A facile formation of 2,5-anhydro sugars by ring contraction in methyl hexopyranoside 2-triflates under conditions of nucleophilic displacement.

Reaction of methyl 3,4-O-isopropylidene-2-O-(trifluoromethylsulfonyl)-alpha-L-fucopyr anoside and its beta anomer with a variety of nucleophilic reagents under mild conditions led to displacement of the triflic ester group, with migration of the ring-oxygen atom to C-2 and entry of the nucleophile at C-1, to give good yields of 2,5-anhydro sugar derivatives. Reagents employed were hydrogen fluoride-triethylamine complex, methanol in the presence of sodium hydrogencarbonate, sodium benzoate, sodium azide, potassium thiocyanate, and sodium borohydride. The same type of substitutive ring-contraction also occurred in methyl 4-O-benzyl-3-O-(4-methoxybenzyl)-2-O-(trifluoromethylsulfonyl)-alp ha-L- fucopyranoside and methyl 4,6-dideoxy-3-O-(4-methoxybenzyl)-2-O-(trifluoromethylsulfonyl)-al pha-L-xylo- hexopyranoside. The reaction is discussed in light of a literature survey of the chemical behavior of hexopyranoside 2-sulfonates in general, and 2-triflates in particular. Direct SN2 displacements, eliminations, and such different kinds of rearrangement as have previously been observed with structural analogs were not encountered in the present study. However, there is some precedent, in hexopyranoside 2-triflates, for the facile rearrangement that the four representatives here investigated have been found to undergo. The synthesis of these triflates from L-fucose is described.

Green tea polyphenols and tannic acid act as potent inhibitors of phorbol ester-induced nitric oxide generation in rat hepatocytes independent of their antioxidant properties.

The deleterious effects of excessive release of nitric oxide (NO) have been implicated in the tissue damage and inflammation. In this study, the effect of various flavonoids and other oxidant scavenging chemical agents have been studied for their ability to inhibit 12-O-tetradecanoyl phorbol 13-acetate (TPA)-induced NO generation in rat hepatocyte. Hepatocytes activated with TPA (25-200 nM) released NO in a concentration- and time-dependent manner. Green tea polyphenols (GTP) and tannic acid (TA) were most effective in inhibiting TPA-induced NO generation (90%). These agents were also effective in inhibiting NO formation when added 2 h following TPA addition. The other oxidant scavengers, such as L-histidine, sodium azide, vitamin E and sodium benzoate, were not found to be effective even up to 1.0 mM concentration. These results suggest that TA and GTP are potent inhibitors of NOS activity and the inhibition of TPA-induced NO generation by these polyphenols is independent of their antioxidant activity. It is tempting to speculate that these agents could be utilized in the pharmacological manipulations of NO-dependent pathophysiological responses.

Comparative first-principles study of structural and optical properties of alkali metal azides.

A comparative first-principles study of the structural and optical properties of the alkali metal azides has been performed with density functional theory within the generalized gradient approximation. The crystal structures of the alkali azides compare well with experimental data. Their ionic character is manifested by the closeness of their internitrogen distances to the calculated N-N bond length for the free azide ion. An analysis of electronic structure, charge transfer, and bond order shows that the alkali azides are all wide-gap insulators and ionic compounds. The energy band and density of states for lithium azide and alpha-sodium azide are very similar, while these for potassium azide, alpha-rubidium azide, and alpha-cesium azide are alike, but some modifications are observed with the increment of alkali metals' electropositivity. These changes are closely related to the differences of the crystal structures. The general shapes of the real and imaginary parts of the dielectric function, adsorption coefficient, and electron energy-loss spectra are quite similar. The peaks originate from the electron transitions from the alkali metal s and p states to the conduction band. Our calculated optical properties for the alkali azides are found to be in good agreement with available experimental data. The absorption spectra of the alkali azides show a number of absorption peaks, which are believed to be associated with different exciton states, in the fundamental absorption region. In general, the electron energy-loss spectra have two plasma frequencies.

