Detection of quantitative trait loci for seminal root traits in maize (Zea mays L.) seedlings grown under differential phosphorus levels.

Suboptimal phosphorus availability is a primary constraint for terrestrial plant growth. Seminal roots play an important role in acquisition of nutrients by plant seedlings. The length and number of seminal roots may be particularly important in acquisition of immobile nutrients such as phosphorus by increasing soil exploration. The objective of this study was to identify quantitative trait loci (QTL) controlling seminal root growth in response to phosphorus stress in maize, and to characterize epistatic interactions among QTL. Seminal root length and number were evaluated in 162 recombinant inbred lines derived from a cross between B73 and Mo17 in seedlings grown in a controlled environment. B73 and Mo17 significantly differed for seminal root length under low phosphorus, but not under adequate phosphorus conditions. Seminal root length of the population grown under low phosphorus ranged from 0 to 79.2 cm with a mean of 32.3 cm; while seminal root length of plants grown under high phosphorus ranged from 0.67 to 59.0 cm with a mean of 23.4 cm. Under low phosphorus, one main-effect QTL was associated with seminal root length and three QTL with seminal root number; under high phosphorus, two QTL with seminal root length and three QTL for seminal root number. These accounted for 11, 25.4, 22.8, and 24.1% of the phenotypic variations for seminal root length and number at low phosphorus, and seminal root length and number at high phosphorus, respectively. Di-genic epistatic loci were detected for seminal root length at low phosphorus (two pairs) seminal root number at low phosphorus (eight pairs), seminal root length at high phosphorus (four pairs), and seminal root number at high phosphorus (two pairs), which accounted for 23.2, 50.6, 32.2, and 20.3% of the total variations, respectively. Seminal root traits observed here were positively yet weakly correlated with shoot biomass in the field under low phosphorus, although no coincident QTL were detected. These results suggest that epistatic interactions are important in controlling genotypic variation associated with seedling seminal root traits.

Cochlear delivery of fibroblast growth factor 1 and its effects on apoptosis and cell cycling in noise-exposed guinea pig ears.

OBJECTIVES: Acidic fibroblast growth factor 1 (FGF-1) is a mitogen and antiapoptotic factor synthesized by cochlear neurons and transported to the organ of Corti. The objectives of this investigation were threefold: (1) to develop an animal model to study the cochlear effects of intratympanic delivery of FGF-1; (2) to determine the distribution, in the mature mammalian cochlea, of FGF-1 and the receptor, FGFR3, to which it binds with high affinity; and (3) to examine the effect of exogenous FGF-1 on cochlear apoptotic and cell-cycling markers in noise and non-noise-exposed guinea pigs ears. METHODS: Fifteen adult Hartley guinea pigs were divided into three groups. Group 1 animals (n = 5) underwent direct placement of FGF-1 in phosphate buffered saline (PBS) (20 pg/mL) soaked Gelfoam pledgets to the right round window membrane. Phosphate buffered saline-soaked Gelfoam pledgets were placed on the left round window membrane as a control. In group 2 animals (n = 5), surgical placement of either FGF-1 or PBS was followed by exposure to 120 dB of white noise for 2 hours. Group 3 animals (n = 5) were subjected to identical noise conditions prior to undergoing round window application of either FGF-1 or PBS. All groups were allowed to recover in a noise-controlled environment for 12 hours following surgery. Anti-FGF-1-stained Western blots and optical densitometry analyses were used to quantitate passage of FGF-1 into cochlear perilymph. Standard in situ immunohistochemical techniques were used to stain each cochlea for FGF-1 and FGFR3, apoptotic markers p53 and p21, Bcl-2, and the cell-cycling marker proliferating cell nuclear antigen (PCNA). Tissue sections were subjected to the terminal deoxynucleotidyl transferase-mediated deoxyuridine triphosphate nick end-labelling technique (TUNEL) for apoptosis. RESULTS: Western blot and optical densitometry analyses of cochlear perilymph showed increased concentrations of FGF-1 in 10 of 14 experimental cochleas. Cochlear perilymph FGF-1 was consistently bound to heparan sulphate proteoglycan (HSPG). Immunoreactivity of both FGF-1 and FGFR3 was observed in spiral ganglion neurons, inner and outer hair cells, pillar cells, and Dieter and Hensen's cells. Specific FGF-1 immunostaining to the distal portion of the pars pectinata of the basilar membrane was noted in noise-exposed animals only. Bcl-2 and PCNA immunostaining was not detected in any group. There was no significant nuclear immunoreactivity to proapoptotic markers, p53 and p21, in any group. Semiquantitative analysis of TUNEL staining in block sections of all cochleas demonstrated a 340% increase in nuclear immunoreactivity of noise-exposed outer hair cells and organ of Corti cells. There was no difference between FGF-1 treated and control ears subjected to TUNEL staining. CONCLUSIONS: Exogenous FGF-1 crosses the round window membrane and is bound to HSPG in cochlear perilymph. The specific immunoreactivity of the pars pectinata to FGF-1 may represent a unique reservoir for cochlear FGF-1 in noise-exposed ears of the guinea pig. Noise induces apoptosis of organ of Corti cells as demonstrated with the TUNEL technique. PCNA, Bcl-2, p53, and p21 in noise-exposed and non-noise-exposed guinea pig cochleas are not affected by exogenous FGF-1. Noise-induced hair cell apoptosis appears to be independent of the p53 pathway. Lack of immunoreactivity to Bcl-2 supports the concept that the apoptotic mechanism is likely to involve C-Jun-N-terminal kinase- or caspase-dependent pathways. Exogenous FGF-1 does not alter apoptosis or cell cycling in the mature guinea pig cochlea within 12 hours of acute acoustic trauma.

