Endoscopic, histologic, and microbiologic evaluations of 21 cynomolgus and 34 rhesus monkeys for naturally occurring Helicobacter pylori infection were done. H. pylori was never isolated from any cynomolgus monkey, but was found in 12 rhesus monkeys. A general correlation existed between a positive culture and a gastric inflammatory response. Inoculation challenges were then undertaken. Four cynomolgus and five rhesus monkeys received two different H. pylori strains isolated from humans. Five rhesus monkeys received an isolate obtained from rhesus monkeys. Evaluation of the cynomolgus monkeys 7 and 14 days later revealed no H. pylori. Endoscopies of the rhesus monkeys were done 7, 14, 21, 28, and 56 days later. One rhesus monkey, which received the isolate from humans, became H. pylori positive at day 21 and remained positive through day 56. Restriction enzyme analysis of genomic DNA at day 56 revealed that the isolate was not identical to the challenge strain isolated from humans. All five rhesus monkeys that received the strain isolated from rhesus monkeys became H. pylori positive by day 14 and remained positive through day 56 Antral inflammation developed in all monkeys. Restriction enzyme analysis of genomic DNA on day 56 confirmed that four of five isolates were identical to the challenge strain isolated from rhesus monkeys. DNA hybridization documented homology between the challenge strains isolated from humans and rhesus monkeys plus those isolated at day 56. In this study, we showed that the rhesus monkey, if given a strain of H. pylori isolated from rhesus monkeys, develops a gastric infection with accompanying histological changes, making this model suitable for further development.
Chorionic gonadotropin (mCG) in serum and urine of monkeys was determined by a radioimmunoassay and hemagglutination inhibition (HAI subhuman primate tube test for pregnancy). HAI-positive pregnancy tests coincided fully with the mCG excretion patterns as determined by RIA (oLH beta system). The least reliable system was the hCG-beta RIA. The HAI test allowed accurate prediction of the parturition date.
M fascicularis monkeys were immunized subcutaneously in the vicinity of the major salivary glands and by retrograde infusion into the parotid duct, with a vaccine containing Formalin-killed S mutans strain 6715 cells and culture-fluid antigens. Indirect immunofluorescent staining was used to titrate and classify antibodies. Subcutaneous immunization induced only a serum response, whereas intraductal infusion stimulated both an IgA antibody response in the parotid fluid and a serum response. Immunized and nonimmunized control groups were orally infected with S mutans strain 6715. The establishment in dental plaque was quantitated by recovery of the infecting organism on selective media and by immunofluorescent staining of plaque smears taken from individual tooth surfaces. The establishment of S mutans strain 6715 was noticeably inhibited in immune monkeys. Immunofluorescent assays for antibody also showed that serum and parotid fluid containing serum IgA antibodies cross reacted with other d serotype and a serotype strains but not representative b and c strains. Immune and control groups were then orally infected with S mutans strain GS-5, a c serotype strain, and no inhibition in establishment was detected of the non-cross-reacting type c organism in the immune group. A latter series of booster immunizations via the intraductal route resulted in a significant decrease in parotid fluid flow. Histological investigations showed inflammatory cell infiltration and replacement of epithelium by connective tissue in the glands from immunized monkeys. A separate group of monkeys, younger than the first, was immunized with the same vaccine via the duct only. In this group, immunizations were given at shorter intervals, but the immunization response was similar to that observed in the first group. The investigations reviewed here and new experiments reported show that immunization of monkeys with S mutan strain 6715 via the parotid duct elicited a reproducible IgA antibody response in the parotid fluid as well as a serum antibody response. Inhibition of colonization on tooth surfaces in immune monkeys showed specificity for the immunizing strain suggesting that inhibition was antibody mediated.
To investigate the nature underlying the process of pattern discrimination learning, a series of seven experiments on seven working assumptions were undertaken. The main findings are as follows. The pattern discrimination learning consists of two time-dependent stages: The initial or first learning stage is the period of performance at chance, and the succeeding or second stage is the period of performance from just above the chance to a criterion level. The duration of the first stage is dependent on the degrees of cue-response separations, whereas that of the second stage is independent of. During the first stage, monkeys do not attend to the discriminative cue even at small cue-response separations, whereas during the second stage, they achieve pattern discrimination, or pattern perception and cognition, regardless of cue-response separations. After having learned the first pattern task with a cue-response separation, they learned new pattern tasks by means of the second stage, showing marked saving of the duration of the first stage. The findings in the present studies indicate that the first stage of learning is predominantly involved in the process of attending to the discriminative cues remote from the response site (a selective attention to the cue), whereas the second stage is concerned with the process of perception and cognition of the discriminative cue.
