Contraction and relaxation of cultured mesangial cells on a silicone rubber surface.

Glomerular mesangial cells (MC) in culture are believed to contract or relax in response to agents such as angiotensin II and cyclic AMP. However, cells grown on glass or plastic surfaces are limited in their response to vasoactive agents because of the rigid surfaces to which they adhere; thus, interpretation of a change in shape as contraction, relaxation, or detachment is difficult. We have grown MC on a flexible silicone rubber (dimethylpolysiloxane) substrate (DMPS), and studied with sequential photographs several models of cell contraction, relaxation, and detachment. When the cells contracted, the DMPS became wrinkled; when the cells relaxed, the DMPS lost wrinkles. In contrast, if the cells detached, the sheet lost wrinkles as the cells became smaller and rounder. Angiotensin II (5 X 10(-7) M), and calcium ionophore A23187 (2 X 10(-6) M) increased wrinkles in more than 30% of cells at 22 degrees C and more than 40% of the cells at 36 degrees C. The earliest effect was visible within five to 10 minutes at 22 degrees C and within one minute at 36 degrees C and increased until 40 minutes; thereafter, the cells relaxed and wrinkles were reduced. 10(-1) M Na azide prevented the increase in wrinkles produced by angiotensin II. Seventy-two percent of the angiotensin II-treated cells whose margins could be seen in their entirety, and 78% of the calcium ionophore-treated cells showed a reduction in surface area at a time when new wrinkles were appearing or wrinkles were increasing in size. Dibutyryl cyclic AMP, a known smooth muscle relaxant, produced a decrease or loss of wrinkles in 90% of the cells, and an accompanying increase in surface area. Untreated control cells, observed in conjunction with the above series, showed little change in wrinkles. Ten percent DMSO, an actin-translocating agent, produced a reversible disappearance of wrinkles. These models of contraction and relaxation could be distinguished from cell detachment; EDTA, for example, in the presence of zero calcium, diminished both cell size and wrinkles, with an accompanying lifting of cells from the surface. Similar results were obtained with cytochalasin B and chlorpromazine. Thus, the silicone rubber system accurately reflects the contraction, relaxation and detachment of cultured mesangial cells in response to a variety of agents.

A defoaming agent should be used with pronase premedication to improve visibility in upper gastrointestinal endoscopy.

BACKGROUND AND STUDY AIMS: The study tested whether pronase can improve endoscopic visibility and alter the accuracy of the CLO test for H. pylori detection. PATIENTS AND METHODS: A total of 160 patients were randomly assigned to receive one of five premedications for endoscopy: group A: dimethylpolysiloxane (DMPS) alone; group B: DMPS plus water (up to 100 ml); group C: pronase only, with 100 ml water; group D: pronase and sodium bicarbonate plus water up to 100 ml; group E: pronase, sodium bicarbonate, and DMPS, plus water up to 100 ml. Endoscopists, who were unaware of the premedication method administered, assessed visibility scores (range 1 - 4) for the antrum, lower gastric body, upper gastric body, and fundus. The higher the score, the less clear the visibility. The sum of scores from the four locations was defined as the total visibility score. A CLO test was also done during the endoscopy. One week after their endoscopy, patients in groups C, D, and E were scheduled for a (13)C-urea breath test (UBT). RESULTS: Group E patients had a significantly lower total visibility score than those in the other four groups ( P < 0.05). Groups C and D had higher total visibility scores than the other three groups ( P < 0.05). The scores did not significantly differ between groups A and B. Based on the UBT results, the sensitivity and specificity of the CLO test were 92.6 % and 96.2 %, respectively. CONCLUSIONS: Premedication as in group E provided the clearest endoscopic visibility. Without the application of DMPS, pronase alone cannot improve endoscopic visibility. Pronase does not influence H. pylori identification using the CLO test.

Histological investigation of the etiology of capsule contracture following augmentation mammaplasty.

