Potent inhibitors of human immunodeficiency virus type 1 integrase: identification of a novel four-point pharmacophore and tetracyclines as novel inhibitors.

A four-point pharmacophore was constructed from energy-minimized structures of chicoric acid and dicaffeoylquinic acid. The search of 206,876 structures in the National Cancer Institute 3D database yielded 179 compounds that contain this pharmacophore. Thirty-nine of these compounds were tested in an in vitro assay specific for human immunodeficiency virus type 1 integrase (IN). Each retrieved structure was fit to the pharmacophore, and the conformation that afforded the best fit was identified. Twenty of the 39 compounds tested exhibited IC50 values of < 20 microM. Among the most potent inhibitors, tetracyclines emerged as a new class of inhibitors. Although the parent tetracycline exhibited marginal potency against purified IN, all substituted tetracyclines tested showed 5-100-fold increased potency. Disintegration assays with truncated IN mutants indicated that tetracyclines inhibit the IN catalytic core domain. To investigate whether chelation of divalent metals is implicated in differential potency of tetracyclines, enzyme assays were performed in the presence of both Mn2+ or Mg2+; no significance difference in potency was observed. Rolitetracycline inhibited IN/DNA complex formation in the presence of EDTA, which suggests that inhibition was metal independent. Rolitetracycline reversed DNA binding of IN after the complex was allowed to form before the addition of drug. Selectivity of tetracyclines was also examined in an assay specific for topoisomerase I, and none of the tetracyclines tested induced topoisomerase I-mediated cleavable complex or inhibited camptothecin-induced cleavable complex. Remarkable potency against the IN in the absence of divalent metals and the core enzyme coupled with water solubility makes tetracyclines potential candidates for X-ray crystal structure determination with IN.

[Peroxidative effects of tetracycline antibiotics].

After six-day administration of rolitetracycline (RTC) in the cardiac mitochondria of old rabbits the rate of oxygen consumption (glutamate-malate as well as 2-oxoglutarate) declines, while the malonic dialdehyde (MDA) rises. This provides evidence of more intense lipoperoxidation due to rolitetracycline. However, the ATP-ase activity increases and therefore this parameter is unsuitable in this experimental set-up (presence of tetracycline antibiotic) as an indicator of impaired integrity of mitochondrial membranes. After administration of RTC the activity of superoxide dismutase (SOD) increases markedly, but in case of concurrent administration of vitamin E it declines which is evidence of the prooxidative action of tetracyclines. Tetracycline itself and its metabolites which are formed during the biotransformation and degradation of epianhydrotetracycline (ETC) do not alter significantly the oxygen consumption nor the ATP-ase activity in cardiac mitochondria of old rabbits in vitro. Of these metabolites only anhydrotetracycline (ATC) and in particular epianhydrotetracycline (EATC) inhibit both parameters. From experiments in vitro ensues the relationship between the action and structure of the antibiotic and its metabolites the structure of cycle C and the nature of the substituents in positions C6 and C1 of this tetracycline cycle, as prerequisites are created for the formation of prooxidative structures.

A high performance liquid chromatographic system for the analysis of tetracycline drug standards, analogs, degradation products and other impurities.

This paper describes the high performance liquid chromatographic (HPLC) analysis of eight parent tetracycline standards: tetracycline, chlortetracycline, rolitetracycline, oxitetracycline, minocycline, doxycycline, democlocycline, methacycline; and three tetracycline epimers: epitetracycline, epianhydrotetracycline, and anhydrotetracycline. The HPLC system employs an octadecylsilane reverse phase column and an isopropanol-diethanolamine-phosphate-ammonium EDTA-water mobile phase. This system produced at least partial resolution of all eight parent compounds and many of their degradation products.

Accidental intra-arterial injection.

Accidental intra-arterial injection of intramuscular antibiotic preparations is described in 3 cases in infants. In 2 benzathine penicillin was injected, and in 1 rolitetracycline. The clinical features are dominated by arteriolar obstruction which produces gangrene of the most severely affected limb. In addition, neuroloigcal involvement occurs when vessels to nerves or spinal cord are involved. The arteriolateral aspect of the thigh is preferable to the buttock as a site for intramuscular injection, and a short (2.5 cm) needle should be used to minimise the risk of intra-arterial injection.

[Penetration of antibiotics into the intervertebral disk].

The role of antibiotics in the treatment and prevention of postoperative spondylodiscitis is still controversial. In a group of 15 patients operated on account of lumbar discopathy, the authors investigated the penetration of antibiotics into the intervertebral disc, in correlation to serum levels, when antibiotics were administered during operation. Despite high serum levels rolitetracycline was not detected in extirpated material of the disc, gentamycin reached 2-6% serum levels. The authors give an account of views on the role of antibiotics in the prophylaxis of postoperative spondylodiscitis and recommend their administration in particular in risk groups. They recommend antibiotics in case of early diagnosis of postoperative spondylodiscitis, in the later stages of the disease the role of antibiotics is controversial.