An investigation of the post-tetanic potentiation of end-plate potentials at a mammalian neuromuscular junction.

1. End-plate potentials (e.p.p.s) were recorded intracellularly from neuromuscular junctions in curarized or Mg-paralysed rat diaphragm-phrenic nerve preparations in vitro. In Mg-paralysed preparations after 1000 impulses at 100/sec the amplitude of e.p.p.s elicited at 1/sec before and after the tetanus was on average greater than the control amplitude for 120 +/- 30 sec.2. The post-tetanic potentiation (P.T.P.) of e.p.p. amplitudes was not thought to be dependent upon post-tetanic hyperpolarization (P.T.H.) of nerve terminals as it lasted longer than the hyperpolarization generated by an identical tetanus; was unaffected by hyperpolarizing currents which reduced P.T.H. or depolarizing currents which prolonged P.T.H.; and was diminished in solutions containing 30% of the normal NaCl concentration or 1% ethyl alcohol, both of which procedures prolong P.T.H. The magnitude and duration of P.T.P. were influenced by the pH of the bathing solution in the range 7-7.5 although there was no change in P.T.H. under these conditions. The inability of polarizing currents to influence P.T.P. was also thought inconsistent with the hypothesis that P.T.P. is due to an increase in available transmitter.3. P.T.P. was not thought to be due to sodium accumulation in nerve terminals, for P.T.P. was reduced or abolished by procedures which would be expected to increase the intraterminal sodium ion concentration. These procedures were: exhibition of metabolic inhibitors (1.8 x 10(-6)M antimycin A, 3-5 mM sodium azide or 1 mM sodium iodoacetate), exhibition of cardiac glycosides (7.7 x 10(-6)M digoxin or 0.42 mM ouabain), and omission of glucose or potassium ions from the bathing solution. Abolition of P.T.P. by potassium-free solutions was also thought to be inconsistent with the hypothesis that P.T.P. is due to a reduction in the potassium concentration in nerve terminals.4. P.T.P. was not thought to be due to terminal volume changes, for no consistent effect upon the quantal content of e.p.p.s could be detected in hypo- or hyperosmotic solutions.5. It was concluded that the only hypothesis for P.T.P. not excluded by our experiments was that P.T.P. is due to some change in ionized calcium at a membrane site important in transmitter release.

Partial characterization of mechanism of insulin accumulation in H35 hepatoma cell nuclei.

The mechanism controlling insulin accumulation in nuclei of H35 hepatoma cells was investigated by incubating intact cells with 125I-labeled insulin in the presence or absence of agents that perturb different intracellular sites involved in the processing of ligand-receptor complexes. Purified nuclei were isolated, and nuclear-associated 125I-insulin was determined. Insulin accumulation in the nuclei was time and temperature dependent. Nuclear accumulation was linear and insulin-concentration dependent between 5 and 50 ng insulin/ml. However, pharmacological concentrations of insulin increased the amount of insulin translocated to the nucleus to a far greater extent than it increased total cell-associated insulin. Chloroquine, an acidotrophic agent, increased total cell-associated and intracellular insulin but had no effect on nuclear accumulation. The monovalent ionophores monensin and nigericin inhibited nuclear accumulation of insulin at low concentrations (0.5-5.0 microM) without affecting total insulin binding or intracellular accumulation. At 10 or 25 microM, monensin and nigericin also acted as acidotrophic agents and increased total insulin binding and intracellular accumulation but inhibited nuclear accumulation by a maximum of 50%. Low concentrations of monensin and nigericin were additive; maximal concentrations were not. A 23187 and valinomycin did not affect insulin binding or intracellular and nuclear accumulation of insulin. Neither depletion of ATP by sodium azide, 2,4-dinitrophenol, sodium cyanide, or oligomycin nor disruption of cytoskeletal elements by cytochalasin D or colchicine had any effect on nuclear accumulation of insulin.(ABSTRACT TRUNCATED AT 250 WORDS)

A modified medium for the recovery of Staphylococcus from water.

