Maintenance of hemopoietic stem cells in diffusion chamber cultures.

When murine bone marrow cells are cultured in DC's in irradiated host mice, there is an initial phase of rapid white cell growth followed by prolonged maintenance of steady-state granulopoiesis. This study examined the role of hemopoietic stem cells in this culture system. During the initial growth phase there was a rapid increase in both pluripotential stem cells, CFU-S, and commmitted white cell precursors, CFU-C. The increase in CFU-C was larger, occurred more rapidly, and was biphasic in nature. After day 7 and until at least day 16, the numbers of CUF-S, CFU-C and total white cells were all maintained at relatively constant plateau levels which were typically well below the maximal capacity of the culture system. During the plateau phase the number of CFU-S was slightly greater than the number initially placed in culture, whereas CFU-C had increased by a factor of 2.3 and total white cells by a factor of 10. This appears to reflect a system of growth amplification, with preferential differentiation of CFU-S into CFU-C and thence into the pathway of white cell development. The plateau phase of culture has received very little attention but appears to represent a model, in a closed culture system, of finely regulated granulopoiesis, with an equilibrium between constant numbers of hemopoietic stem cells and their differentiating white cell progeny.

Proliferation pattern of hamster melanoma cells cultured in diffusion chambers in pre-immunized hosts.

Fortner M Mel 1 melanoma cells from the Golden Hamster were capable of exponential proliferation during intraperitoneal diffusion chamber culture in xenogenic host animals (Balb/c mice, BD IX and Marshall rats). When the host animals had been preimmunized with these melanoma cells, rapid cell lysis was observed in the chambers within 4 hrs after implantation. The cell numbers were reduced to about 1 - 10% of the inoculum. At this level the cell numbers persisted for several days, as opposed to control cultures in untreated hosts, where the cells proliferated rapidly. After 6 - 10 days the cell numbers in chambers from immunized hosts slowly increased in spite of a high level of cytotoxicity in ther serum of the animals. The same pattern was seen after previous immunization with hamster lymphocytes, indicating that the antibodies were directed against a common antigen for melanoma cells and lymphocytes in hamsters. The diffusion chamber method appears to be a useful system for the study of a pure humoral immune reaction against tumour cells, and especially for investigation of the phenomenon that some tumour cells are able to escape a strong humoral cytotoxic effect from host animals.

Effect of the Hewitt keratinizing epidermal carcinoma on cell proliferation in different organs of the host mouse and in human psoriatic skin cultured in diffusion chambers.

Incorporation of 3H-thymidine into different squamous epithelia and other tissues was studied in mice bearing the Hewitt keratinizing epidermal carcinoma with a raised epidermal chalone content in the body. The results showed that DNA synthesis in the tumour-bearing mice was inhibited strongly in the epithelium of the ear (c.80%), moderately in the epithelium of the back skin and the foot (c.55%), and weakly in the epithelium of the tongue and the cornea (c.25%). These inhibitions appear to be specific in nature, because cell proliferation was not detectably affected in the sebaceous gland, intestinal epithelium, bone marrow, and thymus lymphocytes. Thymus size, however, was reduced in the tumour-bearing mice compared with controls, and 3H-thymidine uptake was depressed in the spleen and to some extent also in the lymph nodes, possibly owing to non-specific stress. Labelling index of human psoriatic epidermis was suppressed in diffusion-chamber cultures carried by the tumour-bearing host mice compared with controls. However, this reaction was probably not specific in nature, because the labelling index of mouse bone-marrow cells was also decreased when grown in tumour-bearing animals.

[Enhanced proliferation of human lung cancer cells during culture in diffusion chambers].

A comparative evaluation of the rate of proliferation of lung tumor cells from 23 patients and that of primary diffusion chamber cultures implanted to mice was carried out. Proliferation rate was determined on the basis of 3H-thymidine labeling index. Explanted tumor cells which had undergone further cultivation showed an increase in proliferative rate. The latter effect was particularly marked in adenocarcinoma. The said effect was not observed in small-cell carcinoma. These data suggest that cells of certain human tumors are capable of high proliferation. This property may be used with a view to raising effectiveness of cytotoxic antitumor chemotherapy.
