[The comparison of molecular-biochemical and cytogenetic analyses of blood cells of patients long after the acute radiation sickness].

Molecular-biochemical and cytogenetic analyses were made on blood cells of 17 radiation accident victims who, from 1.7 to 43.8 years previously, had suffered acute radiation sickness (ARS) ranging from severity grades I to IV. Molecular-biochemical data were obtained with patients' leukocytes and with mononuclear cells on their oxidative status by a) the level of an anion-radical O2*- in the 3-(4,5-dimethyl-2-thiazol-2-yl)-2,5-diphenyl-2-tetrazolium bromide (MTT) test and b) the sum of reactive oxygen species in the 2,7-dichlorodihydrofluorescein diacetate (DCFDA) test together with a test for DNA strand unwinding in alkaline conditions by measurements of fluorescence intensity of ethidium bromide. Each parameter was measured in freshly sampled cells and during a 5-hour incubation as absolute means on 5 measurements and in % change from the initial values. Cytogenetic data were obtained from the standard metaphase preparations scored for routine unstable chromosomal aberrations (us-CA)--dicentrics; and stable aberrations (sCA)--translocations. The latter ones were assayed by the FISH method using whole chromosome 2, 4 and 12 fluorescent probes and scaled up to genome equivalence. For all patients reduced oxidative status of about 25-30% was obtained by the MTT-test (p < 0.005), and the DCFDA-test (p < 0.027). The yield of usCA depended on the time after irradiation with higher yields associated with the shorter postirradiation times and reducing almost to expected background frequencies. The yield of sCA was high for all patients, correlating with the severity of ARS whilst the molecular-biochemical parameters showed no relationship with ARS. A correlation was observed between parameters of oxidative status and % of cells with usCA: by the MTT-test r = from 0.50 up to 0.61 (p = from 0.06 up to 0.003), but by the DCFDA-test the strength of correlation was smaller: r = from 0.38 up to 0.48. An inverse correlation was found between initial oxidative state of mononuclear cells and the frequency of CA in lymphocytes. Similarly a marked inverse correlation between degree of DNA unwinding by the ethidium bromide assay on leukocytes and sCA in lymphocytes was also noted. The feasibility of radiation-induced delayed genomic instability in vivo for humans long time after irradiation is discussed.

[Determination of autoantibodies to antigens of thymic epithelial cells of clean up team workers and patients who survived acute radiation sickness at distant periods after irradiation].

A group of patients, suffering from sequelae of acute radiation sickness (ARS), and liquidators was studied 5 years after exposure to a complex of factors resulting from the Chernobyl A.P.S. disaster. Studied were: the antibody titres to antigens of the cytoplasm of thymus epithelial reticulum cells and to Hassall's corpuscles the levels of serum immunoglobulins M, G, A; and the content of serum alpha 1-thymosin. Patients with ARS sequelae and liquidators showed a high level and incidence of autoantibodies to antigens of cytoplasm of thymus epithelial reticulum cells and to Hassall's corpuscles. There were no significant differences between the antibody levels in the blood of patients with ARS sequelae and liquidators. The antibodies were found to belong to IgM class; there was a correlation between the serum IgM titres and the rate of the indirect immunofluorescence reaction with autoantibodies to antigens of the cytoplasm of the thymus epithelial reticulum cells. To identify autoantibodies cryostat sections of human and mouse, (CBA x C57BL/6) F1, thymus as well as the epithelial and stromal cell culture of mouse thymus can equally be used.

EEG patterns in persons exposed to ionizing radiation as a result of the chernobyl accident. Part 2: quantitative EEG analysis in patients who had acute radiation sickness.

Cross-sectional quantitative electroencephalogram (qEEG) study (1996-2001) among Chernobyl accident survivors, who had confirmed acute radiation sickness and were irradiated in dose of 1-5 Gy, revealed the neurophysiological markers of ionizing radiation. Neuropsychological markers were: left fronto-temporal dominant frequency reduction; absolute delta-power lateralization to the left (dominant) hemisphere; relative delta-power increase in the fronto-temporal areas; absolute theta-power decrease in the left temporal region; absolute and relative alpha-power diffusive decrease, which may reflect cortico-limbic dysfunction lateralized to the left, dominant hemisphere, with the fronto-temporal cortical and hippocampal damage. Quantitative electroencephalogram proposed for differentiation of radiation and nonradiation brain damages and as a new biological dosymetry method. High radiosensitivity of the brain, neocortex, and dominant hemisphere higher radiosensitivity are discussed.

[Long-term (45-50 years) follow-up concerning medical and social status of patients with chronic radiation sickness].

The article covers evaluation of medical and social status in 673 subjects who suffered from chronic radiation sickness. By 45-50 years of follow-up, nearly 70% of the subjects had primary disablement established after the age of 60. Main causes of the disablement were cerebrovascular diseases and malignances. Increasing values of primary disablement in chronicradiation sickness patients over able-bodied age match increasing primary disablement among the country's general population and result from not only medical but also social matters.

[The information value of clinico-hematologic criteria for the early diagnosis of acute radiation sickness in pig-tailed macaques].

