The role of dermal irritation in the skin tumor promoting activity of petroleum middle distillates.

Petroleum middle distillates (PMDs), a class of hydrocarbons which boil between 350-700 degrees F, are tumor promoters in mouse skin. The promotional activity is produced under conditions that also result in local changes, including chronic irritation and epidermal hyperplasia. The present study was conducted by comparing equal weekly doses of irritating and minimally or nonirritating test materials, to assess whether tumor promotion was a secondary response to these effects. Four PMDs, C10-C14 normal paraffins (NP), lightly refined paraffinic oil (LRPO), Jet Fuel A (JF), and steam-cracked gas oil (SCGO), were evaluated. Test materials were applied undiluted (2x/week) or as 28.6% (7x/week) or 50% (4x/week) concentrations in mineral oil for 52 weeks following initiation with dimethylbenzanthracene (DMBA). When applied undiluted, all materials produced moderate irritation and significant increase in tumor incidence. When NP, LRPO, or JF were applied in mineral oil diluent, skin irritation was generally ameliorated and few, if any, tumors were produced. SCGO was irritating and produced a significant increase in tumor frequency when administered in mineral-oil diluent. These data indicate that the promotional activity of straight-run PMDs is likely related to chronic irritation at the application site and not to dose. Thus, when used appropriately in the absence of prolonged irritation, these materials should not present a tumorigenic hazard to humans.

Cytotoxic effects of dental desensitizers on human gingival fibroblasts.

The purpose of this study was to evaluate the effects of three different desensitizers on the cell viability and morphology of human gingival fibroblasts (HGF). Human gingival tissues were obtained from individuals who have clinically, healthy periodontium. HGF were grown at 37 degrees C in humidified atmosphere of 5% CO2 in Dulbecco's modified eagle's medium, supplemented with glutamine, penicillin, streptomycin, and 10% fetal bovine serum. The cells were treated with different concentrations (0.1, 0.3, and 0.5 microL/mL) of desensitizers (Gluma Desensitizer, Seal&Protect, and MicroPrime). After 24- and 48-h exposure to the desensitizer solutions, the viable cells were examined using a hemocytometer. To monitor HGF viability, 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl tetrazolium bromide (MTT) colorimetric assay was used and cell morphology was also observed at 48 h. Following exposure to concentrations of 0.1 microL/mL of test materials for 24 h, cell survival rates for Gluma Desensitizer (106%) and Micro Prime (62%) were not significantly different from the control, while it was significant for Seal&Protect (50%). Growing cells were significantly inhibited by all tested materials for 48 h (p < 0.05) in survival rates of 51, 47, and 31%, respectively. On the basis of the MTT assay, the cytotoxic effect of MicroPrime was more prominent, especially at high concentrations, than does Gluma Desensitizer and Seal&Protect. After exposure to Seal&Protect and MicroPrime, HGF became retracted, rounded in appearance and had loss of normal organization, leading to enlargement of intercellular space when compared with Gluma Desensitizer. As a conclusion, taking the limitations of an in vitro experiment into consideration, the cytotoxic effects were varied, depending on the chemical composition and exposure periods of the tested desensitizers.

The bond strengths of resin systems to etched enamel.

This study was undertaken to compare the resin-enamel bond strengths of seven anterior restorative resins using the acid-etch technique. Six of the materials (Restodent, Enamelite, Smile, Nuva-System, Concise, and Adaptic) were composite resins. Only one, Sevriton, was an unfilled resin. Of the seven, only Sevriton and Smile were not designed for the acid-etch technique. Of the materials tested, Sevriton and Enamelite had the weakest bonds. There was no significant difference in the bond strength among Nuva-System, Smile, and Restodent; the bonds of all these were stronger than those of Sevriton and Enamelite. Concise showed a significantly greater bond strength than all the other materials in the study except Adaptic and the Nuva-System. The resin-enamel bond strength of Adaptic was of a greater magnitude than that of all other test materials.

Comparison of synthetic zeolite catalysts and alumina binders administered intratracheally to rats.