Sections obtained from contracted breast capsules were examined by light and electron microscopy. We observed the presence of multiple areas of a particulate foreign material, consistent with the appearance of droplets of dimethylpolysiloxane. An active foreign-body reaction was observed at the interface of the capsule and implant, as well as throughout the tissue subjacent to the deposits of foreign material. Based on these histological observations, we present a theoretical explanation of the etiology of capsular contracture of the breast after augmentation mammaplasty.

Activation of macrophages by silicones: phenotype and production of oxidant metabolites.

BACKGROUND: The effect of silicones on the immune function is not fully characterized. In clinical and experimental studies, immune alterations associated with silicone gel seem to be related to macrophage activation. In this work we examined in vivo, phenotypic and functional changes on peritoneal macrophages early (24 h or 48 h) and late (45 days) after the intraperitoneal (i.p.) injection of dimethylpolysiloxane (DMPS) (silicone). We studied the expression of adhesion and co-stimulatory molecules and both the spontaneous and the stimulated production of reactive oxygen intermediates and nitric oxide (NO). RESULTS: The results presented here demonstrate that the fluid compound DMPS induced a persistent cell recruitment at the site of the injection. Besides, cell activation was still evident 45 days after the silicone injection: activated macrophages exhibited an increased expression of adhesion (CD54 and CD44) and co-stimulatory molecules (CD86) and an enhanced production of oxidant metabolites and NO. CONCLUSIONS: Silicones induced a persistent recruitment of leukocytes at the site of the injection and macrophage activation was still evident 45 days after the injection.

Evaluation of protective ointments used against dermal effects of nitrogen mustard, a vesicant warfare agent.

Mustard, a vesicant warfare agent, has cytotoxic, mutagenic, and cytostatic effects via alkylation of DNA and inhibition of DNA replication. Since symptoms appear following a latent period, it can cause some subacute and chronic effects to occur and delay in the treatment. Therefore, the main approach should be the use of protective preparation to reduce the skin toxicity. Thus, this study was conducted in guinea pigs (350-400 g) shaved in areas of 10 x 10 cm. Mechlorethamine HCl (100 mg), a nitrogen mustard derivative, in ethanol was applied by spraying on hairless regions where previously prepared pharmaceutical topical formulations were medicated before. The experimental regions of the animals were kept preserved from environmental factors. Forty-eight hours after the application of the protective ointments and mechlorethamine consecutively, skin-damaging effects were macroscopically evaluated in terms of erythema formation, ulceration, necrosis, and inflammation occurrences. Then, punch biopsy was performed from these damaged sites for histopathological evaluation. Although numerous topical formulations were prepared and tested for protection, according to microscopic examination of the pathologic sections, tissue specimen treated with the ointment containing the mixture of zinc oxide, zinc chloride, dimethylpolysiloxane in a base of petroleum jelly was determined as being the most effective protective against skin injury caused by the vesicant agent.

[Development of a gas chromatographic column system for the on-line analysis of trace tetrahydrofuran in hexane].

A gas chromatographic system using home-made 7 microns thick cross-linked dimethylpolysiloxane columns for the on-line analysis of trace tetrahydrofuran in hexane in a rubber production facility was developed. The experimental parameters of the column system, including the temperature, flow rate as well as the back-flushing and heart-cutting program were investigated. The total column-switching program was suggested. The system has been successfully operated for more than one year with good resolution, stability and precision (RSD < 5%) in analyzing trace tetrahydrofuran (0-250 x 10(-6), V/V) for industrial process control in the rubber production facility.

Enantiomer separation by pressure-supported electrochromatography using capillaries packed with Chirasil-Dex polymer-coated silica.

Pressure-supported electrochromatography using capillaries packed with permethyl-cyclodextrin covalently linked via an octamethylene spacer to dimethylpolysiloxane and immobilized on silica (Chirasil-Dex silica) has been employed as an efficient and rapid method for the enantiomer separation of various racemic compounds. By comparing this method with micropacked liquid chromatography (LC), employing the same column in a unified instrumental setup, micropacked capillary electrochromatography (CEC) shows higher column efficiencies and hence better resolution factors. The influence of type and concentration of buffer, amount and nature of organic modifier, and pressure support is investigated.