[Facilities, equipment, and biohazards].

The high quality of laboratory animals, the precise and proper humane treatment of animals and the well-controlled environments are the three essential factors for animal experimentation. The defect or disturbance of any of these factors might be a common causation of so-called biohazards, such as a sporadic occurrence of epidemic hemorrhagic fever in Japan. The caging or housing systems, which is the most essential part for proper functioning of animal facilities, were described. In the uninfected animal areas, the air-conditioning system was designed to isolate each room and separate it from unrestricted space in the facility. In the areas for the experimentally infected animals, it was designed to protect the infection among the experimental animals or investigators. The systems of safety handling equipments, containments of infectious agents, and disposal of contaminant materials were discussed. Finally, it was emphasized that the researchers have to be for the protection against the biohazards throughout animal experimentation.

Activity of the triazole antifungal r126638 as assessed by corneofungimetry.

BACKGROUND: R126638 is a novel triazole exhibiting potent in vitro and in vivo antifungal activity against fungal pathogens including dermatophytes and yeasts. OBJECTIVE: To determine the antifungal activity in time in the stratum corneum of healthy volunteers after oral intake of R126638 at a daily dose of 100 or 200 mg for 1 week. METHOD: Sixteen male volunteers were randomly allocated to oral treatment with either 100 or 200 mg of R126638 once daily for 1 week. Five cyanoacrylate skin surface strippings (CSSS) were obtained from the forearm of each subject before drug intake at day 1. CSSS were also collected during treatment at day 2 (24 h after the first drug intake, before the second drug intake), at day 4 (before the fourth drug intake) and at day 7 (10 h after the last drug intake). The post-treatment lingering effect was assessed at day 10 (3 days after treatment) and at day 14 (7 days after treatment). The corneofungimetry bioassay was performed on these CSSS to assess the antifungal profile of R126638. Cells of different fungal species (Trichophyton rubrum, Trichophyton mentagrophytes, Microsporum canis, Candida albicans and Malassezia globosa) were deposited and cultured for 10 days on CSSS in a sterile and controlled environment. The extent of fungal growth on the stratum corneum was determined using computerized image analysis. RESULTS: R126638 clearly reduced the growth of all tested fungal species. The onset of effects of R126638 was evidenced at day 4 when it reached statistical significance for 3 of 5 species. At day 7, significance was reached for 4 of 5 species. During the posttreatment period, R126638 remained effective for 4 of 5 species at day 10, and this activity persisted until day 14 for 2 of 5 species. CONCLUSION: A broad spectrum antifungal activity was rapidly expressed in the stratum corneum after oral intake of R126638. The drug likely reached the upper layers of the stratum corneum by diffusion and persisted in this location for at least 7 days after treatment.

