Out of 189 lambs in the flock, 25 animals suffered from bilateral or unilateral conjunctivitis, or keratoconjunctivitis. By serological examination (RVK), positive levels of antibodies to the group-specific antigen of Chl. psittaci were found in three out of six lambs examined by laboratory methods. Bacteriological examination of eye smears of six lambs showed in four cases the infection by microorganisms of Moraxella bovis. Smears from the conjunctivas of these lambs were after preparation instilled in the yolk sacs of six to seven days old chicken embryos. One strain of Chlamydia psittaci was isolated from the same material as Moraxella bovis.

The effects of a modified-live infectious bovine rhinotracheitis virus vaccine (administered ocularly or intranasally) on experimentally induced infectious bovine keratoconjunctivitis were evaluated. The modified-live infectious bovine rhinotracheitis virus vaccine was administered to 13 male Holstein calves (intranasally in 4 and ocularly in 9; day 0). Five calves were not vaccinated and served as controls. Calves were examined daily and, starting on day 4, Moraxella bovis was administered ocularly to all 18 calves once daily for 4 days. The eyes of all calves were assigned a clinical score, and the ocular secretions were evaluated for presence of infectious bovine rhinotracheitis virus and M bovis daily until day 19. The severity of the ocular lesions was estimated by scoring the lesions clinically and by determining the protein concentration, myeloperoxidase activity, and WBC count in the tears. By day 5, conjunctivitis, chemosis, and epiphora were observed in all of the calves vaccinated ocularly. The calves vaccinated intranasally developed conjunctival plaques, but did not develop chemosis or photophobia. All of the calves developed keratitis after inoculation with M bovis. The median lesion scores were greater in both groups of vaccinated calves than in the controls. Corneal perforations developed exclusively in the vaccinated calves. The frequency of M bovis isolation from ocular secretions was significantly (P less than 0.05) greater in the vaccinated calves than in the controls. The tears from the intranasally vaccinated calves contained the highest myeloperoxidase activity and WBC count. The mean protein concentration in the tears of vaccinated calves was not significantly different from that in tears of controls.(ABSTRACT TRUNCATED AT 250 WORDS)
Infectious bovine keratoconjunctivitis is a common and highly contagious ocular disease affecting cattle worldwide. The tremendous economic losses attributable to this disease warrant continued investigation into methods of prevention. Multiple virulence factors have been linked to the primary aetiologic agent, Moraxella bovis. Efforts to develop an efficacious vaccine have primarily focused upon the use of surface pili or cytolysin to stimulate host immunity; however, M. bovis possesses other virulence determinants that include proteases, fibrinolysins, phospholipases and other cell surface components such as outer membrane proteins. These potentially conserved antigens provide additional possibilities for vaccine development. Examination of appropriate antigen presentation is necessary to attain an adequate immune response. Further, the potential for antigenic diversity as well as epitope conversion requires continuous epidemiological surveillance of isolates recovered from outbreaks. Current work targeting conserved immunogens provides hope for efficacious vaccines that when used in tandem with proper management may control, if not prevent, infectious bovine keratoconjunctivitis.
Infectious bovine keratoconjunctivitis (IBK) was enzootic in the beef cattle herds at Dixon Springs Agricultural Center, University of Illinois. The development of nonhemolytic and hemolytic Moraxella bovis flora in the eyes of 48 calves in a closed cow-calf herd was monitored from late May to October, 1972. The incidence of clinical IBK was recorded each week. In late May, nonhemolytic M bovis was isolated from 26% of calf eyes. The peak incidence of IBK was observed in early September, with 26% of the eyes affected. At that time, nonhemolytic M bovis was isolated from 10% of the eyes, and hemolytic M bovis from 58%. By late October, nonhemolytic M bovis was isolated from 25% of the eyes, and hemolytic M bovis from only 4%. In an attempt to increase the resistance of ocular tissue, 19 calves were vaccinated in each third eyelid with 0.5 ml of an autogenous M bovis bacterin in late May. Vaccination did not provide practical protection against the establishment of hemolytic M bovis in the eyes nor the development of clinical IBK. However, at the peak incidence of IBK, hemolytic M bovis was isolated from the eyes of 48% of vaccinated calves and 73% of nonvaccinated calves; clinical IBK was present in 21% of the eyes of vaccinated calves and 29% of nonvaccinated calves. In evaluating the in vitro proteolytic potential of M bovis isolates, 83 hemolytic and 5 nonhemolytic isolates peptonized litmus milk; 176 nonhemolytic isolates did not peptonize litmus milk. Hemolytic M bovis isolates were more pathogenic for mice than were nonhemolytic isolates. Of 60 mice, 53 (88%) died in 1 to 3 hours after intraperitoneal inoculation of hemolytic isolates; 8 of 32 (25%) mice died in 5 to 15 hours after inoculation of nonhemolytic isolates. Hemolytic M bovis isolates produced testicular swelling and scrotal necrosis after inoculation into the scrotal sac of rabbits; nonhemolytic isolates produced only mild transient testicular swelling.
