Rapid suppression of testosterone secretion after capture in male American alligators (Alligator mississippiensis).

All reptiles studied to date show an increase in circulating corticosterone following capture. This rise in corticosterone has also been shown in a number of instances to result in a decline in reproductive steroids within hours after capture. As a result of these observations it has been considered imperative to collect blood samples as soon as possible after capture to get reliable measures of reproductive hormones. It has been claimed, however, that there is no effect of capture stress on reproductive steroids in juvenile alligators held for 2 h following capture. As we generally reject blood samples that are not collected within 15 min of capture we decided to reinvestigate the effect of short-term capture (2 h) on corticosterone and testosterone in male alligators. Four groups of alligators, ranging in size from 74 to 212 cm total length were captured in a 2-week period in May, the time of year when testosterone levels are highest. Two groups were captured during the day (eight bled at capture and again at 2 h, eight bled at 2 h only) and two at night (10 bled at capture and again at 2 h, 10 bled at 2 h only). Testosterone and corticosterone in alligators bled immediately on capture and at 2 h were not significantly different in the AM and PM samples so the results were combined (Initial bleed: corticosterone, 0.95 +/- 0.09 ng/ml, n=18; testosterone, 6.06 +/- 2.09 ng/ml, n=18. Two-hour bleed: corticosterone 15.68 +/- 1.91, n=18; testosterone, 2.75 +/- 0.79, n=18). Both the increase in corticosterone and the decline in testosterone at 2 h were significant (p<0.05). Corticosterone and testosterone in the alligators sampled only once at 2 h were not significantly different from the 2-h values in alligators sampled twice (corticosterone 15.04 +/- 1.29, n=18; testosterone, 1.85 +/- 0.62, n=18). These results clearly demonstrate that short-term capture stress results in a significant decline in testosterone in male alligators.

Local capture by atrial pacing in spontaneous chronic atrial fibrillation.

BACKGROUND: Atrial fibrillation (AF) is considered to be maintained by multiple reentrant circuits without or with a very short excitable gap. However, the possibility of local atrial capture has been shown recently in experimental AF or induced AF in humans. METHODS AND RESULTS: This study was undertaken to evaluate the feasibility of atrial capture-suggestive of an excitable gap-in spontaneous chronic AF. Decremental pacing was performed in 47 right atrial sites in 14 patients with chronic AF, not taking antiarrhythmic drugs. A Franz catheter (for pacing and monophasic action potential recording) and a recording quadripolar catheter positioned about 10 mm apart were used. Local capture was achieved in 41 (87.2%) sites for a total of 100 captures. In 71 episodes the capture was lost within 15 seconds, while in the remaining 29, pacing was stopped after 15 seconds of stable capture. AF types immediately before capture were type 1 in 83 and type 2 in 17 episodes. Type 3 AF was never captured. Pacing cycle at capture was 175.7 +/- 20.9 ms. The baseline atrial interval (FF) was 185.4 +/- 24.5, significantly longer than the FF recorded during pacing immediately before capture (176.0 +/- 19.8 ms) (P < .02). CONCLUSIONS: During spontaneous chronic AF in humans, (1) local capture by atrial pacing is possible up to at least 15 mm from the pacing site, (2) regional entrainment is possible during type 1 and type 2 AF but not type 3 AF, and (3) pacing before capture accelerates AF, probably by transient or local capture. These findings suggest that an excitable gap is present in chronic AF, therefore supporting the hypothesis that leading circle reentry is not the unique electrophysiological mechanism maintaining the arrhythmia.

Tyrosine-based "activatable pro-tag": enzyme-catalyzed protein capture and release.

