Germ-line affinity and germ-line variable-region genes in the B cell response.

The predominance of germ-line genes in IgM expression was evaluated from the nucleotide sequences of mRNA, derived from 10 hybridoma cell lines, coding for the VH and VL regions of anti-5-dimethylaminonaphthalene-1-sulfonyl (anti-Dns) IgM antibody. At least six germ-line VH gene segments distributed among four families are used in this response. Seven of the 10 independently rear-ranged VH genes were identified as germ line, with the other three possibly germ line. In all of them the D and JH portions retained the germ-line sequences of the D and JH segments from which they were derived. Maximum diversity was found in the D segments and the use of noncoded nucleotides at the VH-D and D-JH junctions. Of the eight cell lines expressing the lambda light chains, all were germ line and involved the three subtypes. Maximum affinity for the homologous ligand was found among the seven cell lines identified as expressing germ-line gene segments. Thus any somatic mutation among the remaining 3 cell lines did not provide enhanced affinity and the observed affinity of each cell line can be described as germ-line affinity. It is further suggested that the anti-Dns selectivity of the IgM antibodies is associated primarily with the CDR3 regions.

Characterization of a tumorigenic murine T-lymphoid-cell line spontaneously derived from an IL-2-dependent T-cell line.

The establishment of IL-2-independent T-cell lines spontaneously derived from long-term IL-2-dependent cytotoxic T-cell lines is described. Two lines (cloned and uncloned) studied in detail have shown the following characteristics: (1) Permanent loss of IL-2 dependence. (2) Partial or complete loss of both cytotoxic activity and the IL-2 receptor. (3) Increased expression of T-cell membrane markers (Thy1.2, Lyt1.2) compared with the parental line. (4) Lower level of DNA methylation than in freshly obtained lymphoid cells. (5) Different karyotypic pattern from the parental IL-2-dependent line, with a mean number of 39-40 chromosomes and a resemblance to T leukemic lines. (6) Leukemia caused in normal syngeneic C57BL/6 mice by the uncloned line, in contrast to the cloned IL-2-independent line or the parental dependent line. Unlike established leukemic lines, however, the independent line gave rise to tumors which regressed in some mice within a few days of their appearance. These findings suggest that T-cell lines maintained with IL-2 for prolonged periods of time (greater than 3 months) can undergo transformation and, therefore, should not be utilized for immunotherapeutic purposes.

Impact of first-line vs second-line antibiotics for the treatment of acute uncomplicated sinusitis.

CONTEXT: Studies suggest little benefit in relief of acute sinusitis symptoms from the use of newer and more expensive (second-line) antibiotics instead of older and less expensive (first-line) antibiotics. However, researchers have failed to include development of complications and cost of care in their analyses. OBJECTIVE: To compare the effectiveness and cost of first-line with second-line antibiotics for the treatment of acute uncomplicated sinusitis in adults. DESIGN, SETTING, AND PATIENTS: Retrospective cohort study using a pharmaceutical database containing demographic, clinical (International Classification of Diseases, Ninth Revision), treatment, and charge information for 29 102 adults with a diagnosis of acute sinusitis receiving initial antibiotic treatment between July 1, 1996, and June 30, 1997. MAIN OUTCOME MEASURES: Absence of additional claim for an antibiotic in the 28 days after the initial antibiotic, presence of a claim for a second antibiotic, serious complications of sinusitis, and direct charges and use for the acute sinusitis treatment. RESULTS: There were 17 different antibiotics prescribed in this study. The majority (59.5%) of patients received 1 of the first-line antibiotics. The overall success rate was 90.4% (95% confidence interval [CI], 90.0%-90.8%). The success rate for the 17 329 patients who received a first-line antibiotic was 90.1% and for the 11 773 patients who received a second-line antibiotic was 90.8%, a difference of 0.7% (95% CI, 0.01%-1.40%; P<.05). There were 2 cases of periorbital cellulitis, one in each treatment group. The average total direct charge for patients receiving a first-line antibiotic was $68.98 and a second-line antibiotic was $135.17, a difference of $66.19 (95% CI, $64.95-$67.43; P<.001). This difference was due entirely to the difference in charge of antibiotics and not other charges, such as professional fees, laboratory tests, or emergency department visits. CONCLUSIONS: Patients treated with a first-line antibiotic for acute uncomplicated sinusitis did not have clinically significant differences in outcomes vs those treated with a second-line antibiotic. However, cost of care was significantly higher for patients treated with a second-line antibiotic.