Sclerotherapy for hydroceles.

Sclerotherapy with 3% sodium tetradecyl sulfate and 3.5% rolitetracycline on an outpatient basis was applied to 55 hydroceles. The over-all cure rate was 96% with an average followup of 13 months. Of the patients 64% were cured after only 1 sclerosant instillation. A post-sclerotherapy operation was necessary in 4% of the patients. Pain of a significant degree occurred after sclerotherapy in only 29% of the patients. Sclerotherapy appears to be an effective, economical and safe form of outpatient therapy for hydroceles.

[Tetracycline in the evaluation of bone turnover].

A new and easy technique for the fluorimetric evaluation of rolitetracycline calcium complex was studied. The study was carried out in 3 different groups of rats; an increase in power of fluorescence in the rats on a low calcium diet (LC) and on a low calcium diet plus vitamin D (LC + D) versus the group of rats on a normal calcium diet (NC) was found. Moreover, the amount of spongy bone showed a significant decrease in bone mass in the first two groups (LC and LC + D) versus control. These data indicated that the bone tie of tetracycline was only related to the high turnover of bone and there was no relationship with bone mass. This new technique, in comparison with the others, (double marking and evaluation of space between the two fluorescent bands) enables to evaluate the kind and the evolution of bone rarefaction by biopsy.

Effect of tetracycline administration on the efficacy of diminazene aceturate therapy and prophylaxis in Trypanosoma brucei infections of mice.

The simultaneous administration, to Trypanosoma brucei infected mice, of rolitetracycline or oxytetracycline and diminazene aceturate appeared to have no effect on the initial trypanocidal action of the diminazene aceturate in that trypanosomes were cleared from the circulation. It also had no effect on the duration of the aparasitaemic period which follows diminazene aceturate treatment and the mice remained free of circulating trypanosomes for some time. However, if used prophylactically, relatively large amounts of tetracyclines (4 x 10 mg kg-1) administered with 40 mg kg-1 diminazene aceturate caused a reduction of the prophylactic period compared with those mice given only diminazene aceturate. This reduction in the prophylactic period is unlikely to have any practical significance in the combination diminazene aceturate/tetracycline treatment of domestic animals as diminazene aceturate is used therapeutically and not prophylactically in the control of trypanosomiasis.

[Tetracycline levels in the sputum in chronic obstructive bronchitis (author's transl)].

After 3 control days, the tetracycline concentration in the sputum of 34 patients was measured daily on 7 days during treatment with the antibiotic. 14 patients took tetracycline (Hostacyclin) in normal doses (2X500 mg or 2X1000 mg) by mouth. 20 patients received rolitetracycline (Reverin) 2X275 mg i.v. daily. With considerable individual fluctuations, sputum concentrations were attained in all series ranging from 1.5 to 2.0 microgram/ml after intravenous administration and from 1.1 to 1.6 microgram/ml after oral dosage. The higher dosage administered orally had no greater influence on the sputum concentration. In each case, sputum levels were attained which lie above the necessary minimum concentrations (0.5 microgram/ml).

Application of various chemotherapeutic agents in experimental bovine anaplasmosis.

A trial was conducted with several chemotherapeutic agents against clinical and carrier cases of experimental bovine anaplasmosis. Dithiosemicarbazone (in combination with oxytetracycline). Chloramphenicol and Rolitetracycline proved to be very efficacious in bringing about the clinical recovery and the elimination of the carrier status. Treatment with Imidocarb caused a clinical recovery but could not completely eliminate the organisms.

[The intraoperative visualization of the bile ducts by the use of fluorescent substances. A feasibility study].

Iatrogenic bile duct injury during cholecystectomy is the most serious complication of this surgical procedure. Initial reports suggest that this complication is particularly problematic during laparoscopic cholecystectomy. Proper identification of biliary anatomy in the subhepatic region is the only way to avoid this severe complication. The potential benefit from a simple, reliable method for intraoperative delineation of biliary anatomy is self-evident. In this experimental work the Authors-study the possibility and the feasibility of intraoperative biliary tree imaging with two fluorescent molecules (rolitetracyclin and fluorescin).

In vitro inhibition of natural killer cell activity by doxycycline.

The effect of four tetracyclines, tetracycline, rolitetracycline, oxytetracycline and doxycycline, on human natural killer (NK) cell-mediated cytotoxicity was examined in vitro. Doxycycline was the only tetracycline which inhibited the NK cell activity. At concentrations of 10 micrograms/ml the drug caused approximately 50% inhibition of NK cell mediated cytotoxicity against K562 target. The inhibition was not a result of a toxic effect of the drug on NK cells. These results support the previous findings that doxycycline shows immunosuppressive properties.

Application of 13C nuclear magnetic resonance spectroscopy to the analysis and structural investigation of tetracycline antibiotics and their common impurities.

