Role of tumor necrosis factor alpha in hyperthermia-induced apoptosis of human leukemia cells.

We used the human myelomonoblastic leukemia cell line PLB-985 to study the effects of temperatures ranging from 37 degrees C to 43 degrees C for 1 h on the induction of apoptosis and cell cycle distribution in leukemia cells. The threshold temperature for the onset of apoptosis was 42 degrees C. Whereas hyperthermia exerted no effect on the expression of Bcl-2 and Bax, heat induced a >30-fold increase of tumor necrosis factor (TNF) alpha mRNA expression and a significant increase in TNF-alpha protein secretion. This endogenous production of TNF-alpha correlated directly with the temperature-induced apoptode effect. Blocking TNF-alpha expression via treatment with pyrrolidinedithiocarbamate or blocking TNF-alpha activity with neutralizing antibodies abrogated heat-provoked apoptosis. In addition, exposure of cell culture supernatant of heat-treated PLB-985 cells to untreated cells induced an apoptotic effect. These data indicate a TNF-a-mediated self eradication of the leukemia cells after heat exposure. Inducing apoptosis with wild-type TNF-alpha or p55 and p75 protein muteins demonstrated that this effect was mediated by the p55 receptor. Interestingly, the autocrine suicidal loop found in immature leukemia cells was lost after granulocytic differentiation with 0.5% N,N-dimethylformamide. These data should be of critical importance for the understanding of the biological impact of fever as well as for developing therapeutic approaches to malignant diseases

Action of ethanol and other solvents on the nerve tissue under in vitro conditions.

Explants and cells of nervous tissue were cultivated in the presence of aethanol, tween 80, dimethylformamid (DMF) and dimethylsulfoxid (DMSO) and the influence upon the index of area, the growth rate and fiber index was observed. They are important to solutions of drugs for tests in vitro. At the beginning of the cultivation aethanol in vitro influenced the regeneration of nerve fibers from explants and cells. A significant increase of the index of growth was observed. After a long term influence of tween 80, DMF and DMSO an inhibition of differentiation of neurons in vitro was observed.

Fluorogenic substrates for assay of chymosin.

The use of fluorogenic substrates with intramolecular fluorescence quenching as substrates for chymosin was studied. It was shown that chymosin hydrolyzes the Phe-Phe peptide bond. The effect of pH on the hydrolysis of substrates by chymosin was investigated. The catalytic characteristics of the hydrolysis of the fluorogenic substrates were obtained at the pH optima. The influence of dimethylformamide on chymosin activity was studied.

Antigen-binding activity of monoclonal antibodies after incubation with organic solvents.

Effects of four organic solvents--methanol, trifluoroethanol, dimethylsulfoxide, and dimethylformamide (DMF)--on the ferritin-binding activity of three monoclonal mouse antibodies of IgG2a and IgG1 subclasses were studied. The ferritin-binding constants of monoclonal antibodies G10 and F11 (the IgG2a subclass) were increased 2-6-fold after incubation with DMF and removal of the organic solvent by gel filtration. The maximum effect on the F11 antibodies was found in the presence of 5-13% DMF and on the G10 antibodies at 11-40% DMF. The effect remained after the removal of DMF from the incubation medium, and this suggests that the incubation with DMF resulted in irreversible conformational changes of the antibodies and in production of active conformers of the G10 and F11 antibodies. These conformations occurred within 15-60 min. The long-term stability and the fluorescence of the antibodies exposed to DMF suggest that the conformational changes were not global, but involved small and relatively independent structural elements of the antibodies, either of hypervariable CDR loops in variable domains or of the hinge region of the antibodies. The affinity of the C5 antibodies of the mouse IgG1 subclass was decreased after incubation with DMF. The activation was a solvent-specific effect because incubation of the G10 antibodies with methanol and dimethylsulfoxide decreased the affinity for the antigen, and incubation with trifluoroethanol virtually did not affect it. Relatively small changes in the antigen-binding activity of the antibodies were found even after the incubation with 5% organic solvent.

Simultaneous determination of alpha and beta hydroxy acids in personal care products by capillary gas chromatography.