An intratracheal (IT) screening assay was performed in rats on a series of aluminosilicate catalysts (synthetic zeolites) and alumina binders to compare their relative ability to cause pulmonary fibrosis and related changes. Before initiation of IT screens, both the uniformity of deposition and residence time of a prototype catalyst in the lung were determined. Subsequently, the test materials were instilled and animals were evaluated 6 mo later for lung volumes, pulmonary pressure-volume curves, pulmonary hydroxyproline (OHPro) content, lung weights, and histopathology. Negative controls were saline and glass beads; the positive control was quartz. The test materials were organic-free ZSM-5 crystals, organic form of ZSM-5 crystals, alumina-bound ZSM-5, nickel/ZSM-5/ Al2O3 binder, used nickel/ZSM-5/Al2O3 binder, nickel-tungsten/ZSM-5/Al2O3 binder, and Bayer pseudoboehmite. For each, groups of 12 male rats were dosed once with either 25 or 50 mg of ground particles (> or =95% less than 2.4 microm). Second to quartz, the sample of alumina caused the most pronounced pulmonary reactions at 6 mo after dosing. Therefore, the effects of four aluminas were subsequently compared (Bayer pseudoboehmite, Bayer gammaAl2O3, Ziegler pseudoboehmite, and Ziegler gammaAl2O3). The results support the idea of a lack of long-term effects from exposure to normal concentrations of these materials in the workplace. Also, the IT assay proved to be a very useful tool for ranking the relative effects of this series of zeolites and aluminas.

Fracture of the acetabulum during insertion of an oversized hemispherical component.

A hemispherical metal-backed acetabular component that was oversized by two or four millimeters was introduced without cement into thirty fresh or embalmed cadaveric acetabula that were fifty, fifty-two, fifty-four, fifty-six, or fifty-eight millimeters in diameter. Manual tapping was used to insert the component into ten fresh and ten embalmed specimens, and a materials testing machine was used for the remaining five fresh and five embalmed specimens. The magnitude of force and the number of pulses necessary to seat the component were determined for the insertions performed with the materials testing machine; 2000 and 3000 newtons of force were required to insert the cups that were oversized by two and four millimeters, respectively. Visual and radiographic assessments were used to identify any resulting acetabular fractures. Over-all, eighteen (60 per cent) of the thirty acetabular specimens fractured. Of the eighteen fractures, only fifteen were detectable radiographically: ten on en face or lateral radiographs of the cup and five only on oblique radiographs of the cup. The remaining three fractures were apparent only after the removal of the acetabular component.(ABSTRACT TRUNCATED AT 250 WORDS)

Defining regional variation in the material properties of human rib cortical bone and its effect on fracture prediction.

This paper presents the results of dynamic material tests and computational modeling that elucidate the effects of regional rib mechanical properties on thoracic fracture patterns. First, a total of 80 experiments were performed using small cortical bone samples from 23 separate locations on the rib cages of four cadavers (2 male, 2 female). Each specimen was subjected to dynamic three-point bending resulting in an average strain rate of 5 +/- 1.5 strain/s. Test coupon modeling was used to verify the test setup. Regional variation was defined by location as anterior, lateral, or posterior as well as by rib level 1 through 12. The specimen stiffness and ultimate stress and strain were analyzed by location and rib level. Second, these material properties were incorporated into a human body computational model. The rib cage was partitioned into anterior, lateral, and posterior segments and the material properties were varied by location using an elastic-plastic material model. A total of 12 simulations with a rigid impactor were performed including 2 separate material assumptions, original and modified rib properties for regional variations, 3 separate impactor velocities, and 2 directions, anterior and lateral. The data from the material tests for all subjects indicate a statistically significant increase in the average stiffness and average ultimate stress for the cortical bone specimens located in the lateral (11.9 GPa modulus, 153.5 MPa ultimate stress) portion of the ribs versus the anterior (7.51 GPa, 116.7 MPa) and posterior (10.7 GPa, 127.7 MPa) rib locations. In addition, the stiffness, ultimate stress, and ultimate strain for all subjects are significantly different by rib level with each variable generally increasing with increasing rib number. The results from the computational modeling for both frontal and lateral impacts illustrate that the location and number of rib fractures are altered by the inclusion of rib material properties that vary by region.

Depth profiling of laser-heated chromophores in biological tissues by pulsed photothermal radiometry.

A solution method is proposed to the inverse problem of determining the unknown initial temperature distribution in a laser-exposed test material from measurements provided by infrared radiometry. A Fredholm integral equation of the first kind is derived that relates the temporal evolution of the infrared signal amplitude to the unknown initial temperature distribution in the exposed test material. The singular-value decomposition is used to demonstrate the severely ill-posed nature of the derived inverse problem. Three inversion methods are used to estimate solutions for the initial temperature distribution. A nonnegatively constrained conjugate-gradient algorithm using early termination is found superior to unconstrained inversion methods and is applied to image the depth of laser-heated chromophores in human skin.

Disruptive effects of auditory signal delay on speech perception with lipreading.