The spectral assignations of all the main resonances in the 13C nuclear magnetic resonance spectra in DMSO-d6 or D2O are presented and reviewed for ten of the principal tetracyclines in therapeutic use: tetracycline, chlortetracycline, 6-demethylchlortetracycline, minocycline, oxytetracycline, methacycline, meclocycline, doxycycline, rolitetracycline and lymecycline. NMR techniques employed include: proton noise-decoupled spectra, off-resonance decoupled spectra, nuclear Overhauser enhancement and the INEPT technique (insensitive nuclei enhanced by polarization transfer). 13C NMR is also examined as a means of detecting and identifying the principal degradation and isomerization products of tetracycline antibiotics. The analytical potential of 13C NMR is discussed with respect to quantitative analysis of impurities, studies on sites of protonation and metal-binding studies.

Polychrome labeling of bone with seven different fluorochromes: enhancing fluorochrome discrimination by spectral image analysis.

Bone formation and remodeling in vivo can be assessed by polychrome labeling using calcium-binding fluorescent dyes. The number of fluorochromes, however, limits this technique due to the fact that with increasing number, fluorescent spectra inevitably overlap, which makes discrimination more difficult. In order to enhance discrimination, we performed spectral image analysis. Non-critical size defects of the femur of male Wistar rats served as a model for bone formation. Eight different fluorochromes (calcein blue, xylenol orange, calcein, alizarine complexone, doxycycline, rolitetracycline, hematoporphyrin, and BAPTA) were administered sequentially subcutaneously every third day starting at day 4 after surgery. Following, bone specimen were embedded in methylmethacrylate and analyzed by spectral image acquisition using a Sagnac type interferometer (ASI, Israel). Seven of the eight applied fluorochromes could be resolved using spectral image examination. With BAPTA, we present a new fluorochrome suitable for bone labeling. Due to the superior sensitivity of the spectral image acquisition, the thickness of the bone sections could be reduced so that 5 mum thick sections could be analyzed. Spectral decomposition and subsequent linear unmixing allows depiction of each individual fluorochrome without interference of any other, enabling a reliable and superior morphometric analysis of labeled regions.

Haemophilus influenzae type B resistant to tetracycline isolated from children with meningitis.

During a three-year-period, 1971-73 inclusive, haemophili isolated from 96 children with severe infections, of whom 73 had meningitis and 19 acute epiglottitis, were serotyped and tested for sensitivity to antibacterial drugs. All strains were identified as Haemophilus influezae type b, and were sensitive to ampicillin, chloramphenicol, and trimethoprim. However, 3 isolates--from a boy aged 11 months and a girl aged 1 year with meningitis, and a girl aged 2 years with epiglottitis--were highly resistant to tetracycline, with a median minimal inhibitory concentration of 50 mug tetracycline hydrochloride per ml (resistance ratio greater than or equal to 50). Resistance was also demonstrated to doxycycline, oxytetracycline, and rolitetracycline and, in one strain, to minocycline. No evidence was obtained that the resistant organisms were capable of inactivating tetracyclines.

Pharmacokinetics and pharmacodynamics of the tetracyclines including glycylcyclines.

The pharmacokinetics of tetracyclines and glycylcyclines are described in three groups. Group 1, the oldest group, represented by tetracycline, oxytetracycline, chlortetracycline, demeclocycline, lymecycline, methacycline and rolitetracycline is characterized by poor absorption after food. Group 2, represented by doxycycline and minocycline, is more reliably absorbed orally, while group 3, represented by the glycylcycline tigecycline, is injectable only, with an improved antibacterial spectrum compared with the tetracyclines. Though incompletely understood, the pharmacodynamic properties of the tetracyclines and glycylcyclines are summarized.

Fluorimetric assay of tetracycline mixtures.

A simple and precise fluorimetric method is described for the simultaneous assay in plasma of a mixture containing chlortetracycline, demethylchlortetracycline and tetracycline. Assay within the therapeutic ranges of 0-5 mg litre(-1) is achieved by formation of strongly fluorescent aluminum/tetracycline complexes, without prior extraction or separation of the individual antibiotics. This is performed by determinations at the peak excitation and fluorescence emission wavelengths of each tetracycline chelate. The method can be similarly applied to the assay of oxytetracycline, rolitetracycline and minocycline.

Susceptibility of Bacterium actinomycetem comitans to 45 antibiotics.

The minimal inhibitory concentrations of 45 antimicrobial agents were determined for 14 strains of Bacterium actinomycetem comitans (Actinobacillus actinomycetem-comitans). All the strains showed good susceptibility to tetracyclines and chloramphenicol, but not to lincomycins. Some strains were clearly resistant to penicillins, cephalosporins, and nitroimidazoles. The lowest minimum inhibitory concentrations were observed with tetracycline, rolitetracycline, methacycline, minocycline, chloramphenicol, and cotrimoxazole.