The effects of rehydration on cycling performance after exercise-induced dehydration.

The effects of 7.6% carbohydrate-electrolyte solution (CES) and placebos (P) on rehydration (R) after exercise-induced dehydration and on a subsequent time-trial (TT) of cycling performance were studied. Thirteen male subjects exercised in a thermally-controlled environment (28 degrees C, 63% RH) until 3% of their body weight was lost. After exercise, the subjects moved to a neutral environment (22 degrees C) and rested for 30 minutes prior to a 2-hour R period. During R, subjects were fed CES or P to a maximum volume of 120% of previous body mass loss at 0, 30, and 60 minutes, in bolus-doses of 50%, 40% and 30% respectively. After R, subjects performed a 1-hour TT with no further fluid intake. % R with CES was significantly higher than with P (70 +/- 3% vs 60 +/- 5%; p < 0.01). During the TT, blood glucose dropped in the CES group but not in the P group. It was found that, despite a more effective R with CES, the performance results did not differ between groups (65.1 +/- 2.2 minutes and 65.2 +/- 2.3 minutes for CES and P respectively). It is suggested that an insulin-mediated rebound effect on CHO metabolism during TT, in which no further CHO was supplied, nullified the benefits of rehydration.

A laboratory evaluation of the palatability of legumes to the field slug, Deroceras reticulatum Müller.

Slugs are major pests of many crops, including winter wheat, in temperate climates, yet current methods of control are often unreliable. The aim of this study is to investigate the potential for common legume species to act as an alternative source of food, or trap crop, for the most damaging agricultural pest species, the grey field slug, Deroceras reticulatum Müller, thereby reducing damage to the wheat crop. A series of three controlled-environment experiments were designed to assess this aim. Individual slugs were fed leaves of one of ten legume species together with winter wheat leaves for a 72-h period. A clear hierarchy of acceptability was shown, with red clover, lucerne, lupin and white clover showing significantly higher Acceptability Indices than the other six species tested. Red clover produced the greatest reduction in mean wheat consumption (78%) from day 1 to day 3. When species were fed individually, red clover was consumed in significantly greater quantities than any of the other treatments: 40% more than white clover and 56% more than wheat. Furthermore, when fed with red clover the amount of wheat consumed was some 50% less than when the latter was fed alone. The results indicate that legumes vary greatly in their acceptability to D. reticulatum and it is essential that a legume with a high Acceptability Index is chosen, which results in the least amount of wheat consumed.

Experimental evidence against diffusion control of Hg evasion from soils.

Elemental Hg (Hg(0)) evolution from soils can be an important process and needs to be measured in more ecosystems. The diffusion model for soil gaseous efflux has been applied to modeling the fluxes of several gases in soils and deserves testing with regard to Hg(0). As an initial test of this model, we examined soil gaseous Hg(0) and CO(2) concentrations at two depths (20 and 40 cm) over the course of a controlled environment study conducted in the EcoCELLs at the Desert Research Institute in Reno, Nevada. We also compared small, spatially distributed gas wells against the more commonly used large gas wells. In this study, two EcoCELLs were first watered (June 2000) and then planted (July 2000) with trembling aspen (Populus tremuloides). Following that, trees were harvested (October 2000) and one EcoCELL (EcoCELL 2) was replanted with aspen (25 April 2001). During most of the experiment, there was a strong vertical gradient of CO(2) (increasing with depth, as is typical of a diffusion-driven process), but no vertical gradient of soil gaseous Hg(0). Strong diel variations in soil gas Hg(0) concentration were noted, whereas diel variations in CO(2) were small and not statistically significant. Initial watering and planting caused increases in both soil gas CO(2) and Hg(0). Replanting in EcoCELL 2 caused a statistically significant increase in soil gas CO(2) but not Hg(0). Calculated Hg(0) effluxes using the diffusion model produced values two orders of magnitude lower than those measured using field chambers placed directly on the soil or whole-cell fluxes. Neither soil gas Hg(0) concentrations nor calculated fluxes were correlated with measured Hg(0) efflux from soil or from whole EcoCELLs. We conclude that (1) soil gas Hg(0) flux is not diffusion-driven and thus soil gas Hg(0) concentrations cannot be used to calculated soil Hg(0) efflux; (2) soil gas Hg(0) concentrations are increased by watering dry soil, probably because of displacement/desorption processes; (3) soil gas Hg(0) concentrations were unaffected by plants, suggesting that roots and rhizosphere processes are unimportant in controlling Hg(0) evasion from the soil surface. We recommend the use of the small wells in all future studies because they are much easier to install and provide more resolution of spatial and temporal patterns in soil gaseous Hg(0).