Second-line chemotherapy for non-small cell lung cancer.

BACKGROUND: The role of second-line chemotherapy for the treatment of patients with non-small cell lung cancer (NSCLC) who have relapsed or failed to respond to first-line treatment was unclear. OBJECTIVES: To determine the effectiveness of any second-line chemotherapy in patients with NSCLC. SEARCH STRATEGY: Bibliographic databases were searched. Handsearching and contact with experts was also performed. SELECTION CRITERIA: Randomised controlled clinical trials in which any second-line chemotherapy was compared with BSC in patients with NSCLC who had previously failed to any previous chemotherapy regimen. DATA COLLECTION AND ANALYSIS: Data was extracted by 2 independent reviewers and revised by all authors. MAIN RESULTS: Only one study was included. It randomised 204 patients to receive either doxetaxel or BSC. Following an unacceptably high toxic death rate the dose of doxetaxel was reduced from 100 mg/m(2) to 75 mg/m(2). Doxetaxel gave an extra 2.4 months of survival - an average of 7.0 months vs 4.6 months on BSC. At 1 year after diagnosis 29% of doxetaxel treated patients were alive compared with 19% of the BSC group. REVIEWER'S CONCLUSIONS: Definitive recommendations cannot be made since evidence is only available from one randomised controlled trial which, though of reasonable quality, had a number of limitations. There is currently no evidence to support second-line treatment of patients with poor performance status. Larger, well-designed controlled trials are needed to further evaluate whether the benefits of second-line chemotherapy to patients with NSCLC outweigh its risks and costs.

Influence of infusion line compliance on drug delivery rate during acute line loop formation.

OBJECTIVE: To determine whether infusion line compliance contributes to irregular drug delivery during vertical displacement of syringe pumps. DESIGN: Five different commercially available infusion lines were studied at infusion rates of 0.5, 1.0, and 1.5 ml/h. Zero drug delivery time was measured after acute line loop formation (70 cm) using an electronic balance. Compliance of each infusion line was calculated using a pressure transducer and measurement of the occlusion release bolus at 300 mmHg occlusion pressure. Finally, the influence of infusion line compliance on drug delivery during acute lowering of the syringe pump was studied using low- and high-compliance infusion lines. RESULTS: Acute line loop formation resulted in zero drug delivery time from 5.1 +/- 1.5 to 44.0 +/- 6.8 s at flow rates of 0.5 ml/h. Increased flow rates significantly reduced loop-induced flow variability. A close correlation was found between zero drug delivery time and calculated infusion line compliance at 0.5 ml/h (linear regression R2 = 0.79). Lowering of the syringe pump 50 cm prolonged zero drug delivery time from 295.8 +/- 20.7 s with the low-compliance tube to 463.3 +/- 24.0 s with the high-compliance infusion line. CONCLUSIONS: Infusion line compliance contributes to irregular drug delivery associated with vertical displacement of syringe pumps. Siphoning of the infusion line during patient care should be avoided, and flow rates of 1 ml/h or higher are recommended. Low-compliance infusion lines are indicated whenever highly short-acting vasoactive drugs at low delivery rates are administered.

Influence of crossing a line selected for increased shank width and a commercial sire line on performance and walking ability of turkeys.