Protein recovery is often achieved by a series of capture and release steps that often involve chromatographic binding and elution. We report an alternative, non-chromatographic, capture and release approach that employs enzymes and the stimuli-responsive polysaccharide chitosan. We capture our protein using the enzyme tyrosinase that oxidizes accessible tyrosine residues of the protein and "activates" these residues for covalent capture (i.e., conjugation) onto chitosan. Using fusions of green fluorescent protein (GFP) we observed that: (i) enzymatic activation is required for protein capture to chitosan; and (ii) capture is enhanced (approximately five-fold) by engineering the protein to have a penta-tyrosine fusion tag that provides additional accessible tyrosine residues for enzymatic activation. Because the fusion tag appears to be the primary site for capture, and capture requires activation, we designate penta-tyrosine as a "pro-tag." The captured GFP-chitosan conjugate possesses the pH-responsive solubility that is characteristic of chitosan. We exploit this pH-responsive solubility to facilitate purification of the captured protein. Two enzymatic methods were explored to release the captured GFP from the chitosan conjugate. The first method employs enterokinase (EK) to cleave the protein at an engineered EK-cleavage site. The second method employs chitosanase to hydrolyze the chitosan backbone. Using GFP as a model protein, we demonstrated that enzymatic capture and release provides a simple, non-chromatographic means to recover proteins directly from cell lysates.

Exploratory investigations of hypervelocity intact capture spectroscopy.

The ability to capture hypervelocity projectiles intact opens a new technique available for hypervelocity research. A determination of the reactions taking place between the projectile and the capture medium during the process of intact capture is extremely important to an understanding of the intact capture phenomenon, to improving the capture technique, and to developing a theory describing the phenomenon. The intact capture of hypervelocity projectiles by underdense media generates spectra, characteristic of the material species of projectile and capture medium involved. Initial exploratory results into real-time characterization of hypervelocity intact capture techniques by spectroscopy include ultra-violet and visible spectra obtained by use of reflecting gratings, transmitting gratings, and prisms, and recorded by photographic and electronic means. Spectrometry proved to be a valuable real-time diagnostic tool for hypervelocity intact capture events, offering understanding of the interactions of the projectile and the capture medium during the initial period and providing information not obtainable by other characterizations. Preliminary results and analyses of spectra produced by the intact capture of hypervelocity aluminum spheres in polyethylene (PE), polystyrene (PS), and polyurethane (PU) foams are presented. Included are tentative emission species identifications, as well as gray body temperatures produced in the intact capture process.

A novel capture-ELISA for detection of anti-neutrophil cytoplasmic antibodies (ANCA) based on c-myc peptide recognition in carboxy-terminally tagged recombinant neutrophil serine proteases.

Testing for antineutrophil cytoplasmic antibodies (ANCA) reacting with proteinase 3 (PR3) is part of the routine diagnostic evaluation of patients with small vessel vasculitis. For PR3-ANCA detection, capture ELISAs are reported to be superior to direct ELISAs. Standard capture ELISAs, in which PR3 is anchored by anti-PR3 monoclonal antibodies (moAB), have two potential disadvantages. First, the capturing moAB may compete for epitopes recognized by some PR3-ANCA, causing occasional false-negative results. Second, the capture of recombinant PR3 mutant molecules becomes unpredictable as modifications of specific conformational epitopes may not only affect the binding of PR3-ANCA, but also the affinity of the capturing anti-PR3 moAB. Here, we describe a new capture ELISA, and its application for PR3-ANCA detection. This new assay is based on the standardized capture of a variety of different carboxy-terminally c-myc tagged recombinant ANCA target antigens using anti-c-myc coated ELISA plates. Antigen used include c-myc tagged human rPR3 variants (mature and pro-form conformations), mouse mature rPR3 and human recombinant neutrophil elastase. This new anti-c-myc-capture ELISA for PR3-ANCA detection has an intra- and inter-assay coefficient of variation of 3.6% to 7.7%, and 15.8% to 18.4%, respectively. The analytical sensitivity and specificity for PR3-ANCA positive serum samples were 93% and 100%, respectively when rPR3 with mature conformation was used as target antigen, and 83% and 100% when the pro-enzyme conformation was employed. In conclusion, this new anti-c-myc capture ELISA compares favorably to our standard capture ELISA for PR3-ANCA detection, enables the unified capture of different ANCA target antigens through binding to a c-myc tag, and allows capture of rPR3 mutants necessary for PR3-ANCA epitope mapping studies.

Lack of consistent hormonal responses to capture during the breeding season of the bearded dragon, Pogona barbata.