Modelling of predator-prey trophic interactions. Part I: two trophic levels.

A class of lumped parameter models to describe the local dynamics in a controlled environment of a two-trophic chain is considered. The class is characterized by a trophic function (functional response of predator to the abundance of prey) depending on the ratio of prey biomass x and a linear function of predator biomass y: f(qx/[(1-rho)k + rhoy]), where q is the efficiency of the predation process, k is a reference biomass, and rho (0 < or = rho < or = 1) specifies the predation model. The trophic function is defined only by some properties determining its shape. A stability analysis of the models has been performed by taking the parameters q and rho as bifurcation parameters: the regions in the (rho,q) plane of existence and stability of nonnegative equilibrium states and limit cycles are determined. This analysis shows that the behaviour of the models is qualitatively similar for 0 < or = rho < 1 (in particular the null state is always a saddle point), while the value rho=1 gives rise to some kind of structural instability of the system (in particular the null state becomes an attractor for sufficiently high predation efficiency).

Comparison of girth materials, girth tensions and their effects on performance in racehorses.

OBJECTIVE: To compare the effect of girth materials and commonly used girth tensions on athletic performance of race-horses and to test the length tension properties of commercially available girths. PROCEDURE: Seven horses were exercised at speeds to produce 95% of maximal heart rates on 15 occasions using a randomised block design, and girthed with 5 different girths at 3 nominal tensions of 6, 12 or 18 kg. The girths used were a standard elastic race girth, an 'American' elastic race girth, an elastic race girth twice the normal width, a standard canvas race girth and a canvas race girth at twice the normal width. Tension in the girth was recorded continuously using an in-line load cell connected to a physiograph. Horses ran to fatigue on a treadmill inclined at 10% slope. Tensions were measured at peak inhalation (T/inh) and exhalation (T/exh), recorded at rest (rest) and during exercise (ex). An analysis of variance was used to compare the mean run to fatigue times (RTFT) between girth types and tensions, multiple pair-wise comparisons were then carried out using Tukey's test where significant differences were found. The length-tension relationships of five commercially available girths for training and racing of Thoroughbred racehorses were studied by the application of standardized weights in series to multiple samples of each type of girth. Measurements were taken in a controlled environment and analysis of variance was used to compare the means for length-tension of each girth type. RESULTS: The elastic and the 'American' elastic girths produced significantly longer RTFT when compared to the standard canvas girth (P = 0.01 and P = 0.001 respectively). Also girths tensioned at Texhrest 6 kg and Texhrest 12 kg produced significantly longer RTFT than when girthed at Texhrest 18 kg (P = 0.03 and P = 0.08 respectively). There were significant differences between the commercially available girth types at each tension (P < 0.05), but differences were not significant between girths of the same type. Girths with an elastic component reached their peak for maximum extension at 14.5 kg and thereafter their extension declined. CONCLUSION: The type of girth and the tension at which it is applied affects athletic performance. Lower girth tensions and the use of elastic materials in the girth would appear to optimise performance. However according to this study and our previous study, none of the commercially available girths studied would adequately protect against the potentially detrimental effects of overtightening on athletic performance.

Controlled exposures of volunteers to respirable carbon and sulfuric acid aerosols.

