[Protease as an immune response enhancer in immunization with an adsorbed staphylococcal anatoxin].

The injection of trypsin into rabbits immunized with adsorbed staphylococcal toxoid was found to have a potentiating effect on the immune response of the body, which was manifested by a considerable increase in the specific antibody titer and by the activation of nonspecific systemic protective factors. The intensity of antistaphylococcal immunity achieved by the moment of challenge (on day 23 of the experiment) in the animals ensured the resistance of the rabbits to staphylococcal test infection. The injection of trielin, a proteolysis inhibiting agent, simultaneously with the immunization of rabbits with adsorbed staphylococcal toxoid produced no such effect.

[Effect of terrilytin on the activity of nonspecific resistance factors during immunization with staphylococcal anatoxin and staphylococcal infection].

Terrilytin and immobilized terrilytin enhance the activity and intensity of phagocytosis and increase the concentration of lysozyme in nonimmunized animals. Both preparations increase the production of antibodies to staphylococcal alpha-hemolysin, the titers of beta-lysins, the activity and intensity of the phagocytosis of bacterial cells by peripheral blood leukocytes in animals immunized with staphylococcal toxoid and challenged with live staphylococcal culture. In healthy animals terrilytin and immobilized terrilytin induce an increase in total proteolytic activity and in the activity of alpha-1-antitrypsin and alpha-2-macroglobulin, decreased as the result of staphylococcal infection.

[Preventive properties of the blood sera from persons vaccinated with a Proteus vaccine made from soluble antigen complexes].

In experiments of the passive protection of mice the protective properties of sera obtained from humans before and after their immunization with Proteus vaccine used as a monopreparation or in combination with staphylococcal toxoid and/or pyoimmunogen were studied. When introduced in a single subcutaneous injection, Proteus vaccine prepared from soluble antigenic complexes ensured an increase in the protective properties of sera. The second injection of the vaccine essentially enhanced the protective potency of the sera of the immunized donors. The therapeutic injection of Proteus vaccine ensured the essential increase of the protective properties of the sera. This increase could be experimentally detected within at least 25-30 days from the beginning of immunization. The immunization of volunteers with Proteus vaccine in combination with pyoimmunogen and adsorbed staphylococcal toxoid ensured the maximum increase of the protective properties of their sera.

Mitogenicity of formalinized toxoids of staphylococcal enterotoxin B.

Staphylococcal enterotoxin B is a potent mitogen for mouse and human lymphocytes. Mitogenic activity was retained after detoxification of the enterotoxin by formaldehyde at pH 5.0, 7.5, OR 9.5. The most active toxoid (pH 7.5) was separated into a monomeric, a dimeric, and a polymeric fraction (1 x 10(5) to 3 x 10(5) molecular weight) by gel filtration, and although each fraction demonstrated mitogenic activity, the polymeric fraction was clearly the most efficacious. These data show that mitogenicity of staphylococcal enterotoxin B does not depend on toxicity. This suggests that the mitogenic and toxic activities are effected by different sites on the molecule.

[Experimental study of the protective activity of a complex staphylococcal antigen and toxoid when administered in combination].

The data obtained in the experimental study of the protective activity of an antigenic staphylococcal complex prepared by the method of aqueous extraction and staphylococcal toxoid (native and adsorbed) are presented. The study carried out on the model of septic staphylococcal infection in rabbits indicated that after immunization with the mixture of the above-mentioned preparations the survival time of the rabbits increased to a greater extent than after immunization with each of these preparations separately. This regularity was especially pronounced under the aggravated conditions of infection with the mixture of strains.

[The creation of specific immunity to staphylococcal infection in newborn infants by the intranasal administration of adsorbed staphylococcal anatoxin].

The possibility of enhancing specific immunity in newborn infants by the intranasal administration of adsorbed staphylococcal toxoid to infants with a high risk of staphylococcal infection in doses of 1 drop (0.05 ml) into each nostril during the first 7-9 years of their life. On days 7-9 the level of anti-alpha-toxin in the blood rose to 3.8 +/- 0.14 I. U./ml and remained sufficiently high 3-6 months later. When this method was used for the simultaneous immunization of mothers, their antitoxic titers were not as high as in newborn infants. No side effects were observed. In the control group, the titers of anti-alpha-toxin were low during the whole period of observation. Infants immunized by the proposed method had no staphylococcal infections both during the newborn period and within the first year of their life. In the control group, 8 cases of minor forms of purulent septic infection were registered during the newborn period, and in 2 infants umbilical staphylococcal sepsis was diagnosed.

[Antigen-specific and antigen-nonspecific reactions of the immunity system in vaccination with a purified concentrated staphylococcal anatoxin].