The bearded dragon (Pogona barbata: Agamidae) is a diurnal, oviparous, multi-clutching lizard from Australia. We examined plasma hormonal responses to capture in males and females during the spring breeding season. Corticosterone concentrations at capture (0 h; < or =3 min after capture) were low (males: 1.81+/-0.63 ng/ml; females 2. 23+/-0.47 ng/ml) and within sexes were unrelated to the time of the day, snout-vent length or, in females, reproductive condition (vitellogenic, gravid, assumed spent). Corticosterone concentrations at capture were significantly and inversely correlated with body condition in males, but not in females. Unexpectedly, neither sex showed significant changes in mean concentrations of corticosterone at 3.5 or 24 h after capture compared with 0 h values. Corticosterone concentrations at 3.5 h after capture did not differ between dragons bled at capture or not. Concentrations of progesterone in both the sexes did not change between 0 h and 3.5 or 24 h after capture. Testosterone concentrations in males at capture were moderate (10.1+/-2.2 ng/ml), and unchanged at 3.5 h after capture. The adrenocortical axis of adult bearded dragons in the breeding season seems remarkably unresponsive to capture compared with many other reptiles. Low adrenocortical responses to capture may be a feature of reptiles known to adjust well to captivity.

Effects of capture on biological parameters in free-ranging bighorn sheep (Ovis canadensis): evaluation of normal, stressed and mortality outcomes and documentation of postcapture survival.

Blood samples and physiological data were collected from 634 bighorn sheep (Ovis canadensis) captured by four different methods between 1980 and 1986 in the western United States. These parameters were evaluated for selected physiological, biochemical and hematological values. Postcapture biological parameters were compared among bighorn sheep according to four different outcomes; normal, stressed or compromised, capture myopathy (CM) mortality, and accidental mortality. Significant differences (P less than 0.05) were noted between outcome groups relative to certain parameters: temperature, respiration, creatinine phosphokinase (CPK), lactic dehydrogenase (LDH), serum glutamic oxaloacetic transaminase (SGOT), blood urea nitrogen (BUN), glucose, white blood cell count (WBC) and plasma pH. Such differences between groups may help in evaluating the clinical status of bighorn sheep at capture, enabling one to predict those animals that might develop CM at a later date, indicate candidates for preventive medical treatment prior to release, and/or which should be followed closely to determine long-term survival. Evaluation of follow-up data (n = 77) related to outcome status and long-term survival of bighorn sheep indicated that less than 4% (3 of 77) were dead within 1 mo of capture (one of these had been classified as normal and two as stressed or compromised at capture); less than 3% (3 of 77) were dead greater than 1 mo, and less than 6 mo after capture two were classified in the stressed outcome and one as diseased. Eighty-eight percent (68 of 77) were alive from 1 mo to 5 yr after capture (53 were classified as normal, 12 as stressed or compromised and 3 as diseased), and 2% (1 of 77) had chronic CM but was still alive (this animal had been classified as normal). Of 77 sheep in the follow-up group, less than 3% (2 of 77) were not observed following capture (one was classified as normal and one as stressed and diseased). Of the fatalities, less than 3% (2 of 40) had been captured by the net-gun and less than 4% (1 of 27) by drive-net. Those two unobserved in the follow-up group also had been caught with the net-gun, 5% (2 of 40). The single surviving CM case had been captured by the net-gun. Although the net-gun appears to be one of the safest methods of capturing individual bighorn sheep, based on evaluation of capture data and biological parameters, it may not be associated with the best long-term survival in some bighorn sheep.(ABSTRACT TRUNCATED AT 400 WORDS)

Spatial extension of local pacing capture in the lateral free wall of the right atrium during clinical chronic atrial fibrillation.

