Detection of single-nucleotide polymorphisms with the WAVE DNA fragment analysis system.

As the Human Genome Project is generating an avalanche of genetic information, molecular researchers and clinical practitioners are setting new criteria for evaluation of the links between newly discovered gene mutations and human disorders. These requirements necessitate the development of highly accurate and yet rapid automated systems for genetic testing. We describe the detection of a single nucleotide polymorphism (SNP) on the human Y chromosome with the fully automated, sensitive, and rapid system WAVE designed for DNA fragment analysis. This new technology, based on temperature-modulated liquid chromatography and a high-resolution matrix, offers new dimensions to the molecular biology research. The versatility of the WAVE makes the equipment a universal molecular biology stations for clinical and research facilities. The key aspects to setting the operating parameters are discussed.

EASY--an Expert Analysis SYstem for interpreting database search outputs.

With the ever-increasing need to handle large volumes of sequence data efficiently and reliably, we have developed the EASY system for performing combined protein sequence and pattern database searches. EASY runs searches simultaneously and distils results into a concise 1-line diagnosis. By bringing together results of several different analyses, EASY provides a rapid means of evaluating biological significance, minimising the risk of inferring false relationships, for example from relying exclusively on top BLAST hits. The program has been tested using a variety of protein families and was instrumental in resolving family assignments in a major update of the PRINTS database.

The development and evaluation of a diluter-ultraviolet spectrophotometric analysis system for the determination of drugs in solutions and suspensions.

An automated system has been developed and validated for the analysis of liquid formulations (solution and suspension). The system comprises a multi-place magnetic stirrer, a diluter and a diode array UV spectrophotometer. The system can handle a wide range of drug formulation concentrations (> 200 mg ml-1) and gives excellent precision and accuracy with no detectable carryover. The analysis acceptance parameters are user-definable and the analysis process, including interpretation and reporting of the data, is fully automated.

An evaluation of the T-Scan computerised occlusal analysis system.

A computerised system has recently become available to assist in occlusal analysis. An evaluation of this system is described, with particular emphasis on the measurement of bite force. A hand-held testing device and a Nene universal testing machine were used to apply force. A limited clinical trial was carried out on two volunteers with known occlusal interferences present. It was concluded that the system does not measure bite force accurately, but is useful as a clinical tool if used with care.

A computer-assisted electrocardiographic analysis system: methodology and potential application to cardiovascular toxicology.

An automated analysis of electrocardiographic (ECG) waveforms, based on a precise one-dimensional analysis of features within a generalized computer-enhanced ECG waveform, has been developed in our laboratory. ECG signals are monitored, amplified, and recorded using standard techniques. The recorder output signal is distributed to a microcomputer system. Software developed for the microcomputer slows the signal playback rate and permits operator review of the slowed signal for arrhythmia analysis. The analysis program identifies and superimposes 10-40 individual ECG complexes, depending on the heart rate, and generates an "ensembled" waveform. Operator interaction permits delineation of specific points on the displayed waveform and calculation of heart rate and duration of components within the ECG complex. The primary advantages of this system include (1) extensive automation--computer support decreases analytical time, increases precision, and permits rapid screening of large numbers of animals; (2) enhanced sensitivity--the use of functional parameters should provide a more sensitive index of toxicity than morphological parameters; (3) broad utility--this system provides the capability to utilize a variety of animals, both anesthetized and unanesthetized, ranging in age from fetuses to geriatrics, and permits studies of block as well as longitudinal design; and (4) ease of replication--standardization of equipment and techniques facilitates replication by other laboratories.

Possible mechanisms of plastic reorganizations of neuronal activity during different schedules of intracellular electrostimulation according to the data of systems analysis.

The influence of rhythmic, random, and associative intracellular electrostimulation has been studied in individual identified and nonidentified neurons of the isolated CNS of the pond snail under the conditions of a controllable experiment. Using the methods of cluster, correlation, and factor analyses, an assessment is given and a comparison made of the changes in the character of the spontaneous spike activity induced by these modes of influence. It has been shown that the associations of the principal frequency-temporal characteristics of the endogenous rhythmicity of the neurons with the parameters of the external influences are identified only during stimulation associated with specific components of the endogenous rhythmicity, and are practically absent in the case of rhythmic and random electrostimulation. Three factors determining the formation of the structure of the spike impulse activity during the application of suprathreshold stimulations were distinguished. With respect to the composition of the variables included in them, they can be interpreted as: 1) the factor of intensity of the spontaneous activity; 2) the factor of the variability of the interimpulse intervals; and 3) the reinforcing factor. The possible endoneuronal mechanisms of the formation of adaptive reorganizations of the spontaneous activity under the conditions of associative influences are discussed.