The effect of auditory signal delay on audio-visual perception of videotaped sentence lists by Ss with normal hearing was measured. The test material consisted of sentences read by a man, with or without a picture representing as context one of the key words in each sentence. In Exper. I, 12 Ss (6M, 6F), inexperienced with lipreading, were presented test material in the audio-only (A), visual-only (V) and audio-visual (AV) modes. Speech signals were presented at 0 or -10 db S/N re a multitalker babble fixed at 60 dbA. The audio signal was delayed (re visual signal) in 6 steps from 0-300 msec. Exper. II repeated Exper. I with 6 Ss (2M, 4F) familiar with lipreading; the speech signal was presented at -5 db S/N and 4 audio delays from 0-240 msec. While these Ss performed better in the V mode than those in Exper. I, the main effects of context and delays were similar. The disruptive effect of the audio delay on speech perception with lipreading (AV mode) was a function of S/N, being relatively more disruptive at the worse S/N, but it was not significant for delays up to a range of 80-120 msec. The results are in agreement with the theory (McGrath and Summerfield, J. Acoust. Soc. Am., 1985, 77, 678-685) that sensitivity to audio-visual desynchrony is significant only at a syllabic level in connected speech. The results further imply that moderate delays of up to 80 msec introduced by speech-processing aids for lipreading, as for cochlear-implanted patients, will not interfere with the advantages of providing the auditory information.

Bioassay of heptachlor for possible carcinogenicity.

A bioassay of technical-grade heptachlor for possible carcinogenicity was conducted by administering the test material in feed to Osborne-Mendel rats and B6C3F1 mice. Groups of 50 rats of each sex were administered low or high concentrations of the heptachlor for 80 weeks, then observed for 30 weeks. Doses for females were first increased, but because of toxic effects the doses were then reduced twice for both male and female rats during the remaining course of the tests. Time-weighted average doses used for the male rats were 38.9 and 77.9 ppm; for the females, 25.7 and 51.3 ppm. Matched controls consisted of groups of 10 untreated rats of each sex; pooled controls consisted of the matched-control groups combined with 50 untreated male and 50 untreated female rats from similar bioassays of five other compounds. All surviving rats were killed at 110-111 weeks. Groups of 50 mice of each sex were administered the test material at low or high concentrations for 80 weeks, then observed for 10 weeks. The low- and high-dose groups were tested at different calendar times, but a concurrent control group was started with each. Because of toxic effects, doses were reduced once for the males at 17-18 weeks after the initiation of tests; twice for the females, at 17 and 30 weeks, after the initiation of tests. The time-weighted average doses used for the male mice were 6.1 and 13.8 ppm; for the females, 9 and 18 ppm. Matched controls consisted of groups of 10 untreated mice of each sex; pooled controls consisted of the matched-control groups combined with 90 untreated male and 70 untreated female mice from similar bioassays of five other compounds. All surviving mice were killed at 90-91 weeks. The effects of heptachlor on body weights and other clinical signs in rats and mice indicated that the dosages used were near the maximum permissible. This was evident in that average body weights of rats treated with high doses were consistently lower than those of untreated controls, while body weights of low-dose rats were unaffected. Body weights of mice given either high or low doses showed little or no differences from those of control mice; however, other adverse clinical signs were found in high-dose mice, predominantly in the females. The effects of heptachlor on survival rates indicated that mortality was dose-related for both female rats and female mice, but not for males of either species. However, a substantial proportion of all groups of animals survived to an age at which tumors could be expected to appear. In mice, hepatocellular carcinoma showed a highly significant dose-related trend in both males (matched controls 5/19, low dose 11/46, high dose 34/47, P=0.001) and females (control 2/10, low dose 3/47, high dose 30/42, P<0.0001). When pooled controls were used for the comparison, the significance of the trend in males increased to P<0.0001. Comparably high levels of significance were attained when the data were subjected to life-table adjustment. No other tumors were found in mice in sufficient numbers to justify analysis. In marked contrast to the findings observed in mice, no hepatic tumors were observed in rats administered heptachlor. There was significant statistical evidence for the induction of proliferative lesions of follicular cells of the thyroid in treated female rats, but this finding was discounted because the rates of incidence were comparatively low and are known to be variable in control rat populations. It is concluded that under the conditions of this bioassay, heptachlor is carcinogenic for the liver in mice.

About variability of Vieussen valve in the adult human heart.

The Vieussen valve is situated at the ostium of the great cardiac vein to the coronary sinus. There are no details about its shape in anatomic literature. The tested material consisted of 150 adult human hearts of both sexes from 18 to 85 years of age, fixed in a formalin/ethanol solution. Classical macroscopic anatomical methods were used. The Vieussen valve was found in about 65% of the tested material. It showed a large variability in terms of morphology.