To elucidate the role of the amygdala in visual perception and cognition, the effects of ablations of the amygdala and inferotemporal cortex on several visual tasks were compared with each other, and also the distribution patterns of the projections between them were investigated. The findings indicate that the inferotemporal cortex plays a critical role in visual perception, cognition and memory, whereas the amygdala is involved fundamentally in controlling emotional and motivational behavior. However, the amygdala is concerned with vision in the following ways: It receives neutral visual information highly processed in the visual cortex, invests the information with emotional and motivational significance through interactions with the cortical and subcortical systems of emotion and motivation, and then it returns the information coded to the visual areas to be re-processed; to be consciously perceived in area TEO, and to be meaningfully cognized, recognized and memorized in areas TE and TEG. Therefore, two channel model regarding the mechanism of visual information processing in the inferotemporal cortex is proposed: A first channel is concerned with processing neutral information, while the second one, with processing meaning information coded emotionally and motivationally in the amygdala. In addition, the present studies demonstrate that area TEG, which is cytoarchitecturally a transitional area between areas TE and TG and whose functional significance has remained unclear, is involved significantly in visual cognition rather than visual perception.
BACKGROUND AND PURPOSE: Q fever is a disease of humans. Vaccines to prevent this disease have demonstrated efficacy in rodents and must also be evaluated for efficacy in a nonhuman primate model. Preliminary to vaccine efficacy experiments, cynomolgus and rhesus monkeys were evaluated as suitable experimental models of acute Q fever. METHODS: Both species of monkeys were challenged with aerosolized 10(5) virulent phase-I Coxiella burnetii Henzerling strain, and clinical and serologic responses were determined. RESULTS: Radiographic changes were observed in seven of eight monkeys of both species; however, changes in cynomolgus monkeys tended to be more significant. Between 7 and 10 days after challenge, all rhesus monkeys and 88% of cynomolgus monkeys were bacteremic. Sequential increases in antibody responses to C. burnetii phase-I and phase-II whole cells and phase-I lipopolysaccharide were observed in both species. Although the maximal rectal temperature increase was similar in both species, duration of fever was slightly longer in rhesus monkeys. Clinical features were similar to those described in human acute Q fever patients. CONCLUSIONS: On the basis of the more pronounced radiographic changes in cynomolgus monkeys, we favor use of this species for future studies of vaccine efficacy.

A developmental comparison of the fields 8, 9, 10, 11, 12, 47 during prenatal life in macacus rhesus and man was performed. Cytomyeloarchitectonic and morphometric methods of investigation were applied: the areas of the fields in question, the transverse thickness of the cortex, separate cortical layers, neuronal dimentions were measured. Alongside, general regularities in the development of these fields in both species (the frontal cortex is laid earlier than other parts of the cortex, the terms of the field differentiation coincide, uneven development of the fields with different phylogenic origin) some important differences are demonstrated. They are seen a) in the rate of the frontal field maturation--in man it takes longer time, b) in a more complex organization of the frontal part in man--during prenatal life new structural features of the fields, 44, 45, 46 and subfields in the field 47 appear. During prenatal life of monkey and man in the development of the frontal area the phenomena of heterochrony (common for both species) and anabolia (peculiar for man only) are observed (the appearance of new more complex structures). These phenomena display themselves in the shift of the laying of the frontal area towards earlier stages of embryogenesis.
Using four female Japanese monkeys, follicular growth was induced by multiple PMSG and single hCG injections, and oocytes were collected from their follicles. PMSG, 200 IU, was injected six or eight times to stimulate follicular growth, and in both cases growth of the follicles was adequately induced. Forty-seven (50.0%) oocytes were recovered from 94 follicles of two of the females and transported from Inuyama-shi to Tsukuba-shi in 10 hours. Five mature oocytes and 42 immature oocytes were observed upon arrival. Thirteen (31.0%) of the 42 immature oocytes released the first polar body in TCM-199 medium containing FCS and PMSG after 24 hour-culture, including transport time. Eighteen mature oocytes were used for in vitro fertilization (IVF). Frozen and thawed cynomolgus monkey spermatozoa preincubated for 2 hours were transferred into the drops containing the Japanese monkey oocytes. Sixteen oocytes (88.9%) were judged to have been successfully fertilized based on the presence of a male pronucleus and/or second polar body, and 12 oocytes then cleaved, and 3 developed into the 6-cell stage. Thus, we succeeded in inducing follicular growth and oocyte maturation, and in achieving IVF using cynomolgus monkey spermatozoa and Japanese monkey oocytes.