The present study was initiated to determine whether walking ability (WA) could be markedly improved without seriously compromising growth rate and body conformation by crossing a relatively unimproved commercial sire line (UC) with a line (FL) selected for increased shank width and backcrossing F1 females to UC line males. Body weights at 8, 16, and 20 wk of age were generally similar for both lines at the time of the initial cross. The UC line had inferior WA, shorter and narrower shanks, and wider breasts compared with the FL line. The F1 birds exhibited overdominance for BW at all ages. Heterosis in BW ranged from 3.2 to 7.8%. At 16 wk of age, the WA scores decreased (indicating improved WA) in the F1 relative to the average of the parental lines. Heterosis of WA scores was -10.5% (P > or = 0.05) for males and -23.5% (P < or = 0.05) for females. Heterosis was observed in the F1 for shank length but not for other shank measurements (width and depth). No heterosis was observed for breast width. Mortality from 8 through 20 wk of age was reduced in the F1 relative to the parental lines. The backcross of the F1 females to UC males still exhibited heterosis for male BW at 16 and 20 wk of age and for WA scores, although the magnitude was reduced 50% or more. Mortality from 8 to 20 wk of age decreased in the backcross compared with the parental lines. The results suggest that a line exhibiting improved leg structure and WA, and poor conformation, such as FL, can be used to greatly improve the WA of a cross involving a sire line with poor WA but excellent conformation. A portion of the gains in BW and breast width obtained in the F1 was retained in a backcross to the sire line, making such a crossing scheme commercially feasible.

Comparison of eggshell surface microbial populations for in-line and off-line commercial egg processing facilities.

The objective of this project was to evaluate the aerobic plate counts (APC) of eggshells at in-line and off-line egg processing facilities at selected sites, throughout the processing procedure. Samples were collected from four sites in the processing plant and five time periods during the daily processing shift. Site 1 was from the conveyor system before the eggs passed through the washing system. Site 2 was after detergent wash but before sanitizer application. Site 3 was immediately after sanitizer treatment. Site 4 was immediately before packaging. Samples were collected from the sites at five equally spaced intervals beginning 15 min after the processing shift began and ending 15 min before the processing shift ended. At each sampling time, eggs were aseptically collected from each site and placed into sterile plastic bags containing 50 mL of PBS that was serially diluted immediately. The dilutions were plated on APC agar within 8 h of collection and were incubated at 37 C for 48 h. APC counts of in-line and off-line eggs were compared within time periods across sites. As the processing shift progressed, off-line APC counts were significantly higher than in-line counts at Site 1. At Site 2, off-line APC counts were significantly higher than in-line counts for Periods 2 through 5. At Site 3, off-line APC counts were significantly higher than in-line counts for Periods 2 through 5. Site 4 off-line counts were significantly higher than in-line counts at all time periods.

Line tension vector thermodynamics of anisotropic contact lines.

Multiphase materials with intersecting diving surfaces give rise to contact lines. A line tension vector thermodynamics formalism is developed and used to analyze contact line problems in the presence of anisotropy, taking into account two elastic modes: change in contact line length and change in contact line orientation. Using this formalism, the contact line-shape equation is derived, and the renormalization of the line tension due to anisotropy is characterized. The correspondence and analogies between the shape equation for anisotropic surfaces (Herring equation) and the shape equation for contact lines is established. Line energies for nematic liquid crystals, representative of generic anisotropic contact lines, are used to derive a shape equation that takes into account ambient orientation effects. It is found that anisotropic line tension may promote bending and chiral modes to avoid unfavorable orientations of the contact line with respect to the ambient nematic ordering.

Markers and characteristics of human SCLC cell lines. Neuroendocrine markers, classical tumor markers, and chromosomal characteristics of permanent human small cell lung cancer cell lines.

