Uredofos, niclosamide, bunamidine hydrochloride, and arecoline hydrobromide were given at various doses to dogs experimentally infected with Mesocestoides corti. The highest efficacies were obtained with bunamidine hydrochloride and uredofos. Variable results were obtained with arecoline hydrobromide and niclosamide.
In long-term experiments with Hooded rats the carcinogenic potential of 1-methyl-3(p-bromophenyl)-1-nitrosourea (Br-MPNU) could be demonstrated for the first time. Br-MPNU is formed also endogenously after combined administration of 1-methyl-3(p-bromophenyl)-urea (Br-MPU) and sodium nitrite. After repeated intragastric administration of 0.33 mmol Br-MPU and 0.73 mmol NaNO2 per kg b.w. papillomas and carcinomas of the forestomach developed in 83%. After repeated administration of 0.28 mmol Br-MPNU per kg b.w. these neoplasms were observed in 88%. The comparison of results obtained in similar experiments with 1-methyl-3-phenyl-1-nitrosourea shows that bromine substitution led to a reduction of the carcinogenic activity. The present paper is part of a complex program studying the interrelationships between structure, physico-chemical properties, mutagenicity and carcinogenicity of nitrosoureas.
Diuredosan has been tested against Echinococcus granulosus and Taenia hydatigena infections in dogs. The drug showed significant activity against T hydatigena, but there was no significant dose response curve against E granulosus in this trial. Two treatments at 50 mg/kg or 100 mg/kg, however, reduced the number of dogs infected with hydatid worms. Worms were still present in some dogs after three treatments at 50 mg/kg. Vomiting and diarrhoea were relatively common sequelae at and above 50 mg/kg.

Polarographic fluoroimmunoassay was developed for estimation of residual amounts of the herbicide isoproturon . The procedure is simple, accurate, highly sensitive and allows 10 preparations to be tested within approximately 7 min; the sample volume was 50 microliters and the low limit of the procedure sensitivity was about 0.2 micrograms/ml. The polarographic immunoassay may be used in screening express controls of isoproturon concentration in the sewage or in various extracts from various sources to control the herbicide contamination.
The teratogenic effects of three herbicides (Flubalex, Fusilade S and Maloran 50 WP) were studied in chicken embryos. Each of the three test substances was administered on days 0 and 12 of incubation. Treatment was followed by evaluation on day 19. The compounds were injected into the air-chamber of eggs at three different concentrations. The medium concentration corresponded to that usually applied in chemical plant protection. In order to determine the combined toxicity of the three herbicides, the medium concentration of Maloran 50 WP and three different concentrations of Flubalex of Fusilade S each were administered simultaneously at a final volume of 0.1 ml per egg, at similar times. Evaluation was done on day 19. In tests of individual toxicity, after injection on day 0 of incubation Maloran 50 WP and Flubalex caused a significant reduction in body mass, while Maloran 50 WP and Fusilade S resulted in marked embryonic mortality. After injection on day 12, the medium and the highest concentration of Flubalex and the highest concentration of Fusilade S caused a marked increase in embryonic mortality. The developmental anomalies were of sporadic nature: their incidence increased only after Flubalex treatment, irrespective of the time of administration. The combined administration of Maloran 50 WP and Flubalex on day 0 resulted in a significant or marked body mass reduction in all groups. Embryonic mortality increased substantially after treatment with the highest dose of Flubalex, while all three concentrations of the other two herbicides led to similar results. When treatment was performed on day 12, the two highest concentrations of Flubalex and the highest concentration of Fusilade S caused expressed embryonic mortality. The developmental anomalies did not show a dose-dependent effect in any of the test series.
On 27 January 1996, about 1 ton of Isoproturone was released into the atmosphere due to an accident in a chemical plant located at Frankfurt-Griesheim, Germany. A nearby residential area was contaminated by up to 500 mg Isoproturon/m2. According to the published toxicological data and the contamination data, model calculations were performed demonstrating that the individual exposure remained below the LOEL. For better exposure and risk assessment urine biomonitoring was offered to the residents. HMEPMH, the main metabolite of Isoproturon was detected in 12% (19/154) of the participants in the test, with a maximum value of 14.2 micrograms/l, i.e. the internal exposure remained even below ADI. Control tests demonstrated the decrease of the soil contamination after some weeks, and by June 1996 Isoproturone was no longer detectable, neither in soil nor in vegetables.
The possibilities of using new chemically bonded anion-exchange (Silasorb DEA) and cation-exchange (Silasorb S) materials for high-performance liquid chromatography of phenylurea herbicide compounds were investigated. The columns packed with Silasorb DEA or with Silasorb S ion-exchange materials can be used for efficient separations in normal-phase systems using n-propanol-n-hexane as mobile phases. Here, specific selectivity effects for phenylurea-type herbicides can be achieved in comparison to chromatography on unmodified silica or on chemically bonded aminosilica or cyanosilica, especially with Silasorb S columns. The influence of the mobile phase composition and of the structure of phenylurea herbicides on the chromatographic behaviour on Silasorb S and Silasorb DEA columns was investigated in detail and the possibilities of separation are illustrated by several examples.
