Methyl-3 hydroxy-6 quinazolin dione-2-4 inhibits plant cell cytokinesis while modifying neither the duration of the cell cycle nor the duration of mitosis. Adding it to a solution of another compound enables one to determine the influence exerted by this compound on the duration of mitosis. This method, applied to phenylurethane, leads to see a slowing down of mitosis followed by a somewhat quick adaptation of the cells.
Thioglycolic acid is, in the Allium test, somewhat less cytotoxic than mercaptoethanol but the characteristics of cytotoxicity are the same in both cases : the mitoclasic effect cannot be separated from a strong mitodepressive effect and is quickly followed by a lethal effect.
Experiments were carried out on the effect of nickel as an inorganic compound (NiSO4.7H2O) and organic Ni(II) complexes (i.e. Ni(II)-Glu and Ni(II)-EDTA) in concentrations of 20, 40 and 85 ?M dm-3 on meristematic cells of root tips of Brassica oleracea L. cv. Sława from Enkhouizen. All three tested chemical forms of nickel had a mitodepressive effect and inhibited root elongation. With respect to the degree of root elongation inhibition and mitodepressive effect, the tested forms of nickel can be put in the following order: Ni(II)-Glu NiSO4.7H2O Ni(II)-EDTA. In all three tested forms, nickel caused disturbances in mitotic divisions, resulting in anaphase bridges and binuclear cells, whose nuclei were joined by a bridge of condensed chromatin or separated. Inorganic nickel and Ni(II)-Glu in higher concentrations damaged nuclei (the amount of condensed chromatin increased), nucleoli (their structure became more condensed and vacuolisation was observed), endoplasmic reticulum (fragmentation, swelling of cisternae) and mitochondria (structure condensation).
For the first time, in altered gravity, with antifibrillarin antibodies and immunogold microscopy fibrillarin that is one of the most important proteins of rRNA processing was localized. The quantitative study of the density of gold particles in the nuclelous, under clinorotation both the transition zone FC-DFC and the dense fibrillar component were less labeled as compared to the control. Obtained data allow us to suppose the lowering of the rRNA processing level in the nucleoli under the influence of altered gravity.
The effect of oryzalin, a microtubule polymerization inhibitor (10 MM), on lectin and mitotic activities (mitotic index and duration of mitotic phases) was studied in unhardened (23 degrees C) and hardened (7 days, 2-3 degrees C) winter wheat seedlings. Three wheat cultivars differing in their frost tolerance were compared. Oryzalin treatment (3 h) decreased activity of soluble lectins, increased activity of cell wall lectin mitotic index. Under these conditions, prolongation of anaphases and disappearance of telophases were detected. Plant hardening reduced the sensitivity of cell wall lectins and mitotic activity to the cytoskeleton inhibitor due, presumably, to the appearance of cold-stable microtubules. Plant growing and hardening with oryzalin stopped mitoses and caused the appearance of polyploid cells and cells with micronuclei. These abnormalities were preserved after hardening. The results obtained demonstrate an important role of microtubules in adaptation of plants to low temperature.
The duration of the cell cycle and its phases after the treatment with herbicide "Chwastox płynny 30" was calculated using 3H-thymidine labelling method. Inhibition of DNA synthesis and marked prolongation of G2 + 1/2 M phase were observed. Tested herbicide caused a significant lowering in the mitotic activity and accumulation of metaphase cells.
The influence of a polish anticancer drug on the cell cycle using Allium test was studied. Methods of aceto-orcein squash slides, curve of labelled mitoses after 3H-thymidine incubation and cytophotometrics after Feulgen's reaction were employed. Ledakrin acts strongly antimitotically, but it does not block the cell cycle completely. The cytostatic activity of ledakrin results from its action on the interphase. The phases G1 and S are prolonged while M is unchanged after 6h incubation with ledakrin. During postincubation in water without ledakrin it was noted, at the beginning, that the mitotic activity decreases and it is brought about the lengthening of S and G2 phases. The duration of the cell cycle phases returns to the control level during further postincubation. The results of analysis of chromatin aberrations and the micronucleus test point to a mutagenic effect of ledakrin.

According to earlier results, cold treatment blocks most of cells in G1 phase; after 3 h of postincubation at 20 degrees C these cells initiate S phase. Simultaneous cytophotometric (DNA stained with Methyl Green) and autoradiographic (3H-uridine, 3H-arginine) analyses of cold pretreated cells have shown that transport of 3H-RNA into cytoplasm is faster in G2 than in G1 cells: radioactivity of cytoplasm is faster in G2 cells becomes higher than that of nucleoli as early as between 2 h and 2.5 h postincubation, while in G1 cells--not until between 2.5. and 3.0 h. Simultaneous cytophotometric measurements of DNA (Feulgen) and protein (Naphthol Yellow S) contents demonstrate the considerable increase in cytoplasmic protein contents during the 2nd h of postincubation at 20 degrees C; therefore it precedes the export of more than 50% of 3H-RNA synthesized after cold treatment. The results of these experiments indicate two separate events in G1 cells: increase in cytoplasmic protein amounts (up to 2nd h of postincubation) and enhanced transport of newly-synthesized ribosome (2.5 h of postincubation), the later event immediately precede the entry of cells into S phase.
Experimental model consisted in blocking cells in G1 phase by cold treatment (12 h, 10 degrees C); following 3 h of postincubation at 20 degrees C, cells initiated S phase. In the present studies it has been shown that 2 h postincubation at 20 degrees C of cold-treated young seedlings of Helianthus annuus L. results in transformation of inactive meristematic nucleoli, characterized by small sizes, reduced amount of dry mass and granular component and by the presence of few and large fibrillar centres into large active nucleoli displaying high dry mass and granular component contents, numerous and small fibrillar centres. After 3 h of postincubation at 20 degrees C, nucleoli lose their granular component, decrease in size and dry mass content. At this moment cytoplasm enriches in ribosomes and its dry mass increases. Maximum of nucleolar activity is preceded by an accumulation of proteins in nucleoli. It is concluded that an enhanced transport of ribosomes is one of the conditions of S phase initiation.
