Three hundred eleven employees of the National Institutes of Health volunteered to take 1 gm of ascorbic acid or lactose placebo in capsules three times a day for nine months. At the onset of a cold, the volunteers were given an additional 3 gm daily of either a placebo or ascorbic acid. One hundred ninety volunteers completed the study. Dropouts were defined as those who missed at least one month of drug ingestion. They represented 44% of the placebo group and 34% of those taking ascorbic acid. Analysis of these data showed that ascorbic acid had at best only a minor influence on the duration and severity of colds, and that the effects demonstrated might be explained equally well by a break in the double blind.
10 Chelating agents were administered to mice by intraperitoneal (i.p.) injection in order to compare their relative effectiveness in preventing death after a single i.p. injection of NaVO3. Ascorbic acid, Tiron (4,5-dihydroxy-1,3-benzene-disulfonic acid) and deferoxamine were the most effective antidotes for acute NaVO3-toxicity. The therapeutic index of ascorbic acid against 0.61 mmol/kg NaVO3 was found to be 95.2. This was 4.6 and 12.2 times greater than those of Tiron and Deferoxamine respectively. L-Cysteine greater than Na2CaEDTA greater than Na3CaDTPA reduced the acute mortality of mice following i.p. injection of NaVO3. Sodium salicylate, D,L-penicillamine, DMSA and DDC were not effective as antidotes for acute NaVO3-toxicity.
Curative potential of riboflavin, niacin and ascorbic acid against tamoxifen mediated endometrial carcinoma was established by studies on carbohydrate metabolizing enzymes. The enzymes investigated were glycolytic enzymes namely, hexokinase; aldolase; phosphoglucoisomerase and the gluconeogenic enzymes namely, glucose-6-phosphatase and fructose-1, 6-biphosphatase in endometrial carcinoma bearing rats. A significant increase in glycolytic enzymes and a subsequent decrease in gluconeogenic enzymes were observed in plasma, liver and kidney of endometrial carcinoma animals. The administration of riboflavin (45 mg/kg bw/day), niacin (100 mg/kg bw/day) and ascorbic acid (200 mg/kg bw/day) along with tamoxifen (45 mg/kg bw/day) caused a significant decrease in the activity of glycolytic enzymes and a significant increase in the activities of gluconeogenic enzymes to near normal levels in experimental animals. Our results suggest that riboflavin, niacin and ascorbic acid have potential combination therapy against tamoxifen mediated secondary endometrial carcinoma in experimental rats. However, there were no deleterious side effects observed in combinants alone treated animals.