Immunization with purified and concentrated staphylococcal toxoid leads to the rapid 12-fold rise of the level of anti-alpha-toxin and to the 17-fold rise of the titres of antibodies to extracellular staphylococcal products. At the period of immunization phasic changes in cell-mediated and humoral immunity characteristics, indicative of the state of the nonspecific resistance system, can be observed. These changes consist in the transient suppression of phagocytosis, including a decrease in the activity of intraleukocytic bactericidal systems (myeloperoxidase and cation protein), a decrease in the activity of the complement and the bactericidal activity of the blood serum, which should be taken into account when using this immune preparation for therapy.

[Immunoprevention of staphylococcal infections at an industrial enterprise].

The immunogenicity, immunological and epidemiological effectiveness of the subcutaneous, enteral and combined immunizations of the workers at a steel plant with staphylococcal toxoid has been studied. The subcutaneous injection of the adsorbed toxoid in a dose of 0.2 ml by means of the jet injector, model, was accompanied by moderate reactogenicity (the frequency of local reactions was 26.8%, and the frequency of systemic reaction 18%) and produced 4-fold increase in antitoxin titers and 12-fold decrease in the morbidity rate of staphyloccal infection. Enteral immunization in 3 administrations proved to be nonreactogenic and only slightly effective (antitoxin titers increased more than 2-fold, morbidity rate fell 2-fold). Combined immunization (subcutaneous and enteral) with the toxoid, preceded by the injection of gamma globulin, reduced the frequency of reaction 2- to 3-fold and stimulated antitoxin production (the titer increased 8-fold), thus decreasing the morbidity rate of staphylococcal infection 3.1-fold.

[The modulation of cellular immunity in vivo and in vitro under the action of purified staphylococcal anatoxin].

Purified staphylococcal toxoid modulates (mainly suppresses) cell-mediated immune response to heterogeneous antigens of animal origin (to sheep red blood cells as shown by the delayed hypersensitivity reaction) or bacterial origin (to BCG as shown by the splenocyte migration test). The direction and manifestation of modulation depend on the strain of mice, dose of the toxoid, dose and nature of the test antigen and the immunization schedule (intervals between the injections of the antigen).

[Changes in the anti-infection immunity indices of esophageal cancer patients undergoing combined immunization against staphylococcal and Pseudomonas aeruginosa infections].

The method of the complex immunization of patients with esophageal cancer against the main causative agents of purulent infections has been developed. Purified concentrated staphylococcal toxoid and polyvalent corpuscular P. aeruginosa vaccine were used for the immunization of 26 patients. Immunization was carried out in 3 injections (11 patients) and 4 injections (15 patients) made at intervals of 7 days. This resulted in the activation of nonspecific immunity factors (the total bactericidal properties of the blood serum, the phagocytic activity of neutrophils, the increase of the IgG level) and in a considerable increase in the factors of specific protection against staphylococcal and P. aeruginosa infections: the titer of staphylococcal anti-alpha-toxin increased 7- to 11-fold and the titer of antibodies to P. aeruginosa antibodies increased 5- to 6.5-fold. The preoperative vaccination of patients decreased the occurrence of postoperative purulent complications from 71% to 11.5%.

[Antigenic similarity of microorganisms to human group-specific blood factors and the immune response of persons with various ABO classifications].

The isohemagglutination test with the introduction of the heat-stable fraction of microbial broth culture into the reacting system was used as a model of the antigen-antibody interaction. In the "A-anti-A" system the inhibition of isohemagglutination in the presence of staphylococcal allergen was observed. This inhibition was caused by the binding of the allergen with anti-A-antibodies, which was indicative of the antigenic similarity between staphylococcus and group-specific factor A. The influence of the similarity thus established on immune response was studied by the determination of antitoxic titers in 292 donors immunized with staphylococcal toxoid and in 86 patients with chronic hematogenic fistulous osteomyelitis with due regard of the ABO phenotype of the examinees. Subjects with group A(II) blood were found to have lesser antibody titers than those with blood of other groups and to be predisposed to the progressive course of staphylococcus-induced diseases.

[Changes in the humoral response to sheep erythrocytes based on data from a determination of the level and classes of antibodies in mice immunized with purified staphylococcal anatoxin].

The intensity and direction of the modulation of humoral immune response, induced by the action of purified staphylococcal toxoid (PST), depend on the dose of the preparation, the dose of the test antigen, the scheme of the experiment, the time of the registration of its results and the genotype of animals. PST induces the modulation of the synthesis of both IgM and IgG antibodies. The injection of small divided doses of PST enhances immune response to heterologous antigen, while a single inoculation of the total dose produces no such effect.

[The patterns of the corrective action of a natural immunomodulator on the secondary immunological defect caused by a corpuscular pertussis vaccine].

Purified staphylococcal toxoid (PST) was shown to be capable of preventing the development of secondary antigen-nonspecific immune deficiency in mice, immunized with whole-cell pertussis vaccine. The immunocorrective action of PST was manifested after its injection before (on day -1), simultaneously with and after (on day +1) the injection of whole-cell pertussis vaccine. Correction was either complete or partial, depending on the scheme of the experiment and the dose of PST.