A simple and rapid procedure is described for the isolation, silylation, and simultaneous capillary gas chromatographic quantitation of alpha hydroxy acids and beta hydroxy acids in various personal care products. The sample is dissolved in acidified N,N-dimethylformamide to simultaneously acidify/extract the hydroxy acids; a portion is then trimethylsilyl derivatized with BSTFA and quantified by capillary gas chromatography (GC) using flame ionization detection.

Nuclear magnetic resonance investigation of the state of water in protein solution.

The line width of the NMR signal of water protons in solutions of native actomyosin and actomyosin denatured by heat, acetone or urea was measured over the temperature range from -10 degrees to below the freezing point. The line widths of the water band which increased exponentially with decreasing temperature were compared with each other and also with those of the corresponding control solution without actomyosin. The line broadening observed for native actomyosin solution on lowering the temperature was significantly smaller than that for heat-denatured actomyosin solution. This difference implies that this signal is sensitive to conformational perturbations of the protein. In addition, the temperature dependence of the line width for heat-, acetone-, or urea-denatured actomyosin solution was similar to that for the corresponding control solution. These phenomena can be interpreted in terms of the state of water associated with the hydrophobic and hydrophilic residues. Similar NMR studies of actomyosin solution containing dimethyl sulfoxide (DMSO) or dimethylformamide (DMF) showed that DMSO and DMF prevent the formation of ice crystals until about -70 degrees, suggesting that the cryoprotective effects of DMSO and DMF are due to the change in the state of water described above. These differences in temperature dependence between the sample and control solutions are well-correlated with the viscosity of the solution. This correlation is useful for elucidation of the mechanism of the protein denaturation.

[Electrospinning and morphology of ultrafine poly (D, L-lactide) fibers].

Ultrafine poly (D, L-lactide) (PLA) fibers with diameter less than 200 nm produced by electrospinning were studied to obtain tissue restoration resembling extracellular matrix. Scanning electron microscopy was used to observe the fiber morphology. Results showed that the solvent was the critical factor to determine the formation of the electrospun PLA fibers. Compared with acetone, N,N-dimethylformamide (DMF) was a better solvent for PLA to electrospin. Entrance of an organic salt, triethylbenzylammonium chlorate, led to a great increase of the conductivity of PLA/DMF solutions, so that the average fiber diameter of the electrospun PLA fibers decreased dramatically from 500 nm to 100-200 nm. The addition of surfactant, Span-80, did not improve the fiber morphology but formed beaded fiber web.

Comparison of the cytidine 5'-diphosphate and adenosine 5'-diphosphate reductase activities of mammalian ribonucleotide reductase.

A comparison of the cytidine 5'-diphosphate (CDP) and adenosine 5'-diphosphate (ADP) reductase activities from Ehrlich tumor cells was made to determine if the properties of the enzyme for these substrates were the same, except for the allosteric effector. It was observed that various purification steps did not result in an enzyme fraction that had a constant ratio of CDP:ADP reductase activities. The optimal Mg2+ ion concentration for CDP reduction was 3 to 4 mM, while the optimal Mg2+ ion concentration for ADP reduction was 0.1 mM inhibited ADP reduction. CDP reduction was relatively insensitive to the presence of dimethylformamide or dimethyl sulfoxide in the reaction mixture, but ADP reduction was decreased in the presence of these two compounds. Periodate-oxidized adenosine 5'-monophosphate, on incubation with the enzyme, had a greater effect on CDP reduction but little or no effect on ADP reduction. The response of the CDP and ADP reductase activities to the same negative effector was essentially the same. Both CDP and ADP reductions showed similar decreases in the presence of various concentrations of deoxyadenosine 5'-triphosphate. These data suggest that the Ehrlich tumor cell reductase enzyme system could consist of at least two different enzymes that may be regulated by the same allosteric protein.

DBU as an N alpha-deprotecting reagent for the fluorenylmethoxycarbonyl group in continuous flow solid-phase peptide synthesis.