[Studies to the in-vitro-effectiveness of Spectacillin (author's transl)].

In this study the sensibility of important disease germs of a clinical and an ambulatory test material has been investigated to the substances "Epicillin" and "Ampicillin". Thereby could be found that 67% (43%) of the E. coli and 55% (36%) of the proteus type, as well as 96% (96%) of the enterococcus type of the ambulatory (clinical) test material were sensitive to Epicillin and Ampicillin. Staphylococcus not producing penicillinase, were well-sensitive, while staphylococcus producing penicillinase, contrarily showed to be insensitive. Salmonellae proved to be well-sensitive up to 97%, and the rest left was limited sensitive. The minimal inhibitory concentration (MIC) resulted in favourable findings for enterococcus, staphylococcus aureus and salmonellae; at a corresponding sensibility the same is valid for E. coli and the proteus. The investigations carried out in-vitro, reveal an equal effectiveness of Epicillin and Ampicillin.

Subchronic inhalation toxicity of amorphous silicas and quartz dust in rats.

The inhalation toxicity of three amorphous silicas (Aerosil 200, Aerosil R 974 and Sipernat 22S) was compared with that of quartz dust. Rats were exposed to 1, 6 or 30 mg Aerosil 200/m3, 30 mg Aerosil R 974/m3, 30 mg Sipernat 22S/m3 or 60 mg quartz/m3 for 6 hr/day, 5 days/wk for 13 wk. Some rats were killed at the end of the exposure period and some were killed 13, 26, 39 or 52 wk after the end of exposure. Clinical signs, body weight, haematology, biochemistry, urinalyses, organ weights, retention of test material in the lungs and regional lymph nodes, collagen content of the lungs, and gross and microscopic pathology were determined in order to disclose possible adverse effects and to study the reversibility, stability or progression of the effects. All test materials induced increases in lung weight, and pulmonary lesions such as accumulation of alveolar macrophages, inflammation, alveolar bronchiolization and fibrosis. In addition, rats exposed to Aerosil 200, Aerosil R 974 or quartz developed granulomatous lesions. Silicosis was observed only in quartz-exposed animals. At the end of the exposure period, Aerosil 200 and quartz had induced the most severe changes. Quartz dust was hardly cleared from the lungs and the changes in the lungs progressed during the post-treatment period, and eventually resulted in lesions resembling silicotic nodules and in one squamous cell carcinoma. Although Aerosil 200 was very quickly cleared from the lungs and regional lymph nodes, the changes in these organs were only partly reversed during the post-exposure period in rats exposed to 30 mg/m3. Aerosil R 974 and the lower levels of Aerosil 200 resulted in less severe, and mostly reversible, changes. The slightest changes were found after exposure to Sipernat 22S, notwithstanding the persistence of this silica in the lungs during the major part of the post-treatment period. The results of this study revealed that only quartz induced progressive lesions in the lungs resembling silicotic nodules. Of the amorphous silicas examined Aerosil 200 induced the most severe changes in the lungs, which only partly recovered, whereas Sipernat 22S induced the least severe, completely reversible lung changes.

Cytotoxicity of a new endodontic filling material.

In vitro cell cultures have been widely used as a means of evaluating cytotoxicity of root canal filling materials. Following ANSI/ADA spec. no. 41, the aim of the present study was to investigate the biological compatibility of a new sealer (FibreFill) and compare it with some commercially available endodontic sealers (Bioseal and Acroseal). Mouse 3T3 fibroblasts were seeded and cultured and subsequently extracts of the cements were added. After 24 hours incubation, the cellular vitality of fibroblasts was evaluated by the neutral red uptake test (NRU), which measures the membrane permeability. Data were collected and statistically analysed. Results showed that all tested materials exhibited mild cytotoxic effects, which are compatible with normal clinical use, and no statistically significant difference was noted between FibreFill and the other tested materials. Therefore, selection amongst these sealers should be based on other factors.

Developing and testing outreach materials on Alzheimer's disease for Asian and Pacific Islander Americans.

Compared to whites, minorities have similar rates of dementia but are less likely to use services. Native language videos and brochures that incorporated culturally specific perceptions of dementia, caregiving, and help-seeking for seven Asian and Pacific Islander American (APIA) groups were developed and tested. Findings suggested that these tools were culturally appropriate and, when used in combination with discussion, helped increase awareness of Alzheimer's disease and sources of help.

Cumulative irritation comparison of adapalene gel and solution with 2 tazarotene gels and 3 tretinoin formulations.