Various contemporary methods available for recovering staphylococci from water were evaluated as to quantitative recovery and degree of selectivity. Each of the methods tested was found to be somewhat inadequate for the selective isolation of staphylococci from water in this area. A modified medium is being proposed which incorporates 0.005% sodium azide into a mannitol salt agar base. This medium when used with a membrane filter procedure and incubated at 32 degrees C for 48 h recovers 85% of Staphylococcus aureus, 88% of Staphylococcus epidermidis, and 100% of Staphylococcus saprophyticus present. Environmental testing in this area indicates that this modified procedure will recover only Gram-positive, catalase-positive cocci. Ninety percent of these cocci are Staphylococcus as ascertained by the lysostaphin susceptibility test.

Increased intraocular pressure following topical azide or nitroprusside.

In rabbits the topical administration of sodium azide (NaNs) or sodium nitroprusside (SNP) increased intraocular pressure in a dose-response manner. These agents, which activate guanylate cyclase, elevated cyclic GMP in the aqueous humor. Systemic blood pressure and pulse were not altered. Tonographic outflow facility was unchanged, suggesting an increase in aqueous humor flow as the mechanism for the elevation of intraocular pressure. Posterior chamber aqueous humor ascorbate concentration was decreased in the eye receiving the NaN3 or SNP. Systemic pretreatment with phenoxybenzamine, an alpha-adrenergic blocking agent, prevented the elevation of intraocular pressure observed following NaN3 and SNP. Pretreatment with systemic indomethacin, propranolol, or acetazolamide or the topical application of atropine or epinephrine failed to alter the elevation of intraocular pressure by either NaN3 or SNP.

Shedding of antibody complexes by Strongyloides ratti (Nematoda) larvae.

Antigens on the epicuticular surface of Strongyloides ratti infective third-stage larvae (L3) could be demonstrated by an indirect fluorescent antibody technique under certain conditions. Infective L3 shed anti-antibody complexes at room temperature, but not at 4 C or in the presence of sodium azide or colchicine. Shedding of antibody did not appear to involve epicuticular antigens, and only occurred when anti-rat IgG was complexed to rat anti-larval antibody. However, parasitic L3 removed from rats did not exhibit this shedding reaction, suggesting that an important developmental change in cuticle physiology occurs during the transition from a free-living existence to a parasitic mode. The ability to shed foreign objects from the epicuticle of free-living infective L3 may be a defensive or protective response to soil microorganisms.

Azide resistance in Rhizobium ciceri linked with superior symbiotic nitrogen fixation.

Isolated azide resistant (AzR) native R. ciceri strain 18-7 was resistant to sodium azide at 10 microg/ml. To find if nif-reiteration is responsible for azide resistance and linked to superior symbiotic nitrogen fixation, transposon (Tn5) induced azide sensitive mutants were generated. Using 4 kb nif-reiterated Sinorhizobium meliloti DNA, a clone C4 that complemented azide sensitivity was isolated by DNA hybridization from genomic library of chickpea Rhizobium strain Rcd301. EcoRI restriction mapping revealed the presence of 7 recognition sites with a total insert size of 19.17 kb. Restriction analysis of C4 clone and nif-reiterated DNA (pRK 290.7) with EcoRI and XhoI revealed similar banding pattern. Wild type strain 18-7, mutant M126 and complemented mutant M126(C4) were characterized for symbiotic properties (viz., acetylene reduction assay, total nitrogen content, nodule number and fresh and dry weight of the infected plants) and explanta nitrogenase activity. Our results suggested that azide resistance, nif-reiteration, and superior symbiotic effectiveness were interlinked with no correlation between ex-planta nitrogenase activity and azide resistance in R. ciceri.

Absence of mitochondrial beta-adrenoceptors in guinea pig myocardium: evidence for tissue disparity.