Potency and cross-resistance of various biorational insecticides, exemplified by the whitefly Bemisia tabaci, have been studied. Bemisia tabaci were exposed to the juvenile hormone mimic pyriproxyfen for the past 12 years resulting in an over 2,000-fold resistance, but there was no appreciable cross-resistance with the benzoylphenyl urea novaluron. Similarly, no cross-resistance was found between pyriproxyfen and the two neonicotinoids, acetamiprid and imidacloprid. On the other hand, a slight cross-resistance of 5-13-fold was observed with another neonicotinoid thiamethoxam. Among the neonicotinoids, a resistant strain of B. tabaci to thiamethoxam (approximately 100-fold) showed no appreciable cross-resistance to either acetamiprid or imidacloprid, while another strain 500-fold resistant to thiamethoxam resulted in a mild of 4-6-fold resistance to acetamiprid and imidacloprid. In other assays, B. tabaci strain resistant to thiamethoxam (approximately 100-fold) had no cross-resistance to pyriproxyfen. Our findings indicate that no appreciable cross-resistance was observed between the benzoylphenyl urea novaluron, the juvenile hormone mimic pyriproxyfen, and the neonicotinoids acetamiprid and imidacloprid. Hence, these compounds could be used as components in insecticide resistance management programs.
The potential to mineralize 2,4-dichlorophenoxyacetic acid (2,4-D), mecoprop, isoproturon and terbuthylazine was studied in soil and aquifer chalk sampled at an agricultural field near Aalborg, Denmark. Laboratory microcosms were incubated for 258 days under aerobic conditions at 10 degrees C with soil and chalk from 0.15-4.45 m below the surface. The [ring-U-14C]-labeled herbicides were added to obtain a concentration of 6 micrograms kg-1 and mineralization was measured as evolved [14C]carbon dioxide. The herbicides were readily mineralized in soil from the plough layer, except for terbuthylazine, which was mineralized only to a limited extent. In the chalk, lag periods of at least 40 days were observed, and a maximum of 51%, 33% and 6% of the added 2,4-D, mecoprop and isoproturon, respectively, were recovered as [14C]carbon dioxide. Large variations in both rate and extent of mineralization were observed within replicates in chalk. No mineralization of terbuthylazine in chalk was observed. As a measure of the general metabolic activity towards aromatic compounds, [ring-U-14C]-benzoic acid was included. It was readily mineralized at all depths.
The uncalibrated predictive ability of four preferential flow models (CRACK-NP, MACRO/MACRO_DB, PLM, SWAT) has been evaluated against point rates of drainflow and associated concentrations of isoproturon from a highly structured and heterogeneous clay soil in the south of England. Data were available for four plots for a number of storm events in each of three successive growing seasons. The mechanistic models CRACK-NP and MACRO generally gave reasonable estimates of drainflow over the three seasons, but under-estimated concentrations of isoproturon over a prolonged period in the first season and over-estimated them in the two remaining seasons. CRACK-NP simulated maximum concentrations of isoproturon over the first two events of each of the three seasons of 156, 527 and 24.4 micrograms litre-1, respectively, and matched the observed data (465, 65.1 and 0.65 micrograms litre-1) slightly better than MACRO (69.1, 566 and 58.5 micrograms litre-1). Automatic selection of parameters from soils information within MACRO_DB reduced the emphasis on preferential flow relative to the stand-alone version of MACRO. This gave a poor simulation of isoproturon breakthrough and simulated maximum concentrations were 0, 50.1 and 35.1 micrograms litre-1, respectively. The capacity model PLM gave the best overall simulation of total drainflow for the first two events in each season, but over-estimated concentrations of isoproturon (967, 808 and 51.3 micrograms litre-1). The simple model SWAT represented total drainflow reasonably well and gave the best simulation of maximum isoproturon concentrations (140, 80.2 and 8.2 micrograms litre-1). There was no clear advantage here in using the mechanistic models rather than the simpler models. None of the models tested was able to simulate consistently the data set, and uncalibrated modelling cannot be recommended for such artificially drained heavy clay soils.