Respirable carbon or fly ash particles are suspected to increase the respiratory toxicity of coexisting acidic air pollutants, by concentrating acid on their surfaces and so delivering it efficiently to the lower respiratory tract. To investigate this issue, we exposed 15 healthy and 15 asthmatic volunteers in a controlled-environment chamber (21 degrees C, 50 percent relative humidity) to four test atmospheres: (i) clean air; (ii) 0.5-microns H2SO4 aerosol at approximately 100 micrograms/m3, generated from water solution; (iii) 0.5-microns carbon aerosol at approximately 250 micrograms/m3, generated from highly pure carbon black with specific surface area comparable to ambient pollution particles; and (iv) carbon as in (iii) plus approximately 100 micrograms/m3 of ultrafine H2SO4 aerosol generated from fuming sulfuric acid. Electron microscopy showed that nearly all acid in (iv) became attached to carbon particle surfaces, and that most particles remained in the sub-micron size range. Exposures were performed double-blind, 1 week apart. They lasted 1 hr each, with alternate 10-min periods of heavy exercise (ventilation approximately 50 L/min) and rest. Subjects gargled citrus juice before exposure to suppress airway ammonia. Lung function and symptoms were measured pre-exposure, after initial exercise, and at end-exposure. Bronchial reactivity to methacholine was measured after exposure. Statistical analyses tested for effects of H2SO4 or carbon, separate or interactive, on health measures. Group data showed no more than small equivocal effects of any exposure on any health measure.(ABSTRACT TRUNCATED AT 250 WORDS)

The influence of age upon opioid analgesic use in the patient-controlled analgesia (PCA) environment.

It is often asserted that older patients are more sensitive to opioid analgesics than younger patients but experimental evidence for this assertion remains sparse. Two studies were conducted investigating the relationship between age and opioid analgesic use in the patient-controlled analgesia environment. In study I, the relationship was analysed subsequent to our publication of a study investigating patients' responses to opioid use with patient-controlled analgesia. Fifty-five postoperative patients, stratified into 'older' and 'younger' patients by median age, received morphine or pethidine or fentanyl patient-controlled analgesia. A strong inverse relationship was found between age and fentanyl and morphine use but not between age and pethidine use. Study II was a retrospective study of the medical records of 199 patient-controlled analgesia patients who had received morphine or pethidine patient-controlled analgesia; there were insufficient patients who had used fentanyl for a reasonable sample. There was a difference in morphine use with the younger patients using significantly more morphine than the older patients (< 60 years). Findings were less clear for patients receiving pethidine but there was an inverse correlation between age and pethidine use as well. Overall, the findings of these two studies supported the common clinical belief that older patients require less opioids than younger patients.

Seasonal variation in respiration of 1-year-old shoots of scots pine exposed to elevated carbon dioxide and temperature for 4 years.

Sixteen 20-year-old Scots pine (Pinus sylvestris L.) trees growing in the field were enclosed for 4 years in environment-controlled chambers that maintained: (1) ambient conditions (CON); (2) elevated atmospheric CO2 concentration (ambient + 350 micro mol mol-1; EC); (3) elevated temperature (ambient +2-6 degrees C; ET); or (4) elevated CO2 and elevated temperature (ECT). The dark respiration rates of 1-year-old shoots, from which needles had been partly removed, were measured over the growing season in the fourth year. In all treatments, the temperature coefficient of respiration, Q10, changed with season, being smaller during the growing season than at other times. Respiration rate varied diurnally and seasonally with temperature, being highest around mid-summer and declining gradually thereafter. When measurements were made at the temperature of the chamber, respiration rates were reduced by the EC treatment relative to CON, but were increased by ET and ECT treatments. However, respiration rates at a reference temperature of 15 degrees C were reduced by ET and ECT treatments, reflecting a decreased capacity for respiration at warmer temperatures (negative acclimation). The interaction between season and treatment was not significant. Growth respiration did not differ between treatments, but maintenance respiration did, and the differences in mean daily respiration rate between the treatments were attributable to the maintenance component. We conclude that maintenance respiration should be considered when modelling respiratory responses to elevated CO2 and elevated temperature, and that increased atmospheric temperature is more important than increasing CO2 when assessing the carbon budget of pine forests under conditions of climate change.

Circadian Rhythm of Leaves of Phaseolus angularis Plants Grown in a Controlled Carbon Dioxide and Humidity Environment.

Leaf movements of primary leaves of Phaseolus angularis Wight. were studied in an environment with controlled levels of CO(2), relative humidity, temperature, light, nutrient concentrations, and water tension. Rhythmic circadian movements and irregular short period movements were evident as the leaves unfolded and persisted during development of the leaves. The mean period in rhythmic circadian movement was 27.3 hours with no significant differences in period between plants of the same or different experiments. The leaf movements of separate plants were not closely synchronized.A system is described for growing plants for extended periods while collecting data with time lapse photography. The system was developed to minimize disturbances to the plants. The occurrence of leaf movements in plants grown from the time of seeding in an environment free of any recognized fluctuation which could stimulate leaf movements supports the hypothesis that leaf movement rhythms originate spontaneously within the plant.