1. Binding sites in guinea pig myocardial tissue labelled by (-)-[125I] cyanopindolol (CYP) were investigated using differential centrifugation and autoradiographic techniques. Autoradiographs of myocardial sections (0.1 microns) indicated (-)-[125I]CYP binding to sarcolemmal membrane. A low density of binding sites was observed to mitochondria. 2. Binding studies were performed in subcellular fractions. The density of binding sites in the mitochondrial fraction (36.1 +/- 9.4 fmol/mg protein) was less than 10% that in the sarcolemmal membrane (371.7 +/- 38.2 fmol/mg protein). The beta 1/beta 2-adrenoceptor subtype ratio in the mitochondrial fraction (83.3/16.7) was similar to that in the sarcolemmal fraction (87.1/12.9). 3. Ouabain (100 microM), in the presence of sodium azide (0.4 mM), inhibited a Na+K+ stimulated ATPase activity (1.0 +/- 0.2 mumol Pi/mg protein/hr reduction), indicating a low but significant level of sarcolemmal contamination of the mitochondrial fraction. 4. The study showed beta-adrenoceptors in guinea pig heart are located primarily on the sarcolemmal membrane of myocardium. No evidence was obtained for beta-adrenoceptors over mitochondria, as has been suggested in other tissues and species, but that this binding was to sarcolemmal inclusions in the mitochondrial fraction.

Induction by glucose of an antimycin-insensitive, azide-sensitive respiration in the yeast Kluyveromyces lactis.

Increasing the glucose concentration from 0.1 to 10% in exponentially growing cultures of Kluyveromyces lactis CBS 2359 does not repress the antimycin-sensitive respiration (QO2 of 80 microliter O2 . h-1 . mg-1 dry weight) but raises the antimycin-insensitive respiration from 3 to 12 microliter O2 . h-2 . mg-1 dry weight. Antimycin A inhibits the growth of K. lactis on a variety of substrates with the exception of glucose at concentrations equal to or higher than 1% where substantial antimycin-insensitive respiratory rates are induced. It can be concluded that a minimal antimycin-insensitive QO2 is necessary for cellular growth when the normal respiratory pathway is not functional. The antimycin-insensitive respiration elicited by growth in high glucose concentrations is poorly inhibited by hydroxamate and is inhibited by 50% by 90 microM azide or 1 mM cyanide. These concentrations are much higher than those necessary to inhibit cytochrome c oxidase which is not involved in the antimycin-insensitive respiration as was demonstrated by spectral measurements. A pigment absorbing at 555 nm is specifically reduced after addition of glucose to antimycin-inhibited cells. The same pigment is reoxidized by further addition of high concentrations of sodium azide indicating its participation in the antimycin-insensitive, azide-sensitive respiration.

2,4-Dichlorophenoxyacetic acid influx is mediated by an active transport system in Chinese hamster ovary cells.

The influx of 2,4-dichlorophenoxyacetic acid (2,4-D) into Chinese hamster ovary (CHO) cells was studied. The cells mainly took up but did not metabolize the undissociated form of the herbicide. The uptake of 2,4-D was carried out against a concentration gradient and was inhibited by sodium azide and dinitrophenol. The results presented here show that the herbicide influx was an active, energy dependent process. (Na+ + K+)ATPase does not seem to be involved because ouabain, an inhibitor of the enzyme, did not affect the 2,4-D uptake.

[Antimutagenicity of propionic acid bacteria].

The antimutagenicity of the cell extracts of Propionibacterium shermanii VKM-103, P. pentosaceum CCM 1859 and P. acnes CCM 3322 against mutagenicity of sodium azide and N-methyl-N'-nitro-N-nitrosoguanidine was demonstrated for the first time. The extracts of propionic acid cocci didn't show such effect. The antimutagenic factor acts as a desmutagen, has polypeptide nature and evidently is an enzyme (enzymes). The inhibitory effect of the extract is due to the presence of more than one protein factor in it.

Effect of glutathione L-cystein and L-djenkolic acid in the synthesis and mutagenicity of azide metabolite in Bacillus subtilis ATCC 6633 strain.