Forty-two subjects with normal skin were enrolled in a single-center study to assess the cumulative irritancy potential of adapalene (Differin gel 0.1% and Differin solution 0.1%) compared with tazarotene (Tazorac gels 0.05% and 0.1%), tretinoin (Retin-A Micro gel 0.1%, Avita cream 0.025%, and Avita gel 0.025%), and white petrolatum (negative control). All test materials were applied randomly, under occlusion, to sites located on either side of the midline--the mid thoracic area of the subjects' backs. All patches were applied daily, Monday through Friday, to the same sites, unless the degree of reaction to a test product or adhesive necessitated removal (grade 3). Thirty-eight of the 42 subjects (90.5%) completed the study. Thirty-four of those 38 subjects (89.5%) had to discontinue using both tazarotene concentrations due to intolerance. Patch discontinuations for the remaining test materials were as follows: 7 subjects discontinued use of tretinoin microsphere gel 0.1%, 3 discontinued tretinoin cream 0.025%, 1 discontinued tretinoin gel 0.025%, and 1 discontinued adapalene gel 0.1%. None of the subjects discontinued use of the white petrolatum or the adapalene solution 0.1%. Adapalene gel and solution 0.1% were statistically (P<.01) less irritating than both tazarotene gels 0.1% and 0.05%, tretinoin microsphere gel 0.1%, and tretinoin gel 0.025%, and they were not statistically different from tretinoin gel 0.025%.

Cytotoxic effects of gingival retraction cords on human gingival fibroblasts in vitro.

The objective of this study was to determine the cytocompatibility of three different extracts of gingival retraction cords and to compare the cytotoxic effect of these materials on human gingival fibroblasts. Gingival retraction cords impregnated with aluminium sulphate (Gingi-Aid), dl-adrenaline HCl (Gingi-Pak) and non-drug-impregnated cord (Gingi-Plain) were eluted with culture medium for 10 min and 24 h. Cytotoxicity was judged using a tetrazolium bromide reduction assay. Our data demonstrated that gingival retraction cords applied alone almost completely inhibited cell viability (P < 0.05). In addition, the results also showed that the eluates from aluminium sulphate-impregnated cord, dl-adrenaline HCl-impregnated cord and non-drug-impregnated cord were cytotoxic to primary human gingival fibroblast cultures (P < 0.05). The cell viability of incubation of gingival fibroblasts containing 10-min eluates of aluminium sulphate, dl-adrenaline HCl and non-drug-impregnated cord was 61, 21 and 70%, respectively. The cell viability of incubation of gingival fibroblasts containing 24 h eluates of aluminium sulphate, dl-adrenaline HCl and non-drug-impregnated cord was 68, 58 and 72%, respectively. It was found that dl-adrenaline HCl-impregnated gingival retraction cord was the most toxic gingival retraction cord among the materials tested in all cultures (P < 0.05). The cytotoxicity decreased in an order of dl-adrenaline HCl-impregnated cord > aluminium sulphate-impregnated cord > non-drug-impregnated cord. The extent or degree of the cytotoxicity depended on the materials tested. Gingival retraction cords have significant potential for gingival toxicity. Careful management of gingiva retraction cords would lower the risk of potential gingival tissue damage during clinical application procedure and thus increase the success of prosthodontic procedures.

Microleakage of three cement bases.

The purpose of this study was to evaluate the ability of a glass-ionomer cement-base material to prevent bacterial penetration along the dentin interface and to compare it with two conventional cement-base materials. A total of 107 Class 5 restorations was placed in Rhesus monkey teeth by means of three test materials [zinc oxide-eugenol (ZOE), copalite varnish + zinc phosphate cement base (V + ZP), and a glass-ionomer lining cement (GI)], with unetched and unbonded resin composite used alone as a control material and as a final restoration over the test base materials. Following disinfection, Class 5 cavities were prepared on the buccal surfaces of the teeth to the inner one-half of dentin. A sterile filter-paper disk was then placed on the axial wall and covered with a Teflon disk. Next, the cavities were based to the dento-enamel junction with one of the test base materials and finally restored with unetched and unbonded resin composite. After five and 16 weeks, the filter-paper disks were retrieved and cultivated for the presence and type of bacteria. The five-week results showed positive growth in two groups: the composite-only controls and the V + ZP group. The 16-week results showed growth in all of the test groups, but only one of nine teeth showed growth in the zinc oxide-eugenol group and one of 16 teeth in the glass-ionomer group. The results of this study indicate that under the conditions tested, a glass-ionomer base was capable of minimizing bacterial penetration along the material-tooth interface.