The aim of this study was to investigate the spatial variability in degradation and mineralization of atrazine and isoproturon in subsurface samples taken from sandy loam soils overlying gravel terraces which form part of a groundwater protection zone. Percussion drilling was used to obtain samples from 11 boreholes (maximum depth 3 m). Unlabelled atrazine or isoproturon, and ring-14C-labelled atrazine or isoproturon were added to samples, incubated at 25 degrees C for up to 16 weeks, and analyzed for the residual herbicide or [14C]carbon dioxide. All samples showed the potential to degrade these herbicides, although the percentage degradation decreased by a factor of 2-3 from the surface soil to a depth of 3 m. This was associated with a decrease in organic matter content, but there was no change in the potential to mineralize acetate, indicating that specific changes in the catabolic ability of the microbial population occurred with depth. The capacity of samples to mineralize atrazine and isoproturon to carbon dioxide decreased markedly with depth, with no mineralization potential observed at a depth of 80 cm.
Black-grass (Alopecurus myosuroides Huds) is a major grass weed in winter cereals in Europe. It reduces yields and can act as a secondary host for a range of diseases. Herbicide resistance in this species was first detected in the UK in the early 1980s, and has now been reported in thirty counties. To successfully manage herbicide resistance it is vital that suspect populations are tested so that appropriate action can be taken. Ideally, a test will be quick, cheap and easy to use. Furthermore, it should provide an unequivocal result before post-emergence herbicides are to be applied, allowing alternative strategies to be adopted where necessary. This paper reports the development of new tests for herbicide resistance based on our observation that the resistant black-grass biotype Peldon contains approximately double the activity of the enzyme glutathione S-transferase (GST) compared with susceptible biotypes. Data are presented on the production of a monoclonal antiserum to a novel 30 kDa GST polypeptide purified from the biotype Peldon. An ELISA using this antiserum is described and the utility of this assay to detect resistant black-grass biotypes in plants grown under glass and in the field is presented. In addition, a microtitre assay for GST activity is described, which allows the rapid assessment of GST activities of plants. Both abundance and activity of GSTs are discussed as markers for herbicide resistance in black-grass.
Metallothioneins (MTs) are low-molecular-weight proteins mainly involved in metal ion detoxification. Recently it has been demonstrated that MTs participate in several cellular functions such as regulation of growth and antioxidative defenses. Moreover, pesticides can induce their synthesis. The aim of the current work was to determine the effects of isoproturon, either pure or formulated as Matin (suspension containing an isoproturon concentration of 500 g. L(-1)), on the metallothionein and total protein contents of the aquatic worm Tubifex tubifex. MT levels in exposed worms increased significantly after 7 and 15 days of exposure to a concentration of the herbicide of 50 mg. L(-1). Isoproturon reduced the metal (Cu, Zn, and Cd) content of metallothioneins, and it also increased the total protein content of the worms. These results suggest that MT induction may not be considered a specific biomarker of metal exposure but that it can be used as a nonspecific biomarker of the effect of isoproturon effect in aquatic worms.
Preliminary results of a new method to investigate the OH radical reactivity of semi-volatile organic compounds (e.g., pesticides) are presented. Terbuthylazine, simazine, sodium benzoate, and bromoxynil were adsorbed on highly disperse silicon dioxide powder as an unreactive carrier at a thickness well below one monolayer. The coated material was suspended in air as an aerosol, sampled on filters, and exposed in an 840-liter Duran chamber to OH radicals, produced by photolysis of hydrogen peroxide in the gas phase. Sunlamps on top of the chamber were used as cold light sources [T(aerosol) approximately 25 degreesC]. OH radical concentrations (10(5)<cOH (cm-3)<3x10(6)) were monitored in the gas phase using a set of four hydrocarbons with well-known OH reactivities as reference compounds. The triazine terbuthylazine (kOH=1.1x10(-11) cm3 s-1) was used as a reference compound in the filter samples. Simazine and isoproturon were found to react with a comparable OH rate constant with respect to terbuthylazine. Sodium benzoate reacts about a factor of 3 slower. Rapid degradation was observed for bromoxynil, explained mainly by photolysis. Besides the characterization and discussion of the experimental setup used, the rate constants obtained are discussed and compared with estimated values.
Using ring-(14)C-labeled isoproturon (1 microg/L), the uptake into spawn and tadpoles of Bombina bombina and Bombina variegata was investigated. Two percent of the applied radioactivity was found per gram fresh weight in the embryo after 24h. Results indicate that the jelly mass of the spawn does not act as a sufficient physical barrier for protection against the uptake and influence of isoproturon (IPU) on the embryo. In vivo metabolism of ring-14C-labeled IPU by the cytochrome P-450 system was analyzed in tadpoles. Different metabolites of IPU, such as N-demethylated and C-hydroxylated derivatives, and the olefinic metabolite were detected. In tadpoles of B. variegata, the activity of microsomal and soluble glutathione-S-transferase (sGSTs) toward different model substrates was measured after treatment with IPU. Activities of sGST increased corresponding to elevated stress by IPU dependent on exposure time and dose. Compared to the pure active ingredient IPU, the commercial phenyl-urea herbicide Tolkan Flo, consisting of IPU and an emulsifier, also caused significantly elevated enzymatic response.