Permanent human small cell lung cancer (SCLC) cell lines established in our laboratory were investigated for their expression of the enzymatic neuroendocrine markers L-DOPA decarboxylase (DDC), neuron-specific enolase (NSE), and creatine kinase (CK), including its BB isoenzyme (CK-BB), the classical tumor markers carcinoembryonic antigen (CEA), the alpha and beta subunits of human chorionic gonadotropin (alpha-HCG, beta-HCG), and alpha-fetoprotein (alpha-FP), and their chromosomal characteristics. DDC activities were detectable in 5/6 SCLC cell lines and absent in non-SCLC. NSE levels ranged from 160 to 1422 ng/mg soluble protein and were less than 290 ng/mg soluble protein in non-SCLC. Activities of CK and levels of CK-BB clearly distinguished SCLC from non-SCLC with CK activities greater than 1000 munits/mg soluble protein and CK-BB levels greater than 3000 ng/mg soluble protein in SCLC and less than 300 munits/mg soluble protein and less than 2000 ng/mg soluble protein in non-SCLC. CEA was detectable in 5/6 SCLC cell lines but absent in non-SCLC, and its level seemed to correlate with those of DDC, NSE, and CK. One cell line, SCLC-16H, lost some of its neuroendocrine properties and CEA after 1 year of in vitro cultivation. Generally, marker levels were low in fast growing cell lines and high in slow growing cell lines. HCG alpha and beta subunit and alpha-FP were not detectable in SCLC cell lines. All SCLC cell lines examined had near diploid DNA indices and modal chromosome numbers. Double minute chromosomes and homogeneously staining regions were found in 2/5 and 4/5 SCLC cell lines respectively. With respect to chromosomal aberrations, we found a deletion of the short arm of at least one chromosome 3 in all SCLC cell lines (5/5). These data show that SCLC expresses neuroendocrine markers and CEA; CK is the most sensitive marker, and DDC and CEA are the most specific markers for SCLC in vitro; individual marker levels correlate with each other and the in vitro malignancy of SCLC; and SCLC cell lines have relatively uniform chromosomal characteristics. Our results suggest that patients whose tumors have high levels of DDC, NSE, CK-BB, and CEA have a better prognosis than those with low marker levels. This hypothesis could be proved by comparing pairs of patients that are matched for all known prognostic parameters, in particular tumor spread, for their serum and tumor marker levels with respect to the patients' outcome and prognosis.

Secretion of a suppressor cell inducing factor by an interleukin 3-dependent cell line with natural cytotoxic activity. III. Comparison with other interleukin 3-dependent cell lines.

We previously described the interleukin 3 (IL-3)-dependent cell line, M1-A5, which has both natural cytotoxic (NC) and suppressor cell activities, the latter of which is mediated, in part, by the release of two cytokines which activate suppressor cells from unprimed lymphoid precursor cells. In this study we have compared the M1-A5 cell line with four other IL-3-dependent cell lines to determine whether these dual activities are universally associated with IL-3 dependence and to test the hypothesis that there is a direct relationship between the cytotoxic and the suppressive activities. The cell lines tested were a bone marrow derived Dexter culture derived line (FDC-P1), two Moloney leukemia virus induced leukemias (DA-1 and DA-3), and a mast cell line (PT18(A17]. All lines were dependent on IL-3 for survival but FDC-P1, DA-1, and DA-3 showed varying degrees of short-term proliferation in granulocyte-macrophage colony stimulating factor (GM-CSF). The cell lines all expressed asialo GM1 and Ly-5 surface markers but differed with respect to other markers. DA-1 expressed MAC-1, FDC-P1 and DA-3 expressed Thy-1, and PT18(A17) expressed receptors for the Fc portion of IgE. The cell lines varied greatly in their cytotoxic activity against WEHI-164. FDC-P1, DA-1, and PT18(A17) had low NC activity. DA-3 had consistently high activity, greater than that seen with M1-A5 cells. However, none of the cell lines secreted constitutively a suppressor cell inducing factor (SIF). In addition, it was demonstrated that recombinant murine TNF did not activate suppressor cells capable of inhibiting antibody synthesis and that anti-TNF did not block SIF activity, thus suggesting that TNF contamination of the M1-A5 derived SIF preparation is not responsible for the induction of suppressor cells. We conclude that suppressor cell inducing factors are not universally secreted by IL-3-dependent cell lines, that there is no correlation between NC and SIF activity, and that the dual activities of M1-A5 cells are not mediated by TNF.

Comparison of MHC antigen expression on PHA- and MLC-induced T cell lines with that on T and B lymphoblastoid cell lines by cell cycle dependency.