Enantiomer separation by capillary electrochromatography on a cyclodextrin-modified monolith.

A chiral monolithic stationary phase was prepared by packing a capillary with bare porous silica and sintering the silica bed at high temperature. The resulting silica monolith was polymer-coated with Chirasil-Dex, a permethylated beta-cyclodextrin covalently linked via an octamethylene spacer to dimethylpolysiloxane. Subsequently, Chirasil-Dex was thermally immobilized on the silica support and a chiral monolith of very high stability (30 kV, more than 400 bar pressure) was obtained. The enantiomer separation of various chiral compounds by monolithic (rod) capillary electrochromatography (rod-CEC) was feasible. This method was compared with capillary liquid chromatography (LC) in a single-column mode using unified equipment. About two to three times higher efficiency was found in the rod-CEC mode as compared to rod-LC. The influence of pressure-driven flow support on efficiency, resolution, elution time and baseline stability was investigated. The amount and nature of organic modifier strongly influences efficiency and resolution.

Dual chiral recognition system involving cyclodextrin derivatives in capillary electrophoresis II. Enhancement of enantioselectivity.

The enantiomer separation of hexobarbital was investigated by open tubular electrochromatography (OTEC) using the chiral stationary phase (CSP) CHIRASIL-DEX (a permethylated beta-cyclodextrin covalently linked to a dimethylpolysiloxane) and by cyclodextrin-electrokinetic chromatogaphy (CD-EKC) using anionic beta-cyclodextrin-sulfo-n-propyl ether (SPE-beta-CD) and cationic beta-cyclodextrin-2-hydroxy-3-trimethylammoniumpropyl ether chloride (HTAP-beta-CD) added to the running buffer. By employing two chiral selectors, the enantiomer separation of hexobarbital was then studied simultaneously by OTEC with CHIRASIL-DEX and by CD-EKC with either SPE-beta-CD or HTAP-beta-CD in the dual chiral recognition mode. In conjunction with CHIRASIL-DEX, anionic SPE-beta-CD decreased the chiral separation factor alpha due to compensation of enantioselectivity whereas the cationic additive HTAP-beta-CD increased the chiral separation factor alpha due to enhancement of enantioselectivity. It is concluded that CHIRASIL-DEX imparts an opposite enantioselectivity to the enantiomers of hexobarbital as compared to the charged CDs SPE-beta-CD and HTAP-beta-CD. Unusual peak broadening phenomena are observed in the dual chiral recognition system comprised of CHIRASIL-DEX and HTAP-beta-CD. The possible consequences of accidental dual chiral recognition systems caused by wall stacking effects of the mobile phase additives onto the inner surface of the capillary column are discussed.

Induction of plasmacytomas in genetically susceptible mice with silicone gels.

Silicone gels injected intraperitoneally into strains of mice related to BALB/c develop plasmacytomas in approximately the same numbers and with similar phenotypes as previously obtained with pristane. Silicone gels produce few side effects and are well tolerated for long periods. Silicone gels contain several components that are potentially biologically active: residual vinyl groups and platinum. Microscopic and histological evidence suggests the silicone gel is degraded over a long period of time. Preliminary studies with long chain liquid dimethylpolysiloxanes with viscosities of 1000 cSt and 12,500 cSt have not produced plasmacytomas as yet. The plasmacytomagenic action of the gel appears to be due to the release of liquids from the gel matrix.

Plasmacytoma development in mice injected with silicone gels.

