C gave an alkynyl beta-phosphonio-alkenyl complex (E)-[TpRu(C[triple bondCPh){CH=C(PPh3)Ph}(NO)]BF4(8.BF4) as a sole product, of which additional hydration in the presence of HBF4.Et2O afforded a [small beta]-phosphonio-alkenyl ketonyl (E)-[TpRu{CH2C(O)Ph}{CH=C(PPh3)Ph}(NO)]BF(.9BF4). Five complexes, 2-5 and 7 were crystallographically characterized.