Infectious bovine keratoconjunctivitis (IBK) is one of the most economically important diseases in preweaned calves. This study examined the health records of 45,497 calves over a 20-yr period to determine environmental and genetic factors influencing the incidence of IBK. Three data sets were analyzed with an animal model. The first data set (n = 41,986) evaluated environmental factors and genetic differences among nine purebred (Angus, Braunvieh, Charolais, Gelbvieh, Hereford, Limousin, Pinzgauer, Red Poll, and Simmental) and three composite breeds (MARC I, MARC II, and MARC III). Weaning weights of calves diagnosed with IBK were 8.9 kg lighter (P < 0.05) than weights of healthy calves. Incidence of IBK was related to age of the calf and the seasonal life cycle of the face fly (Musca autumnalis). Incidence of IBK increased in the spring (June), peaked during the summer months (July to September), and then decreased in the fall. Herefords were the most susceptible breed (P < 0.05) compared with all other purebreds and composites. Estimates of direct heritability for the incidence of IBK were generally low and ranged from 0.00 to 0.28 by breed. The maternal permanent environmental and genetic effects of the dam on the incidence of IBK were not significant for most breeds. The second data set (n = 9,606) was used to estimate heterosis for the incidence of IBK from a Hereford and Angus diallel design. The heterosis effect for the incidence of IBK in reciprocal Hereford/Angus crossbred calves was slightly negative (P = 0.12) but not large. The higher incidence of IBK in Angus x Hereford calves compared with Hereford x Angus calves (13.3 vs. 8.9%) suggests a maternal effect related to the incidence of IBK. Incidence of IBK in crossbred calves sired by tropically adapted breeds (Brahma, Boran, Tuli) compared with purebred and crossbred Bos taurus types was investigated in the third data set (n = 2,622). Crossbred calves sired by tropically adapted breeds had a lower incidence of IBK than most Bos taurus types (P < 0.05), but they were not different than either reciprocal crosses of Hereford and Angus or purebred Angus calves. Response to selection for decreasing the incidence of IBK is likely to be slow because of low heritability and low incidence in most breeds. Significant breed differences for incidence of IBK may be important to some producers and management systems.
Two commercially available infectious bovine keratoconjunctivitis (IBK) vaccines were evaluated for their effectiveness in protecting cattle from disease caused by experimental challenge exposure and natural transmission of Moraxella bovis infections. The study was conducted as 2 experiments, using a total of 81 cattle that were culture-negative for M bovis prior to vaccination. In each experiment, young adult cattle were randomly allotted to 4 groups. Each calf in groups 1 and 2 was vaccinated according to the vaccine manufacturer's directions. Groups 3 and 4 were unvaccinated controls. Three weeks after the last vaccination, each calf in groups 1 and 3 was experimentally challenge exposed by dropping a suspension of viable cells of a virulent strain of M bovis directly onto the corneal surface of each eye. Calves in all 4 groups were then commingled in open pastures so that calves in groups 2 and 4 could be naturally exposed to the calves with experimentally induced infections. Each calf was examined for signs of ocular disease on a regular basis by 2 experienced clinicians who scored each eye for severity of disease on the basis of a prearranged scale. Neither clinician was aware of the vaccination or exposure status of the calf nor to which experimental group they belonged. Lacrimal secretions were collected regularly to determine the number of eyes in which the virulent organism became established. Moraxella bovis with bacterial cultural characteristics similar to those of the virulent strain placed in the eyes of groups 1 and 3 was cultured from greater than or equal to 83% of the eyes of calves in all groups.(ABSTRACT TRUNCATED AT 250 WORDS)
Out of a total of 224 bovine eye secretions, 126 Moraxella bovis and 64 Neisseria ovis strains were isolated. The pathogenesis and histological lesions caused by Neisseria ovis have been studied on the eyes of three calves naturally affected with IBK, using electron microscopy. Neisseria ovis caused in 1-12 weeks old calves acute, transient and mostly benign serous conjunctivitis with only slight affection of the cornea. More rarely erosions and even ulceration of the cornea have been observed. Moraxella bovis and Neisseria ovis strains proved nearly unanimously sensitive in vitro to chloramphenicol, neomycin, oxytetracyclin, nitrofurantoin, erythromycin and cefoperazone. Other antibiotics and chemotherapeutics inhibited the growth of these agents only partly or were ineffective. Experimental therapy has been carried out using a single i.m. injection of Terramycin/LA inj. (Pfizer) in a dose of 20 mg/kg body mass, repeated if necessary after 72-96 h. This formulation proved more effective and practical than treatments used earlier.

Protection conferred by a cell-free preparation from a haemolytic Moraxella bovis isolate, UQV 148NF, was compared to an equivalent fraction from a non-haemolytic M. bovis isolate, Gordon 26L3, and to a recombinant DNA-derived pili vaccine. Three groups of ten calves were vaccinated twice with one of the three preparations and, together with ten non-vaccinated calves, challenged with virulent M. bovis isolate Dal 2d. Compared to the control group, significant protection was observed in the group receiving the pili vaccine and the group receiving the preparation from haemolytic isolate, UQV 148NF.