Silicone gels derived from commercially obtained implants induce plasmacytomas in 60-70% of highly susceptible BALB/cAn.DBA/2-Idh1-Pep3 congenic mice. In contrast, dimethylpolysiloxane (DMPS) silicone oils with viscosities of 5, 1000 and 12,500 cs fail to elicit these tumors. 1000 cs vinylmethylpolysiloxane is also inactive. Silicone gels, in contrast to the oils, induce a highly inflammatory silicone granuloma. Silicone gels contain chemical components not found in the oils. The chemical component responsible for inducing the permissive environment for plasmacytoma formation has not yet been identified. Silicone gels are well tolerated for long periods of time in mice without adverse effects other than plasmacytoma formation. The response to different gel preparations varies; some are associated with relatively rapid formation of plasmacytomas resembling that seen with pristane, while in others the plasmacytoma formation is extended nearly over a two year period.

Effect of hydrophilic coating on microorganism colonization in silicone tubing.

BACKGROUND: Shunt infections are one of the major causes of mortality and morbidity of patients with hydrocephalus. The aim of this research is to compare the bacterial colonization characteristics of a regular silicone elastomer shunt material coated with polyvinylpyrrolidone and dimethylpolysiloxane (silicone). METHOD: Regular coated shunt materials were compared by in-vivo and in-vitro methods. In the in-vitro experiment, silicone and coated material immersed and not immersed in vancomycin solution was treated with a certain concentration of Staphylococcus epidermidis. In the in-vivo study, silicone and coated material specimens were treated with Staphylococcus epidermidis and they were stereotactically placed in the lateral ventricles of the rats. One week after the inoculation, shunt pieces were removed and the colonies were counted by using a scanning electron microscope. FINDINGS: There was a statistically significant difference of colonization in the in-vitro groups in coated material vs. silicone, coated material vs. vancomycin treated silicone, vancomycin treated coated material vs. silicone, vancomycin treated coated material vs. vancomycin treated silicone. There was no statistically significant difference for colonization in in-vitro groups of coated material and vancomycin treated coated material. With in-vivo experiments we can say that, coated material catheters are superior than the silicone catheters in respect to colonization but after the bacterial colonization has occurred, the amount of colonization did not differ. INTERPRETATION: Coated material catheters are superior to silicone catheters and they prevent bacterial colonization in some respect.

Analysis of the seed oil of Heisteria silvanii (Olacaceae)--a rich source of a novel C18 acetylenic fatty acid.

Besides some usual fatty acids (FA), two conjugated ene-yne acetylenic FA [trans-10-heptadecen-8-ynoic acid (pyrulic acid) (7.4%), and trans-11-octadecen-9-ynoic acid (ximenynic acid) (3.5%)], a novel ene-yne-ene acetylenic FA [cis-7, trans-11-octadecadiene-9-ynoic acid (heisteric acid) (22.6%)], and 9,10-epoxystearic acid (0.6%) could be identified in the seed oil of Heisteria silvanii (Olacaceae). Two further conjugated acetylenic FA [9,11-octadecadiynoic acid (0.1%) and 13-octadecene-9,11-diynoic acid (0.4%)] were identified tentatively by their mass spectra. The FA mixture has been analyzed by gas chromatography/mass spectrometry (GC/MS) of their methyl ester and 4,4-dimethyloxazoline derivatives. The structure of heisteric acid was elucidated after isolation via preparative silver ion thin-layer chromatography and by various spectroscopic methods [ultraviolet; infrared; 1H, 13C nuclear magnetic resonance (NMR); 1H-1H and 1H-13C correlation spectroscopy]. To determine the position of the conjugated ene-yne-ene system, the NMR spectra were also measured after addition of the lanthanide shift reagent Resolve-Al EuFOD. Furthermore, the triyglyceride mixture was analyzed by high-temperature GC and high-temperature GC coupled with negative chemical ionization MS. A glass capillary column coated with a methoxy-terminated 50%-diphenyl-50%-dimethylpolysiloxane was used for the separation of the triacylglycerol (TAG) species. No evidence of decomposition of the TAG species containing conjugated ene-yne-ene FA was observed. Twenty-six species of the separated TAG were identified by means of their abundant quasi molecular ion [M - H]- and their corresponding carboxylate anions [RCOO]- of the fatty acids, respectively. The major molecular species of the TAG were found to be 16:0/18:1/18:1, 16:0/18:1/18:3 (heisteric acid), 17:2 (pyrulic acid)/18:1/18:1, 18:1/18:1/18:3 (heisteric acid). The TAG containing acetylenic FA showed an unexpected increase of the retention time in comparison to the TAG containing usual FA, thus making the prediction of the elution order of lipid samples containing acetylenic FA difficult.