The Bacillus subtilis ATCC 6633 strain synthesizes a mutagenic metabolite from sodium azide and O-acetylserine. Mutagenicity of azide was decreased in growth media containing 10(-4) M glutathione, L-cysteine or L-djenkolic acid whereas dithiothritol (DTT) added at the same concentration did not reduce the mutagenicity of azide. Likewise, glutathione, L-cysteine, L-djenkolic acid, and DTT were found to have no effect in reducing the mutagenicity of the in vitro produced metabolite using bacterial cell-free extract. These results suggest that O-acetyl-serine sulfhydrylase catalyzes the reaction of azide and O-acetylserine to form a mutagenic metabolite, which is ninhydrin positive and migrates in TLC to an Rf value similar to that of azidoalanine in both acidic and basic solvent systems.

Ecto-nucleotidase activities in the fat body of Rhodnius prolixus.

In this study, we describe the ability of intact fat body of an insect, Rhodnius prolixus, to hydrolyze extracellular ATP. In these fat bodies, the ATP hydrolysis was low in the absence of any divalent metal, and was stimulated by MgCl(2). Both activities (in the absence or presence of MgCl(2)) were linear with time for at least 30 min. In order to confirm the observed nucleotidase activities as ecto-nucleotidases, we used an impermeant inhibitor, DIDS (4, 4'-diisothiocyanostylbene 2'-2'-disulfonic acid). This reagent inhibited both nucleotidase activities and its inhibitory effect was suppressed by ATP. Both ecto-nucleotidase activities were insensitive to inhibitors of other ATPase and phosphatase activities, such as oligomycin, sodium azide, bafilomycin, ouabain, vanadate, molybdate, sodium fluoride, levamizole, tartrate, p-NPP, sodium phosphate, and suramin. Concanavalin A, activator of some ecto-ATPases, was able to stimulate the Mg(2+)-independent nucleotidase activity, but not the Mg(2+)-dependent one. The Mg(2+)-independent nucleotidase activity was enhanced with increases in the pH in the range between 6.4-8.0, but the Mg(2+)-dependent nucleotidase activity was not affected. Besides MgCl(2) , the ecto-ATPase activity was also stimulated by CaCl(2),() MnCl(2), and SrCl(2), but not by ZnCl(2). ATP, ADP, and AMP were the best substrates for the Mg(2+)-dependent ecto-nucleotidase activity, and CTP, GTP, and UTP produced very low reaction rates. However, the Mg(2+)-independent nucleotidase activity recognized all these nucleotides producing similar reaction rates, but GTP was a less efficient substrate. The possible role of the two ecto-nucleotidase activities present on the cell surface of fat body of Rhodnius prolixus, which are distinguished by their substrate specificity and their response to Mg(2+), is discussed.

Production and characterization of a thermostable chitinase from a new alkalophilic Bacillus sp. BG-11.

An alkalophilic, environmental micro-organism, Bacillus sp. BG-11, has been isolated and characterized. It produced 76 U ml-1 of chitinase in liquid batch fermentation after 72 h of incubation at 50 degrees C using chitin-enriched medium. The molecular weight of purified chitinase was estimated to be 41 kDa by SDS-PAGE. The pH and temperature optima of chitinase immobilized on chitosan and calcium alginate were 8.5 and 50 degrees C, respectively, which were same as that of free enzyme. The pH and thermostability of immobilized chitinase were enhanced significantly. The chitinase immobilized on chitosan was stable between pH 5.0 and 10.0, and the half-life of chitosan-immobilized enzyme at 70, 80 and 90 degrees C was 90, 70 and 60 min, respectively. The end-products formed during the enzyme-substrate reaction were identified by 13C-NMR, and N-acetyl-D-glucosamine was found to be the major end-product. GlcNAc (GlcNAc)2 and (GlcNAc)3 inhibited the chitinase activity by 32, 25 and 18%, respectively, at a concentration of 10 mmol l-1. The shelf-life of chitinase (retained 100% activity) at 4 degrees C was 8 weeks in the presence of either sodium azide (100 microgram ml-1), sodium metabisulphite (0.1% w/v) or KCl (15% w/v). The enzyme was resistant to the action of proteases and allosamidin.