Although it is well known that the expression of major histocompatibility complex (MHC) antigens on the surface of lymphoblastoid cell lines are cell cycle dependent, the way in which the MHC antigen expression on activated T cells varies with cell cycle phase has not previously been described. Using 11 lymphoblastoid cell lines from malignant and nonmalignant tissues (B cells, T cells, and myeloid cells) and five activated T cell lines (two cell lines activated by phytohemagglutinin and three alloreactive T cell clones), MHC antigen expression was quantitatively studied by dual-beam flow cytometry. Correlated measurements of surface antigen quantity (immunofluorescence), DNA content (Hoechst 33342), and cell size (light scatter), uninfluenced by induction synchrony and cell fixation, were performed. The data indicate that cell surface antigen quantity and cell surface area demonstrate specific values at each phase of the cell cycle when the cells are in logarithmic growth. Examining cells in logarithmic growth, it was confirmed, for all lymphoblastoid cell lines, that the quantity of MHC antigens on G2 (S + G2 + M) cells was greater than that on G1 cells. In addition, it was found, by analyzing antigen quantity and surface area, that class I antigen density in the G2 phase is 17% less than that in the G1 phase in leukemic T cell lines, and that both class I and class II antigen densities in the G2 phase were 21% less than that in the G1 phase in lymphoblastoid B cell lines. In activated T cells, class I antigen density in the G2 phase was 11% less than that in the G1 phase, while class II antigen density in the G2 phase was 12% greater than that in the G1 phase. We describe four important observations in this report. In both G1 and G2 phases, activated T cells express: quantitatively fewer class I antigens than lymphoblastoid B cell lines; similar quantity of class I antigens as that of leukemic T cell lines; and similar quantity of class II antigens as that of lymphoblastoid B cell lines. Also, class II antigens are expressed in greater density in the G2 phase than in the G1 phase in activated T cells. In contrast, lymphoblastoid B cell lines express greater density of class II antigens in the G1 phase than in the G2 phase of the cell cycle. These findings differ from previous reports, suggesting that G1 phase cells may have a more significant role than G2 phase cells as target cells for MHC restricted cytotoxic cells.

Human lymphoid cell lines and glucocorticoids: I. Characterization and cytolethal responses of lymphoblastoid, leukaemia and lymphoma lines.

Using various genotypic and phenotypic markers 20 human lymphoid cell lines have been classified as lymphoblastoid, leukaemia or lymphoma subtypes. Each cell line type exhibited characteristic morphological and behavioural properties in suspension culture. Whilst most lymphoblastoid and lymphoma cell lines manifested B-cell phenotypes and contained Epstein Barr virus (EBV) genome, leukaemia lines demonstrated T-cell markers and lacked EBV genome. Individual cell lines demonstrated unique isoenzyme profiles for the seven polymorphic enzymes studied without subtype specificity. None of the cell lines studied was entirely homogeneous although lymphoblastoid lines contained only a minor subpopulation of other cell line types. The mixed population of cells indicate the need for caution in the use of these cell lines as in vitro models of lymphoid cancer and suggests further refinement of classification methods is required. Incubation of different cell line types with prednisolone for 48-168 h revealed most were highly resistant to cytolethal and cytostatic effects of glucocorticoids in vitro. Suprapharmacological doses of steroid (10(-3) M) were required in most instances before significant cytolethal responses occurred. Only one lymphoblastoid, one lymphoma and two leukaemia lines responded to pharmacological doses (10(-5)-10(-6) M) of prednisolone.

Establishment and characterization of 12 uterine cervical-carcinoma cell lines: common sequence variation in the E7 gene of HPV-16-positive cell lines.

