High pressure liquid chromatographic determination of the rodenticide brodifacoum in rat tissue.

An HPLC method was developed to determine residues of individual isomers of brodifacoum (3-[3-4'-bromo[1,1'-biphenyl]-4-yl)-1, 2, 3, 4-tetrahydro-1-naphthalenyl]-4-hydroxy-2H-1-benzopyran-2-one) in rat tissue. The compound was extracted twice with 10% methanol in chloroform, filtered, and cleaned up by using automated gel permeation chromatography. A final cleanup on a silica gel SEP-PAK was added to protect the analytical column from irreversible adsorption and to reduce analysis time. The analysis was done on a microPorasil column; a UV detector was used for quantitation. Recoveries of brodifacoum added to rat tissue in concentrations of 0.12-5.0 ppm were greater than 90%.

[Treatment with anticoagulants in glomerulonephritis (author's transl)].

Four patients with different forms of glomerulonephritis were treated with the anticoagulant heparin (Liquemin) or phenprocoumon (Marcumar). Following the results in these cases and the experiences of other research groups, which have been published in the last few years, we have tried to lay down a scheme for treatment with anticoagulants in glomerulonephritis. Such a treatment is recommended in peracute or rapid progressing glomerulonephritis, in forms of the disease of which is known that they have a poor prognosis, and in situations in which the risk of simultaneous thromboembolic complications as a result of a nephrotic syndrome represents a further argument for the use of anticoagulants.

[Effect of an anticoagulant rodenticide on the female albino rat with offspring].

Lactating female albino rats have been treated with coumatetralyl rodenticide, having anticoagulant activity. The letal effect on mothers was of 40% (using a maximum concentration of toxic substance) while it was of 54% in offsprings. Among the probable causes of death in offsprings, the authors suggest the hypothesis that an interference in the relationship mother-offsprings is due to the influence of coumatetralyl on parental behavior.

[Coagulation-physiological studies on the prevention of thromboembolism with dextran 60. II. Changes in coagulation and fibrinolysis].

The postoperative behavior of different parameters of coagulation and fibrinolysis have been studied in three groups of 20 patients each. Group A got only 4-hydroxycumarin, group B 500 ml Dextran 60 intraoperatively in addition to that, group C 500 ml Dextran 60 during surgery and on the 1., 2., 4., 7. and 10. postoperative day. The results show that the hypercoagulability which occurs immediately after surgery can be prevented by infusion of Dextran 60. On the other hand there is no evidence that Dextran 60 may cause a consumptive coagulability. There is only little influence on fibrinolysis, so that no defect of haemostasis can be expected. So it is possible to use Dextran 60 both for bridging over the phase of early embolism after surgery until the change to other anticoagulants and as a substance which can be applicated for a longer period of time in prophylaxis of thromboembolism. It is referred to the additional properties of Dextran 60 in prevention of thrombosis due to reduction of blood viscosity and improvement of microcirculation.

[Does oxazepam (ADUMBRAN) interfere with anticoagulant treatment. A controlled study. (author's transl)].

Newer communications dealing with drug interaction between tranquillizers and anticoagulants and the frequent combined intake of these drugs have induced us to perform a series of coagulation-and platelet parameters in 21 antiocoagulated patients with and without oxazepam (Adumbran). Control of adherence to the prescriptions was stressed upon: all cases were controlled by analysis of urin and nonadherent patients were excluded from statistical evaluation of hemostatic parameters. In the obtained well defined group no differences were observed in the effect of constant doses of anticoagulants and different tests of plasmatic coagulation and platelet aggregability.

[Studies on the interaction of Phenprocoumon with Diazepam and with Nitrazepam (author's transl)].

The influence of Diazepam and Nitrazepam on the anticoagulating activity of Phenprocoumon was studied in rats. It was found that the repeated oral application of Diazepam or Nitrazepam during the treatment with Phenprocoumon did not change the adjusted prothrombin time. The prothrombin time was considerably shortened when the application of Diazepam or Nitrazepam was stopped although Phenprocoumon was still applied. Furthermore, the influence of Chlordiazepoxid, Diazepam and Nitrazepam on the normal prothrombin time was studied. It was found that the treatment with benzodiazepine derivatives had no influence upon the prothrombin time, not only during the treatment, but also after the treatment was stopped.

Individualization of drug dosage during long-term treatment with phenprocoumon (Falithrom).

Pharmacokinetic studies were carried out with 3H-phenprocoumon (3H-PPC) during long-term treatment with phenprocoumon (PPC, Falithrom) in 10 female and male patients with thromboembolic diseases. Additionally, 12 mg 3H-PPC were administered per os to 6 male volunteers. Half-life and pattern of urine metabolites were measured and compared with those in patients. The radioactivity decreased independently of the total PPC plasma concentration, which was estimated fluorometrically. An exponential regression with a high coefficient of determination was found between the mean maintenance dose (PPC) and the area under plasma concentration curve of radioactivity. This procedure is considered to be a method to improve long-term treatment with PPC.