The versatility and efficiency of the N alpha-fluorenylmethoxycarbonyl method of solid-phase peptide synthesis have been further enhanced by recent and continuing refinements in side-chain protecting group, linker and amino acid-coupling technology. In contrast, comparatively little work has been carried out on studying and further improving the N alpha-deprotection step. This report demonstrates that, in low concentrations, the non-nucleophilic amidine 1,8-diazabicyclo[5.4.0]undec-7-ene is a preferred alternative to the more commonly employed piperidine for the rapid and efficient cleavage of the N alpha-fluorenylmethoxycarbonyl group in the continuous flow method of solid-phase peptide synthesis. In the cases studied, use of this base did not cause cyclization of Asp(OBut)-Gly or Asn-Gly sequence to succinimide derivatives. At the recommended concentration of 2% 1,8-diazabicyclo[5.4.0]undec-7-ene in dimethylformamide, racemization of resin-bound C-terminal S-trityl cysteine was substantially reduced compared with standard piperidine-mediated deprotection conditions. Additionally, use of the base appears to reduce the extent of broadening of ultraviolet fluorenylmethoxycarbonyl-deprotection peaks occasionally observed when piperidine is employed, indicating that its reaction kinetics and mode of action are different.

Transdermal absorption of radioprotectors using permeation-enhancing vehicles.

Radioprotectors are not currently used clinically due to concerns regarding toxicity and uncertainties regarding tumor protection. Topical radioprotection of skin might find clinical applications with protectors such as WR-2721, but laboratory studies in which protectors have been applied in water have not been promising. We have studied the absorption of 14C-WR-2721 and [14C]cysteine dissolved in skin permeation-enhancing vehicles through the skin of hairless mice and compared the absorption to that in water. Skin concentration of WR-2721 was increased most by dimethylformamide (DMF), but only propylene glycol increased absorption as far as the dermis, as measured by plasma concentration. Skin concentration of cysteine was improved by DMF, 2-pyrrolidone (2-P), and methyl-2-pyrrolidone (M-2-P); only dimethylsulfoxide (DMSO) resulted in increased plasma levels of the protector. Pretreating skin with DMSO before application of WR-2721, irrespective of the vehicle, improved its concentration within the skin. Plasma levels were improved (10 and 12 times) only with 2-P and DMF. Therefore, by choosing the appropriate vehicle, it is possible to breach the barrier of the stratum corneum and enhance the presence of the protector in all layers of the skin.

Transdermal absorption of radioprotectors in the rat using permeation-enhancing vehicles.

Topical radioprotection of rat skin with WR-2721 has not been effective presumably because the drug does not cross the stratum corneum to reach the epidermis and dermis. Earlier, we showed in the mouse that WR-2721 and cysteine dissolved in permeation-enhancing vehicles passed through the skin more readily than when in water. However, the most effective vehicles in the mouse were not necessarily as effective in the rat. Here we report that the most effective transport vehicles in the rat were (1) water with WR-2721, (2) water and dimethylformamide (DMF) with cysteine, and (3) water and DMF with prostaglandin E2 (PGE2). Pretreatment of the skin with dimethylsulfoxide (DMSO) further improved the transfer of the radioprotectors across the skin in most cases. After pretreatment with DMSO, the most effective vehicles were (1) water for WR-2721, (2) water and methyl-2-pyrrolidone (M-2-P) for cysteine, and (3) DMF for PGE2.

[The effect of modification of lactate and malate dehydrogenases on their stability and activity].

The inactivation of lactate and malate dehydrogenases (LDH and MDH) modified by progesterone in the water-dimethylformamide (DMF) medium is described by the first-order equation up to large conversion degrees. The MDH modification is accompanied by the increase of its stability by 7-14%, while LDH modification leads to the enzyme stability decrease by 67%. The enzymes catalytic activities are changed simultaneously. The main factors of the stability and activity changes are the DMF influence upon the quaternary structure of the proteins at the modification and a hydrofobization of the external and internal protein sites by progesterone.

Simple enzymatic determination of total cholesterol in gallstones.