OBJECTIVES: The aims of our study were to define the spatial extension of local atrial pacing capture (LAPC) in the right lateral atrial wall and to verify the effects of antiarrhythmic drugs on capture extension. BACKGROUND: Recently, LAPC during spontaneous chronic atrial fibrillation (AF) has been demonstrated directly in humans, but no systematic attempt has been made to evaluate its spatial extension. METHODS: The study was conducted in 14 consecutive patients with chronic AF, nine of whom were taking antiarrhythmic drugs. A Franz catheter for pacing and monophasic action potential recording was positioned in the lateral right atrial wall. An octopolar recording lead was positioned about 10 mm apart. The presence of atrial capture and its extension (number of captured bipolar pairs) were evaluated during decremental pacing. RESULTS: Forty-eight atrial capture attempts were made. LAPC was achieved in all the performed attempts. The AF types immediately before capture were type 1 in 35 (73%) and type 2 in 13 (27%) episodes. In 32 episodes, the capture was lost within 15 seconds and in the remaining 16 pacing was stopped after 15 seconds of stable capture. In 26 episodes, capture extended radially up to 4 cm from the pacing site, in 18 up to 3 cm, in 3 up 1 cm and in 1 up 1 cm. The mean extension in patients being treated with antiarrhythmic drugs was significantly greater than what was seen in untreated patients (3.8 +/- 0.4 vs 2.9 +/- 0.7 cm, p < 0.00001). Capture loss was less frequently observed when all four of the bipolar recording sites had been captured (13/25, 52% vs 19/23, 82%, respectively, p = 0.05). CONCLUSIONS: LAPC in the lateral right atrial wall extends over a radius of 30-40 mm in the majority of capture episodes and is greater in patients treated with antiarrhythmic drugs. Our results suggest that, at least theoretically, simultaneous multisite pacing could be used to terminate AF. Moreover, a reduction in the atrial defibrillation threshold could be expected performing low-energy internal cardioversion during LPC.

A serotonin receptor antagonist, but not melatonin, modulates hormonal responses to capture stress in two populations of garter snakes (Thamnophis sirtalis parietalis and Thamnophis sirtalis concinnus).

Hormonal and behavioral responses to a stressor depend on many factors, including the influence of other hormones. We examined the role of melatonin in modulating hormonal responses to capture stress in two populations of male garter snakes, Thamnophis sirtalis. Studies of red-sided (T. sirtalis parietalis) and red-spotted (T. sirtalis concinnus) garter snakes were conducted in the field with free-living snakes. Populations of red-sided garter snakes in south-central Manitoba, Canada undergo a period of winter dormancy for approximately 8 months each year followed by an attenuated mating season (4-5 weeks) in early spring. In contrast, the mid-latitude red-spotted garter snake in western Oregon, USA has an extended breeding season and can be active during 10-12 months of the year given appropriate environmental conditions. We chose to study these two populations of garter snakes to investigate possible variation in melatonin function among snakes with different suites of environmental adaptations. To better address these questions, we also examined the effects of 5-hydroxytryptophan (a precursor of melatonin synthesis) and ketanserin (a serotonergic type 2A receptor antagonist) on hormonal responses to capture stress. We observed a trend of increased corticosterone and decreased androgen concentrations in northern-latitude red-sided garter snakes (T. sirtalis parietalis) subjected to 4 h of capture stress during the spring. However, these differences were not statistically significant. During the fall, red-sided garter snakes showed no change in corticosterone or androgen concentrations in response to the capture stress treatments. We speculate that northern-latitude red-sided garter snakes suppress hormonal responses to capture stress during preparation for winter dormancy. Treatment with melatonin, 5-hydroxytryptophan, or ketanserin did not significantly influence corticosterone or androgen concentrations of northern-latitude red-sided garter snakes during the spring or fall. Mid-latitude red-spotted garter snakes (T. sirtalis concinnus) from Oregon showed a statistically significant increase in corticosterone concentrations in response to 4 h of capture stress; treatment with melatonin, 5-hydroxytryptophan, or ketanserin prior to capture stress had no significant influence on plasma corticosterone concentrations. Androgen concentrations of mid-latitude red-spotted garter snakes in response to capture stress were significantly lower than those of non-stressed control snakes. Neither melatonin nor 5-hydroxytryptophan influenced the change in androgen concentrations during capture stress. However, androgen concentrations of snakes treated with ketanserin prior to 4 h of capture stress did not differ significantly from those of non-stressed control snakes. These studies suggest that melatonin does not modulate hormonal responses to capture stress in this ectothermic model. Our results also suggest that a serotonin-regulated system may play a role in modulating the activity of the hypothalamic-pituitary-gonadal axis during physiological stress responses.