Unified enantioselective capillary chromatography on a Chirasil-DEX stationary phase. Advantages of column miniaturization.

Immobilized Chirasil-DEX (mono-6-O-octamethylenepermethyl-beta-cyclodextrin chemically linked to dimethylpolysiloxane) can be employed as a versatile chiral stationary phase in chromatography. The chiral polymer has a long lifetime and is configurationally and thermally stable. The concept of unified enantioselective chromatography has been demonstrated for the enantiomer separation of hexobarbital by gas chromatography, supercritical fluid chromatography, liquid chromatography and capillary electrochromatography on a single open-tubular column (1 m x 50 microns I.D.) coated with Chirasil-DEX. The advantages of miniaturization in contemporary chromatographic enantiomer separation are demonstrated. Chirasil-DEX coated on porous silica is also useful for enantiomer separation in high-performance liquid chromatography.

Silicone plate-induced granuloma presenting pituitary apoplexy-like symptoms: case report.

BACKGROUND: Granuloma induced by silicone plate is rare. We present a case of silicone plate-induced granuloma in the sellar and suprasellar region confirmed by histologic examination at surgery. CASE DESCRIPTION: A 31-year-old woman suddenly complained of severe headache, polyuria, and visual disturbance 8 years and 9 months after an initial transsphenoidal surgery for pituitary adenoma. At surgery, granuloma and hematoma around the dimethylpolysiloxan (silicone) plate, which had been placed on the sella floor at the initial surgery, were found. Postoperative course was uneventful. CONCLUSIONS: Our presented case showed one of the disadvantages for silicone placement for repairing the sella floor, and the diagnostic possibility of silicone-induced granuloma in the sellar region.

Gas chromatographic determination of residual levels of tert.-butanol from lyophilized liposomal formulations.

The use of liposomal formulations is rapidly gaining popularity in pharmaceutical research and development. Their preparation often involves the use of organic solvents such as tert.-butanol to dissolve lipophilic lipids. To improve the physicochemical stability of the liposomes, lyophilizing the product is one of the best available means. A gas chromatographic method for the determination of tert.-butanol residual levels in lyophilized liposomes, employing sec.-butanol as internal standard, using a flame ionization detector (FID) detector and a cross-linked dimethylpolysiloxane capillary column, was developed. The method described is simple, sensitive, rugged, reliable and reproducible and requires less time than other reported methods for the quantitation of tert.-butanol; it also has pharmaceutical applications.

Qualitative evaluation of thermal desorption-programmable temperature vaporization-comprehensive two-dimensional gas chromatography-time-of-flight mass spectrometry for the analysis of selected halogenated contaminants.

The separation of 38 toxic and predominant polychlorinated biphenyl (PCB) congeners, 11 persistent halogenated pesticides, 1 brominated biphenyl (BB), and 8 polybrominated diphenyl ethers (PBDEs) has been optimized using comprehensive two-dimensional gas chromatography coupled to time-of-flight mass spectrometry (GC x GC-TOFMS). A thermal desorption-programmable temperature vaporization (TD-PTV) step was used for the injection. Different column sets were investigated, and a 100% dimethylpolysiloxane (15 m x 0.25 mm i.d. x 0.25 microm film thickness) narrowbore capillary column coupled to a high temperature (8% phenyl)-polycarborane-siloxane (2 m x 0.10 mm i.d. x 0.10 microm film thickness) microbore column set was selected. Of the 58 compounds investigated, only one pair of PCBs was not resolved. All other analytes were either baseline separated into the chromatographic plane or were virtually separated using the deconvolution capability of the TOFMS.