In this simple method for rapid enzymatic determination of total cholesterol in human gallstones, gallstone powder is dissolved in an 80/20 by vol mixture of N,N-dimethylformamide and dimethyl sulfoxide and reacted directly with the aqueous enzymatic reagent, without further treatment. The organic solvents do not interfere with enzymatic activities of cholesterol oxidase, esterase, and peroxidase in the assay mixture. The method is reproducible (CVs 3.7% to 6.6%), analytical recoveries ranged from 98.6% to 102%, and linearity is good. Furthermore, results correlated well with those obtained by infrared spectroscopic measurements.

Modulation of the expression of a multidrug resistance gene (mdr-1/P-glycoprotein) by differentiating agents.

Acquired resistance to multiple natural products in vitro is mediated by P-glycoprotein (Pgp). Expression of this protein has been demonstrated in some normal tissues and in tumor samples obtained from both untreated and treated patients. In situ hybridizations with RNA probes have demonstrated higher levels of expression of mdr-1/Pgp in well-differentiated tumors and in well-differentiated areas in tumors with mixed histologies. Expression of mdr-1/Pgp in human colon carcinoma cell lines was increased by the differentiating agents sodium butyrate, dimethyl sulfoxide, and dimethylformamide. In the SW-620 cell line addition of sodium butyrate resulted in a rapid induction of mdr-1/Pgp mRNA that was sustained for the duration of the exposure. The levels of P-glycoprotein were measured by immunoblotting and were also increased. Similar results were obtained in three other cell lines including the HCT-15 line. This induction occurred without alterations in nuclease sensitivity. Discontinuation of sodium butyrate was followed by a rapid fall in the levels of mRNA. The levels of P-glycoprotein returned to normal with a half-life of about 24 h. In spite of a 20-25-fold increase in the level of mdr-1/Pgp mRNA and P-glycoprotein, the SW-620 cell line did not demonstrate increased resistance to doxorubicin and vinblastine or decreased accumulation of vinblastine. In contrast, in the HCT-15 cell line, a 5-fold increase of mdr-1/Pgp was accompanied by a comparable fall in vinblastine accumulation which was reversed by verapamil. In the SW-620 cell line, the induced protein could be photolabeled using [3H]azidopine. Expression of mdr-1/Pgp appears to correlate with the degree of differentiation. However, its induction is not always accompanied by expression of the multidrug-resistance phenotype.

Heterogeneous response to differentiation induction in different clonal subpopulations of a rat rhabdomyosarcoma cell line (BA-HAN-1).

Three clonal subpopulations (A, B, C) isolated from the same rhabdomyosarcoma of the rat were tested and compared for their susceptibility to differentiation induction using retinoic acid (RA), dimethylformamide (DMF), and N-monomethylformamide (NMF). These subpopulations differ in that a block to spontaneous differentiation is imposed at different stages which are characteristic for each subpopulation. Whereas tumor cell proliferation was significantly inhibited (P less than 0.001) in all three subpopulations, the effects of RA, DMF, and NMF on tumor cell differentiation were strikingly heterogeneous. The response was most marked in subpopulation C, as evidenced by a significant increase in the number of terminally differentiated myotube-like giant cells (P less than 0.001) and in biochemical differentiation, as indicated by the creatine kinase activity (P less than 0.05). Between 5% (DMF and NMF) and 30% (RA) of the mononuclear cells in subpopulation C exhibited thick and thin myofilaments, which were never observed in the mononuclear cells of the control. In contrast, subpopulation A and B responded to RA, DMF, and NMF quite heterogeneously with an increase in biochemical differentiation, whereas terminally differentiated myotube-like giant cells were never observed. These results demonstrate that the therapeutic potential of differentiation induction in malignant tumors may be impaired by tumor heterogeneity.

[Status of reproductive function and the development and health of children of female workers in the synthetic leather industry].

The study was undertaken to analyze the course of pregnancy and childbirth along with the state of the lactation function in 48 female workers occupationally exposed to dimethylformamide (DMFA) during 1-5 years of service. Physical development and health status of 48 children born to DMFA-exposed mothers were examined. It was demonstrated that DMFA had adverse effect on pregnancy and childbirth, characterized by a reliable increase of toxicosis rates, birth complications, manual elimination of afterbirth. DMFA-exposed females experienced weakening of the lactation function and some deviations in the development and health status of infants.