Antibody microarray detection of Escherichia coli O157:H7: Quantification, assay limitations, and capture efficiency.

A sandwich fluorescent immunoassay in a microarray format was used to capture and detect E. coli O157:H7. Here, we explored quantitative aspects, limitations, and capture efficiency of the assay. When biotinylated capture antibodies were used, the signal generated was higher (over 5-fold higher with some cell concentrations) compared to biotinylated protein G-bound capture antibodies. By adjusting the concentration of reporter antibody, a linear fluorescent response was observed from approximately 3.0 x 10(6) to approximately 9.0 x 10(7) cells/mL, and this was in agreement with the number of captured bacteria as determined by fluorescence microscopy. Capture efficiency calculations revealed that, as the number of bacteria presented for capture decreased, capture efficiency increased to near 35%. Optimization experiments, with several combinations of capture and reporter antibodies, demonstrated that the amount of bacteria available for capture (10(6) versus 10(8) cells/mL) affected the optimal combination. The findings presented here indicate that antibody microarrays, when used in sandwich assay format, may be effectively used to capture and detect E. coli O157:H7.

On using simple time-of-travel capture zone delineation methods.

As part of its Wellhead Protection Program, the U.S. EPA mandates the delineation of "time-of-travel capture zones" as the basis for the definition of wellhead protection zones surrounding drinking water production wells. Depending on circumstances the capture zones may be determined using methods that range from simply drawing a circle around the well to sophisticated ground water flow and transport modeling. The simpler methods are attractive when faced with the delineation of hundreds or thousands of capture zones for small public drinking water supply wells. On the other hand, a circular capture zone may not be adequate in the presence of an ambient ground water flow regime. A dimensionless time-of-travel parameter T is used to determine when calculated fixed-radius capture zones can be used for drinking water production wells. The parameter incorporates aquifer properties, the magnitude of the ambient ground water flow field, and the travel time criterion for the time-of-travel capture zone. In the absence of interfering flow features, three different simple capture zones can be used depending on the value of T . A modified calculated fixed-radius capture zone proves protective when T < 0.1, while a more elongated capture zone must be used when T > 1. For values of T between 0.1 and 1, a circular capture zone can be used that is eccentric with respect to the well. Finally, calculating T allows for a quick assessment of the validity of circular capture zones without redoing the delineation with a computer model.

Performance of indirect immunoglobulin M (IgM) serology tests and IgM capture assays for laboratory diagnosis of measles.

As progress is made toward elimination of measles, the laboratory confirmation of measles becomes increasingly important. However, both false-positive and false-negative results can occur with the routinely used indirect measles immunoglobulin M (IgM) serology tests. The measles IgM capture assay is considered to be more specific, and therefore, its use is indicated for confirmatory testing, but its relative performance has not been fully assessed. Four commercial indirect measles IgM serology test kits (the Behring, Clark, Gull, and PanBio assays) and a commercial IgM capture assay (the Light Diagnostics assay) were evaluated for their abilities to detect measles virus-specific IgM antibody with a total of 308 serum samples from patients involved in a measles outbreak and with confirmed cases of measles and 454 samples from subjects without measles. The Centers for Disease Control and Prevention (CDC) IgM capture assay was also used in a part of the evaluation. Among the indirect assays, the overall sensitivities ranged from 82.8% (Clark assay) to 88.6% (Behring assay) and specificity ranged from 86.6% (PanBio assay) to 99.6% (Gull assay). These rates were 92.2 and 86. 6%, respectively, for the Light Diagnostics capture assay and 87.0 and 94.8%, respectively, for the CDC capture assay. While the Light Diagnostics capture assay had the best detection rate (80%) with the acute-phase samples compared with those for the rest of the tests (CDC capture assay, 77%; Behring assay, 70%; Gull assay, 69%; PanBio assay, 58%; and Clark assay, 57%), all tests showed a significantly improved sensitivity in the range of 92% (Clark and PanBio assays) to 97% (Light Diagnostics and CDC capture assays) with the convalescent-phase samples, as expected. The best seropositivity rates (in the range of 92 to 100%) were observed with samples collected 6 to 14 days after the onset of symptoms. The Gull assay showed the highest positive predictive value (99.6%), followed by the Behring assay (97.8%) and the CDC capture assay (96.1%). Overall, the Gull and Behring assays were found to be as good as or better than the capture assays. In conclusion, laboratory diagnosis of measles based on IgM serology varies depending on the timing of specimen collection and the test used, and the case for the use of the IgM capture assay as the confirmatory test appears to be uncertain.