A total of 12 carcinoma cell lines of the human uterine cervix were established from 5 keratinizing and 5 nonkeratinizing squamous-cell carcinomas, and 2 small-cell carcinomas. Of these, 10 lines grew as adherent cells and 2 as floating aggregates. All lines showed (i) similarity in morphology to the primary tumor from which they were derived; (ii) high viability with relatively long doubling times (48-96 hr); (iii) absence of Mycoplasma and other bacteria, apart from one Mycoplasma-contaminated line; (iv) genetic heterogeneity by DNA-fingerprinting analysis; (v) absence of p53 mutation from exon 4 through 9; and (vi) the presence of HPV DNA sequence. Among the lines, 7 were infected by HPV-16, 3 by HPV-18, 1 by HPV-31, and 1 by HPV-33; the 2 cell lines derived from small-cell carcinomas contained HPV-18. Interestingly, 6 of the 7 cell lines containing HPV-16-type DNA harbored the same alteration of E7 at nucleotide position 647 (amino acid 29, AAT --> AGT, Asn --> Ser), whereas the 3 HPV-18-positive lines did not; 3 cell lines proved to have intact E1/E2 of HPV, suggesting the presence of episomally replicating HPV DNA as well as the integrated form, whereas the other 9 lines were shown to have integrated HPV. Taken together, these cell lines would be very useful for studying the biology of uterine cervical carcinoma.

Establishment and characterization of 7 ovarian carcinoma cell lines and one granulosa tumor cell line: growth features and cytogenetics.

The characteristics of 7 newly established ovarian carcinoma cell lines and one granulosa tumor cell line obtained from tumor samples of 7 patients with varying histology of the primary tumor are reported. The cell lines were isolated from 5 serous carcinomas, a mucinous carcinoma, an endometrioid carcinoma and a granulosa cell tumor. All cell lines were passaged at least 25 times and showed stable growth rates. Colony-forming efficiency varied on plastic from 2 to 57% and in agar from 0.01 to 9.30%. The DNA index of the granulosa tumor cell line was diploid, while the ovarian carcinoma cell lines were all aneuploid. In 2 cell lines polyploidisation occurred during culturing. A thorough cytogenetic analysis of 7 cell lines revealed that the granulosa tumor cell line has only minor cytogenetic abnormalities (+5, 22q+). In contrast, the epithelial ovarian-cancer cell lines gave very complex karyotypes with numerous markers and structurally rearranged chromosomes. The chromosomes most often in excess were 15 and 20. Structural rearrangements of chromosomes 1, 3, 7 and 11 were prominent in all ovarian cell lines. In addition, we found changes in chromosomes X, 5, 8 and 13 that have rarely been described before.

A dotted line assimilates in visibility to a solid line.

This study tested the theory that a context-produced increase in visibility of a target is due to its assimilation in visibility to the context. A context + target and a context are discriminated better than are a target and background. This occurs for two different context + targets in which the context is a solid line and the target is a dotted line. But it does not occur when solid lines replace these dotted lines. The dotted lines are much less visible than the solid lines. Therefore, the dotted lines increase in similarity in visibility to the solid lines, which is assimilation, but for visibility, rather than for a typical part. Assimilation does not occur between perceptually equal parts. Consequently, the reason why the two context + targets with only solid lines do not result in increases in visibility may be that these lines are sufficiently equal in visibility that assimilation in visibility is precluded. So, the theory is supported. This theory is consistent with evidence that one group (phenomenal whole) is associated with both assimilation and an increase in visibility. Accordingly, a stimulus with a relatively large distance between its solid and dotted lines is apprehended as a relatively weak group, and does not result in an increase in visibility.

False leukemia-lymphoma cell lines: an update on over 500 cell lines.

Human leukemia-lymphoma (LL) cell lines represent an extremely important resource for research in a variety of fields and disciplines. As the cell lines are used as in vitro model systems in lieu of primary cell material, it is crucial that the cells in the culture flasks faithfully correspond to the purported objects of study. Obviously, proper authentication of cell line derivation and precise characterization are indispensable requirements to use as model systems. A number of studies has shown an unacceptable level of LL cell lines to be false. We present here the results of authenticating a comprehensively large sample (n = 550) of LL cell lines mainly by DNA fingerprinting and cytogenetic evaluation. Surprisingly, near-identical incidences (ca 15%) of false cell lines were observed among cell lines obtained directly from original investigators (59/395: 14.9%) and from secondary sources (23/155: 14.8%) implying that most cross-contamination is perpetrated by originators, presumably during establishment. By comparing our data with those published, we were further able to subclassify the false cell lines as (1) virtual: cross-contaminated with and unretrievably overgrown by other cell lines during initiation, never enjoying independent existence; (2) misidentified: cross-contaminated subsequent to establishment so that an original prototype may still exist; or (3) misclassified: unwittingly established from an unintended (often normal) cell type. Prolific classic leukemia cell lines were found to account for the majority of cross-contaminations, eg CCRF-CEM, HL-60, JURKAT, K-562 and U-937. We discuss the impact of cross-contaminations on scientific research, the reluctance of scientists to address the problem, and consider possible solutions. These findings provide a rationale for mandating the procurement of reputably sourced LL cell lines and their regular authentication thereafter.