Effect of fibrin on morphologic characteristics of retinal pigment epithelial cells.

Human retinal pigment epithelial (RPE) cells in culture, when overlaid by a fibrin clot, lose their normal epithelial morphologic features and migrate into the overlying clot as fibrocytelike cells. The behavior of human RPE cells on exposure to fibrin correlates well with the observed response of RPE in several ocular disorders in which fibrin deposition within the eye is an important feature. The mechanism of recognition and interaction between fibrin and RPE cells is unknown. The in vitro system used in our current studies allows the investigation of this interaction in a controlled environment. Further study of the interaction between human RPE cells and fibrin offers the possibility of improving our understanding and treatment of several blinding ocular disorders, including the disciform phase of senile macular degeneration, proliferative vitreoretinopathy, and the sequelae of ocular trauma.

Changes in mood, craving, and sleep during short-term abstinence reported by male cocaine addicts. A controlled, residential study.

We examined changes over 28 days in mood states, craving for cocaine, and sleep during short-term abstinence reported by 12 male, predominantly intravenous-using, cocaine-addicted subjects residing in a research facility. For comparison, we examined 10 nonaddicted control subjects. There were no significant differences between cocaine addicts and controls regarding demographics and selected DSM-III-R diagnoses other than psychoactive substance use disorder and antisocial personality disorder. There were significantly higher scores of psychiatric symptoms reported by cocaine addicts 1 week before admission. Mood-distress and depression scores recorded at admission and during short-term abstinence were significantly greater than those reported by controls. Addicts' mood-distress scores and craving for cocaine were greatest at admission and decreased gradually and steadily during the 28-day study. There were no significant differences between groups regarding reports of sleep other than difficulty falling asleep and clearheadedness on arising. Although there were significant differences in resting heart rate at admission and over time, there were no significant differences in weight gain or blood pressure. Given the absence of a classic "withdrawal" pattern, "short-term abstinence" may be a more appropriate classification of psychological and physical phenomena experienced by cocaine addicts who initiate abstinence in a controlled environment.

Rat prostate explants in serum-free organ culture: a comparison of two media and gas mixtures.

BACKGROUND: Urologists are looking for a way to easily discriminate between aggressive and very slow-growing prostate tumors. A sound way to appreciate such developing activities would be to identify an appropriate cell marker in prostate explants maintained in a defined culture system. METHODS: Different biological parameters were compared in rat prostate explants cultured for 5 days in rich CMRL or basic Leibovitz's L-15 medium, unsupplemented with serum, under a mixture of either 95% air/5% CO2 or 50% N2/45% O2/5% CO2. RESULTS: DNA synthesis was somewhat similar with the two-gas combination, but was higher in explants maintained in L-15 medium than in CMRL. Hence, L-15 medium and the 95% air/5% CO2 mixture were selected. Under these defined conditions for 5 days, cells were still able to synthesize DNA and proteins while preserving their morphological integrity and maintaining alkaline and acid phosphatase activities. CONCLUSIONS: Since the present culture system works well in a controlled environment and under such minimal conditions, it appears to be a reliable and promising model that will provide basic data and allow the study of hormones and growth factors involved in prostatic tissue growth. It might eventually permit the identification of a cell marker.

Methodologic approaches to quality care evaluation.

A number of problems inherent to quality of care evaluation have been described above. Although issues such as limited resources and clinical staff resistance may not be specific to cancer care, they certainly impact on the evaluation of the quality of care offered to cancer patients. Other issues such as complexity of care and the time span over which care can occur are more applicable to the cancer patient population alone. In response to these issues, I have offered a number of solutions, such as reorganizations of tasks, the use of statistical techniques such as sampling and power analysis, and various feedback mechanisms which have been implemented at MSKCC. The data presented indicate that these mechanisms have been at least partially successful; however, given the absence of a controlled environment, drawing firm conclusions about the relationship between cause and effect is not possible. I hope that in the future these results may be replicated at other institutions who elect to implement any of the suggestions I have made.