Efficacy of 1,4-diaminobutane (putrescine) in a food-based synthetic attractant for capture of Mediterranean and Mexican fruit flies (Diptera: Tephritidae).

Field trials were conducted in Guatemala to evaluate the importance of 1,4 diaminobutane (putrescine) in traps baited with ammonium acetate, trimethylamine, and putrescine. For the Mediterranean fruit fly, Ceratitis capitata (Wiedemann), there were no differences in percentage of females captured in coffee and citrus or in percentage of males captured in citrus in traps with ammonium acetate and trimethylamine lures (females in coffee, 26.4 +/- 6.27%; females in citrus, 35.7 +/- 5.35%; males in citrus, 37.7 +/- 7.48%) versus ammonium acetate, trimethylamine, and putrescine lures (females in coffee, 36.6 +/- 9.64%; females in citrus, 41.1 +/- 5.18%; males in citrus, 37.1 +/- 6.09%). Percentage of males captured in coffee was reduced significantly when putrescine was not used with the ammonium acetate and trimethylamine (39.9 +/- 4.34 versus 31.6 +/- 5.29%). Lower percentages were captured in traps baited with ammonium acetate and putrescine, and the lowest percentages were captured in traps baited with putrescine and trimethylamine. When population level as indicated by capture in traps baited with ammonium acetate, trimethylamine, and putrescine was considered, a higher percentage of C. capitata males were captured in traps baited with all three components when one or more flies per trap per day were captured in coffee, and a higher percentage of females were captured when less than one fly per trap per day was captured in citrus. Percentage of the Mexican fruit fly, Anastrepha ludens (Loew), captured was significantly higher in traps baited with ammonium acetate and putrescine and significantly lower in traps baited putrescine and trimethylamine than in all other treatments. Results indicate that putrescine may be deleted when monitoring established populations of C. capitata but should be used in traps used to monitor A. ludens or to detect new infestations of C. capitata.

Impact of trap design, windbreaks, and weather on captures of European corn borer (Lepidoptera: Crambidae) in pheromone-baited traps.

Pheromone-baited traps are often used in ecological studies of the European corn borer, Ostrinia nubilalis (Hübner) (Lepidoptera: Crambidae). However, differences in trap captures may be confounded by trap design, trap location relative to a windbreak, and changes in local weather. The objectives of this experiment were, first, to examine differences in 0. nubilalis adult (moth) captures among the Intercept wing trap, the Intercept bucket/funnel UNI trap, and the Hartstack wire-mesh, 75-cm-diameter cone trap (large metal cone trap) as well as among three cone trap designs. Second, we examined the influence of the location of the large metal cone trap relative to a windbreak on the number of moths captured. Third, we examined the relationship between nightly mean air temperature, relative humidity, wind speed, precipitation, and the number of moths captured in large metal cone traps. The number of moths captured was significantly influenced by trap design, with large metal cone traps capturing the most moths. Wing and bucket traps were ineffective. Differences among trap captures were significant among trap locations relative to a windbreak. Under strong (>14 kph) or moderate (7 <14 kph) wind speeds, traps located leeward of the windbreak captured the most moths, but when wind speeds were light (<7 kph), traps not associated with windbreaks captured the most moths. The multiple regression model fitted to the relationship between number of moths captured per Julian date and nightly weather patterns was significant. Nightly mean air temperature was the most influential parameter in the model, and its relationship with moth capture was positive.