Does attention have different effects on line orientation and line arrangement discrimination?

Visual search and texture segregation studies have led to the inference that stimuli differing in the orientation of their component line segments can be distinguished without focal attention, whereas stimuli that differ only in the arrangement of line segments cannot. In most of this research, the locus of attention has not been explicitly manipulated. In the first experiment presented here, attention was directed to a relevant peripheral target by a cue presented near the target location or at the fovea. Effects of attention on orientation discrimination were assessed in a two-alternative forced-choice task with targets that were either: (1) lines that slanted obliquely to the right or left, or were horizontal or vertical, or (2) Y-like targets that had a short arm leading obliquely right or left of a vertical line. In some groups, a four-alternative forced-choice test with lines at 0 degree, 45 degrees, 90 degrees, and 135 degrees orientations was used. Discrimination of these targets (i.e. targets that differ in the orientation of component line segments) was only minimally facilitated as the time between the onset of the valid cue and the onset of the target (cue-target stimulus onset asynchrony, SOA) was increased from 0 or 17 msec to 267 msec. In contrast, discrimination of targets that did not differ in the orientation of component line segments but differed in line arrangement (T-like characters), was greatly facilitated by longer cue-target SOAs. In Experiment 2, a cue misdirected attention on 20% of the trials. A decrement occurred on incorrectly cued trials in comparison to correctly cued trials for both types of stimuli used (lines and Ts). The results from these experiments suggest that discrimination of line orientation benefits less from focal attention than does discrimination of line arrangement, but that both discriminations suffer when attention must be disengaged from an irrelevant spatial location.

Genetic variation in pure lines and crosses of large-bodied turkey lines. 2. Carcass traits and body shape.

An experimental line (F) of turkeys was reciprocally crossed with sire lines (designated A and B) from each of two commercial breeders in order to study the inheritance of carcass traits and body shape. The birds were weighed and killed at 17 wk of age, and various measurements of muscling, leg bones, and body shape were made. Additive genetic variation, as indicated by line differences, was an important source of variation in most traits. The only traits that did not exhibit a line difference in any comparison were weight of the drumstick muscles, tarsometatarsal width, keel length, and Body Depth 2 (body depth at a point 2.54 cm anterior to the posterior end of the keel). Heterosis of live BW was greater in males than in females. Heterosis in the weight of the pectoralis major and p. minor muscles were similar to those of live BW, but heterosis in the weight of the leg muscles were higher than that of live BW. The average heterosis for bone measurements (length of the femur, tarsometatarsal, and tibiotarsal bones and tarsometatarsal width) was very low and ranged from -0.3 to 1.4%. For measurements of body shape [keel length, Body Depth 1 (body depth measured at the cranial process of the keel), Body Depth 2, ratio of Body Depth 1 to Body Depth 2, length, width, and height of the body cavity, and body cavity volume index), heterosis was low and the average ranged from -2.7 to 2.6%. Based on an analysis of both sexes combined, the commercial sire lines differed in BW, weights of the p. minor and drumstick muscles, weights of the tarsometatarsal, femur, and tibiotarsal bones, lengths of the tarsometatarsal and femur bones, and various measurements of body shape (Body Depth 1, body depth ratio, body cavity length and height, and body cavity volume index). Relative to the commercial sire lines, the F line was smaller, had less breast and leg muscling, and, in general, larger leg bones when sexes were combined. The carcass of the F line was deeper than that of the commercial sire lines when measured at the cranial process of the keel but not at 2.54 cm anterior to the posterior portion of the keel. Body cavity height and body cavity volume index were larger in the F line than in the commercial sire lines.