Ten pig-tailed monkeys (Macaca nemestrina) were subjected to 60Co radiation at a dose of 6.0-6.5 Gy and a dose rate of 1.2 Gy/min. Acute radiation sickness has developed in the monkeys causing their death on the 16-20 day. In spite of this, the initial reaction was weakly expressed and according to its manifestation it was impossible to evaluate severity and possible outcome of the lesion. At an early stage of the disease (6-24 hours) insufficient was uranin fluorescence in blood plasma, but more informative were the changes in adhesive properties of leukocytes the dynamics of lymphocytes (lymphopenia), reticulocytes (reticulocytopenia) and shifts in reticulograms (increased per cent of juvenile forms).

[Morphofunctional characteristics of the cells of the APUD system of the adrenal glands and the duodenum in the cerebral form of acute radiation sickness].

Changes of a phase nature develop in the APVD system cells of rats, dogs, and monkeys exposed to electron radiation of doses inducing a cerebral form of radiation sickness. These changes in the morphofunctional status of APVD system cells may be considered as a syndrome displaying acute radiation apudopathies.

[Characteristics of the development of radiation sickness in guinea pigs exposed to non-uniform irradiation in highly lethal doses].

Certain peculiarities have been revealed in the development of radiation sickness in guinea pigs subjected to nonuniform irradiation with superlethal doses. With electron and gamma-neutron radiation LD50/1.5 was 116.11 and 114.48 Gy respectively, when delivered to whole body, and 133.40 and 111.72 Gy when delivered to the posterior part of the body.

[The RBE of 0.85-MeV neutrons in the intestinal form of radiation sickness in guinea pigs].

Relative biological effectiveness (RBE) coefficient of 0.85 MeV neutrons was 1.87 in comparison with 0.66 MeV gamma radiation (137Cs) when estimated by the death rate of guinea pigs with intestinal form of radiation sickness. LD50/5 was 5.9 and 11.06 respectively. Features of the mortality rate dynamics, clinical picture and pathoanatomical changes are discussed.

[Thyroid diseases and the dynamics of their development in those convalescing from acute radiation sickness].

Patternture was studied together with the time course of development of thyroid disorders in 81 patients who had survived acute radiation sickness (ARS) in 1986, 88 irradiated individuals with unconfirmed ARS diagnosis, and 365 control, by results of clinical and laboratory examination carried out in 1989-1993. Increase in the incidence of functional disorders, especially that of hypothyrosis in the irradiated subjects (4.97% to 6.82%) can be regarded as realization of thyroidal non-stochastic effects in the immediate postaccident period.

[Free-radical processes and blood antioxidant systems in the late period following acute radiation sickness].

Some indices of free radical processes and antioxidant blood protection systems were investigated in a group of persons exposed to ionizing radiation at the Chernobyl accident. Lipid peroxidation was shown to undergo changes against a background of decreased antiperoxide protection of erythrocytes 22-24 mos. after exposure in patients after acute radiation sickness, I degree. As compared to donors, the examinees demonstrated a tendency to the accumulation of primary products of lipid peroxidation--hydroperoxides (by 32.2%) and a significant decrease in erythrocyte peroxide stability.

[Bone marrow hematopoiesis in those who worked in the cleanup of the aftermath of the accident at the Chernobyl Atomic Electric Power Station with a history of acute radiation sickness (based on myelogram data)].

On the basis of myelograms an evaluation was done of medullary hemopoiesis in 15 subjects who had been taken ill with acute radiation sickness (ARS) as a result of Chernobyl accident in 1986. 6-12 years following the exposure to ionizing radiation the bone marrow of ARS convalescents still shows a tendency toward hypocellularity (the number of myelokaryocytes in the examinus ranged from 40 to 80 x 10(9)/l), high index of neutrophil maturation (0.99) was recordable, which fact was related to depletion of granulocyte reserve and increase among the elements of granulopoiesis of share of proliferating cells, together with a rise in the number of lymphoid elements (19%).

[The status of the endocrine function of the thymus in acute radiation sickness resulting from the accident at the Chernobyl Nuclear Power Station].

Data are presented on the role of inhibition of endocrine function of the thymus in the pathogenesis of acute radiation sickness resulted from the direct and indirect (via the increased glucocorticoid production) effects of ionizing radiation. The complex treatment, including nonspecific active immunotherapy, permitted to normalize the thymic hormone level and certain parameters of the immune system.

[Radiation sickness; prospective study of incidence and course].

In a prospective inventory analysis of 169 irradiated patients concerning the incidence and course of Acute Radiation Sickness (fatigue, loss of appetite, nausea and vomiting), it was found that 2/3 of the patients had these complaints. The complaints started during the first week of treatment and mostly even within a few hours after each radiotherapy session. All complaints were progressive during the radiotherapy course, but disappeared spontaneously within one week after completion of the treatment. The possible occurrence of these symptoms soon after the start of radiotherapy should be kept in mind, especially since they can be reduced by simple regimens.

[Change in the fractional composition of thrombocytic proteins during experimental acute radiation sickness].

Thrombocytic proteins of healthy and irradiated rats were divided into 13 fractions by anode electrophoresis in the polyacrylamide gel. In rats with acute radiation sickness there was a reduction in the content of thrombocytic proteins with an electrophoretic mobility corresponding to the prealbumin, albumin, gamma-globulin and fibrinogen; an increase in the protein content with the mobility corresponding to the alpha- and beta-globulins was seen.
